Expression of Mosquitocidal Bacillus sphaericus Binary Toxin and B. thuringiensis cry11B Genes in B. thuringiensis 407

  • Wild type Bacilus thuringiensis subsp. israelensis and B. sphaericus toxins have been used separately as active in ingredients for bacterial insecticides to control mosquito larvae due to their comparable toxicity to chemical insecticides. Cry11B, recently cloned from B. thuringiensis subsp. jegathesan, shows higher toxicity against three major species of mosquito larvae than Cry11A, one of the major component of B. thuringiensis subsp. israelensis inclusion body. To determine whether the combination of cry11B and B. sphaericus binary toxins is as toxic as B. thuringiensis subsp. israelensis parental strain, cry11B and B. sphaericus binary toxins genes were co-expressed as an operon using cytlA promoters/STAB-SD hybrid expression system in B. thuringiensis subsp. israelensis acrystalliferous strain 4Q7. However, unexpectedly, B. sphaericus binary toxins were barely produced, whereas relatively large amount of Cry11B was produced. When this strain was grown in four different media, NB+G and Peptonized Milk produced more toxin proteins and spores per unit of media than GYS and G-Tris. Toxicity of this strain against fourth instar Culex quinquefasciatus was ranged from of 8.3 to 45.7 ng/ml, with NB+G culture being the highest, and GYS culture was the lowest.

Structural nature of chemically inequivalent borons in the nonlinear optical material β-BaB2O4 studied using 11B MAS NMR and 11B single-crystal NMR

  • Detailed information about the structural nature of three-coordinate borons in ${\beta}-BaB_2O_4$ is obtained through $^{11}B$ MAS NMR and $^{11}B$ single-crystal NMR. The three-coordinate $BO_3$ of the two borons B(1) and B(2) in ${\beta}-BaB_2O_4$ were distinguished. The spin-lattice relaxation time in the laboratory frame $T_1$ for B(1) and B(2) slowly decreases with increasing temperature, whereas the spin-lattice relaxation time in the rotating frame $T_{1{\rho}}$ for B(1) and B(2), which differs from $T_1$, is nearly constant. The B(1) and B(2) of the two types were distinguished by $^{11}B$ MAS NMR and $^{11}B$ single-crystal NMR.

The Antioxidant Effect of Cheonggukjang, Fermented Using the New Strain, Bacillus amyloliquefaciens NBF11-1 (신 균주 Bacillus amyloliquefaciens NBF11-1을 이용하여 발효한 청국장의 항산화효과)

  • This study aims to compare and analyze the antioxidant effect of Cheonggukjang's traditional fermentation strain, Bacillus subtilis NG24 which was a control of the study, in order to see the biological activity effect of the new strain, Bacillus amyloliquefaciens NBF11-1 that was first found in the surface of the bamboo stem, but hasn't been insufficiently researched. In the antioxidant activity experiment of the Cheonggukjang extract, the B.amyloliquefaciens NBF11-1 sample showed a significant increase in the total polyphenol extract content, compared to B.subtilis NG24(p=0.032). Also, compared to B.subtilis NG24, the sample containing B.amyloliquefaciens NBF11-1 showed a significant increase in SOD-like Activity, DPPH radical scavenging, and NO radical scavenging, as the concentration rose(p<.05). Additionally, $IC_{50}$ in each antioxidant activity experiment significantly decreased in the B.amyloliquefaciens NBF11-1 sample like SOD-like Activity(p=0.045), DPPH radical scavenging(p=0.041), and NO radical scavenging(p=0.019), compared to B.subtilis NG24.

Cloning and Characterization of Xylanase 11B Gene from Paenibacillus woosongensis (Paenibacillus woosongensis의 Xylanase 11B 유전자 클로닝과 특성분석)

