Effect of Dietary Calcium on Cell Proliferation and Colonic Mucosal Levels of Eicosanoid and 1,2-diacylglycerol in Colon Carcinogenesis of Rats (쥐에서 식이 Calcium이 대장 암화관정의 세포증식과 대장점막의 Eicosanoid 및 1,2 -diacylglycerol 수준에 미치는 영향)

  • The objective of this study was to observe the effect of dietary calcium(Ca) level on colonic mucosal levels of cell proliferation, 1, 2-diacylglycerol(DAG), TXB2, PGE2 and phospholipid fatty acid composition which have been known as biomarkers for colon cancer. One hundred male Sprague Dawley rats, at 7 weeks of age, were divided into two fat type groups. Each group of which was further divided into two Ca level groups. Each rt was intramuscularly injected with 1, 2,-dimenthylhydrazine(DMH) for 6 weeks (total dose of 180mg/kg body weight) and simultaneously fed one of four experimental diets containing 15% dietary fat(corn oil or perilla oil )and 0.3% or 1.0% Ca by weight for 20 weeks. Compared to corn oil, perilla oil significantly reduced cell proliferation by decreasing labeling index, proliferating zone, crypt length in colonic mucosa and colonic mucosa and colonic mucosal levels of DAG, TXB2 . PGE2 and phospolipid (PL) arachidonic acid distribution. The effect of Ca on biomarketrs was different depending on the type of dietary fat comsumed . Ca effect of Ca on biomarkers was different depending on the type of dietary fat comsumed. Ca effect was not significantly shown in the PO group, but it was significant in the CO group in which high Ca(1.0%) decreased the levels of levels of PL-C20 : 4(%), DAG and PGE2 . However , high Ca supplementation had shown only the trends of improving cell proliferation. Overall , high dietary Ca significantly reduced cell proliferation by inhibiting the synthesis of eicosanoid and DAG with reduced distribution of PL-C20 : 4 , which may have resulted in lower activation of PKC through reduced signal transduction. Since a high level of dietary Ca was more effective in reducing the risk factor against colon cancer in corn oil fed rats, it could be suggested that a higher amount of dietary Ca be consumed , especially when more vegetable oil rich in linoleic acid is included in the diet.

  • According to previous studies, the effect of intrinsic motivation related variables such as cognitive absorption (CA) on the use of information technology(IT) should be mediated by the conscious intention. However, CA, which is a thought of previous emotional experience, a deep involvement with a software, may be one of environmental cues stimulating the use of IT independently of conscious intention. The objective of this study is to show the direct effect of CA on the automatic use of IT. For this purpose, as well as the relation between CA and use, two relations, CA-intention and intention-use, were also suggested as hypotheses, which will show that conscious intention would not fully mediate the relation between CA and automatic use. Moreover, for seeing the effects of CA as time passes, the proximal effect, which is short term and transitory effect, and the distal effect, which is long term and lingering, were simultaneously included in hyptheses. Finally, the effect of past use was also suggested as a hypothesis which aimed to show that automatic use would be controlled mostly by habit rather than not only intention but also CA. For the empirical validation of the hypotheses, 174 users of 'Cyworld', one of most influential social network services, were longitudinally surveyed, which showed that the hypotheses were accepted.

Effect of Calmodulin on Ginseng Saponin-Induced $Ca^{2+}$-Activated $Cl^{-}$ Channel Activation in Xenopus laevis Oocytes

  • We previously demonstrated the ability of ginseng saponins (active ingredients of Panax ginseng) to enhance $Ca^{2+}$-activated $Cl^{-}$ current. The mechanism for this ginseng saponin-induced enhancement was proposed to be the release of $Ca^{2+}$ from $IP_{3}-sensitive$ intracellular stores through the activation of PTX-insensitive $G\alpha_{q/11}$ proteins and PLC pathway. Recent studies have shown that calmodulin (CaM) regulates $IP_{3}$ receptor-mediated $Ca^{2+}$ release in both $Ca^{2+}-dependent$ and -independent manner. In the present study, we have investigated the effects of CaM on ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current responses in Xenopus oocytes. Intraoocyte injection of CaM inhibited ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement, whereas co-injection of calmidazolium, a CaM antagonist, with CaM blocked CaM action. The inhibitory effect of CaM on ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement was dose- and time-dependent, with an $IC_{50} of 14.9\pm3.5 {\mu}M$. The inhibitory effect of CaM on saponin's activity was maximal after 6 h of intraoocyte injection of CaM, and after 48 h the activity of saponin recovered to control level. The half-recovery time was calculated to be $16.7\pm4.3 h$. Intraoocyte injection of CaM inhibited $Ca^{2+}$-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement and also attenuated $IP_{3}$-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement. $Ca^{2+}$/CaM kinase II inhibitor did not inhibit CaM-caused attenuation of ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement. These results suggest that CaM regulates ginseng saponin effect on $Ca^{2+}$-activated $Cl^{-}$ current enhancement via $Ca^{2+}$-independent manner.

