The objective of this study was to investigate the antioxidative capacity of ethanol extracts from Sanguisorbae officinalis L. root (Sanguisorbae radix) in vitro. The concentration of Sanguisorbae radix extract at which DPPH radical scavenging activity was inhibited by 50% was 0.33 mg/mL, which was similar to $IC_{50}$ of ${\alpha}$-tocopherol (0.40 mg/mL), as compared to 100% by pyrogallol as a reference. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of ${\alpha}$-tocopherol. Superoxide scavenging activities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of catechin. Oxygen radical absorbance capacities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of ascorbic acid. Cupric reducing antioxidant capacities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of ${\alpha}$-tocopherol. Sanguisorbae radix extract prevented supercoiled DNA strand breakage induced by hydroxyl radical and peroxyl radical. Total phenolic contents of Sanguisorbae radix extract at concentrations of 0.5 and 5 mg/mL were 0.50 and 3.33 mM gallic acid equivalents, respectively. Sanguisorbae radix extract at concentration of 0.01, 0.1 and 0.5 mg/mL inhibited 0.2 mM tert-butyl hydroperoxide-induced cytotoxicity by 33.8, 79.1 and 96.9%, respectively, in HepG2 cell culture system. Thus, strong antioxidant and cytotoxicity-ihnibiting effects of Sanguisorbae radix extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents.

Adenophora remotiflora (Mosidae) is a perennial plant in the Campanulaceae family and a wild plant that only inhabits in Korea. This research analyzed the volatile compounds in Mosidae and their antioxidant activities. The volatile compounds in fresh, shady air-dried, and freeze-dried Mosidae were isolated by steam-distillation extraction (SDE) method using diethylether as a solvent. Volatile compounds were analyzed by gas chromatography-mass spectrometry (GC/MS). Antioxidant activities were determined using the linoleic acid system and 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. Sixty and seventy-two volatile compounds were identified from fresh and shady air-dried Mosidae, respectively. In fresh Mosidae, the most abundant compounds were ethyl acetate and heptyl acetate, while ethyl acetate and limonene were the most abundant in the shady air-dried sample. Inhibition of peroxide formation by fresh Mosidae was higher than that of ${\alpha}-tocopherol$, and inhibition by shady air-dried Mosidae was same as that of ${\alpha}-tocopherol$. Furthermore, volatile compounds from shady air-dried Mosidae had higher free radical scavenging activity than ${\alpha}-tocopherol$. The freeze-dried sample showed lower antioxidant activity in both the linoleic acid system and DPPH method.
To examine the effects of feed additives on the expression of perpheral blood cell surface molecules, phagocytosis and antigen specific antibody formation, broilers were randomly assigned to $T_{1}$ , $T_{2}$ , $T_{3}$ , and $T_{4}$ groups. $T_{1}$ group was fed diet without any additives for 13 weeks, $T_{2}$ was fed diet with full fat flax, $T_{3}$ was fed diet with full fat flax containing $\alpha$-tocopherol, and $T_{4}$ was fed diet with full- fat flax containing $\alpha$-tocopherol and selenium. Since 5 weeks feeding the data were examined by flow cytometry using a panel of monoclonal antibodies. The expression of monocyte in all treated groups was significantly increased, in which the ratio of expression in $T_{3}$ group was especially evident. B cell expression of all treated groups was increased more than 2 fold. The expression of CD4+(helper T cell) cell and CD8+(cytotox$ic^pressor T cell) cell of all treated groups also was increased.ed.

