장기간 알콜투여가 생쥐 비장의 세포성 면역 저해에 미치는 면역조직화학적 연구 : T 림프구, IL-2 수용기 및 NK세포의 변화를 중심으로 (Imunohistochemical study on the inhibition of cell mediated immunity in spleen of mouse by chronic alcohol administration : Based on the change of T lymphocytes, IL-2 receptors, and NK cells)

  • As a mood-altering drug, long-term alcohol consumption have significant harmful effects on the human body and people's mental functioning. This study observed that the suppression of cell mediated immunity induced in spleen of ICR mouse by long-term alcohol administration. After 8% alcohol voluntary administered for 120 days, the splenic tissue irnmunohistochemically stained by following ABC method that used monoclonal antibody including L3T4(CD4), Ly-2(CD8), IL-2 receptor(CD25R) and NK-1.1(CD56) after embedding with paraffin. The results were as follows. 1. The size of marginal zone in splenic white pulp was diminished and the number of macrophage in marginal zone was decreased in test group than control group. 2. After alcohol administration, the number of Helper T lymphocyte, cytotoxic T lymphocyte, and IL-2 receptor were decreased in periarterial lymphatic sheaths of white pulp and penicilla artery of red pulp and the degree of CD4, CD8, and CD25R positive reaction were soften. 3. In test group, the number of NK cell were decreased. These results indicated that the secretion of lymphokine as IL-2 was inhibited by long-term alcohol administration and subsequently prevent to activate and proliferate splenic T lymphocytes and NK cells as cell mediated immunity component.

  • Cell separation from peripheral blood was investigated using surface-modified polyurethane (PU) membranes with different functional groups. Both red blood cells and platelets could pass through unmodified PU and PU-SO$_3$H membranes, while the red blood cells preferentially passed through PU-N(C$_2$H$_{5}$ )$_2$ and PU-NHC$_2$H$_4$OH membranes. The permeation ratio of T and B cells was less than 25% for the surface-modified and unmodified PU membranes. CD34$^{+}$ cells have been recognized as various kinds of stem cells including hematopoietic and mesenchymal stem cells. The adhesiveness of CD34$^{+}$ cells on the PU membranes was found to be higher than that of red blood cells, platelets, T cells or B cells. Overall, the adhesiveness of blood cells on the PU membranes increased in the following order: red blood cells $\leq$ platelets < T cells $\leq$ B cells < CD34$^{+}$ cells. Treatment of PU-COOH membranes with a human albumin solution to detach adhered blood cells, allowed recovery of mainly CD34$^{+}$ cells in the permeate, while both red blood cells and platelets could be isolated in the permeate using unmodified PU membranes. The PU membranes showed different permeation and recovery ratios of specific cells depending on the functional groups attached to the membranes.mbranes.

Associations Between Infiltrating Lymphocyte Subsets and Hepatocellular Carcinoma

  • Aims: We aimed to analyze the phenotype of tumor-infiltrating lymphocytes (TILs) and non-tumor infiltrating lymphocytes (NILs) in HCC and non-tumor tissues, and evaluate relationships between changes in these cells and the prognosis of HCC. Methods: Lymphocytes were isolated from HCC and corresponding non-tumor tissues and tested by flow cytometry. For comparison, clinical parameters were analyzed. Results: Compared with the non-tumor tissue, tumor tissue had a lower intensity of NK, NKT andCD8+T cell infiltration. TILs had higher intensity of CD4+CD25+Foxp3+regulatory T cell (Treg cells) infiltration compared with that in NILs. The prevalence of Treg cells was associated with fewer CD8 + T lymphocytes in the HCC immune microenvironment. The frequencies of NK cells and CD8+T cells in TILs of HCC patients with metastasis less than 12 months were lower than those without metastasis. However, the frequency of Treg cells was higher than those without metastasis. Conclusion: These results suggest that the frequencies of CD8+T, NK and NKT cells as well as Treg cells in the tumor tissue of HCC are significantly associated with patient survival, and could be applied as predictive indicators for HCC prognosis.