  • A gene coding for the xylanase predicted from the partial genomic sequence of Paenibacillus woosongensis was cloned by PCR amplification and sequenced completely. This xylanase gene, designated xyn11B, consisted of 1,071 nucleotides encoding a polypeptide of 356 amino acid residues. Based on the deduced amino acid sequence, Xyn11B was identified to be a modular enzyme, including a single carbohydrate-binding module besides the catalytic domain, and was highly homologous to xylanases belonging to glycosyl hydrolase family 11. The SignalP4.1 server predicted a stretch of 26 residues in the N-terminus to be the signal peptide. Using DEAE-Sepharose and Phenyl-Sepharose column chromatography, Xyn11B was partially purified from the cell-free extract of recombinant Escherichia coli carrying a copy of the P. woosongensis xyn11B gene. The partially purified Xyn11B protein showed maximal activity at $50^{\circ}C$ and pH 6.5. The enzyme was more active on arabinoxylan than on oat spelt xylan and birchwood xylan, whereas it did not exhibit activity towards carboxymethylcellulose, mannan, and para-nitrophenyl-${\beta}$-xylopyranoside. The activity of Xyn11B was slightly increased by $Ca^{2+}$ and $Mg^{2+}$, but was significantly inhibited by $Cu^{2+}$, $Ni^{2+}$, $Fe^{3+}$, and $Mn^{2+}$, and completely inhibited by SDS.

A STUDY ON THE RANGE OF MANDIBULAR MOVEMENT OF NORMAL AND CLASS III MALOCCLUSION CHILDREN (정상교합과 III급 부정교합아동의 하악운동 범위에 대한 연구)

  • The purpose of this study is to obtain the normal range of mandibular movement for 13 year old boys and girls, and to compare it to the range of mandibular movement of class III patient and normal adult for diagnosis and prognosis of class III malocclusion and TMJ dysfunction. The subjects were 20 normal boys 20 normal girls 22 class III boys and 18 class III girls. The author measured the ranges and shapes of movement of the mandible in the frontal, sagittal and horizontal trajectories using Saphon visitrainer C II (Tokyo Shizaisha Inc.) for the subjects. The results obtained are as follows: 1) The mean for maximum right laterotrusion in the frontal trajectory were 11.96mm in N.B., 11.10mm in N.G., 11.32mm in III. B., 11.24mm in III G. The mean for maximum left laterotrusion were 11.48mm, 11.0mm, 10.91mm, 10.44mm respectively, Area of border movement were 7.16cm in N.B., 6.59cm in N.G., 7.29cm in III. B., 7.50cm in III. G. 2) The mean for maximum protrusion in the sagittal trajectory were 11.7mm in N.B., 11.4mm in N.G.,11.87mm in III B., and 11.02mm in III. G. 3) The mean for maximum protrusion in the horizontal trajectory were 10.20mm in N.B.,10.00 mm in N.G., 9.12mm for III. B. and 9.36mm in III. G. 4) The mean for maximum protrusion of Class III subjects were shorter than those of normal subjects. 5) There was no sexual difference in the range of mandibular movement for 13 year old subjects.   • 3-Carbomethoxy-1(3H)-isobenzofuranone(9) underwent condensation with ${\alpha}{\beta}-unsaturated$, esters 3a-b to produce the corresponding naphthacene-6,7-diones 11a-b with high yields in one pot procedure. Among the naphthacene-6,7-diones formed, compound 11a without an ethyl group at C-9 position was oxidized to give the naphthacene-5,12-dione 13a, while compound 11b containing the ethyl group was oxidized to give a 3:2 mixture of the naphthacene-5,7,12-trione 12b and naphthacene-5,12-dione 13b under the same experimental conditions.

[ $^{11}B$ ] Nuclear Magnetic Resonance Study of Spin Structures in Terbium Tetraboride

  • [ $^{11}B$ ] nuclear magnetic resonance (NMR) measurements were performed on the single crystals of $TbB_4$ to investigate local electronic structure and 4f spin dynamics. $^{11}B$ NMR spectrum, Knight shift, spin-lattice and spin-spin relaxation rates were measured down to 4K at 8T. $^{11}B$ NMR shift and linewidth are huge and strongly temperature dependent due to the 4f moments. In addition, both are proportional to magnetic susceptibility, indicating that the hyperfine field at the boron site originates from the 4f spins of Tb. Below $T_N$, the single broad resonance peak of $^{11}B$ NMR splits into several peaks reflecting the local magnetic fields due to antiferromagnetic spin arrangements. The longitudinal and the transverse relaxation rates, $1/T_1\;and\;1/T_2$, independent of temperature above $T_N$, decreases tremendously confirming huge suppression of spin fluctuation below $T_N$.

