Anti-Inflammatory Effects of Water Chestnut Extract on Cytokine Responses via Nuclear Factor-κB-signaling Pathway

  • Water chestnut (Trapa japonica Flerov.) is an annual aquatic plant. In the present study, we showed that the treatment of water chestnut extracted with boiling water resulted in a significant increase 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity and decrease the intracellular $H_2O_2$-induced accumulation of reactive oxygen species. In addition, water chestnut extract (WCE) inhibited lipopolysaccharide (LPS)-induced nitric oxide production and suppressed mRNA and protein expression of the inducible nitric oxide synthase gene. The cytokine array results showed that WCE inhibited inflammatory cytokine secretion. Also, WCE reduced tumor necrosis factor-${\alpha}$- and interleukin-6-induced nuclear factor-${\kappa}B$ activity. Furthermore, during sodium lauryl sulfate (SLS)-induced irritation of human skin, WCE reduced SLS-induced skin erythema and improved barrier regeneration. These results indicate that WCE may be a promising topical anti-inflammatory agent.

귀비탕이 Glutamate에 의한 성상세포의 손상에 미치는 영향 (Effects of Gwibitang on Glutamate-induced Death in Rat Neonatal Astrocytes)

  • Objectives: This study was designed to investigate effects of Gwibitang on the glutamate-induced toxicity of primary rat neonatal astrocytes. Methods and Results: Gwibitang significantly recovered the glutamate-induced apoptosis and inhibited the generation of $H_2O_2$ in astrocytes. In addition, both Gwibitang and antioxidants such as GSH reduced the glutamate-induced cytotoxicity in astrocytes, indicating that Gwibitang possibly had an antioxidative effect. Moreover, Gwibitang also inhibited the glutamate-induced degradation of Bcl-2 protein and poly(ADP)-ribose polymerase (PARP) in astrocytes. Conclusions: We suggest that Gwibitang has protective effects on glutamate-induced cytotoxicity via an antioxidative mechanism.

새싹 추출물의 항산화 작용과 미량 성분 (Anti-oxidative Activity and Trace Component of a Sprout Serum)

  • In this study, the anti-oxidative effects and components of sprout serum were investigated. In the buds, high levels of zinc, iron and manganese were analyzed in addition to copper, magnesium, and potassium. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of sprout serum was evaluated with $IC_{50}$. $IC_{50}$ of sprout serum was $58.0\;mgmL^{-1}$, and that of vitamin C was $3.7\;mgmL^{-1}$. In the test of superoxide scavenging activity of sprout serum, the activity was dependent on the concentration of serum. In this case, the $IC_{50}$ was 2.0 wt%. Cell viability was detected by using the MTT method. Cultured human fibroblast was treated with 15 mM $H_2O_2$ and cell viability was 70% in case of control. However, the effect of treating 0.5% of sprout serum was similar to that of 0.0001% of vitamin C.

  • The purpose of this study was to develop the new microbial bioflocculant available in a food and fer-mentation industal. This study was reported the results of the composition for optimum culture medium and elemental characteristic of crude purification bioflocculant following the previous report(I). The maximum production of the flocculant from Bacillus megaterium was observated in the culture medium containing 2% sucrose 0.3% NaNO3 0.01% tryptone 0.01% beef extract 0.05% MgSO4 ·7 H2O 0.005% CaCO3 Addition of the sucrose as carbon sources and inorganic salt such as MgSO4, CaCO3 significantly increased the production of flocculant more than nitrogen sources. In the result of color reaction of the crude purified bioflocculant it was investgated that anthrone was positive and benedict burette and nin-hydrin was negative. These result were indicated that the flocculant produced from Bacillus megaterium was a kind of exopolysaccharide.

