Changes of Free Radical Concentrations with Irradiation Dose and Storage Time in Gamma-Irradiated Sesame and Perilla Seeds (감마선 조사된 참깨, 들깨의 조사선량과 저장기간에 따른 free radical 농도의 변화)

  • Electron spin resonance (ESR) spectroscopy was used to investigate the effect of irradiation dose and storage time on the free radical concentrations in gamma-irradiated sesame (Korean and Sudan) and perilla seeds (Korea). The samples were sealed with packed pyrex tube (50 g respectively) and then irradiated with doses of 0, 1, 5 and 10 kGy at room temperature using a Co-60 irradiator. Each sample in ESR quartz tube were calculated with ESR signals by Bruker-EPR spectrometer for 4 weeks. Free radicals detected for irradiated and unirradiated samples were higher than those of unirradiated ones. Since the free radical concentrations linearly incresed with the applied doses $(1{\sim}10\;kGy)$, highly positive correlation coefficients $(R^2=0.9926{\sim}0.9994)$ were obtained between irradiation doses and free radical concentrations right after irradiation. Although the free radical concentrations decreased with storage time, the signals of free radical in irradiated samples were observed even after 4 weeks of storage at room temperature.

EFFICACY AND BIOLOGICAL ACTIVITIES OF A NEW ANTI-AGING AGENT OBTAINED FROM ARECA CATECHU

  • Inhibitory effects of the new material obtained from Areca catechu seed (CC-516) according to a special process, and its applicability to the skin as a cosmetic raw material in terms of its efficacy were presented. Areca catechu extract out of 150 medicinal plants, exhibited high inhibitory effect on the porcine pancreatic elastase ($IC_{50}$ : $40.8{\mu}$g/ml). It also had an inhibitory effect on the human leukocyte elastase ($IC_{50}$ : 48.1$\mu$g/ml), hyaluronidase ($IC_{50}$ : $416{\mu}$g/ml), antioxidative activity ($IC_{50}$ : $45.4\mu$g/ml) and free radical scavenging activity ($SC_{50}$ : $10.2{\mu}$g/ml). The cream contained 3% of CC-516 improved skin hydration above 16.5%. Especially, the skin elasticity increases more than 35% and skin wrinkles decreased more than 23%. The CC-516 was designed to be utilized in cosmetology. The cream containing 3% of this product has not only protecting effect on the skin mechanical properties provided by the collagen and the elastin in the derm but also restructuring effect of scarring or aging tissue.

Skin Anti-aging Effect of Forsythia viridissima L. Extract (연교추출물의 피부 항노화 효과)

  • Skin anti-aging effect of Forsythia viridissima L. extract was evaluated by using antioxidant assay, expression of type I procollagen, and UVA-induced matrix metalloproteinase-1 in human dermal fibroblasts. Matairesinol-rich Forsythia viridissima L. extract was showed the scavenging activity of radicals and reactive oxygen species with the $IC_{50}$ values of $4.50\;{\mu}m/ml$ against 1,1-diphenyl-2-picrylhydrazly radical and $542.43\;{mu}m/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. The type I procollagen was increased 33.76% by treatment with matairesinol-rich Forsythia viridissima L. extract, and UVA-induced MMP-1 was reduced 35.78% in a dose dependent manner. In the human skin irritation test, 2% matairesinol-rich Forsythia viridissima L. extract did not show any adverse effect. Also, the clinical study indicated that a cream group treated with 0.2% matairesinol-rich Forsythia viridissima L. extract significantly reduced skin wrinkles, as compared with a non-treated cream group (p < 0.05). These results suggest that Forsythia viridissima L. extract may be useful as a potential source of functional anti-aging cosmetics.