The changes of the physico-chemical properties and fatty acid compositions of cooked soybean oil, 30 min at $180{\pm}5^{\circ}C$, were investigated to compare the antixidative effects of some antioxidants such as BHA, ${\alpha}-tocopherol$ and sesamol according to various storage conditions(room temperature, room temperature with a air tight in the dark and low temperature with air tight) for 4 weeks. The order of antioxidative effects according to different storage conditions was low temperature with air tight, room temperature with air tight in the dark and room temperature. Acid values, peroxide values and carbonyl values of soybean oil stored under low temperature with air tight after ${\alpha}-tocopherol$ treatment were similar to those of soybean oil stored under room temperature after BHA treatment. The relative contents of linoleic acid and linolenic acid decreased during storage, wheras those of oleic acid and palmitic acid increased. The contents of linoleic acid in soybean oil treated with ${\alpha}-tocopherol$ and BHA under low temperature with air tight storage were $53.61{\sim}50.29%$ and $53.78{\sim}50.68%$, respectively. These contents were very high in comparison with those in untreated oil under room temperature storage, $52.09{\sim}43.96%$.

To develop a natural antioxidative peptide, the gelatin was extracted from fish (Yellowfin sole) skin by hot $water(50^{\circ}C)$ extraction method and hydrolyzed with Alcalase, pronase and collagenase through a continuous 3-step membrane reactor. Each step enzymatic hydrolysates were determined the antioxidative activity and their synergistic effects, compared with $\alpha-tocopherol$ and butylated hydroxytoluene (BHT). Also, we tried to investigate the antioxidative disposition of peptide which was successfully separated by gel filtration, ion-exchange chromatography, and HPIC in cultured rat hepatocytes intoxicated with tert-butyl hydroperoxide (TBHP). Second step enzymatic hydrolysate (SSEH) among all hydrolysates and $\alpha-tocoperol$ was showed the strongest antioxidative activity. The optimum concentration of antioxidative activity for SSEH was $1\%(w/w)$ in linoleic acid. The synergistic effects were increased in using the hydrolysate with tocopherol and BHT. In the presence of the peptide isolated from SSEH, supplemented hepatocytes exposed to TBHP showed that delayed cell killing and decreased significantly the lipid peroxidation, compared with hepatocytes not cultured with isolated peptide.

In this study, we investigated the cellular protective effects and mechanisms of nicotiflorin and its aglycone kaempferol isolated from Annona muricata. The protective effect of these components against $^1O_2$-induced cell damage was also studied by using L-ascorbic acid and (+)-${\alpha}$-tocopherol as controls. Kaempferol exhibited the most potent protective effect, followed by (+)-${\alpha}$-tocopherol and nicotiflorin. L-Ascorbic acid did not exhibit any cellular protective effects. To elucidate the mechanism underlying protective effects, the quenching rate constant of the singlet oxygen, free radical-scavenging activity, ROS-scavenging activity, and uptake ratio of the erythrocyte membrane were measured. The results showed that the cell membrane penetration is a key factor determining the cellular protective effect of kaempferol and its glycoside nicotiflorin. The result from L-ascorbic acid demonstrated that the cellular protective effect of a compound depends on its ability to penetrate the cell membrane and is independent of its antioxidant capacity. In addition, it is suggested that cellular protective effects of kaempferol and (+)-${\alpha}$-tocopherol depend not only on the cell permeability, but also on free radical- and ROS-scavenging activities. These results indicate that the cell permeability and free radical- and ROS- scavenging activities of antioxidants are major factors affecting the protection of cell membranes against the oxidative damage induced by photosensitization reaction.
