Transcriptional repression of ANGPT1 by histone H3K9 demethylase KDM3B

  • Here we report that the H3K9 demethylase KDM3B represses transcription of the angiogenesis regulatory gene, ANGPT1. Negative regulation of ANGPT1 by KDM3B is independent of its Jumonji (JmjC) domain-mediated H3K9 demethylase activity. We demonstrate that KDM3B downregulates ANGPT1 via interaction with SMRT, and suggest that the repressor complex is formed at the promoter area of ANGPT1. Using MTT and wound healing assays, depletion of KDM3B was found to increase cell proliferation and cell motility, indicating that KDM3B has a role in angiogenesis. Taxonomy of Botryotrichum luteum sp. nov. based on Morphology and Phylogeny Isolated from Soil in Korea

  • In this study, a fungal strain KNUF-22-025 belonging to the genus Botryotrichum was isolated from the soil in Korea. The cultural and morphological characteristics of this strain differed from those of closely related species. On malt extract agar, strain KNUF-22-025 showed slower growth than most of the related species, except B. domesticum. The conidia size (9.6-21.1×9.9-18.4 ㎛) of strain KNUF-22-025 was larger than those of B. piluliferum, B. domesticum, and B. peruvianum but smaller than those of B. atrogriseum and B. iranicum. Conidiophores in strain KNUF-22-025 (137 ㎛) were longer than those in other closely related species but shorter than those in B. atrogriseum. Multi-locus analysis of molecular markers, such as ITS, 28S ribosomal DNA, RBP2, and TUB2 revealed that strain KNUF-22-025 was distinct from other Botryotrichum species. Thus, this strain is proposed as a novel species based on morphological characteristics along with molecular phylogeny and named Botryotrichum luteum sp. nov.

Influential Factor of Life Satisfaction based on Types of Baby Boomer Generation (베이비부머세대의 유형별 삶의 만족 영향 요인)

  • The purpose of this study was to identify the multidimensional factors that affect the satisfaction of life by type of baby boomer generation. The subjects of this study were 1,895 households who were born from 1955 to 1963 among the respondents in the Korea welfare panel (the 10th year). The baby boomer generation was classified into four types such as gender, income status, male and female as socioeconomic status. The male type IV (B=.074, t=1.992, p<.05), health status (B=.233, t=7.268, p<.001), household income (B=.174, t=4.476, p<.001), depression (B=-.182, t=-5.242, p<.001), self-esteem (B=.255, t=7.027, p<.001), trust (B=.064, t=2.036, p<.05), etc. were found to have a significant effect on the life satisfaction of the male baby boomer generation. The female type IV (B=.100, t=2.598, p<.05), health status (B=.239, t=6.790, p<.001) (P<.001), depression (B=-.181, t=-5.142, p<.001), and self-esteem (B=.320, t=8.600, p<.001), etc. were found to have a significant effect on the life satisfaction of the female baby boomer generation.

  • This research presents the reliability analysis of the pneumatic actuator within the tape feeder that is used to transfer the correct force to linked parts during l.0E+7 cycles. First, the degradation analysis for thrust and air leakage is executed to obtain the failure data of a product based on its performance over time. Second, once the parameters has been calculated using the weibull 2-parameter distribution and MLE(Maximum Likelihood Estimation), information related to life such as reliability, failure rate, probability density function is estimated. Finally, MTTF(Mean Time To Failure) and $B_{10}$ life of actuators are calculated. MTTF means the mean life at the confidence level and $B_{10}$ life refers to the time by which 10% of the product would fail. In this study, failure causes and solutions are examined using the reliability analysis.

Screening of the Antigen Epitopes of Basic Fibroblast Growth Factor by Phage Display

  • In order to investigate the epitope of basic fibroblast growth factor (bFGF) and its immunogenicity, the epitopes of bFGF were screened from the phage display library with monoclonal antibody GF22, which can neutralize the bio-activity of bFGF. By three rounds of screening, the positive phage clones with bFGF epitopes were selected, which can effectively block the bFGF to bind with GF22. Sequence analysis showed that the epitopes shared a highly conservative sequence (Leu-Pro-Pro/Leu-Gly-His-Phe/Ile-Lys). The sequence of PPGHFK was located at 22-27 of the bFGF. The specific immuno-response of mouse could be highly induced by phage clones with the epitopes. And the anti-bFGF activity induced by LPGHFK was 3 times higher than the original sequence, which showed that the mimetic peptide LPLGHIK might be used as a tumor vaccine in the prevention and treatment of tumor.

The Effect of Inhibition of Uncaria rhynchophylla as an Inhibitor of Melanogenesis and an Antioxidant in B16F10 Melanoma Cells (B16F10세포에서 멜라닌 생성 억제제 및 항산화제로서 조구등의 억제 효과)

  • Many people of all ages wish to have lighter skin for cosmetic reasons, and natural products attract more attention than chemically synthesized compounds. Uncaria rhynchophylla is widely used in Asia as a traditional herbal medicine. In order to find novel skin whitening agents, the present study evaluated the antioxidant activity and potential tyrosinase-inhibiting properties of U. rhynchophylla. Specifically, this study analyzed the antioxidant capacity of a 70% ethanolic extract of U. rhynchophylla as well as its effects on tyrosinase activity and melanin synthesis. Total mRNA levels were examined using reverse transcription polymerase chain reaction. The results revealed that U. rhynchophylla extracts exhibit great antioxidant capacity and significant levels of polyphenol and flavonoid compounds. U. rhynchophylla extracts can also powerfully inhibit tyrosinase activity. This same capacity was observed in melanoma B16F10 cells; that is, U. rhynchophylla extracts suppressed intracellular tyrosinase activity and reduced the amount of melanin in treated cells. In addition, a 1 mg/ml concentration of U. rhynchophylla extract significantly reduced the mRNA expression levels of tyrosinase. U. rhynchophylla extracts decrease tyrosinase and inhibit melanogenesis in B16F10 cells. This finding suggests that U. rhynchophylla has great potential as a natural whitening agent in skincare products.

