Study on the Anti-cancer, Anti-metastasis and Immune response improvement of Aqua-acupuncture with Cistanches Herba infusion solution (육종용약침의 항암작용(抗癌作用) 및 면역효과(免疫效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement of aqua-acupuncture with Cistanches Herba infusion solution, we used Cistanches Herba infusion solution(taken by water-alcohol method) put into Chung-wan(CV12) and Chok-Samni(S36) of BALB/c or C57BL/6 which are corresponding to human body. We observed the cytotoxicity, the effect on the expression of MMP-9 gene, the ability to control cancer cell proliferation, change of body weight, surviving number, MST, ILS, changes in amount of WBC, RBC, PLT, total protein, creatinine, glucose and LDH, weight of spleen, number of pulmonary colony, histological analysis on tissue metastasis of lung and liver, splenic cell proliferation, the expression of cytokine gene, the number of $CD4^+$, $CD8^+$, $CD19^+$ and NK cell, and concluded like this. The results were obtained as follows : 1. The cytotoxicity about B16-F10 cell line of $2^0$, $2^{-1}$, $2^{-2}$, $2^{-5}$ diluent groups in Cistanches Herba infusion solution treatment was inhibited significantly, compared with control group. 2. The cytotoxicity about HT1080 cell line of $2^0$, $2^{-1}$, $2^{-2}$, $2^{-3}$, $2^{-5}$, $2^{-8}$ diluent groups in Cistanches Herba infusion solution treatment was inhibited significantly, compared with control group. 3. The effect on expression of MMP-9 gene was decreased in all the sample groups, compared with control group. 4. The effect on the control-ability on the cancer cell proliferation showed cytotooicity significantly in $2^0$, $2^{-1}$, $2^{-2}$, $2^{-3}$, $2^{-4}$, $2^{-5}$ diluent groups. 5. S-180 cancer cell line transplants in BALB/c mice were inhibited significantly in weight increase in all the sample groups, compared with control group. The surviving number increased in almost sample groups, except one group put into Chok-Samni(S36) with 20% Cistanches Herba infusion solution treatment group that showed same number of the control group. 6. S-180 cancer cell line transplants in BALB/c mice showed high MST and ILS significantly in almost sample groups, compared with control group. But one group put into Chok-Samni(S36) with 20% Cistanches Herba infusion solution treatment group showed low MST and ILS than control group. 7. The sample group injected in vein with B16-F10 cancer cell line in C57BL/6 mice showed increased ILS compared with control group significantly in anti-metastasis test. 8. The sample group injected in vein with B16-F10 cancer cell line in C57BL/6 mice were increased significantly in the number of WBC and glucose, and decreased significantly in the amount of LDH, compared with control group. However, there's no significant increase or decrease in number of RBC, PLT, total protein and creatinine. 9. We couldn't find any significant relation in spleen weight of the sample group. 10. In pulmonary colony, sample group was decreased significantly, compared with control group. 11. Histological analysis of sample group inhivited compared with that of control group in both of lung and liver. 12. In immune system, all the sample groups showed having more relevancy to the effect on splenic cell proliferation than normal group. 13. The effect on cytokine gene expression of all the sample groups were increased than control group. 14. In flow cytometry there's no significant relation in number of $CD8^+$, $CD19^+$ cell, however, the number of $CD4^+$ cell and NK cell in sample groups were increased than in control group. Above the results showed that aqua-acupuncture of Cistanches Herba infusion solution has effects of anti-cancer, anti-metastasis and immune response improvement.