The purpose of this study was to evaluate the intake of antioxidant vitamins and plasma concentrations of those in 60 maternal-infant pairs (30 in normal term delivery group, NT; 30 in preform delivery group, PT). We also investigated the relationship between vitamin levels of maternal-umbilical cord plasma and pregnancy outcome. Mean energy intakes of NT and PT pregnant women were 93.2% and 85.4%, and their protein intakes were 113.3% and 110.9% of the recommended dietary allowance (RDA), respectively. While vitamin A intakes were only 51.2% and 39.6% of the RDA in NT and PT pregnant women. The vitamin E intake was about 50% of the RDA (NT 6.27 mg, PT 7.78 mg). The levels of retinol in maternal plasma of NT and PT were $1.51\mumol/\ell\;and\;1.43\mumol/\ell$, respectively. The retinol levels in umbilical cord plasma in NT and PT were $0.72\mumol/\ell\;and\;0.61\mumol/\ell$, respectively. The level of $\beta-carotene$in maternal plasma of NT was 0.49 $\mu$mol/$\ell$, significantly (p < 0.01) higher than that of PT ($0.31\mumol/\ell$).The $\beta-carotene$ of umbilical cord plasma of NT and PT were $0.702\mumol/\ell\;and\;0.01\mumol/\ell$, respectively. The plasma $\alpha$-tocopherol of maternal of NT and PT were $0.72\mumol/\ell\;and\;0.01\mumol/\ell\;29.51 /mumol/\ell\; and 27.17\mumol/\ell,\;respectively.\; The $\alpha$-tocopherol of umbilical cord plasma of NT and PT were $4.16\mumol/\ell\;and\;3.80\mumol/\ell$, respectively. The antioxidant vitamin levels (retinol, $\beta-carotene,\;and\;\alpha$-tocopherol) in maternal plasma were significantly higher (p<0.0001) than those in umbilical cord plasma. However, there was no correlation between the vitamin levels in maternal plasma and those in umbilical cord plasma. The maternal plasma $\beta$-carotene level showed a positive correlation to gestational age. Also Apgar score at 1 min produced a positive correlation to maternal plasma $\beta$-carotene level.
The characteristics of sesame oil containing one of natural antioxidant, ' $\gamma$-tocopherol', were studied with the supercritical $CO_2$ extraction. Although $\gamma$-tocopherol has a lower vitamin E value in biological systems than $\alpha$-tocopherol, it is a more potent antioxidant with in oils. For the research of various factors influence to the $\gamma$-tocopherol contents increment, we have checked roasting time and temperature, as well as pressure, temperature and flow rate of supercritical fluid. As a result, we found that the $\gamma$-tocopherol content was maintained constant under the condition of roasting temperature over $200^{\circ}C$. With the longer roasting time, $\gamma$-tocopherol content was increased. Except 250 bar, the $\gamma$-tocopherol content was maintained constant under the condition of the various pressure of supercritical fluid. But $\gamma$-tocopherol content was increased with lower flow rate of supercritical fluid from 1 $m{\ell}$/L to 3 $m{\ell}$/L. When the extraction performance with the supercritical fluid was compared to the conventional compressed extraction, $\gamma$-tocopherol content was increased up to 1.6 times.
The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.
The effects of smoking and physical exercise on the plasma concentrations of lipid-soluble antioxidants were investigated in 62 healthy males, aged 34-65 years. Current smokers (n=21) and ex-smokers(n=16) had significantly lower plasma levels of carotenoids ($\alpha$-carotene, $\beta$-carotene, cryptoxanthin and lycopene), $\alpha$-tocopherol and ${\gamma}$-tocopherol than non-smokers (n=25). Plasma concentrations of retionl and ubiquinone (coenzyme Q10) were lower among ex-smokers and current smokers than among non-smokers, but the differences were not statistically significant. Regular physical exercise was associated with increased plasma levels of lipid-soluble antioxidants. Plasma concentrations of crytoxanthin, retinol and ubiquinone were significantly elevated in the group engaging in moderate amounts of exercise (more than 20 minutes per day) compared to the group engaging in small amounts of exercise (less than 10 minutes per day). Plasma $\alpha$-carotene, $\beta$-carotene, lycopene levels in the subjects were affected more by smoking than by exercise. However, plasma levels of cryptoxanthin, retinol and ubiquinone in the subjects were affected more by exercise than by smoking. These findings suggest than smoking may cause a decrease in plasma lipid-soluble antioxidants during neutralization of reactive oxygen species present in cigarette smoke and that poor exercise habits may accelerate this imbalance of oxidant/antioxidant homeostasis in middle-aged Korean men.