Analysis of the Effect of Mordants on the Degradation of Alizarin in Silk Dyed with Natural Madder Dye

  • This research investigated the effect of mordants on the degradation of madder dye in silk when silk was treated by the H2O2/UV condition as a laboratory simulation of burial induced degradation. Alum, iron, and alum/iron composite mordanting methods were applied to silk before dyeing with madder dye. Dye extracted from silk was examined using HPLC-DAD-MS analysis. The abundance of the chromatogram peak at 8.88 min retention time was used as the concentration of alizarin pigment in silk. K/S values, CIE $L^{\ast}a^{\ast}b^{\ast}$ values; in addition, Munsell HVC values were obtained using a spectrocolorimeter. The findings indicated that alizarin degraded most severely in silk mordanted by alum/iron composite mordanting than alum mordanting or iron mordanting. Mordanting with alum alone provided a relatively lower dye fixation at the point of dyeing; however, it provided a better survival of alizarin after 12 hours of degradation treatment.

Effects of Dietary Olive Oil on Growth Performance, Carcass Parameters, Serum Characteristics, and Fatty Acid Composition of Breast and Drumstick Meat in Broilers

  • This experiment was conducted to evaluate the effects of dietary olive oil on growth performance, carcass parameters, serum characteristics, and fatty acid composition of breast and drumstick meat in broiler chickens. A total of 480 broilers were randomly allotted into three dietary treatments, including T (basal diet, 5% tallow), O1 (2% olive oil+3% tallow), and O2 (5% olive oil). During d 0 to 21, broilers fed the diet supplemented with 5% olive oil showed lower (p<0.05) body weight gain (BWG) and feed intake (FI) compared with those fed the T diet. Serum triglyceride concentration was reduced (p<0.05), while high density lipoprotein (HDL)-cholesterol concentration was increased (p<0.05) in the O2 treatment group compared with the T and O1 treatment groups. The addition of olive oil to the diets induced a reduction (p<0.05) in the total saturated fatty acid (SFA) contents in breast and drumstick meat, and increased (p<0.05) the total unsaturated fatty acid (USFA) contents and USFA/SFA ratios. In conclusion, a diet with 5% olive oil could decrease BWG and FI of broilers during the starter period (wk 0 to 3), and cause an increase in the serum HDL-cholesterol level, while decreasing the serum triglyceride concentration. Furthermore, USFA level and USFA/SFA ratios in breast and drumstick meat were increased by dietary supplementation of 2 or 5% olive oil.

  • Amorphous $MnO_{2}{\cdot}nH_{2}O$ in 1M KOH aqueous electrolyte proves to be an excellent electrode for a faradic electrochemical capacitor cycled between -0.5 and +1.0 versus Ag/AgCl. The effect of thermal treatment on the crystalinity, particle structure, and corresponding electrochemical properties of the resulting xerogel remained amorphous as Mn(OH)2 up to 160$^{\circ}C$. With an increase in the temperature above 200$^{\circ}C$, both the surface area and pore volume decreased sharply, because the amorphous Mn(OH)2 decomposed to form MnO that was subsequently oxidized to form crystalline Mn3O4. In addition, the changes in the crystallinity, and particle structure all had significant but coupled effects on the electrochemical properties of the xerogels. A maximum capacitance of 160.6F/g was obtained for an electrode prepared with the MnOx Xerogel calcined at 150$^{\circ}C$, which was consistent with the maxima exhibited in both the surface area and pore volume. This capacitance was attributed solely to a surface redox mechanism.