Induction of Regulatory Dendritic Cells by Lactobacillus paracasei L9 Prevents Allergic Sensitization to Bovine β-Lactoglobulin in Mice

  • Supplementation with probiotics can protect against the development of food allergies by modulating the host immune response; however, the mechanisms are not fully understood. The objective of this study was to investigate the allergy-reducing effects of regulatory dendritic cells (regDCs) induced by Lactobacillus paracasei L9 (L9) in β-lactoglobulin (BLG)-sensitized mice. The L9 supplement suppressed the aberrant balance of Th1/Th2 responses to BLG in mice, via upregulation of the CD4+CD25+Foxp3+Treg cell responses. The amount of CD4+CD25+Foxp3+Treg cells in mesenteric lymph nodes increased by 51.85%. Furthermore, administration of L9 significantly induced the expression of CD103 and reduced the maturation status of DCs in mesenteric lymph nodes, Peyer's patches, and spleen. Bone marrow-derived dendritic cells (BM-DCs) were activated by L9 in vitro, with an approximate 1.31-fold and 19.57-fold increase in expression of CD103 in CD11c+DCs and the level of IL-10 production, respectively, while the expression of CD86 did not change significantly. These data demonstrate that L9 reduced the BLG allergic sensitization, likely through regDCs mediated active suppression.

  • 인간면역결핍바이러스(Human immunodeficiency virus; HIV), B형 간염 바이러스(Hepatitis B virus; HBV), 그리고 C형 간염 바이러스(Hepatitis C virus; HCV)는 만성 감염질환을 일으키는 대표적인 바이러스들이다. 인체내 감염시 임상적 진행경과에 따른 바이러스 특이 T림프구의 항바이러스 기능변화 및 바이러스의 체내 지속성과 T림프구에 발현되는 다양한 면역인자(e.g., CD28, CD25, FoxP3, PD-1, CTLA-4)들과의 구체적인 상관관계는 최근 많은 국내외 연구진들을 통해 연구되고 있다. 그 중 FoxP3 (forkhead box P3), PD-1 (programmed death-1) 그리고 CTLA-4 (cytotoxic T lymphocyte-associated antigen 4)는 T림프구에서 발현되는 면역조절인자로 만성 바이러스성 감염시 그 발현이 증가되는 것으로 관찰되었으며, 항바이러스 작용을 가지는 T림프구의 기능결핍과 밀접한 상관관계가 있는 것으로 알려져 있다. 본 총설에서는 만성적인 HIV, HBV, 그리고 HCV 감염에서 바이러스 특이 T림프구에서 발현되는 FoxP3, PD1, 그리고 CTLA-4의 발현변화와 각 질환의 임상적 진행경과와의 상관성, 그리고 이들 발현이 T림프구의 항바이러스 기능에 미치는 영향 등을 중심으로 기술하였다.

벼메뚜기(Oxya japonica japonica Thunberg) 물 추출물 LCT-CT의 항암면역 활성에 관한 유세포 분석학적 연구 (A Flow Cytometrical Analysis of the Antitumor and Immunostimulatory Effects of LCT-CT, a Cold-water Extract Prepared from Rice Grasshopper Oxya japonica japonica Thunberg)

  • Water extracts of rice grasshopper (Oxya japonica japonica Thurnberg) were prepared and their antitumor and immunostimulatory activities were investigated using a flow cytometer. When LCT-CT was ip injected into ICR mice at the dose of 33.3 mg/kg before and after the implantation of $4{\times}10^5$ cells/mouse of sarcoma 180 tumor cells, it inhibited the growth of the tumor cells by 96.6%, showed lymphoblstogenic activities on the splenic lymphocytes and increased the expression of CD25 molecules on the splenic T lymphocytes. When co-cultured with the splenic lymphocytes of a BALB/c mouse, LCT-CT showed strong immunostimulatory activities at the concentration of $25{\sim}100{\mu}g/ml$ by significantly increasing lymphoblasts ratio and CD25 expression.

The Differential Staging of Murine Thymic Lymphoma Cell Lines, Scid.adh, R1.1 and EL-4

