Anti-Microbial and Anti-Wrinkle Effect of Kaempferol and Kaempferol Rhamnosides isolated from Hibiscus cannabinus L. (양마에서 분리한 Kaempferol과 그 배당체의 항균 및 주름억제 효과)

  • In this study, kaempferol and its rhamnosides (${\alpha}$-rhamnoisorobin, afzelin, kaempferitrin) were isolated electively by bioconversion technology from Hibiscus cannabinus L. leaves to evaluate the anti-wrinkles effects and anti-microbial effects. In order to evaluate anti-wrinkles activity, reduction of expression matrix metalloproteinase-1 (MMP-1) protein and proliferation/ pro-collagen production were investigated. Kaempferol and ${\alpha}$-rhamnoisorobin showed inhibition activity of MMP-1 generated to compared to positive control. In HaCaT cell proliferation assay, kaempferol and ${\alpha}$-rhamnoisorobin significantly promoted cell proliferation in a concentration-dependent manner. In addition, procollagen synthesis assays (by HDF-N cell) showd that TGF-${\beta}$ induced procollagen production and also, all four kinds of experimental significantly promoted procollagen synthesis in a concentration-dependent manner. Kaempferol and ${\alpha}$-rhamnoisorobin exhibited strong antimicrobial activities on five of microbes, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger.

Mechanical Property and Corrosion Resistance of Mg-Zn-Y Alloys Containing Icosahedral Phase (준결정상을 포함한 Mg-Zn-Y 합금의 기계적 특성 및 부식 저항성)

  • Mechanical and property corrosion resistance of Mg-Zn-Y alloys with an atomic ratio of Zn/Y of 6.8 are investigated using optical microscopy, scanning electron microscopy, transmission electron microscopy, uniaxial tensile test and corrosion test with immersion and dynamic potentiometric tests. The alloys showed an in-situ composite microstructure consisting of ${\alpha}$-Mg and icosahedral phase (I-phase) as a strengthening phase. As the volume fraction of the I-phase increases, the yield and tensile strengths of the alloys increase while maintaining large elongation (26~30%), indicating that I-phase is effective for strengthening and forms a stable interface with surrounding ${\alpha}$-Mg matrix. The presence of I-phase having higher corrosion potential than ${\alpha}$-Mg, decreased the corrosion rate of the cast alloy up to I-phase volume fraction of 3.7%. However further increase in the volume fraction of the I-phase deteriorates the corrosion resistance due to enhanced internal galvanic corrosion cell between ${\alpha}$-Mg and I-phase.

A Study on the Aging Behavior of a Mg-8.5Li-4.5Al alloy by Differential Scanning Calorimetry (열분석법에 의한 Mg-8.5Li-4.5Al합금의 시효거동 연구)

  • Precipitation and strengthening mechanisms in squeeze cast Mg-8.5wt%Li-4.5wt%Al have been investigated by differential scanning calorimetry(DSC), scanning electron microscopy(SEM), in-situ and ex-situ X-ray diffraction analysis and hardness measurement. Special emphasis was placed on the investigation of the precipitation behavior by the DSC technique. Microstructural and calorimetric analysis showed that ${\theta}$ and ${\delta}$ precipitates in the b.c.c. ${\beta}$ phase matrix, forming two exothermic peaks at the temperature ranges of $130^{\circ}C{\sim}180^{\circ}C$ and $236^{\circ}C{\sim}280^{\circ}C$. ${\theta}$ and ${\delta}$ dissolve into the matrix forming an endothermic peak at the temperature range of $280^{\circ}C{\sim}352^{\circ}C$. The as-cast microstructure consists of ${\alpha}$, ${\beta}$ and ${\delta}$. Peak strength was obtained after aging for 1 hour at $50^{\circ}C$. The aging time required for the peak strength decreased as the aging temperature increases. The hardness decrease during overaging was due to the coarsening of ${\theta}$ precipitates. Microhardness measurement showed that variation of the hardness of ${\beta}$ matrix was more pronounced than that of the ${\alpha}$ phase, indicating that the ${\beta}$ phase is more responsible for the strengthening of the Mg-8.5wt%Li-4.5wt%Al alloy.

