사람의 세포질 Superoxide Dismutase 유전자의 클로닝과 대장균내에서의 대량발현에 관한 연구 (Molecular Cloning and High-Level Expression of Human Cytoplasmic Superoxide Dismutase Gene in Escherichia coli)

  • 생체내의 유해산소를 제거하는 superoxide dismutase (superoxide : superoxide oxidoreductase E.C.1.15.1.1) 중 세포질내에서 그 활성을 지니는 인체의 세포질 superoxide dismuta~ie (SODl) 유전자를 사람의 간 cDNA library로부터 동위원소로 표지된 oligonucleotide probe를 이용, in situ plaque hybridization 방법으로 선별 분리하여 내장균 벡터로 클로닝하였다. 이 클론은 SOD1 유전자의 5"L"TR과 3’UTR을 포함한 1.6 kb 정도의 cDNA였다 SOD1 구조유전자만을 선택적으로 분리하기 위해서 ATG를 포함하는 sense strand primer와 3’UTR 부위의 antisense strand primer를 이용하여 중합효소연쇄반응(Polymerase Chain Reaction) 방법을 써서 SOD1 구조유전자 부위만을 선택적으로 증폭시켰다. Taq DNA polymerase에 의해 증폭된 DNA를 벡터 pUCl9의 multiple cloning site (MCS) 내의 Hinc II 위치에 넣였으며 이 insert DNA를 M13 mp19으로 옮겨 dideoxy chain termination 방법으로 sequenase를 사용하여 염기서열을 결정하였다. 클론닝된 cDNA는 153개의 아미노산을 포함하고 있는 하나의 open reading frame (ORF)을 가셨다. 중합효소연쇄반응에 의해 이때 증폭된 SOD1 구조유전자를 $\lambda P_{L}$ 프로모터를 포함하고 있는 발현 벡터 pUPL에 옮긴 후 대장균에서 대량으로 발현시켰다. 이때 발현된 단백질 SOD1은 고유의 효소활성을 가지고 있었다.

  • Sulfametrole, $C_9H_{10}N_4O_3S_2$, crystallizes in the monoclinic system, space group $P2_1/n$ , with a = 8.145(2), b = 16.505(4), c = 9.637(1)${\AA},{\beta}=103.72(1)^{\circ},D_m=1.52gcm^{-3}$,Z=4.Intensities for 3594(2143 observed) unique reflections were measured on a four-circle diffractometer with Mo $K{\alpha}$ radiation $({\lambda}=0.71069{\AA})$. The structure was solved by direct method and refined by full-matrix least squares to a final R of 0.070. The geometrical features of the thiadiazole ring indicate some ${pi}$-electron delocalization inside the ring. The least squares planes defined by the benzene and thiadiazole rings are nearly perpendicular to each other(dihedral angle; $93.9^{\circ}$ ). All the potential hydrogen-bond donor atoms in the molecule, N(1) and N(2), are included in the hydrogen bonding. The molecules through hydrogen bonding form three dimensional network.

  • This paper proposes a Chaos Pulse Width Modulation(CPWM) technique. For generating the chaotic numbers by chaos phenomenon, chaos area $\lambda$=0.99 in bifurcation tree of the proposed double tent mapping is used. A micro-controller is used for the generation of chaos numbers and triangular carrier with chaotic frequency is obtained through the process of frequency modulation according to the generated chaos numbers. The experiments are executed with the 1.5kw induction motor coupled with a 2.5A load. The experimental results show that the voltage / current spectra are spread to a chaotic range, and the switching noise of motor is reduced by the proposed method compared to the fixed frequency PWM method.

  • The problem of thermal stability of an exothermic reactive viscous fluid between two parallel walls in the plane Poiseuille and Couette flow configurations is investigated for different thermal boundary conditions. Neglecting reactant consumption, the closed-form solutions obtained from the momentum equation was inserted into the energy equation due to dissipative effect of viscosity. The resulting energy equation was analyzed for criticality using the variational method technique. The problem is characterized by two parameters: the Nusselt number(N) and the dynamic parameter($\Lambda$). We observed that the thermal and dynamical boundary conditions of the wall have led to a significant departure from known results. The influence of the variable pre-exponential factor, due to the numerical exponent m, also give further insight into the behavior of the system and the results expressed graphically and in tabular forms.

THE PEETRE K-METHOD IN STRONG $\lambda$-K-MONOTONE SPACES

  • In [5], M. Cwikel and J. Peetre showed that some Banach couples are Calderon pairs and every K-monotone cones in that couples can be constructed by the Peetre K-method. With the help of theorem 4 in [3], we can extend the above result to the K-monotone spaces. In [4], M. Cwikel provided the proof of therem 4 in [3] and also mentioned that some theorems in [5] which were stated in terms of K-monotone cones can be extended to K-monotone spaces. But M. Cwikel and J.Peetre's results only cover the case when over bar A = over bar B. We extend this result to the case when over bar A.neq. over bar B. In this case, we need a condition stronger than that the K-monotone space.