Thermal Stability and Behavior of Isothermal Crystallization in Fe-P-C-B-(AI-Ge) Amorphous Alloys (Fe-P-C-B-(AI-Ge)계 비정질합금의 열적 안정성과 등온결정화 거동)

  • Thermal properties of Fe- base amorpous alloys were investigated. $Fe_{80}P_6C_{12}B_{12}$ and $Fe_{73}P_{11}C_6B_4AI_4Ge_2$ amorphous alloys were fabricated by melt spinning method and thermal analysis was done by differential scanning calorimeter. After isothermal crystallization. the Avrami exponents of $Fe_{80}P_6C_{12}B_{12}$ and $Fe_{73}P_{11}C_6B_4AI_4Ge_2$ amorphous alloys were 1.8-2.2 and 2.5-4.0, respectively. It means the former alloy shows diffusion controlled growth and the latter one shows interface controlled growth. For $Fe_{80}P_6C_{12}B_{12}$ and $Fe_{73}P_{11}C_6B_4AI_4Ge_2$ amorphous alloys. the activation energies of isothermal crystallization was 353 and 371kJlmol. Also the activation energies of nucleation and growth were 301, 324kJlmol and 273. 30lkJ/mol, respectively. Thus $Fe_{73}P_{11}C_6B_4AI_4Ge_2$ amorphous alloy is considered to be more stable than $Fe_{73}P_{11}C_6B_4AI_4Ge_2$ amorphous alloy.

Review and proposed improvements for Romanization and English expressions of rubrics in the WHO ICD-11 beta version traditional medicine chapter (세계보건기구 국제질병분류 11판 베타버전 중 한의학 고유 상병의 로마자 표기 및 영문표현 검토연구)

  • Objectives : The purpose of this study is to review and propose improvements for the Romanization and English expressions in the WHO international classification of diseases 11th revision beta version (ICD-11b) traditional medicine chapter. Methods : ICD-11b as of October 5, 2015, was reviewed. Romanization and English expressions were analyzed with reference to existing standards such as the Basic Principles of Romanization stipulated by the National Institute of Korean Language, and the Korean Standard Classification of Diseases (KCD), suggested improvements followed. Results : Following the Basic Principles of Romanization, 131 ICD-11b rubrics need improvement in the Romanization of Korean. When compared to KCD-6 comparable rubrics, 161 ICD-11b rubrics are the same and 64 are different. When compared to KCD-7 comparable rubrics, 118 ICD-11b rubrics are the same, and 51 are different. In KCD-6, there are 127 rubrics that do not match with items in ICD-11b. In KCD-7, there are 123 rubrics that do not match with items in ICD-11b. Conclusions : ICD-11b may be improved by correcting the Romanization and consideration of English expressions suggested in this study.

CD11b Deficiency Exacerbates Methicillin-Resistant Staphylococcus aureus-Induced Sepsis by Upregulating Inflammatory Responses of Macrophages

  • Macrophages are important for the first line of defense against microbial pathogens. Integrin CD11b, which is encoded by Itgam, is expressed on the surface of macrophages and has been implicated in adhesion, migration, and cell-mediated cytotoxicity. However, the functional impact of CD11b on the inflammatory responses of macrophages upon microbial infection remains unclear. Here, we show that CD11b deficiency resulted in increased susceptibility to sepsis induced by methicillin-resistant Staphylococcus aureus (MRSA) infection by enhancing the pro-inflammatory activities of macrophages. Upon infection with MRSA, the mortality of Itgam knockout mice was significantly higher than that of control mice, which is associated with increased production of TNF-α and IL-6. In response to MRSA, both bone marrow-derived macrophages and peritoneal macrophages lacking CD11b produced elevated amounts of pro-inflammatory cytokines and nitric oxide. Moreover, CD11b deficiency upregulated IL-4-induced expression of anti-inflammatory mediators such as IL-10 and arginase-1, and an immunomodulatory function of macrophages to restrain T cell activation. Biochemical and confocal microscopy data revealed that CD11b deficiency augmented the activation of NF-κB signaling and phosphorylation of Akt, which promotes the functional activation of macrophages with pro-inflammatory and immunoregulatory phenotypes, respectively. Overall, our experimental evidence suggests that CD11b is a critical modulator of macrophages in response to microbial infection.