  • Cardiac chambers serve as mechanosensory systems during the haemodynamic or mechanical disturbances. To examine a possible role of fluid pressure (FP) in the regulatien of atrial $Ca^{2+}$ signaling we investigated the effect of FP on L-type $Ca^{2+}$ current $(I_{Ca})$ in rat ventricular myocytes using whole-cell patch-clamp technique. FP $(\sim40cm\;H_2O)$ was applied to whole area of single myocytes with electronically controlled micro-jet system. FP suppressed the magnitude of peak $I_{Ca}$ by $\cong25\%$ at 0 mV without changing voltage dependence of the current-voltage relationship. FP significantly accelerated slow component in inactivation of $I_{Ca}$, but not its fast component. Analysis of steady-state inactivation curve revealed a reduction of the number of $Ca^{2+}$ channels available for activity in the presence of FP. Dialysis of myocytes with high concentration of immobile $Ca^{2+}$ buffer partially attenuated the FP-induced suppression of $I_{Ca}$. In addition, the intracellular $Ca^{2+}$ buttering abolished the FP-induced acceleration of slow component in $I_{Ca}$ inactivation. These results indicate that FP sup-presses $Ca^{2+}$ currents, in part, by increasing cytosolic $Ca^{2+}$ concentration.

Effect of $Ca^{2+}$ on Phvsiological Activites of the Acrosome Reaction on Spermatozoa ($Ca^{2+}$이 정자 첨체반응의 생리적 활성에 미치는 영향)

  • It has been known that spermatozoa should obtain their fertilizing ability through capacitation and acrosome reaction, and that in these processes of fertilization, Ca2+ platys an important role for their conjugation. Therefore the present study has been designed in order to clarify the effect of fluctuation of the media Ca2+ level and the intracellular concentration of the spermatozoa on the acrosomes. During the incubation of spermatozoa, a considerable fluctuation in the media Ca2+ level has been observed after the BSA administration and the media concentration of Ca2+. It is deduced that these fluctuation rates may have an effect on the acrosome reaction. The fluctuation of K+ flux has been observed in accordance with the incubation period over time, and it's concentration seems to be closely related with the acrosomal reaction. The respiratory exchange rate (RERI of the spermatozoa is kept more regular in the BSA and Cacl2 administration groups than the non-administration group. Based on the experimental findings, it is possible to deduce a hypothesis from these findings that physiological activities of the acrosome reaction are not functionally related to the media Ca2+ level and the intracellular influx of Ca2+ concentration, although Ca2+ platys an important role as a stimulating factor in the acrosome reaction.

  • Effect of MgO, CaSO4, and CaF2 addition on the formation of clinker minerals in portland cement have been investigated by measuring the amounts of free-CaO and C3S in the fired specimens and analyzing the Mg and S concentration in C3S and C2S. It was found that CaSO4 inhibited C3S formation but MgO addition offset this effect of CaSO4. MgO addition also enhanced the mineralizing effect of CaSO4+CaF2, resulting in the acceleration of C3S formation. It was suggested that Mg might inhibit the formation of sulphate compounds rim around C2S and thus C2S+CaOlongrightarrowC3S reaction was facilitated.

The Effect of Ghrelin on $Ca^{2+}$ Concentration in Thyroid FRTL-5 Cells

  • Ghrelin is a newly discovered peptide, which is released from the stomach and neurons in the hypothalamic arcuate nucleus (ARC), and potently stimulates growth hormone release and food intake. In the present study, we investigated the effect of ghrelin on $[Ca^{2+}]_i$ in thyroid FRTL-5 cells. Ghrelin (5 nM) increased $[Ca^{2+}]_i$ and TSH (1 unit/l) had an additive effect on $[Ca^{2+}]_i$ when extracellular normal solution was 1.1mM $Ca^{2+}$ containing Coon's modified Ham's F12 medium. When $Ca^{2+}-free$ medium containing 2 mM EGTA replaced the above normal solution, ghrelin also induced a similar rise in $[Ca^{2+}]_i$. In the middle of $[Ca^{2+}]_i$ increment by ghrelin, nifedipine $(1\;{\mu}M)$, nickel $(100\;{\mu}M)$ and $La^{3+}\;(100\;{\mu}M)$ had no effect on $[Ca^{2+}]_i$. After endoplasmic reticulum was depleted by cyclopiazonic acid $(CPA;10\;{\mu}M)$, ghrelin caused no visible change on $[Ca^{2+}]_i$ in $Ca^{2+}-free$/2 mM EGTA solution. These results suggest that ghrelin can increase $[Ca^{2+}]_i$ through endoplasmic reticulum in thyroid FRTL-5 cells.