  • The present study was performed to investigate an antibacterial activity of specific bacteriophage (phage) and Bacillus subtilis KM-1 (B. subtilis) mixture against Edwardsiella tarda (E. tarda). An appropriate number of phage showing the most effective antibacterial activity was $2{\times}10^5$ PFU/ml with $1{\times}10^7$ CFU/ml of B. subtilis 36 h post incubation. On the other hand, B. subtilis showed a dose dependant manner in inducing antibacterial activity in the presence of phage ($2{\times}10^5$ PFU/ml). The phage and B. subtilis mixture showed higher antibacterial activity against E. tarda than phage or B. subtilis only. These results suggest that the phage and B. subtilis mixture could be utilized as an alternative to antibiotics in the control of fish diseases caused by E. tarda.

Development of mcyB-specific Ultra-Rapid Real-time PCR for Quantitative Detection of Microcystis aeruginosa (Microcystis aeruginosa의 정량을 위한 mcyB 특이 초고속 실시간 유전자 증폭법의 개발)

  • A mcyB-specific Ultra-Rapid quantitative PCR was developed for the quantitative detection of Microcystis aeruginosa, which is often a dominant species in green tide. McyB-specific UR-qPCR was optimized under extremely short times of each step in thermal cycles, based on the specific primers deduced from the mcyB in microcystin synthetase of M. aeruginosa. The M. aeruginosa strain KG07 was used as a standard for quantification, after the microscopic counting and calculation by mcyB-specific UR-qPCR. The water samples from the river water with the Microcystis outbreak were also measured by using both methods. The $1.0{\times}10^8$ molecules of mcyB-specific DNA was recognized inner 4 minutes after beginning of UR-qPCR, while $1.0{\times}10^4$ molecules of mcyB-specific templates was detected inner 7 minutes with quantitative manner. From the range of $1.0{\times}10^2$ to $1.0{\times}10^8$ initial molecules, quantification was well established based on $C_T$ using mcyB-specific UR-qPCR (Regression coefficiency, $R^2=0.9977$). Between the numbers of M. aeruginosa cell counting under microscope and calculated numbers using mcyB-specific UR-qPCR, some differences were often found. The reasons for these differences were discussed; therefore, easy compensation method was proposed that was dependent on the numbers of the cell counting. Additionally, to easily extract the genomic DNA (gDNA) from the samples, a freeze-fracturing of water-sample using liquid nitrogen was tested, by excluding the conventional gDNA extraction method. It was also verified that there were no significant differences using the UR-qPCR with both gDNAs. In conclusion, the mcyB-specific UR-qPCR that we proposed would be expected to be a useful tool for rapid quantification and easy monitoring of M. aeruginosa in environmental water.

Generation of a Specific Marker to Discriminate Bacillus anthracis from Other Bacteria of the Bacillus cereus Group

  • Bacillus anthracis is a soil pathogen capable of causing anthrax that is closely related to several environmental species, including B. cereus, B. mycoides, and B. thuringiensis. DNA homology studies showed that B. anthracis, B. cereus, B. mycoides, and B. thuringiensis are closely related, with a high sequence homology. To establish a method to specifically detect B. anthracis in situations such as environmental contamination, we initially performed RAPD-PCR with a 10-mer random primer and confirmed the presence of specific PCR bands only in B. anthracis species. One region specific for B. anthracis was cloned and sequenced, and an internal primer set was designed to amplify a 241-bp DNA fragment within the sequenced region. The PCR system involving these specific primer sets has practical applications. Using lyses methods to prepare the samples for PCR, it was possible to quickly amplify the 241-bp DNA segment from samples containing only a few bacteria. Thus, the PCR detection method developed in this study is expected to facilitate the monitoring of environmental B. anthracis contamination.

The Parallel Multiple Mediating Effect of Emotional Intelligence and Self-Efficacy in the Relationship between Interpersonal Ability and College Life Adjustment among Nursing Students since COVID-19 (COVID-19 이후 간호대학생의 대인관계능력과 대학생활적응의 관계에서 정서지능과 자기효능의 병렬다중매개효과)

  • This study was conducted to confirm the effect of interpersonal ability on college life adjustment of nursing students and the parallel multiple mediating effect of emotional intelligence and self-efficacy. Participants in this study were 130 nursing students at a college. Data were collected from 24 July to 1 August 2023, and analyzed by descriptive statistics, Pearson's correlation, and PROCESS macro model 4 using SPSS/WIN 27.0. As a result of the study, interpersonal ability was positively correlated with emotional intelligence, college life adjustment and self-efficacy. Interpersonal ability had a significant total effect(B=0.48, p<.001) on college life adjustment. And the mediating effect of emotional intelligence(B=0.20, 95% bootstrap CI=0.06~0.36) and self-efficacy(B=0.17, 95% bootstrap CI=0.05~0.31) was also significant. Therefore, it will be necessary to develop and research various training programs that can enhance emotional intelligence and self-efficacy together in developing programs to help nursing students improve their college life adjustment.
