The Effect of Mugwort Extracts on the Benzo(a)pyrene-induced Hepatotoxicity in Rats (Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 쑥의 효과)

  • Mugwort has been used as a Korean folk medicine in treating liver diseases acting as an analgesics, sedative, diuresis, choleretics. This study was perfomed to evaluate the effect of mugwort extracts on the changes of enzyme activities, lipid accumulation of the serum and liver, when hepatotoxicity was induced by benzo(a)pyrene. The results are as follows: 1. Mugwort water extract administration prevented the increase of serum and liver AST, ALT, LDH, ${\gamma}$-GTP, liver ALP activities and bilirubin content caused by B(a)P injection. 2. The increase of serum and liver ALT, LDH, ${\gamma}$-GTP, serum AST activities and liver bilirubin contents in B(a)P treated group were decreased by mugwort methanol extract treatment. 3. Serum and liver total cholesterol, phospholipid, triglyceride level and serum HDL-cholesterol level were increased by B(a)P treatment. After combined treatment of mugwort water and methanul extracts, these lipid content were significantly decreased. 4. The hepatotropic effect of mugwort water extract and after-treatment against B(a)P induced hepatotoxicity was superior to that of methanol extract and pretreatment.

Changes in Some Metabolites and Ovarian Response to Super-ovulation Treatment after the Induction of Inflammation in Sheep (면양에 있어서 염증유발후의 혈중대사물의 동태 그리고 그후의 과잉배란처치에 대한 난소반응)

  • The effects of inflammation on the changes in some blood metabolites and ovarian response to super-ovulation treatment in sheep were investigated. Inflammation was induced artificially with LPS(Lipopoly-saccharide E. coli; 0.2mg/kg). Changes in the levels of NEFA, GOT, ${\gamma}$-GTP and body temperature after LPS injection were observed. Superovulation was carried out about 2 months after LPS injection. The body temperature, the levels of NEFA and GOT increased, and the level of total cholesterol decreased transiently. However, the level of ${\gamma}$-GTP did not change significantly. Superovulation results were not different between LPS injection and control groups. It was indicated that superovulation results did not decrease in the sheep which showed regular estrous cycle even after the induction of inflammation.

An Investigation on the Metabolic Syndromes and Health-Related Risk Factors among Male Workers (근로자들의 대사증후군과 건강위험 요인과의 관련성 연구)

  • This study is performed to estimate the prevalence of metabolic syndrome among male workers and to identify the relationships with many related factors including anthropometry, hematological index, serum lipid level, dietary-related behaviors and health-related behaviors. According to the age groups, the 20s are significantly higher in normal and risk groups than in the metabolic syndrome (MS) group, the 30s are significantly higher in MS group than the other groups. The levels of AST and ${\gamma}$-GTP both show significant differences in the order of MS group ($30.3{\pm}8.8U/l$, $91.1{\pm}40.2U/l$) > risk group ($25.7{\pm}8.1U/l$, $41.8{\pm}20.2U/l$) > normal group ($22.8{\pm}6.0U/l$, $26.6{\pm}10.7U/l$). For the frequency of breakfast consumption, the response of 'Every day' is significantly higher in MS group than normal and risk groups, but the response of 'Not at all' is significantly higher in normal group than MS and risk groups. The drinking amount is positively correlated with ${\gamma}$-GTP in normal group, and it is negatively correlated with the hematocrit level, but it is positively correlated with the systolic blood pressure in MS group. AST is positively correlated with glucose concentrations of the MS group. The ALT is positively correlated with waist circumferences and systolic blood pressure in the risk group. The results of this study show that breakfast frequency, education level, drinking amount, drinking frequency, exercise frequency, AST, ALT and ${\gamma}$-GTP levels are all important risk factors of MS. Therefore, it is very important to maintain a healthy life style for the prevention of MS incidence.

  • The classical type of transient receptor potential (TRPC) channel is a molecular candidate for $Ca^{2+}$-permeable cation channels in mammalian cells. Because TRPC4 and TRPC5 belong to the same subfamily of TRPC, they have been assumed to have the same physiological properties. However, we found that TRPC4 had its own functional characteristics different from those of TRPC5. TRPC4 channels had no constitutive activity and were activated by muscarinic stimulation only when a muscarinic receptor was co-expressed with TRPC4 in human embryonic kidney (HEK) cells. Endogenous muscarinic receptor appeared not to interact with TRPC4. TPRC4 activation by $GTP{\gamma}S$ was not desensitized. TPRC4 activation by $GTP{\gamma}S$ was not inhibited by either Rho kinase inhibitor or MLCK inhibitor. TRPC4 was sensitive to external pH with $pK_a$ of 7.3. Finally, TPRC4 activation by $GTP{\gamma}S$ was inhibited by the calmodulin inhibitor W-7. We conclude that TRPC4 and TRPC5 have different properties and their own physiological roles.

