Immune-Enhancing Effects of Phellinus linteus Fruit Body and Mycelium Cultured in Cudrania tricuspidata (꾸지뽕나무 배양 상황버섯 자실체 및 균사체의 면역증진 효과)

  • Polysaccharides produced in microorganisms and plants are known to increase the immune response in the body. We proposed analysis of beta-glucan contents of phellinus linteus fruit body (FB) and mycelium (MC) cultured in cudrania tricuspidata. Also, we examined whether fruit body and mycelium can increase the immune response in cyclophosphamide-induced immunosuppression animal models. We injected cyclophosphamide (50 mg/kg) twice to produce immunosuppression mice. Then, FB (200 mg/kg) and MC (200 mg/kg) were oral administered for 14 days. In order to confirm the immune-enhancing effect of FB and MC, we analyzed spleen weight, the number of immune cells, cytokines, and immunoglobulins levels. Cyclophosphamide decreased the weight of spleen, the number of immune cells. However, FB and MC have significantly increased the weight of spleen, the number of white blood cell, lymphocyte and monocyte. In addition, they have significantly increased immune-related cytokines (IL-2 and IFN-${\gamma}$) and immunoglobulins (IgA, IgG, IgM) levels. As a results, phellinus linteus fruit body (FB) and mycelium (MC) cultured in cudrania tricuspidata can be used as effective natural materials for immune-enhancing.

Structural and physicochemical properties of starch by barley cultivars

  • The objective of this study was to investigate the structural and physicochemical properties of starch by barely cultivars. Hwanggeumchal had a moisture content and ${\beta}$-glucan content of 12.02 and 6.23%, respectively. Hyegang had higher protein contents than those of the other cultivars. As a result of observing the particle size of starch, Hwanggeumchal and Hyegang had smaller particles of starch compared with the other cultivars at 15.7 and $15.9{\mu}m$, respectively. The analysis results on the content of damaged starch showed that Dahan and Hyegang had a damaged starch content of 1.14 and 1.20%, whereas Boseokchal and Hwanggeumchal were 0.76 and 0.49% respectively, showing low waxy cultivars. As for the content of amylose, the results show that Dahan and Hyegang had an amylose content of 37.07 and 37.75%, and Boseokchal and Hwanggeumchal were at 11.22 and 37.75%, respectively. As for the degree of amylopectin polymerization, all four cultivars had the highest degree of polymerization (DP) content of 13 - 24 at more than 54%, whereas the DP content ${\geq}37$ was the lowest at less than 5.35%. The results for the soluble and resistant starch content show that the content of soluble starch ranged from 93.90 to 95.76%, and resistant starch was 0.17 - 0.40%. After analyzing the gelatinization properties of barley starch, the value of the setback was low in Hwanggeumchal and Hyegang; thus, it is considered that the aging process of those cultivars will be slower than that of the others.

Effect of Continual Application of Liquid Pig Manure on Malting Barley Growth and Soil Environment in Double Cropping System of Rice-Malting Barley (벼.맥주보리 작부체계에서 돈분액비 연용이 맥주보리 생육과 토양 환경에 미치는 영향)

  • To investigate the effect of continual pre-plant application of liquid pig manure (LPM) on malting barley growth, quality and soil environment in double cropping system of rice and malting barley, the liquid pig manure was applied after harvesting rice and malting barley for 3 years. Field experiment was designed with non-fertilizer, chemical fertilizer (CF) recommended by soil testing, rice (LPM 50%+CF 50%)+malting barley (CF 100%), rice (LPM 50%+CF 50%)+malting barley (LPM 50%+CF 50%), rice (LPM 100%)+malting barley (CF 100%) and rice (LPM 100%)+malting barley (LPM 100%). By continuous application of LPM 100%, the contents of available $P_2O_5$ and exchangeable K in the soil were increased. The available $P_2O_5$ increased from 243 to 350 mg $kg^{-1}$ and exchangeable K was changed 0.31 to 0.44 $cmol_{c}\;kg^{-1}$. However, the contents of available $P_2O_5$ and exchangeable K were not significant changes in rice (LPM 50%+CF 50%)+malting barley (LPM 50%+CF 50%) plot. Bulk density of soil was not affected by application of LPM. The microbial density was high in order of bacteria > actinomycetes > fungi. The population of aerobic bacteria in rice (LPM 100%)+malting barley (LPM 100%) plot was higher than other plots. The ratio of aerobic bacteria/fungi and biomass C content were the highest in rice (LPM 100%)+malting barley (LPM 100%) plot. The yield of malting barley was increased 22% by increasing culm length, panicle length, No. of panicle and 1,000 grains as 358 kg $10a^{-1}$ in rice (LPM 100%)+malting barley (LPM 100%) plot compared with 294 kg $10a^{-1}$ in rice (CF 100%)+malting barley (CF 100%) plot. The content of ${\beta}$-glucan was low by 4.5 and 4.4% in non-fertilizer and rice (CF 100%)+malting barley (CF 100%) plot, respectively. The content of crude protein was the lowest by 8.2% in non-fertilizer and rice (CF 100%)+malting barley (CF 100%) plot and the quality of malting barley was good as within 11%.

