In this study, the anti-oxidant, anti-tumorigenic, anti-hypertensive, anti-thrombic, anti-diabetic, and anti-inflammatory properties of 18 different species of genus Pleurotus were investigated. In addition, the amino acid, $\beta$-glucan, and polyphenol content were also measured. All species contained more than 20 mg% of polyphenol with the highest contents found in Pleurotus cornucopiae var. citrinopileatus (yellow pleurotus) ($39.13{\pm}0.82\;mg%$). The $\beta$-glucan contents was also the highest in yellow Pleurotus ($37.67{\pm}0.22%$) followed by Won-Hyeong1 (C, $28.75{\pm}0.61%$) and Jang-an PK (A, $27.95{\pm}0.33%$). The yellow Pleurotus exhibited the highest antioxidant activity as assessed by the DPPH scavenging rate with an $IC_{50}$ value of $2.94{\pm}0.44\;mg/mL$. Ethanol extracts from the yellow Pleurotus treated at 1% concentration showed cytotoxic activity up to 36.9% in the human embryonic kidney 293T cell lines. The yellow Pleurotus also showed the highest inhibitory effects on ACE activity ($60.52{\pm}0.2%$). Finally, the yellow Pleurotus exhibited anti-diabetic and anti-inflammatory properties as shown by inhibition of $\alpha$-amyloglucosidase activity ($50.5{\pm}0.8%$) and nitric oxide production ($68.4{\pm}0.3%$). Taken together, our data indicate the yellow pleurotus is a promising functional food ingredients.

This study investigated the changes in functional components of barley powder produced from different grain varieties (Dahan, Hinchalssalbori, Heukgwang, Huknuri and Boseokchal) and varying milling degrees (27, 23, 19, and 15%). Total polyphenol contents increased with a decrease in the milling degree, with content ranges of 0.97-1.40, 1.19-1.66, 1.22-1.77, 1.30-1.93, and 1.46-2.12 mg/g, respectively. Total flavonoids content also increased with a decrease in the milling degree. The total polyphenol and flavonoids contents were the highest in barley powder from Huknuri and Boseokchal grains. Total dietary fiber, arabinoxylan, and GABA contents increased with a decrease in the milling degree. As the milling degrees decreased, ${\beta}$-glucan contents, which was the highest in Hinchalssalbori and Boseokchal, decreased with ranges of 4.98-7.29, 5.26-7.03, 4.84-7.17, 4.84-700, and 4.66-6.33 mg/100 g, respectively. These results provide useful data for selection of an appropriate variety and milling degree to achieve a high quality in barley processing.
A new six-rowed naked barley cultivar "Jinjuchal" with high whiteness after cooking and high $\beta$-glucan content was developed from the cross between 'Jinmichapssalbori' with high winter hardiness, lodging tolerance, grain whiteness and pearling yield, and 'Suwon 333' with waxy endosperm by the Honam Agricultural Research Institute (HARI), NICS, RDA in 2007. An elite line, SB962002G-B-B-B-84-4 was selected in 2002 and designated as 'Iksan 79' It showed good agronomic performance in the regional yield trials (RYT) from 2005 to 2007 and was released with the name of "Jinjuchal" possessing high whiteness and low proanthocyanidin waxy endosperm. The average heading and maturing dates of "Jinjuchal" were April 27 and June 1 in paddy field, which were same and one day later than those of the check cultivar 'Saechalssalbori' respectively. The new cultivar, "Jinjuchal" had 81 cm of culm length that was 1cm longer than that of 'Saechalssalbori' and 4.9 cm of spike length. It showed 673 spikes per $m^2$, 56 grains per spike, 27.0 g of 1,000-grain weight, and 752 g of test weight. "Jinjuchal" showed stronger hardiness and better resistance to powdery mildew and BaYMV (Barley yellow mosaic virus) than those of the check cultivar, 'Saechalssalbori' It showed higher $\beta$-glucan content(8.4%) and water absorption rate than those of the check cultivar, 'Saechalssalbori' Its average yield of the pearled grain in the regional yield trial was 3.79 MT/ha in upland, and 3.73 MT/ha in paddy field, which were 1% lower and 3% higher than those of the check cultivar, respectively. Total phenol and proanthocyanidin contents were 4.2 and 0.2 mg/g, respectively. This cultivar is suitable for the area of the daily minimum temperature above $-6^{\circ}C$ in January in Korean peninsula.