Anti-aging Effect of Asterosaponin P1 Isolated from Asterina pectinifera (별불가사리로부터 분리된 Asterosaponin P1의 항노화 효능)

  • The starfish, Asterina pectinifera Muller and Troschel (Asterinidae) is an indigenous species commonly found in all coasts of Korea causes damages to shellfish farms. In order to exterminate A. pectinifera, they are dried and used as fertilizer. Although various studies have been conducted to create high added value from the retrieved A. pectinifera, their actual utilization is relatively low. Accordingly, this study aimed to find new practical uses of starfish by identifying useful ingredients for skin anti-aging. Two polyhydroxysteroids and one asterosaponin were isolated from the A. pectinifera. The structures of these compounds were identified as $5{\alpha}$-cholestane-$3{\beta},6{\alpha},7{\alpha},8,15{\alpha},16{\beta},26$-heptol, $5{\alpha}$-cholestane-$3{\beta},4{\beta},6{\alpha},7{\alpha},8,15{\beta},16{\beta},2$6-octol, and asterosaponin $P_1$ on the basis of chemical and spectroscopic analysis. Among these compounds, we have found that asterosaponin $P_1$ increased epidermal stem cell proliferation and the expression of hyaluronan synthase-2 and hyaluronan synthase-3 gene, which are enzymes that synthesize water-binding matrix hyaluronic acids in keratinocytes. In addition, asterosaponin $P_1$ increased synthesis of pro-collagen type I, a major dermal collagen in fibroblasts. As a result, asterosaponin $P_1$ isolated from A. pectinifera could be used as a useful cosmetic ingredient that improves skin symptoms accompanying skin aging.

  • A theory of the helix/coil transition for $\alpha$ helical dimer such as $\alpha$ tropomycin and paramycin is developed. The treatment differs from those formulated previously for oligopeptide dimer which is explained by the matrix method using Zimm-Bragg parameter: In the present treatement, it is explained by the zipper model which can account for the dangling H-bond. We calculate the fractional helicity in $\alpha$ helical dimer as a function of helix initiation $constant(\sigma)$, helix stability constant(${\xi}$) and hydrophobic interaction parameter(w). For $\alpha$ tropomycin, the helix stability profile is also calculated. The transitions of this oligomer due to the change of temperature and the concentration of oligopeptide involve simultaneous dissociation of the dimer. The transitions of dimers which have cross-linked S-S bonds or have long chains don't occur, because they keep always helical structures. The transitons due to the concentration of the oligopeptides are steeper than those due to the chain length or temperature.

  • A lot of methods are proposed to provide services for object informations in distributed domain to satisfy the recent increase of user-centered services. This paper proposed a method called fuzzy drop manager for the service of reliable distribution application domain objects. The proposed system accesses the domain using replica parameter ci,j and access matrix Z, and evaluates the reference relatedness inside the domain using the relatedness, given by the mapping of intra-domain fuzzy relevance, between fuzzy sets. Objects in the domain generated an $\alpha$-level set according to the reference relatedness obtained by applying $\alpha$-level to extend queries. Simulation results showed that the proposed method has better performance than the other methods.

Effect of Heat Treatment on the Mechanical Properties of a Ti-15Mo-3Nb-3Al-0.2Si Alloy (β-type Ti-14Mo-3Nb-3Al-0.2Si 합금의 열처리 조건에 따른 기계적 특성)