Comparison of the physiological activities of Korean and Chinese Auricularia auricula and Tremella fuciformis extracts prepared with various solvents (한국 및 중국산 목이 및 흰목이의 추출용매에 따른 생리활성 성분 비교)

  • The aim of this study was to evaluate the physiological activities of hot-water and 70% ethanol extracts of three edible mushroom species (Auricularia auricula, Tremella fuciformis, and Lentinus edodes) cultivated in Korea and China. The DPPH radical scavenging activities of hot-water extracts of Korean A. auricula, T. fuciformis, and L. edodes were significantly higher than those of the Chinese counterparts (p < 0.01). However, the nitrite scavenging activities of the hot-water extracts did not significantly differ among the mushrooms regardless of their origins. Chinese A. auricula showed the highest nitrite scavenging activity in the ethanol extract. The total polyphenol contents of the ethanol extracts of the three edible mushroom species from both of the countries were higher than those of the hot-water extracts. Korean A. auricula and T. fuciformis yielded a high polyphenol content with hot-water extraction. The ${\beta}-glucan$ contents of the Chinese A. auricula, T. fuciformis, and L. edodes extracts were 20.49%, 31.43%, and 30.09%, respectively, which were higher than those of the Korean counterparts. From the results of this study, it can be deduced that the maximum yield of nutritional ingredients can be extracted by using the optimal solvent for each mushroom species. These results are expected to be useful in appreciating the difference among extracts of Korean and Chinese edible mushrooms in terms of their physiological activities in the solvents used.

  • Improvements in understanding the effects of dietary fermentable carbohydrates and their interaction with supplemental feed enzymes and the feed manufacturing process may lead to reductions in volatile organic compound (VOC) emissions from poultry manure. Starch digestibility has been improved by replacing ground wheat or barley with whole wheat or barley, but there was no consistent effect of cereal species or feed form on the pH value of the gizzard contents. Pelleting results in improvements in feed conversion from 0 to 12%. Starch digestibility has been reported to account for up to 35 % of the improvement in available metabolic energy as a result of xylase supplementation. Factors which affect starch utilization and non-starch polysaccharide (NSP) absorption include the presence of anti-nutrient facto. (ANF) in grains, the nature of grain starch, NSP and the digestive capacity of animals. Improvements in feed production technology have been made in enzyme stabilization, allowing some dry enzyme products to be pelleted after conditioning at up to $87.69^{\circ}C$ and liquid enzymes to be stored in the feed mill for up to low months prior to use. The soluble NSP, arabinokylans and beta-glucans are partially degraded into smaller fragments by enzymes. With fragmentation, the water holding capacity is decreased, which leads to a reduction in digesta moisture, wet feces, and dirty eggs from hens fed diets containing viscosity-inducing ingredients.

Changes of biological activities and nutrition contents by different extraction conditions in the mixtures of roasted edible mushrooms and grain additives for the development of mushroom tea (버섯차 개발을 위한 로스팅 식용버섯류와 곡물첨가물의 혼합비율에 따른 추출온도 및 시간별 생리활성 및 영양성분 변화)

  • This study aimed to investigate the biological activities and amino acid content in the mixtures of roasted edible mushrooms (Pleurotus ostreatus, and Lentinula edodes) and grain additives (oat and brown rice) obtained under different extraction conditions for the development of mushroom tea. The total amount of polyphenol and β-glucan in edible mushrooms was increased with the roasting treatment compared to that observed with the air-drying treatment. In addition, DPPH radical scavenging activity and the amount of all amino acid components, including glutamic acid (Glu), were higher in the oat extracts than those observed in the brown rice. The biological activities and nutrient content were examined based on extraction temperatures and thetime required to obtain the mixtures of edible mushrooms and grain additives. The extract of a 1:1 mixture of L. edodes + oat (LE+O) obtained at 100℃, 3 min showed high DPPH radical scavenging activity (33.5%), and the extract of 3:1 mixture of P. ostreatus + brown rice (PO+B) obtained at 100℃, 3 min showed high nitrite scavenging activity (49.9%). The total polyphenol content of the extract of 3:1 mixture of P. ostreatus + brown rice (PO+B) was the highest when obtained at 16.2 mg GAE/g and+ brown rice (PO+B) was the highest when obtained at 16.2 mg GAE/g and 70℃, 10 min. The essential amino acid content was higher in the 1:1 mixing ratio of P. ostreatus + oat (PO+O) and P. ostreatus + brown rice (PO+B) and 3:1 mixing ratio. L. edodes + oat (LE+O) and L. edodes + brown rice (LE+B) compared to that observed in other mixing ratios.