Flavonol Glycosides from the Aerial Parts of Aceriphyllum rossii and Their Antioxidant Activities

  • The methanol extract obtained from the aerial parts of Aceriphyllum rossii (Saxifragaceae) was fractionated into ethyl acetate (EtOAc), n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel column chromatography of EtOAc and n-BuOH layers afforded six flavonol glycosides. They were identified as kaempferol 3-O-$\beta$-D-glucopyranoside (astragalin, 1), quercetin 3-O-$\beta$-D-glucopyranoside (isoquercitrin, 2), kaempferol 3-O-$\alpha$-L-rhamnopyranosyl $(1{\to}6)-\beta$-D-glucopyranoside (3), quercetin 3-O$\alpha$-L-rharnnopyranosyl $(1{\to}6)-\beta$-D-qlucopyrano-side (rutin, 4), kaempferol 3-O-[$\alpha$-L-rharnnopyranosyl $(1{\to}4)-\alpha$-L-rhamnopyranosyl $(1{\to}6)-\beta$-D-glucopyranoside] (5) and quercetin 3-O-[$\alpha$-L-rhamnopyranosyl $(1{\to}4)\alpha$-L-rhamnopyranosyl $(1{\to}6)\beta$-D-glucopyranoside] (6) on the basis of several spectral data. The antioxidant activity of the six compounds was investigated using two free radicals such as the ABTS free radical and superoxide anion radical. Compound 1 exhibited the highest antioxidant activity in the ABTS $\{2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)\}$ radical scavenging method. 100 mg/L of compound 1 was equivalent to $72.1\pm1.4\;mg/L$ of vitamin C, and those of compounds 3 and 5 were equivalent to $62.7\pm0.5\;mg/L$ and $54.3\pm1.3\;mg/L$ of vitamin C, respectively. And in the superoxide anion radical scavenging method, compound 5 exhibited the highest activity with an $IC_{50}$ value of $17.6{\pm}0.3{\mu}M$. In addition, some physical and spectral data of the flavonoids were confirmed.

  • Let M, N be modules over a ring R and $[M,N]=Hom_R(M,N)$. The concern is study of: (1) Some fundamental properties of [M, N] when [M, N] is regular or semipotent. (2) The substructures of [M, N] such as radical, the singular and co-singular ideals, the total and others has raised new questions for research in this area. New results obtained include necessary and sufficient conditions for [M, N] to be regular or semipotent. New substructures of [M, N] are studied and its relationship with the Tot of [M, N]. In this paper we show that, the endomorphism ring of a module M is regular if and only if the module M is semi-injective (projective) and the kernel (image) of every endomorphism is a direct summand.

Antioxidative and Anti-inflammatory Effects of Petasites japonicus (머위추출물의 항산화와 항염증 효과)

  • Antioxidative and anti-inflammatory activities of Petasites japonicus extract were evaluated. P. japonicus extract showed 70.1% inhibition on peroxidation of linoleic acid. In the experiment using the cell permeable dye, 2',7'- dichlorofluorescein diacetate (DCFDA) as an indicator of reactive oxygen species (ROS) generation, intracellular oxidative stress in UVB-irradiated keratinocytes was shown to be decreased by P. japonicus extract. Also, UVB-induced production of interleukin-$1{\alpha}$ and prostaglandin $E_2$ in human HaCaT keratinocytes was reduced in a dose-dependent manner by treatment with P. japonicus extract. All these results suggest that P. japonicus extract can be effectively used for prevention of UV-induced adverse skin reactions such as radical production and inflammation.

Preparation and Characterization of Graft Copolymer/$TiO_2$ Nanocomposite Polymer Electrolyte Membranes (가지형 공중합체/$TiO_2$ 나노복합 고분자 전해질막의 제조 및 분석)

  • A graft copolymer, i.e. poly(vinylidene fluoride-co-chlorotrifluoroethylene )-g-poly(styrene sulfonic acid) (P(VDF-co-CTFE)-g-PSSA) with 47 wt% of PSSA was synthesized via atom transfer radical polymerization (ATRP). This copolymer was combined with titanium isopropoxide (TTIP) to produce graft copolymer/$TiO_2$ nanocomposite membranes via sol-gel process. $TiO_2$ precursor (TTIP) was selectively incorporated into the hydrophilic PSSA domains of the graft copolymer and grown to form $TiO_2$ nanoparticles, as confirmed by FT-IR and UV-visible spectroscopy. Water uptake and ion exchange capacity (IEC) decreased with TTIP contents due to the decrease in number of sulfonic acid in the membranes. At 5 wt% of TTIP, the mechanical properties of membranes increased while maintaining the proton conductivity.