  • The mechanical properties of the various heat treatment conditions on Ti-15Mo-3Nb-3Al-0.2Si alloy plates were examined. XRD patterns from the surface of Ti-15Mo-3Nb-3Al-0.2Si were analyzed as a solution-treated Ti alloy has the single-phase ${\beta}$ structure whereas the aged Ti alloys have the ${\beta}$ matrix embedded with ${\alpha}$ needles. High strength (~1500 MPa) with decent ductility (7%) was obtained by the Ti alloy double aged at $300^{\circ}C$ and $520^{\circ}C$ for 8 hours each. The double-aged alloy exhibits the finer structure than the single-aged alloy at $300^{\circ}C$ for 8 hours because of the higher nucleation rate of ${\alpha}$ needles at an initial low aging temperature ($320^{\circ}C$). TEM observation revealed that the fine nanostructure with ${\alpha}$ needles in the ${\beta}$ matrix ensured the excellent mechanical properties in the double aged Ti-15Mo-3Nb-3Al-0.2Si alloy. In the solution treated alloy, the yield drop, stress-serrations and the ductility minimum typically associated with dynamic strain aging can be attributed to the dynamic interaction between dislocations and oxygen atoms. The yield drop and the stress serration were not observed in aged samples because the geometrically introduced dislocations due to phase precipitates suppressed the dynamic strain aging.

A Proteomic Screen for Presynaptic Terminal N-type Calcium Channel (CaV2.2) Binding Partners

  • N type calcium channels (CaV2.2) play a key role in the gating of transmitter release at presynaptic nerve terminals. These channels are generally regarded as parts of a multimolecular complex that can modulate their open probability and ensure their location near the vesicle docking and fusion sites. However, the proteins that comprise this component remain poorly characterized. We have carried out the first open screen of presynaptic CaV2.2 complex members by an antibody-mediated capture of the channel from purified rat brain synaptosome lysate followed by mass spectroscopy. 589 unique peptides resulted in a high confidence match of 104 total proteins and 40 synaptosome proteome proteins. This screen identified several known CaV2.2 interacting proteins including syntaxin 1, VAMP, protein phosphatase 2A, $G_{o\alpha}$, G$\beta$ and spectrin and also a number of novel proteins, including clathrin, adaptin, dynamin, dynein, NSF and actin. The unexpected proteins were classified within a number of functional classes that include exocytosis, endocytosis, cytoplasmic matrix, modulators, chaperones, and cell-signaling molecules and this list was contrasted to previous reports that catalogue the synaptosome proteome. The failure to detect any postsynaptic density proteins suggests that the channel itself does not exhibit stable trans-synaptic attachments. Our results suggest that the channel is anchored to a cytoplasmic matrix related to the previously described particle web.

A Promising Serum Autoantibody Marker, Anti-Heat Shock Protein 90α, for Cholangiocarcinoma

  • The present study was designed to investigate cholangiocarcinoma (CCA) antibodies in hamster serum. Hamster CCA cell lines were processed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. A candidate biomarker was confirmed by immunoprecipitation and western blot, and was further analyzed using ELISA and sera from normal control hamsters, hamsters with opisthorchiasis and hamsters with various stages of CCA, as well as from CCA patients and healthy individuals. One candidate marker was identified as $HSP90{\alpha}$, as indicated by a high level of anti-$HSP90{\alpha}$ in hamster CCA sera. It was found that the levels of anti-$HSP90{\alpha}$ were specifically elevated in the sera of hamsters with CCA compared with other groups and progressively increased with the clinical stage. At the cut-off point of 0.4850 on the receiver operating characteristic curve, anti-$HSP90{\alpha}$ could discriminate CCA from healthy control groups with a sensitivity of 76.2%, specificity of 71.4% and total accuracy 75.5%. In the present study, we have shown that anti-$HSP90{\alpha}$ may be a potential useful serum biomarker to discriminate CCA cases from healthy persons.

  • Analysis of interferon alpha (IFN) modified with high molecular weight branched PEG was performed by capillary electrophoresis (CE) and MALDI-TOF mass spectrometry (MALDI-TOF MS). IFN was modified by the reaction of amine residues with an active ester of monomethoxy polyethylene glycol at various molar ratios. As a CE method. capilary electrophoresis sodium dodecyl sulfate nongel sieving (CE-SDS NGS) was performed using an uncoated capilary filled with a hydrophilic replaceable polymer network matrix.