Protective Effect of Processed Saengmaek-san(SM) on Cell Damage in UV-exposed HaCaT Cell (생맥산(生脈散)이 자외선에 의한 피부각질세포의 상해에 미치는 영향)

  • Objectives: This study was performed to assess the protective effect of Saengmaek-san (SM) on UVB-induced HaCaT cell damage. Methods: The protective effects of Saengmaek-san(SM) were determined by UVB-induced HaCaT assay. We assessed protective effects of Saengmaek-san (SM) on LDH release and nitrite release from HaCaT. And COX-2, Bcl-2, Bax, $TNF{\alpha}$, c-jun, c-fos, NF-kB, iNOS, Bcl-xL gene expression were determined in HaCaT using real-time PCR method. Results: 1. SM inhibited LDH-release, nitrite production in UVB-exposed HaCaT. 2. SM suppressed the gene expression of COX-2, $TNF{\alpha}$ in UVB-exposed HaCaT. 3. SM increased the gene expression of Bcl-2, Bax, Bcl-xL family protein in UVB-exposed HaCaT. 4. SM suppressed the gene expression of c-jun, c-fos, NF-kB in UVB-exposed HaCaT. Conclusions: The study showed SM inhibited the cell damage in UVB-exposed HaCaT.

Cell proliferation and migration mechanism of caffeoylserotonin and serotonin via serotonin 2B receptor in human keratinocyte HaCaT cells

  • Caffeoylserotonin (CaS), one derivative of serotonin (5-HT), is a secondary metabolite produced in pepper fruits with strong antioxidant activities. In this study, we investigated the effect of CaS on proliferation and migration of human keratinocyte HaCaT cells compared to that of 5-HT. CaS enhanced keratinocyte proliferation even under serum deficient condition. This effect of CaS was mediated by serotonin 2B receptor (5-HT2BR) related to the cell proliferation effect of 5-HT. We also confirmed that both CaS and 5-HT induced G1 progression via 5-HT2BR/ERK pathway in HaCaT cells. However, Akt pathway was additionally involved in upregulated expression levels of cyclin D1 and cyclin E induced by CaS by activating 5-HT2BR. Moreover, CaS and 5-HT induced cell migration in HaCaT cells via 5-HT2BR. However, 5-HT regulated cell migration only through ERK/AP-1/MMP9 pathway while additional Akt/NF-${\kappa}B$/MMP9 pathway was involved in the cell migration effect of CaS. These results suggest that CaS can enhance keratinocyte proliferation and migration. It might have potential as a reagent beneficial for wound closing and cell regeneration.

The Development of Early Mouse Embryos Depend on $Ca^{2+}$ Concentration (Ca$^{2+}$ 농도에 따른 생쥐 초기배의 발생)

  • Objective: This study was to determine the effect of different concentration of calcium III medium on the preimplantational development of zygotes and early 2 cell embryos. Methods: Female mice of ICR strain ($5{\sim}8$ weeks old) were superovulated and mated with fertile males. Zygotes or early 2-cell embryos were collected by flushing the oviducts $31{\sim}32$ hours after hCG injection. The embryos were cultured in various concentrations of $Ca^{2+}$ in medium or with EDTA, EGTA and $Ni^{2+}$. Result and Conclusion: Treatment of high concentraion of $Ca^{2+}$ (3.42 mM $(2X){\sim}17.l$ mM (10X)) in medium didn't develop well compared to the control. Low concentrations of $Ca^{2+}$ (0.214mM $(1/8X){\sim}0.855$ mM (1/2X) were deterimental to development beyond 2-cell stage. EDTA, $Ca^{2+}$ chelating agent was treated with ranged concentrations of EDTA (0.014 $mM{\sim}0.107$ mM) to medium contaning 1.71 mM $Ca^{2+}$ showed beneficial effect to development to blastocyst compared to the control. EGTA, extracellular $Ca^{2+}$ chelator, was treated with ranged concentrations of EGTA ($0.014{\sim}0.107$ mM) to the medium contaning 1.71 mM $Ca^{2+}$. There is no significant difference with the control. $Ni^{2+}$ (50 ${\mu}M$), T-type $Ca^{2+}$-channel blocker was treated to medium contaning low concentration of $Ca^{2+}$. It overcame 2-cell block significantly. Rate of degenerated embryos decreased and developmental rate to morula and blastocyst increased more than low $Ca^{2+}$ concentration alone. Further studies are needed for the overcoming effect of 2-cell block by $Ni^{2+}$.