A Study for the Correlations between Obesity and Blood Parameters at the Worksite (직장인에서 비만과 혈액지표와의 상관성에 관한 연구)

  • Obesity, a state of having excessive body fat enough to attack one's health, is most blamed for causing chronical degenerative illness such as hypertension, diabetes mellitus as well as arteriosclerosis. According to many studies, the program for the weight control was the most demanded at the worksites. This study was designed to investigate the correlation between the degree of obesity and blood parameters of the employees at the worksites. The results of periodic medical examination for 571 employees(male: 418, female: 153) were analyzed; Age, BMI, systolic blood pressure(SBP), diastolic blood pressure(DBP), hemoglobin(Hb), fasting blood sugar(FBS), total-cholesterol(TC), serum glutamic oxaloacetic transaminase (sGOT), serum glutamic pyruvic transaminase(sGPT) and $gamma$-glutamyl transpeptidase($gamma$-GTP). Pearson correlation coefficient was used to find out the correlation between BMI and each blood parameters. In the case of male, 27.3% of the subjects were over-weight. The subject's age and their blood levels of uric acid, FBS, sGOT and sGPT did not show any significant difference regardless of BMI's, while SBP, DBP, Hb, TC and $gamma$-GTP levels of the overweight group were significantly higher than those of normal group. In the case of female, 9.2% of the subjects were over-weight. This study shows that the implementation of weight control program is required at the worksites

Effect of Zinc-enriched Yeast FF-10 Strain on the Alcoholic Hepatotoxicity in Alcohol Feeding Rats

  • The possible protective effects of highly zinc-containing yeast Saccharomyces cerevisiae, FF-10 strain, isolated from tropical fruit rambutan on acute alcoholic liver injury in rats were evaluated. Zinc concentration in this strain was 30.6mg%. The activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) were highly increased when alcohol was treated, relative to the normal rats. Also, a highly significant increase in the blood alcohol and acetaldehyde levels by alcohol treatment was observed. Administration of FF-10 strain markedly prevented alcohol-induced elevation of the activities of serum ALT, AST, and $\gamma$-GTP, and the levels of blood alcohol and acetaldehyde, and these reduced levels reached to that of normal rats. As compared with alcohol treated control rats, the FF-10 strain supplementation showed highly decreased the triglyceride concentration in serum. Alcohol treatment induced the marked accumulation of small lipid droplets, hepatocytes necrosis, and inflammation, but FF-10 strain administration attenuated to alcohol-induced accumulation of small lipid droplets and hepatocyte necrosis in the liver. Therefore, the current finding suggests that zinc-enriched yeast FF-10 strain isolated from tropical fruit rambutan may have protective effect against alcohol-induced hepatotoxicity.

Effects of Hot Water Extracts from Lentinus edodes on Hepatic Functional Enzyme Activities in the Rat Fed Butter Yellow(p-Dimethylaminoazobenzene) (표고버섯 열수 추출물이 발암원을 급여한 흰쥐의 간 기능 관련 효소활성에 미치는 영향)

  • This study was designed to observe the effect of hot water soluble polysaccharides extract(PS) from Lentinus edodes on the enzyme activities related with hepatic function and peroxidation in the rats fed better yellow. The four groups of male SD rats were fed with the diets contained 15% casein(basal diet; NO group), added butter yellow(BO group) or /and PS(NP, BP group) for 6 weeks. The activities of ${\gamma}$-GTP and GPT in BP were significantly lower compared with BO. The activities of glutathione peroxidase, catalase and lactate dehydrogenase were not significantly different between NP and NO, while those activities were significantly lower value in BP than BO. The activities of glutathione S-transferase of the microsomal and cytosol fractions were significantly lower in BP than in BO. The contents of glutathione and malondialdehyde in the liver were considerably low value in BP. In a view of these results the PS of Lentinus edodes prevents the lipid peroxidation and diminishes the liver toxicity caused with better yellow. The superoxide dismutase activity in cytosolic fraction of liver was not found any effect in all groups. But hepatic function enzyme activities such as catalase and glutathione peroxidase, LDH activities were remarkably decreased in the groups 2(basal diet + PS) and the ${\gamma}$-GTP, GOT and GPT activities, too. In liver, the contents of glutathione decreased by PS supplementation but HDL-cholesterol and total cholesterol ratio in plasma decreased at the groups 3, 4. The ${\gamma}$-GTP, GOT and GPT in plasma were remarkably higher in the rats fed the p-DAB than the control group, too. But above enzyme activities significantly decreased in the groups fed PS.