Quality characteristics of yogurt with added colored barely flour was investigated during fermentation by lactic acid bacteria. Chemical properties such as moisture, crude protein, starch, ash and ${\beta}$-glucan contents was measured. pH, acidity, brix, Hunter color value and growth of lactic acid bacteria in yogurt was investigated during fermentation by L. acidophilus, L. bulgaricus, and S. thermophilus mixed culture. Crude protein contents of Daeanchal and Boseokchal was 16.16 and 12.17%, respectively. Starch contents of daeanchal were shown lower score. The pH of yogurt by addition of barley flour (Daeanchal) addition 0 and 20% were 6.66 and 6.40, respectively. The pH of yogurt supplemented with barley flour tended to be lower than before control which was not added barely flours and oligosaccharide in yogurt. Titratable acidity of yogurt added barley flour was higher compared with that of control. Brix of yogurt was decreased during fermentation by lactic acid bacteria. Lightness of yogurt added barley flour (Daeanchal) addition 0 and 20% were 83.25 and 69.83, respectively. The original microbial population of the yogurt during 0, 5, 8, and 15 hr fermentation were 7.48, 7.79, 8.15, and 8.71 Log CFU/g, respectively. Moreover, the addition of colored barley flour was to promote the proliferation of lactic acid bacteria in yogurt. In our research, addition of colored barley flours added into the yogurt may also have contributed to growth of lactic acid bacteria.
Studies have been conducted on fermentation products known to increase biological activity through bioconversion of mycelium. In this study, ethanol extract of waxy sorghum (WS) and ethanol extract of waxy sorghum fermented with Phellinus linteus mycelium (WSPM) were prepared, and functional component contents, antioxidant activity, and cytotoxicity were analyzed. Total polyphenol contents and total flavonoid contents of WSPM were higher than those of WS. In addition, the ${\beta}-glucan$ content of WS was higher than that of WSPM. DPPH and ABTS radical scavenging activities showed that WSPM had higher antioxidant activity than WS at all concentrations. Analysis of SOD-like activity also showed higher antioxidant activity in WSPM. MTT assay demonstrated that WSPM exhibited high inhibitory activity in all cancer cells, and in particular, in HeLa cells with the highest inhibition. A concentration-dependent increase in anticancer activities of WS and WSPM was detected in all cancer cells, which was identical to the SRB assay result. MTT and SRB assay showed the increased cytotoxicity of WSPM in cancer cells. Therefore, it is expected that WSPM can be used as a functional food material.
This study suggests a method of making cheese using soybeans for healthy food for people that are allergic to animal proteins, vegetarians, people on a diet, infants and toddlers. Additionally, the study was conducted to provide basic data that can contribute to the development of a quality control class of Korean cheese and underdeveloped cheese industry. Soybean cheeses have a high protein content and low fat. The free amino acids of soybean cheeses contained 11.48 mg of arginine per 100 g, 9.33 mg of glutamate, and leucine 4.91 mg, in that order. The free amino acids of Company A's milk cheese contained 20.95 mg of glutamate, 8.95 mg of proline and 8.02 mg of lysine per 100 g. In soybean cheeses, there were 2.21 mg of tryptophan and 0.73 mg of cysteine, which were not analyzed in the milk cheese of company A. The contents of the constituent amino acids was 1,070.22 mg of glutamate, 467.30 mg of aspartate and 446.30 mg of leucine in 100 grams of soybean cheeses per 100 grams. The milk cheese of Company A was 1,715.97 mg of glutamate, 798.72 mg of leucine and 685.31 mg of proline. The mineral contents of the soybean cheese were 120.29 mg/100 g of calcium, 0.92 mg/100 g of iron, 0.78 mg/100 g of zinc and 0.40 mg/100 g of selenium, respectively. The contents of vitamin $B_1$ and ${\beta}$-glucan in soybean cheese were higher than that of the milk cheese in Company A at 0.08 mg/100 g and 13.73 mg/g, respectively. Therefore, it is considered that the soy cheese is excellent in nutritional aspect and will contribute to health promotion. It is also suitable for people that are allergic to animal proteins, vegetarians, people on a diet and healthy foods for infants.