Cooking Properties and Antioxidant Activity of Cooked Rice According to the Addition of Glutinous and Non-glutinous Sorghum (찰수수 및 메수수 첨가 밥의 취반특성과 항산화활성)

  • The purpose of this study was to evaluate the physicochemical characteristic of the cooked rice added with glutinous and non-glutinous sorghum. The sorghum cultivars were Sorghum bicolor L. Moench cv. Hwanggeumchal, Nampungchal (glutinous), and Donganme (nonglutinous), and rice cultivar was Ilpum rice. The cooking properties and pasting characteristics of cooking rice adding with sorghum according to varieties and different addition rates evaluated. The cooking properties and pasting characteristics had significant changes with the varieties and different addition rates of sorghum. With increased addition rates of sorghum, the pasting temperature, peak viscosity, trough viscosity, breakdown viscosity, and final viscosity were decreased. With increased addition rates of sorghum, the total polyphenol and flavonoid contents before and after cooked rice were increased. Total polyphenol contents of 30% addition rates before cooking rice with Hwanggeumchal, Nampungchal, and Donganme sorghum were 1,693.30, 1,890.98 and $2,386.11{\mu}g/g$ sample, whereas those after cooking rice with sorghum were 1,189.28, 1,190.42 and $1,397.87{\mu}g/g$ sample, respectively. The high level of DPPH radical scavenging activity before and after cooking rice with sorghum were 126.29 and 70.58 mg TE/100g sample in the Donganme in 30% addition rates. Also, ABTS radical scavenging activity was 135.56 and 83.12 mg TE/100g sample, respectively. The results of this study show that the addition of sorghum can make cooked rice improved antioxidant activity.

Multi-residue Pesticide Analysis in Cereal using Modified QuEChERS Samloe Preparation Method (곡물류 중 잔류농약 다성분 분석을 위한 개선된 QuEChERS 시료 정제법의 개발)

  • This study explored an efficient modified Quick, Easy, Cheap, Effective, Rugged and safe (QuEChERS) method combined with liquid chromatography-electrospray ionization with tandem mass spectrometric detection for the analysis of residues of 76 pesticides in brown rice, barley and corn including acidic sulfonylurea herbicides. Formic acid (1%) acid in acetonitrile and dispersive solid phase extractions used for extraction of pesticides and clean-up of the extract respectively. Two fortified spikes at 50 and 200 ng $g^{-1}$ levels were performed for recovery test. Mean recoveries of majority of pesticides at two spike levels ranged from 73.2 to 132.2, 80.9 to 136.8, 66.6 to 143.5 for brown rice, barley and corn respectively with standard error (CV) less than 10%. Good linearity of calibration curves were achieved with $R^2$ > 0.9907 within the observed concentration ranged. The modified method also provided satisfactory results for sulfonylurea herbicides. The method was applied to the determination of residues of target pesticides in real samples. A total of 26 pesticides in 36 out of 98 tasted samples were observed. The highest concentration was observed for tricyclazole at 1.17 mg $kg^{-1}$ in brown rice. This pesticide in two brown rice samples exceeded their MRLs regulated for rice in republic of Korea. Except tricyclazole none of the observed pesticides' concentration was higher than their MRLs. The results reveal that the method is effectively applicable to routine analysis of residues of target pesticides in brown rice, barley and corn.

Responses to 1-MCP during Storage of Kimchi Cabbage Ryouckgwang Cultivar (배추 력광 품종의 저장 중 1-MCP에 대한 반응)

  • The effect of 1-methylcyclopropene (1-MCP) in the storability of kimchi cabbage at cold storage condition was investigated. Kimchi cabbage (Brassica campestris L. cv Ryouckgwang) was divided four groups, forced air cooling (FAC), FAC + 0.03 mm linear low density polyethylene liner (Liner), $FAC+2{\mu}L{\cdot}L^{-1}$ 1-MCP (1-MCP), and FAC + 1-MCP + Liner. After each treatment kimchi cabbage was stored at $2^{\circ}C$, 95% RH. Quality parameters were weight loss, soluble solids content (SSC), firmness, and color ($CIE\;L^*$, $a^*$, $b^*$, chroma, hue angle). Weight loss during storage was showed significant difference by Liner treatment. In particular FAC + 1-MCP + Liner treatment showed 12.5% reduction after 6 weeks of storage period and minimized the weight loss rate compared to other treatments. SSC of kimchi cabbage was $2.5^{\circ}Brix$ at harvest and FAC + 1-MCP + Liner treatment maintained the SSC until 3 weeks, while in other treatments gradually were increased. The firmness of kimchi cabbage was 24.0 N immediately after harvest and the firmness at harvest time tended to be maintained at 22.6 N after 6 weeks of storage in FAC + 1-MCP + Liner treatment. During the storage period, the color change of the kimchi cabbage leaf can be confirmed by $CIE\;a^*$ and hue angle value. 1-MCP treatment alone did not affect the color change, however 1-MCP + Liner treatment was able to maintain the chromaticity at harvest time while minimizing the change of $CIE\;a^*$ and hue angle. These results suggest that 1-MCP treatment is not effective for the storage of kimchi cabbage but can be maintained for up to 6 weeks when treated with Liner.