Determination of cable force based on the corrected numerical solution of cable vibration frequency equations

  • The accurate determination of cable tension is important to the monitoring of the condition of a cable-stayed bridge. When applying a vibration-based formula to identify the tension of a real cable under sag, stiffness and boundary conditions, the resulting error must not be overlooked. In this work, by resolving the implicit frequency function of a real cable under the above conditions numerically, indirect methods of determining the cable force and a method to calculate the corresponding cable mode frequency are investigated. The error in the tension is studied by numerical simulation, and an empirical error correction formula is presented by fitting the relationship between the cable force error and cable parameters ${\lambda}^2$ and ${\xi}$. A case study on two real cables of the Shanghai Changjiang Bridge shows that employing the method proposed in this paper can increase the accuracy of the determined cable force and reduce the computing time relative to the time required for the finite element model.

투과곡선을 이용한 La 농도에 따른 PLT 박막의 광학적 특성에 관한 연구 (A study on the optical properties of PLT thin films with varying the La concentration by using the transmission spectrum)

  • We have measured the optical properties, thickness, and energy band gap of the P $b_{1-x}$/100/L $a_{x}$100/ $Ti_{1-}$0.25x/100/ $O_{3}$ (PLT(x)) thin film prepared by the sol-gel method with varying the La concentration, x, fyom 15 nto 33 mol%. We have obtained the values from the tranmission spectrum and employed the envelope method in anayzing the spectrum. We have also performed the simulation of the transmission spectrum on the PC (personal computer) to verify the accuracy of the values 15 to 33mol%, the refractive index (at .lambda.=632.8nm) increases from 2.39 to 2.44. The extinction coefficient does not depend on the la concentration but mainly on te wavelength, and has the values between 0.2 and 0.5 at the wavelength shorter than 330nm and between 0.001 and 0.008 at the wavelength longer than 700nm. The energy band gap of the PLT (x) thin film has been obtained on the assumption of the direct band-to-band transition. It decreases from 3.28 to 3.17eV as the La concentration increases from 15 to 33 mol%. The thickness of the PLT(x) thin film has been also obtained in high accuracy by the envelope method..

골프장 농약 검사를 위한 다성분동시시험방법 확립에 관한 연구

  • The possibility of simultaneous analysis of 24 pesticides out of 30 residual pesticides which are subjected to test in the golf courses was examined. The utility of the simultaneous analysis of multi-residue pesticides was evaluated by recovery test through a standard addition method of pesticides in water, soil, and lawn grass. The experimental results of the recovery rates for individual pesticides are as follows : The number of pesticide of which average recovery was over 70% regardless of medium was 16 pesticides. These pesticides were composed of 8 organophosphorus pesticides(Chlorpyrifos, Chlorpyrifos-methyl, Diazinon, EPN, Fenitrothion, Phenthoate, Phosalone, and Toclofos-methyl). 4 organochlorinated pesticides(Daconil, Captan, Endosulfan, and Tetradifon), 2 pyrethroid pesticides(Fepropathrin, Lambda-cyhalothrin) and 2 other pesticides(Bromopropylate, Pendimethalin). On the other hand, in case of Dicofol, average recovery by medium was over 70% for water and lawn grass but was only 53.3% for soil. Therefore, the simultaneous analytical method applied in this experiment is not appropriate for analysis of Dicofol in soil. Furthermore, among 7 pesticides, 2 pesticides(Amitraz and Pyraclofos) showed that theirs average recovery rates deviated from criteria(70~130%) at almost all media, while 5 pesticides(Bensulide, Deltamethrin, Iprodione, Phosphamidon and Tralomethlin) were not detected from all media by selected GC detector(NPD or ECD).

Initial Growth Mode and Epitaxial Growth of AIN Thin Films on $Al_2O_3(0001)$ Substrate by DC Faced Target Sputtering

  • Using DC faced target sputtering method we grow AIN the films on the $Al_2O_3$(0001) substrate with varying thickness(17$\AA$-1000$\AA$). We measured x-ray diffraction(XRD) profiles by synchrotron radiation($\lambda$=1.12839 $\AA$) with four circle diffractometer. The full width half maximum(FWHM) of rocking curve for the AIN (0002) diffraction of the film grown at $500^{\circ}C$ was $0.029^{\circ}$. Also, we confirmed that the stress between AIN thin film and $Al_2O_3$(0001) substrate was reduced as increasing AIN film thickness, and the critical thickness of 400~500 $\AA$, defined as a lattice constant in the film agrees with that in a bulk without stress, was obtained.

임의의 분배비를 갖는 새로운 이중 대역 가지 선로 결합기 (A New Dual Band Branch Line Hybrid Coupler with Arbitrary Power Division Ratio)

  • 본 논문에서는 두 개의 대역에서 서로 다른 임의의 분배비를 갖는 이중 대역 가지 선로 결합기를 제안한다. 가지 선로 결합기의 선로들을 $\pi$-형 등가회로로 변환할 때 이 등가회로가 임의의 두 주파수에서 서로 다른 임피던스 특성을 갖고 두 주파수에 대하여 선로의 전기적 길이는 중심 주파수의 $\lambda/4$가 되도록 설계한다. 제안된 설계법의 타당성을 입증하기 위해서 0.9 GHz와 2 GHz에서 서로 다른 분배비를 갖는 결합기를 설계하였다. 분배비가 두 주파수에서 각각 1:1과 1:3이 되는 결합기를 설계, 제작 및 측정하였다. 측정 결과, 중심 주파수에서 삽입 손실은 0.33 dB 이내의 오차 범위를 보였고, 반사 손실과 격리 특성은 -18.07 dB 이상으로 매우 우수한 특성을 보였다.