  • Background: Scid.adh is a recently developed murine thymic lymphoma cell line, which has been used as in vitro model for the study of double negative stage III thymocytes. In this study, we compared the expression profile of a number of genes and proteins, which are tightly related to T cell development and apoptosis, in thymic lymphoma cell lines, R1.1, EL-4, and Scid.adh for the developmental staging. Methods: We examined the expression of development marker genes and proteins in three lymphoma cell lines by flow cytometry and RT-PCR. In addition, the expression of apoptosis-related molecules including bcl-2, bax and Fas was also investigated. Results: As previously reported, Scid.adh cell line expressed CD8 and CD25 but not TCR ${\alpha}$ chain, while R1.1 cells expressed TCR ${\alpha}$ chain and both CD4 and CD8 transcripts. These suggest that R1.1 might be in double positive stage, and low level of CD44 expression and the absence of CD25 support this suggestion. In contrast, EL-4 cells showed high level of TCR ${\alpha}$ chain transcript, and low-level of CD4 expression, suggesting that EL-4 is in more mature stage than R1.1. Further, this suggestion was supported by the lack of mT-20 in EL-4 cells, which is expressed in the immature thymocytes, and Scid.adh and R1.1 cell lines, but not in the terminally differentiated thymocytes and peripheral T cells. Among the apoptosis-related gene, transcripts of bcl-2 gene were detected in both R1.1 and EL-4 but not in Scid.adh cells, while bax was expressed in all cell lines. Fas expression was the highest in EL-4 cells and low in Scid.adh cell line. Conclusion: R1.1 cell may represent double positive stage, and EL-4 is more differentiated cell line. In addition, Scid.adh and EL-4 cell lines are suspected to be useful for the study of function of bcl-2 family and Fas during the thymocyte development, respectively.

보존된 동종동맥편 조직의 면역성 변화에 관한 연구 (Changes in Immunogenicity of Preserved Aortic Allograft)

  • 동종동맥판의 보존기법이 발전하면서 상당한 정도의 생육성이 보존되며, 특히 면역반응의 주된 요인인 내피세포 생육성이 약 50%이상 보존되기 때문에 보존처리된 동종동맥판 내피세포의 면역능력을 평가하는것이 동종동맥판의 임상적변화의 원인을 규명하는데 필요할 것이다. 실험은 200~250gm의 Sprague-Dawley Rat를 사용하였다. Rat로부터 적출한 동맥벽을 현재 임상적으로 사용하고 있는 냉장보존법과 냉동보존법을 사용하여 2주일간 보존하였으며 보존처리전(No treat)과 멸균처리후(sterile),냉장 보존후 1(1day), 2(2day), 7(7day), 14일째(14day), 2주간의 냉동보존후(cryo)에 표본을 채취하여 보존시간에 따른 변화를 관찰하였다. 면역표현에 대한 연구를 위하여 혈관조직으로부터 내피세포를 분리한 뒤 면역조직화학검사(Immunohistochemical study)를 하였다. 혈관내피세포의 항원 표현정도를 정량적으로 분석하기 위하여 anti-MHC class I antibody(MRC OX-18)과 anti-MHC class II antibody(MRC OX-6), anti-ICAM antibody를 사용하였다. 처리된 내피세포를 Flow cytomwtry로 분석하여 항체가 부착된 내피세포의 비율을 알아냄으로써 내피세포의 항원성(antigenic expression)을 조사하였다. 또한 보존처리된 동종동맥판에 의한 생체내에서의 면역반응을 평가하기 위하여 위에서와 같은 방법으로 보존처리전(No treat), 멸균처리 후 2일 보존후(2 day), 7일 보존후(7 day), 14일 보존 후(14 day), 냉동보존(cryo)된 동종동맥판을 Mouse에 이식한 후 일정기간(1, 2, 3, 4, 6, 8주)이 경과된 시점에서 혈중의 CD4$^{+}$, CD8$^{+}$ T cell분포를 측정하였다. 이를 위하여 Mouse의 미정맥에서 채취한 혈액에 monoclonal antibody를 처리한 뒤 flow cytometry를 이용하여 lymphocyte중의 CD4$^{+}$, CD8$^{+}$ T cell 비율을 측정하였다. 내피세포의 MHC Class I 표현정도는 No treat에서 23.95%였고, sterile에서 48.08%로 증가한 뒤 14day 까지 36.02%로, cryo에서도 35.53% 로 증가되어 있었다(p=0.0183). MHC Class II 표현정도는 No treat에서 9.72%, sterile에서 10.13%이였고 14day 에서 10.27%, cryo 에서 13.39% 였다(P=0.1599). ICAM-1 표현정도는 No treat에서 15.02%, sterile에서 19.85%였고, 14day에서 35.33%, cryo에서 34.67% 로 증가하였다(P=0.001). 정상 Mouse에서 CD4$^{+}$, CD8$^{+}$ T-cell분포는 각각 42.13%, 25.57% 였고 CD4$^{+}$/CD8$^{+}$ ratio는 1.64였다. 동종동맥을 이식받은 Mouse의 정맥혈중 CD4$^{+}$ T-cell분포는 No treat군에서 1주에서 8주사이에 49.23% 에서 36.8%사이로 변화를 보이지 않았고(p=0.955), 2 day군에서는 30.36%로 감소하였고(p=0.0001), 7day군에서는 32.8%로 감소하였고(p=0.008), 14 day 군은 26.92%로 감소(p=0.0001), cryo군은 29.56%로 감소하였다(p=0.0018). CD8$^{+}$T-cell은 모든 군에서 1주에서 8주 사이에 42.32%에서 58.92%사이로 증가하였다(p=0.0001~0.0002). CD4$^{+}$/CD8$^{+}$ ratio는 모든 군에서 1주에 1.22 에서 2.28 사이에 있었으나 8주후에는 모든 군에서 0.47에서 0.95 사이로 감소하였다(p=0.0001). 즉, 보존처리된 동종동맥판의 내피세포는 보존처리과정의 초기에는 MHC class I과 II항원효과를 동시에 보이고, 보존기간이 길어지면서 MHC class II항원효과는 변함이 없으나 MHC class I 항원효과는 증가함을 알 수 있다. 또한 CD4$^{+}$ T-cell은 보존처리 기간 중 소폭의 변환를 보임에 반하여 CD8$^{+}$ T-cell은 보존처리된 기간에 관계없이 이식된 후 8주간에 걸쳐 지속적으로 증가함을 알 수 있다. 4$^{\circ}C$에 냉장보존한 군과 냉동보존한 군간에는 차이가 없었다. 이와같은 결과를 볼 때 동종동맥판을 체내에 이식할 경우 내피세포에 의한 MHC class I 항원효과가 지속적으로 유지되고 있음을 추측할 수 있다.