Agaricus brasiliensis, one of edible mushroom belonging to Basidiomycota, has been used for curing gastric ulcer and stomach cancer of human beings and also known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were prepared from fruiting body of the mushroom. ${\beta}$-glucan and total protein contents were identify from fractions of edible mushrooms extract. The ${\beta}$-glucan and protein contents of all fractions of the mushrooms ranged from 21.54~32.31% and 0.16~9.34%, respectively. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HT-29, NIH3T3 and RAW 264.7 at the concentration of 10~2000 ${\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of 18.8~50.6% in mice previously inoculated with Sarcoma 180. Fr. HW increased the numbers of spleen cell by 1.2 fold at the concentration of 200 ${\mu}g/ml$ compared with control. Fr. MeOH and Na improved the immuno-potentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.6 fold compared with control at the concentration of 50~500 ${\mu}g/ml$. Fr. Na generated 15.9 ${\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7 at the concentration of 200 ${\mu}g/ml$, while lipopolysaccharide, a positive control, produced 3.7 ${\mu}M$. The Fr. NaCl, Fr. HW and Fr. MeOH increased the secretion of TNF-${\alpha}$, IL-$1{\beta}$, Il-2 and IL-6 by 2.2 times compared with the control group. Fr. Na increased the numbers of peritoneal exudate cells by 4 folds at the concentration of 50mg/kg compared with control. Circulating leukocytes increased by 2.7 folds when Fr. HW from A. brasiliensis was inoculated at the concentration of 50 mg/kg body weight. The hematological and blood chemical analysis of the 3 fractions did not show any difference in blood compositions and enzyme activities compared with the control group (p<0.05). Therefore, the experimental results suggested that crude polysaccharides extracted from A. brasiliensis contain antitumor and immuno-potentiating activities against Sarcoma 180 in ICR mice.
We examined the effects of biological activity Phellinus linteus mycelium culture with cassiae semen extract. Firstly, the optimal temperature, initial pH and culture period for mycelial growth in a liquid culture of P. linteus were determined, and they were $30^{\circ}C$, pH 5.0 and 8 days respectively. The five herbal materials were examined against several health functional efficacies, and, as a result, Cassiae semen was chosen, with its superior inhibitory effects in ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities(95.3%, 80.9%, 96.1 and 24.2%, respectively). P. linteus fruit body was investigated on ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities, and they were 54.7%, 81.9%, 30.0% and 20.1%, respectively. Accordingly, C. semen was used in the following experiment, to give an additive functional effect on the P. linteus. As the amount of C. semen in the cultural media increased, mycelial weight and ${\beta}$-glucan contents also increased, but final pH was not influenced. In addition, the ${\beta}$-glucuronidase inhibitory activity, electron donating activity, and ${\alpha}$-glucosidase inhibitory activity increased. P. linteus mycelium culture showed higher activities in the other three tests above, except for electron donating activity, when C. semen was added to the medium before cultivation.
A total of 24 processed barley samples produced in Korea, 9 split and 15 pressed, were analyzed for protein, $\beta$-glucan, and amylose content whiteness cooking characteristics (water absorption and expansibility) and pasting properties, with respect to the areas in which they were grown. Split and pressed barley products from Japan were compared. Both forms of barley products had similar contents of protein and $\beta$-glucan. Whiteness was higher in pressed barley than in split barley. Split barley produced in Korea had a higher water absorption (342-436%) and expansibility (449-608%) than did pressed barley. Japanese processed barley products were much lower in protein (4.4-6.6%, w/w) and showed a higher whiteness grade than did Korean products. Whiteness of pressed barley ranged from 57.2-68.3 and was higher than that of split barley, but split barley product showed better cooking characteristics than did pressed barley. Japanese products were similar to Korean materials in this respect. A negative correlation was observed between protein content in and whiteness of processed barley products (r=-0.5112, p<0.01). Waxy barley products had a lower pasting temperature, and showed higher breakdown and lower setback than did non-waxy barley products. The pasting properties of Japanese products were different from those of Korean materials. The pasting temperatures of Japanese split and pressed barley were lower, and the setback much higher, than seen with Korean products. The results show that both splitting and pressing efficiently improve the cooking characteristics of barley. The processing of waxy barley was particularly effective.

The antihyperlipidemic and glycemic control effects of mycelia of Inonotus obliquus including protein-bound polysaccharides extract were investigated. In high fat diet-induced obese C57BL/6J mice, the mycelia of Inonotus obliquus including protein-bound polysaccharides extract showed significant decrease in epididymal fat tissue weight, blood triglyceride and VLDL level, triglyceride and total cholesterol level in liver, serum insulin and HOMA-IR level and AUC. Oral glucose tolerance test glucose level reduction were 4.9% (M150L; ${\beta}$-glucan 1.303 mg/kg bw), 9.5% (M150; ${\beta}$-glucan 2.606 mg/kg bw) after 180 min of glucose loading compared to H-C group. Besides, the mycelia of Inonotus obliquus including protein-bound polysaccharides extract treatment significantly increased glycogen contents in liver and adiponectin level in high fat diet-induced obese mice. In conclusion, the results showed that the mycelia of Inonotus obliquus including protein-bound polysaccharides extract possesses significant antihyperlipidemic and glycemic control effects in C57BL/6J mice.