Effect of Ganjeonggyeok Acupuncture and Saengkankunbi-tang Treatment in the Changes of LFT and Lipid-obesity Index Values (간정격 시술 및 생간건비탕 투여가 간기능 및 비만도 변화에 미치는 영향)

  • Objectives : This study was aimed to figure out how effectively acupuncture treatment and herb-medicine work on patients with elevation of LFT value and lipid-obesity index. Methods : The subject were 23 patients visited to Dept. of Acupuncture and Moxibustion, Kwangdong Oriental Medicine Hospital, who complained chronic fatigue and wandering pain from Oct. 2007. to Oct. 2008. Before treatments, they were tested for LFT, total cholesterol, triglyceride, body fat ratio and obesity index. They were treated with Ganjeonggyeok acupuncture weekly for 1 month. During acupuncture treatments, we applied Sangkankunbi-tang to all patients twice a day. After 1 month, they were retested for LFT, total cholesterol, triglyceride, body fat ratio and obesity index. Results : 1. After treatments, we could get the result of dropping means of AST, ALT and $\gamma$-GTP value. Statistical significance of ALT and $\gamma$-GTP were recognized before and after treatments, but AST was not recognized. 2. After treatments, we also could get the result of dropping means of total cholesterol, triglyceride, body fat ratio and obesity index. Statistical significance of all values were recognized before and after treatments. 3. Among 23 patients, 10 cases were reported that they were not within normal limits at least one in AST, ALT and $\gamma$-GTP. After treatments of those patients, we could get the result of dropping means of AST, ALT and $\gamma$-GTP value. Statistical significance of all values were recognized before and after treatments. 4. Among 23 patients, 19 cases were reported that they were not within normal limits at least one in total cholesterol, triglyceride, body fat ratio and obesity index. After treatments of those patients, we could get the result of dropping means of total cholesterol, triglyceride, body fat ratio and obesity index. Statistical significance of all values were recognized before and after treatments. Conclusions : Though further study is necessary, our finding suggest that the conduction of Ganjeonggyeok acupuncture and Sangkankunbi-tang treatments have helped to decrease the figures of LFT and lipid-obesity index in patients.

Analysis of Hematological Factor to Predict of the Gallbladder Stone in Abdominal Ultrasound Images (복부초음파 영상에서 담낭담석을 예측하는 혈액학적 수치의 분석)

  • This study investigated the risk factor of Gallbladder stone in Busan and Kyungnam area. The subjects of the experiment was performed with patients by abdominal ultrasonography among the patients who came to the P hospital from June 2016 to December 2016. Among them, risk factors were analyzed on 353 people at the same time when abdominal ultrasonography and hematological test. The statistical analysis of risk factors related to the Gallbladder stone was performed by independent t-test and chi-square test. In consider of difference verification result for calculations odds ratio about independent variables, multiple logistic regression analysis to conduct verify adequacy by calculating forecasting model from variable. As a result, Gallbladder stone risk factors have relevance to age ${\gamma}GTP$ with probability model and values to calculated. Age was showed sensitivity 49.7%, specificity 82.2%, receiver operating characteristic area under curve 0.724. Forecasting probability sensitivity 69.3%, specificity 62.4%, receiver operating characteristic area under curve 0.699 showed, ${\gamma}GTP$ confirmed validity of forecasting model.

Clinical Usefulness of Ultrasonography in the Diagnosis of Fatty Liver (지방간 진단에 있어 초음파 검사의 임상적 유용성 연구)

  • The purpose of this study was to evaluate the clinical significance of ultrasonographic classification of fatty liver in three grades. From June 2018 to April 2019, 1047 patients (818 males and 229 females) diagnosed as fatty liver among 3607 patients who underwent abdominal ultrasonography at Busan screening center. Ultrasonography was classified into three grades: Grade I (mild fatty liver), Grade II (moderate fatty liver), and Grade III (severe fatty liver) according to the degree of parenchyma texture, acoustic attenuation, Obesity index, hematological test, and metabolic syndrome. The average age of men in each sex increased with the increase of the fatty liver. Body mass index (BMI) and waist circumference were significantly increased in both men and women (p=.000). hematological analysis showed that AST, ALT, ${\gamma}-GTP$, TG, fasting blood sugar, and glycated hemoglobin were significantly different from each other (p<.05). In women, ALT, ${\gamma}-GTP$ and TG showed a significant difference with increasing fatty liver (p<.05). The prevalence of metabolic syndrome was significantly increased in both sexes as the grade of fatty liver increased (p=.000). Based on the results of this study, it is suggested that the use of ultrasound - guided fatty liver according to severity may be useful for the treatment and follow - up of fatty liver if the liver grade is divided in consideration of hematological variables and metabolic syndrome.