Lateral Bud Suppression and Runner Plants Growth of 'Maehyang' Strawberry as Affected by Application Method and Concentration of IBA (IBA 처리방법과 농도에 따른 딸기 '매향'의 측아 발생 억제와 자묘 생육)

  • This study was conducted to examine the effect by application method and concentration of the indole-3-butyric acid (IBA), which is auxin-based plant growth regulator, on the growth and runner plants production of strawberry in the greenhouse. The seedlings of strawberry were transplanted in the pot (150 ×135 × 90 mm) filled with coir medium on April 12, 2019. The IBA was applied with a foliar spray or drench as 50, 100, 150, and 200 mg·L-1 (50 mL per plant), respectively. The treatment was started on April 29, 2019. The foliar spray and drench treatment of IBA were repeated at 2-week intervals for 9 weeks from the start date of treatment. At 9 weeks after treatment, the petiole length of mother plants was the shortest in the control. The number of runner plants showed a tendency to decreased in the foliar spray. The number of lateral buds showed a tendency to decreased in the IBA treatment, and the least in the foliar with 100 mg·L-1. There was not significantly difference in the fresh and dry weights of the first and second runner plants. However, in the third runner plants, the fresh and dry weights were the greatest in the drench with 100 mg·L-1. Therefore, when considering the growth of third runner plants and lateral bud suppression, the drench with the 100 mg·L-1 could be better application method and concentration of IBA treatment for growth of the third runner plants and runner plants production of strawberry, and the results can be used as a basic research of plant growth regulator application to save the labor force and enhance the seedling quality in strawberry seedling stage.

Soil Characteristics of the Saprolite Piled Upland Fields at Highland in Gangwon Province (강원도 고랭지의 석비레 성토지 토양 특성)

  • As one of the typical farming practices in the sloped upland in Pyeongchang and Hongcheon area, application of piling with coarse saprolite materials has been practiced by farmers for several reasons such as reduction of damage by monocropping, better development of plant roots, and better drainage. However, adverse effect on application of coarse saprolite soil materials to environmental aspects should not be ignored. Therefore, this research was conducted to evaluate the physicochemical properties of coarse saprolite materials in upland fields in Pyeongchang area. According to particle size distribution of coarse saprolite materials, averaged gravel contents for Pyeongchang and Hongcheon county were 16.7 and 25.3%, respectively. There was no significant difference in gravel contents by soil depth, and CV values for each particle size ranged from 20 to 40%, which implied that application of coarse material with similar properties. When we compared CEC values of dressed soil with or without considering gravel content, CEC values decreased as increasing gravel contents. The penetration resistances were 0.04-7.48 MPa at the 0 to 10 cm surface soil, and 0.10 to 8.80 MPa at the depth below 11 cm. The bulk density of the soil was $1.15g\;cm^{-3}$ at the surface soil and 1.29 to $1.35g\;cm^{-3}$ at the soil depth below 10 cm. The organic matter content, cation exchange capacity, and avaliable $P_2O_5$ concentrations of soil in upland where piling with saprolite materials of Pyeongchang area applied were $12.4g\;kg^{-1}$, $7.1cmol_c\;kg^{-1}$, and $526mg\;kg^{-1}$, respectively. Cation exchange capacity was lower than that of averaged Korean upland soil, while available $P_2O_5$ concentration was relatively higher than that of averaged Korean upland, which indicated high input of various fertilizers.