A Study on the Laminar Burning Velocity and Flame Structure with H2 Content in a Wide Range of Equivalence Ratio of Syngas(H2/CO)/Air Premixed Flames (넓은 당량비 구간에서 수소함유량에 따른 합성가스(H2/CO)/공기 예혼합화염의 연소속도 및 화염구조에 관한 연구)

  • In this study, the laminar burning velocity of syngas fuel($H_2/CO$) and flame structure with various hydrogen contents were studied using both experimental measurements and detailed kinetic analysis. The laminar burning velocities were measured by the angle method of Bunsen flame configuration and the numerical calculations including chemical kinetic analysis were made using CHEMKIN Package with USC-Mech II. A wide range of syngas mixture compositions such as $H_2$ : CO = 10 : 90, 25 : 75, 50 : 50, 75:25 and equivalence ratios from lean condition of 0.5 to rich condition of 5.0 have been considered. The experimental results of burning velocity were in good agreement with previous other research data and numerical simulation. Also, it was shown that the experimental measurements of laminar burning velocity linearly increased with the increment of $H_2$ content although the burning velocity of hydrogen is faster than the carbon monoxide above 10 times. This phenomenon is attributed to the rapid production of hydrogen related radicals such as H radical at the early stage of combustion, which is confirmed the linear increase of radical concentrations on kinetic analysis. Particular concerns in this study are the characteristics of burning velocity and flame structure different from lean condition for rich condition. The decrease of OH radicals and double peaks are observed with $H_2$ content in rich condition once $H_2$ fraction exceeds over threshold.

Effect of Irradiation Dose and Storage Time on the Free Radical Concentrations in Gamma-reradiated Dried Seasoning Powder

  • Electron spin resonance (ESR) spectroscopy was used to investigate the effect of irradiation dose and storage time on the free radical concentrations in gamma-irradiated dried seasoning powder. Seasoning powders of dried squid flavor, shrimp flavors, kimchi flavor, spicy beef soup flavor and soy sauce flavor were irradiated with doses of 0, 1, 3, 5, 7 and 9 kGy at room temperature using a Co-60 irradiator. Characteristic signals of free radicals were observed in all the irradiated samples of this experimental while these signals were not detected in non-irradiated samples. Since the free radical concentrations linearly increased with the applied doses (1~9 kGy), highly positive correlation coefficients ($R^2$ = 0.9285~0.9965) were obtained between irradiation doses and free radical concentrations during all the storage times. Free radical concentrations of the irradiated flavored seasoning powder did not change even at 16 weeks of storage at room temperature, while those of dried squid, shrimp and spicy beef soup flavors decreased until 2 weeks of storage after irradiation with 5 kGy or over, and these of soy cause flavors slowly decreased until 8 weeks of storage after irradiation with 3 kGy or over. Although the free radical concentrations decreased with storage times, the characteristic signals of the irradiated seasoning powders of dried squid, shrimp, spicy beef and soy sauce flavors were observed even after 16 weeks of storage at room temperature.

Convenient Assay of O2- Generated on Potato Tuber Tissue Slices Treated with Fungal Elicitor by Electron Spin Resonance - No Secondary Oxidative Burst Induction by H2O2 Treatment

  • Since the discovery of generation of $O_2^-$ in plant, many evidence for the oxidative burst (OXB) has been accumulated in various combinations of plant and pathogen or elicitor systems. $O_2^-$ generating system responsible for the OXB was coupled with oxidation of reduced nicotinamide adenine dinucleotide phosphate (NADPH) in microsomal fraction isolated from sliced aged potato tuber slices which were treated by hyphal wall components elicitor from Phytophthora infestans (HWC). We developed new assay method for quantitative measurement of oxygen radical $O_2^-$ by using electron spin resonance (ESR) analysis during elicitor­induced OXB on the surface of plant tissues. The ESR analysis using an $O_2^-$ trapper, Tiron (1,2-dihydroxy-3,5­benzenedisulfonic acid), provided a convenient assay for detecting only $O_2^-$ during elicitor-induced OXB producing various active oxygen species (AOS) on plant tissue surface. Tiron was oxidized to Tiron semiquinon radical by $O_2^-$. Quantity of the radical signal was measured by specific spectra on ESR spectroscopy. The level of $O_2^-$ was high in from surface of potato tuber tissue treated with hyphal cell wall elicitor (HWC) from Phytophthora infestans. There was no secondary OXB induction by $H_2O_2$ treatment in plant.