Low Cost Alcoholic Breath Sensor Based on SnO2 Modified with CNTs and Graphene

  • In this work, $SnO_2$ modified with reduced graphene oxide (rGO) and carbon nanotubes (CNTs) separately and combined sensitized by using the co-precipitation method and their sensing behavior toward ethanol vapor at room temperature were investigated. An interdigitated electrode (IDE) gold substrate is very expensive compared to a fluorine doped tin oxide (FTO) substrate; hence, we used the latter to reduce the fabrication cost. The structure and the morphology of the studied materials were characterized by using differential thermal analyses (DTA) and thermogravimetric analysis (TGA), transmission electron microscope (TEM), X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, Brunauer-Emmett-Teller surface area and Barrett-Joyner-Halenda (BJH) pore size measurements. The studied composites were subjected to ethanol in its gas phase at concentrations from 10 to 200 ppm. The present composites showed high-performance sensitivity for many reasons: the incorporation of $SnO_2$ and CNTs which prevents the agglomeration of rGO sheets, the formation of a 3D mesopourus structure and an increase in the surface area. The decoration with rGO and CNTs led to more active sites, such as vacancies, which increased the adsorption of ethanol gas. In addition, the mesopore structure and the nano size of the $SnO_2$ particles allowed an efficient diffusion of gases to the active sites. Based on these results, the present composites should be considered as efficient and low-cost sensors for alcohol.

애기장대 H+-pyrophosphatase 발현 형질전환 배추의 건조스트레스에 대한 생리적 반응 (Physiological responses to drought stress of transgenic Chinese cabbage expressing Arabidopsis H+-pyrophosphatase)

  • 한발에 대한 식물체의 내성은 수분이 부족한 환경에서도 안정적으로 작물의 생산성을 유지하기 위해 필요한 유용한 특성 중 하나이다. 우리는 애기장대의 $H^+$-pyrophosphatase (AVP1)를 발현하도록 형질전환된 배추가 내건성과 관련되어 있는 몇몇 생리적 척도에 있어 향상됨을 검증하였다. 관수중단 처리로 조성된 토양수분 결핍 조건에서 AVP1 발현 식물체는 비형질전환체와 비교하여 비록 외형적 위조의 정도로는 그 차이를 구별할 수 없었지만 형질전환체가 재식된 토양의 수분포텐셜이 비형질전환체 재식 토양에 비해 더 빠르게 낮아졌다. 이는 형질전환체 잎의 상대적 수분함량이 비형질전환체에 비해 더 높은 것과 연관되어 있는 것으로 사료된다. 또한 건조스트레스 환경에서 비형질전환체에 비해 형질전환체는 광계II 양자수율이 높은 반면 전해질누출과 활성산소족의 하나인 $H_2O_2$와 3,3'-diaminobenzidine의 반응산물이 적었다.

Gpx3-dependent Responses Against Oxidative Stress in Saccharomyces cerevisiae

  • The yeast Saccharomyces cerevisiae has defense mechanisms identical to higher eukaryotes. It offers the potential for genome-wide experimental approaches owing to its smaller genome size and the availability of the complete sequence. It therefore represents an ideal eukaryotic model for studying cellular redox control and oxidative stress responses. S. cerevisiae Yap1 is a well-known transcription factor that is required for $H_2O_2$-dependent stress responses. Yap1 is involved in various signaling pathways in an oxidative stress response. The Gpx3 (Orp1/PHGpx3) protein is one of the factors related to these signaling pathways. It plays the role of a transducer that transfers the hydroperoxide signal to Yap1. In this study, using extensive proteomic and bioinformatics analyses, the function of the Gpx3 protein in an adaptive response against oxidative stress was investigated in wild-type, gpx3-deletion mutant, and gpx3-deletion mutant overexpressing Gpx3 protein strains. We identified 30 proteins that are related to the Gpx3-dependent oxidative stress responses and 17 proteins that are changed in a Gpx3-dependent manner regardless of oxidative stress. As expected, $H_2O_2$-responsive Gpx3-dependent proteins include a number of antioxidants related with cell rescue and defense. In addition, they contain a variety of proteins related to energy and carbohydrate metabolism, transcription, and protein fate. Based upon the experimental results, it is suggested that Gpx3-dependent stress adaptive response includes the regulation of genes related to the capacity to detoxify oxidants and repair oxidative stress-induced damages affected by Yap1 as well as metabolism and protein fate independent from Yap1.