Stimulatory Effects of Ginsan on the Proliferation and Viability of Mouse Spleen Cells

  • Ginsan is an acidic polysaccharide purified from Panax ginseng, a famous oriental herb. Although a variety of biological activities of ginsan have been studied, the effects of ginsan on spleen cells are not fully elucidated. We investigated the effect of ginsan on the viability and proliferation of spleen cells. Using Cell Counting $Kit-8^{(R)}$ solution and trypan blue solution, we found that ginsan significantly enhanced viability and proliferation. Multiple clusters, indicating proliferation, were observed in ginsan-treated spleen cells and, carboxyfluorescein succinimidyl ester and surface marker staining assay revealed that ginsan promoted proliferation from $CD19^+$ B cells rather than $CD4^+$ or $CD8^+$ T cells. In addition, ginsan decreased the percentage of late apoptotic cells. Ginsan increased the surface expression of CD25 and CD69 as well as production of interleukin-2 from spleen cells, suggesting increased activation. Taken together, these results demonstrate that ginsan increases the viability and proliferation of spleen cells via multiple mechanisms, valuable information for broadening the use of ginsan in clinical and research settings.

이습활통탕(利濕活通湯)이 관절염(關節炎) 병태(病態) 동물(動物)모델에서 비장내(脾臟內) 면역세포(免疫細胞) 변화(變化)에 미치는 영향(影響) (The Effect of Esubwhaltongtang (ESWTT) on the changes of lymphocyte composition in spleen of collagen-induced arthritis mice model)

  • To investigate the effects of ESWTT on rheumatoid arthritis in collagen-induced arthritis mice model, the animals were orally administrated with the extract following by analyzing the changes of lymphocyte composition in spleen with flow cytometry. These results suggest as follows; 1. ESWTT decreased the total cell number of spleen at 400 and 200 mg/kg about 41% and 32%, respectively. 2. ESWTT reduced the number of CD19+ cells in spleen at 400 and 200 mg/kg about 51% and 46%, respectively. 3. ESWTT down-regulated the number spleen CD3+ cells approximately 26% at 400 mg/kg-treated group. 4. ESWTT decreased the number of CD3+/CD69+ cells in spleen at 400 and 200 mg/kg about 79% and 76%, respectively. 5. ESWTT reduced the number of CD4+ and CD8+ cells in spleen, however, the results were not significant. 6. ESWTT decreased the number of CD4+/CD25+ cells in spleen at 400 and 200 mg/kg about 60% and 56%, respectively. Taken together these results, ESWTT has an efficacy on rheumatoid arthritis through inhibiting the proliferation of lymphocytes such as B and T cells.