Development of a Method for Analyzing the Nicotine Content in Synthetic Flavoring Substances as Unauthorized E-cigarette Liquid by Using HPLC (전자담배 액상 충진제와 유사한 합성착향료 중 니코틴의 HPLC 분석법 개발)

  • A simple, sensitive, and specific method for quantifying the nicotine content of synthetic favoring substances (SFS) was developed using high performance liquid chromatography (HPLC) with a photo-diode array detector (PDA). Nicotine was extracted from SFS samples by using an acid-base liquid-liquid extraction method with dichloromethane and distilled water. The nicotine content was quantified by HPLC/PDA (261.9 nm) with a $C_{18}$ column under a gradient of 10% acetonitrile with 20 mM ammonium formate (ammonia solution adjusted to pH 8.7) to 100% acetonitrile. The calibration curve, analyzed from concentration standards between 0.1 to 2 mg/L, presented linearity with a correlation coefficient ($r^2$)>0.9999. The limit of quantitation (LOQ) of nicotine in SFS was 0.4 mg/kg, and the average recoveries ranged from 76.4% to 96.3%. The repeatability of measurements, expressed as the coefficient of variation (CV%), ranged from 1.74 to 5.12%. This newly developed method for nicotine quantification in SFS can be considered an analytical method with an acceptable level of sensitivity and repeatability.

The Characteristics of Anatomical Structure and Fruit Quality According to Fruit Developmental Stage of Pyrus pyrifolia Nakai cv. Manpungbae ('만풍배'의 생육기별 해부학적 구조와 과실품질)

  • This study was carried out to understand the physiological characteristics of 'Manpungbae' (Pyrus pyrifolia Nakai) pears through the seasonal changes of pericarp structure and anatomical differences between bagging and non-bagging treatment, and also fruit quality and peel coloration characteristics at the harvest time. The pericarp at full bloom was consists of outer epidermis, hypodermis, parenchyma cell, and inner epidermis from the exterior. The cell layers from the outer epidermis to vascular bundle increased rapidly 7-10 layers to 18-26 layers from full bloom (FB) to 77 days after full bloom (DAFB) and did not change significantly until maturity. Thus, the cell division period of 'Manpungbae' pear was until 77 DAFB and during this period, the thickness from hypodermis to vascular bundle increased from $73.1{\mu}m$ to $195{\mu}m$ in this period. Stone cells were formed from seven to 21 DAFB and stone cell clusters were formed around 49 DAFB. The cork cell layer was formed between 49 and 77 DAFB. 'Manpungbae' fruit pericarp was consists of 4.5 layers of the cork cell layers and seven layers of hypodermis which has the tannin at harvest time (161 DAFB). Comparison of the fruit enlargement and fruit structure development by bagging or non-bagging showed that 'Manpungbae' fruits without bagging had more than three cork cell layer than those with bagging at maturity. The size of stone cell clusters were varied in two treatments. Fruit weight was higher in the non-bagging treatment but there was no difference in soluble solid contents (SSC) between two treatments. The weight of the 'Manpunbae' fruit was distributed from 301 g to more 900 g and the average fruit weight was 677.2 g at harvest time, and fruits in the range of 551-800 g accounted for 71.6% of total production. The SSC, acidity and SSC/acidity ratio was $10.2-12.1^{\circ}Brix$, 0.10-1.24% and 9.76-14.31 respectively, and the SSC was higher in bigger fruit which had a very higher positive correlation with a fruit weight. However, the fruit firmness tended to be lower with fruit size which had a very higher negative correlation with the fruit weight and SSC. The cork cell layer numbers between yellowish brown and green pericarp were not different significantly, in 3.8 and 3.5 respectively.