Development of Functional Food Materials from Acanthopanax senticosus-Fermented Mushroom Mycelia (가시오가피 버섯균사체 발효물의 기능성 식품 소재 개발)

  • Three mushroom mycelia, Ganoderma lucidum, Hericium erinaceum, and Phellinus linteus, were separately diluted with the natural culture media Acanthopanax senticosus. Solid-state fermentation was used to produce three different A. senticosus-fermented mushroom mycelium groups: G. lucidum mycelia, H. erinaceum mycelia, and P. linteus mycelia. The resulting mycelia were analyzed to assess their efficacies as health functional foods. Optimized fermentation conditions were determined by considering the density and growth speed of mycelia in each A. senticosus-fermented mushroom mycelium group. The cultured mushroom mycelia under the optimized conditions were extracted using water and 70% ethanol. Extraction was followed by filtration, concentration and freeze-drying to produce extract powder of A. senticosus-fermented mushroom mycelia: Water extracts (FM-5111, FM-5121, and FM-5131) and 70% ethanol extracts (FM-5112, FM-5122, and FM-5132). Analysis of extract powder of A. senticosus-fermented mushroom mycelia was performed using the maker compounds eleutheroside B and eleutheroside E. Analysis of ${\beta}$-glucan contents was performed by enzymatic procedures.

Antioxidant and Immunological Activities of Sparassis crispa Fermented with Meyerozyma guilliermondii FM (Meyerozyma guilliermondii FM을 이용한 꽃송이버섯 발효물의 항산화 효과 및 면역 활성)

  • The effects of Sparassis crispa extracts fermented with isolated strain from S. crispa on antioxidant and immunological activities were determined. S. crispa extracts fermented with Meyerozyma guilliermondii FM showed significantly higher total phenol contents and DPPH radical scavenging activities compared to those fermented with lactic acid bacteria. In methotrexate-induced immunosuppressed rats, reduced levels of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-2, and immunoglobulin E (IgE) and increased levels of IL-10 were detected in S. crispa extract injected groups regardless of fermentation. We confirmed that rats treated with S. crispa fermented with M. guilliermondii FM showed higher blood leukocyte contents compared to other treatments. These results suggest that M. guilliermondii FM has high potential as a starter culture for fermentation of S. crispa extracts with increased antioxidant and immunological activities.

Physicochemical Properties of Hull-less Barley Flours Prepared with Different Grinding Mills (제분방법에 따른 쌀보리가루의 이화학적 특성)

  • During the pearling process of hull-less barley, protein, lipid, ash and insoluble dietary fiber (IDF) contents decreased, while soluble dietary fiber (SDF) and ${\beta}-glucan$ contents slightly increased. Depending on milling methods and types of grinding mills used, there were differences in particle size distribution of barley flour. Flour particle size was smaller in the following order of Fitz mill, Ball mill, Pin mill, Cyclotec sample mill and Jet mill. Color (brightness) was closely related to the particle size of barley flour. Damaged starch (%) in pearled barley flour was the highest in Jet mill among different mills. Flours prepared with Cyclone mill and Pin mill had a reasonable amount of damaged starch. Flour produced by Fitz mill showed the lowest amount of damaged starch. Scanning electron microscopy (SEM) of the flour samples demonstrated different sizes and shapes of particles consisting of starch granules and cell wall materials. Damaged starch tended to increase water absorption index (WAI), water solubility index (WSI), and water retention capacity (WRC). Pasting viscosity determined by amylograph was relatively high in Pin-milled and Cyclone-milled flours. Viscosity was the lowest in coarsely ground flour by Fits mill.

Changes in Physicochemical Characteristics of Immature Barley Kernels during Roasting (미숙보리곡립의 볶음중 이화학적 특성변화)

  • Changes in physicochemical characteristics were investigated for immature barley kernels roasted at $160{\sim}220^{\circ}C$ for $1{\sim}12$ min. Only small differences in chemical constituents including starch, protein, fat, ash, total dietary fiber, and ${\beta}-glucan were observed between immature and mature barley kernels. The amounts of 75% ethanol-soluble sugars and amino acids present in immature barley kernels were considerably higher than those in mature kernels, and gradually decreased in the process of roasting. Of free sugars, sucrose, raffinose, glucodifructose($GF_{2}$) and maltose were reduced by roasting. Glucose and fructose, simple reducing sugars, decreased at the early stage of roasting, followed by a slight increase at the later stage. Starch and nitrogen contents decreased slowly, while TDF(total dietary fiber) had a tendency to increase slightly. Stacking volume of immature barley kernels increased markedly, especially at the higher temperatures. L value of immature barley decreased throughout roasting, and a, b values increased at the early stage of roasting but b value decreased with continued roasting. The degree of roasting was strongly affected by the roasting temperature. Darkness of immature barley kernel, depending on the degree of roasting, was highly associated with concentrations of brown pigments extracted from roasted immature barley kernels.