Single Laboratory Validation and Uncertainty Estimation of a HPLC Analysis Method for Deoxynivalenol in Noodles (면류에서 HPLC를 이용한 데옥시니발레놀 분석법의 검증과 불확도 산정)

  • An isocratic high performance liquid chromatography (HPLC) method for routine analysis of deoxynivalenol in noodles was validated and estimated the measurement uncertainty. Noodles (dried noodle and ramyeon) were analyzed by HPLC-ultraviolet detection using immunoaffinity column for clean-up. The limits of detection (LOD) and quantification (LOQ) were 7.5 ${\mu}g$/kg and 18.8 ${\mu}g$/kg, respectively. The calibration curve showed a good linearity, with correlation coefficients $r^2$ of 0.9999 in the concentration range from 20 to 500 ${\mu}g$/kg. Recoveries and Repeatabilities expressed as coefficients of variation (CV) spiked with 200 and 500 ${\mu}g$/kg were $82{\pm}2.7%$ and $87{\pm}1.3%$% in dried noodle, and $97{\pm}1.6%$ and $91{\pm}12.0%$ in ramyeon, respectively. The uncertainty sources in measurement process were identified as sample weight, final volume, and sample concentration in extraction volume as well as components such as standard stock solution, working standard solution, 5 standard solutions, calibration curve, matrix, and instrument. Deoxynivalenol concentration and expanded uncertainty in two matrixes spiked with 200 ${\mu}g$/kg and 500 ${\mu}g$/kg were estimated to be $163.8{\pm}52.1$ and $435.2{\pm}91.6\;{\mu}g$/kg for dried noodle, and $194.3{\pm}33.0$ and $453.2{\pm}91.1\;{\mu}g$/kg for ramyeon using a coverage factor of two which gives a level of statistical confidence with approximately 95%. The most influential component among uncertainty sources was the recovery of matrix, followed by calibration curve.

Differential Tolerance of Rice Cultivars to Mesotrione-Contained Herbicides (Mesotrione 함유 제초제에 대한 벼 생태형간 약해반응 차이)

  • Greenhouse experiment was conducted to evaluate tolerance of six rice cultivars, three Indica${\times}$Japonica rice (long grain, cv. Dasanbyeo, Segejinmi and Hanareumbyeo) and three Japonica rice (short grain, Nampyung, Ilpumbyeo and Junamjosaeng) cultivars, to mesotrione+pretilachlor (MP) and bensulfuron-methyl+mesotrione+pretilachlor+pyriftalid (BMPP) in transplanting rice. Two herbicides were applied at 90 g and 180 g a.i. $ha^{-1}$ to three to four leaf stage rice at 5 and 15 days after transplanting, respectively. Related study was also conducted to compare $GR_{50}$ for Indica${\times}$Japonica and Japonica rice cultivars at different MP rates. Response to two herbicides varied with respect to rice cultivars and herbicide rates. All Indica${\times}$Japonica rice cultivars exhibited susceptible to both MP and BMPP as reflected by increased visual injury, shorter plant height and higher plant dry matter reduction when compared with nontreated rice cutlivars. The degree of foliar chlorosis by MP at 90 g a.i. $ha^{-1}$ was 5~6 at 7 days after treatment(DAT) but it was decreased to 3~6 at 14DAT. The degree of leaf chlorosis treated with 180 g a.i. $ha^{-1}$ MP was 6~7 at 7DAT and it was also decreased to 3~8 at 14 DAT. The plant hight of Indica${\times}$Japonica rices was inhibited by 18~43% at application of 90 g a.i. $ha^{-1}$ MP and 30~50% at 180 g a.i. $ha^{-1}$ MP. The dry matter reduction was greater than that of plant height inhibition, showing 46~73% at 90 g a.i. $ha^{-1}$ MP and 65~82% at 180 g a.i. $ha^{-1}$ MP. Similar leaf chlorosis injury and growth inhibition of rice cultivars was observed in the BMPP treatment. The injury and growth inhibition by MP and BMPP increased with increase in herbicide rate from 90 g a.i. $ha^{-1}$ to 180 g a.i. $ha^{-1}$. However, most of the Japonica cultivars are tolerant to MP and BMPP at both rates. There was no visible leaf chlorosis but plant height and dry matter production were slightly reduced at 180 g a.i. $ha^{-1}$. Based on $GR_{50}$ value determined in reduction of shoot dry weight for MP, the Indica${\times}$Japonica rice showed 12.9 fold lower compared with the Japonica rice. The result indicates that rice cultivars vary in tolerance to herbicides of MP and BMPP and Indica${\times}$Japonica rices were more susceptible than the Japonica rices to the MP and BMPP.
