Skin Whitening Effect of Ethyl Acetate Fraction of Adenophora triphylla var. japonica Sprout (잔대(Adenophora triphylla var. japonica)순 아세트산에틸 분획물의 피부 미백 효과)

  • To investigate skin-whitening effect of Adenophora triphylla var. japonica sprout extract, antioxidant activity, inhibitory effect on tyrosinase and melanin synthesis in B16/F10 melanoma cell were examined. Total phenolic content (246.25 mg GAE/g) and total flavonoid content (303.94 mg RE/g) of ethyl acetate fraction from Adenophora triphylla sprout (EFAT) showed the highest contents than other fractions (n-hexane, chloroform and distilled water). Antioxidant activities of EFAT has been evaluated using ABTS, DPPH radical scavenging activities, FRAP and inhibitory effect of lipid peroxidation. EFAT showed excellent radical scavenging activity and inhibitory effect on MDA production. Inhibitory effect of tyrosinase as a major enzyme of melanin synthesis was also measured. In these results, EFAT showed higher inhibitory effect against L-DOPA (51.27%) than L-tyrosine. $IC_{50}$ value on ${\alpha}-glucosidase$ was $41.93{\mu}g/ml$. In B16/F10 melanoma cells, EFAT inhibited melanin synthesis at $200{\mu}g/ml$ concentration (about 42% decrease). Finally, main physiological compounds of EFAT were identified as a rutin and a chlorogenic acid using high performance liquid chromatography.

Biological activity of Euonymus alatus (Thunb.) Sieb. wing extracts (화살나무 날개 추출물의 생리활성)

  • Euonymus alatus (Thunb.) Sieb., also known as the arrow tree in Korea, is a plant in East Asia used in traditional medicine and food. In particular, the wings of E. alatus are rich in phenolic compounds. This study evaluated the antioxidant, α-glucosidase inhibition, and anti-cancer activities of E. alatus wing extracts. The radical and hydrogen peroxide scavenging acitvities and reducing the power of 1,000 ㎍/mL E. alatus wing extracts, were similar to those of the positive control (0.1% BHT, 0.1% α-tocopherol). In addition, ethanol and methanol extract at 250 ㎍/mL showed 95.70 and 94.99% of α-glucosidase inhibition activity, respectively. The ethanol extract of E. alatus wings had the highest total polyphenol and flavonoid contents (867.8 mg% and 521.7 mg%, respectively). The E. alatus wing extracts significantly decreased the cell viability of LNCaP human prostate cancer cells (p<0.001), MDA-MB-231 human breast cancer cells (p<0.001), and HT-29 human colon cancer cells (p<0.001) in a dose-dependent manner. However, there was no significant effect on B16 mouse melanoma cells. Notably, the ethanol extracts showed higher cancer cell growth inhibitory activity in LNCaP and HT-29 cells than the other extracts. These results suggest that E. alatus wing extracts could have significant clinical applications, and our results can be used as basic data for future functional food material development.

Antioxidant Effects and Physiological Activities of Pumpkin (Cucurbita moschata Duch.) Extract from Different Aerial Parts (늙은 호박 부위별 추출물의 항산화 효과 및 생리활성에 관한 연구)

  • This study investigated the antioxidant effects and physiological activities of extract from various parts of pumpkin (Cucurbita moschata Duch.) such as leaf, flesh, skin and seed. Different dried parts of C. moschata were boiled in ethanol for three hours, evaporated, and lyophilized with a freeze-dryer. Total polyphenol contents were the highest in leaf ($29.62{\pm}0.88mg$ GAE/g DM). The same result was observed in the measurement of the flavonoid content. The leaf extract of C. moschata had stronger DPPH radical-scavenging activities and ABTS-radical scavenging activities than any other part. Leaf extract of C. moschata was found to have the highest ferric-reducing antioxidant power (FRAP) value. Meanwhile, at 0.1 mg/mL of all extracts higher degree of tyrosinase inhibitory effects than kojic acid was observed (leaf; 49.58%, flesh; 40.20%, skin; 41.30%, seed; 36.79% and kojic acid; 20.70%). The glycation inhibitory activity decreased in the order leaf>skin>flesh>seed. The ${\alpha}$-glucosidase inhibitory activity of seed extract was the highest with a value of 63.58% at the concentration of 1 mg/mL.

Changes of Antioxidant Activities on Cultured Ginseng with Mushroom Mycelia During Cultivation (배양기간에 따른 버섯균사체 인삼배양물의 항산화활성)

  • This study was conducted to investigate the antioxidant activities of the cultured ginseng with mushroom mycelia (Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE)) during cultivation periods of 10, 20, 30, 40, and 50 days. The lyophilized powder from the cultured ginseng with mycelia was extracted with 80% ethanol, and then evaluated for antioxidant activities. Total phenolic contents ranged from 149.63 to 205.91 mg/g, and the highest value was 80% EtOH extract from the cultured ginseng with GL at 30 days. The highest antioxidant activity ($IC_{50}$) for DPPH was 1.16 mg/mL in the cultured ginseng with HE at 40 days, and total antioxidant activity for ABTS was the highest value of 4.03 mg AA eq/g in PL cultivation at 30 days. $\alpha$-Glucosidase inhibitory activity was the highest value of 92.51% in EtOH extract from the cultured ginseng with PL at 50 days, and tyrosinase inhibitory activity was highest value of 13.21% in GL cultivation at 40 days. These results suggest that mushroom mycelium cultivation period for enhancement of antioxidant activity might be 40 days.

Antioxidant and α-Glucosidase Inhibitory Activities of Tradescantia pallida (Rose) Hunt Leaf Extract and Fractions (팔리다자주닭개비 잎 추출물 및 분획물의 항산화 및 α-글루코시다아제 저해 활성)

  • Background: The biological activities of Tradescantia pallida grown in Korea have not been well determined, thus the aim of this study was to investigate the possibility of using it as a medicinal plant. Methods and Results: To investigate the antioxidant activity, ${\alpha}$-glucosidase inhibitory effect and antimicrobial activity of T. pallida, we performed the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity and reducing power assay. This assay for T. pallida leaf extract showed the highest antioxidant activity for the ethyl acetate fraction ($RC_{50}=14.55{\pm}0.16{\mu}g/m{\ell}$ and Abs = 0.613 at $300{\mu}g$). Further, the ethyl acetate fraction exhibited higher ${\alpha}$-glucosidase inhibitory effect with an $IC_{50}$ value of $14.1{\pm}0.1{\mu}g/m{\ell}$ and showed antimicrobial activity against gram positive bacteria (minimum inhibitory concentration = $1,000{\mu}g/m{\ell}$). Conclusions: The ethyl acetate fraction of the crude methanol extract of T. pallida showed remarkable antioxidant activity, ${\alpha}$-glucosidase inhibitory effects and antimicrobial activity. These activities might be related to the flavonoid content in the T. pallida leaf extract.

Characteristics and Physiological Functionalities of Unrecorded Yeasts from Wild Flowers of Seonyudo in Jeollabuk-do, Korea (전라북도 선유도일대 야생화에서 분리한 국내 미기록 효모들의 특성 및 생리기능성)

  • Six kinds of newly recorded yeasts such as Rhodosporidium diobovatum SY4-2, Cryptococcus bestiolae SY7-1, Kazachstania unispora SY14-1, Kazachstania servazzii SY14-3, Pichia holstii SY20-2 and Cryptococcus tephrensis SY26-1 were screened from sixty one yeasts derived from wild flowers found in Seonyudo, Gogunsanyeoldo, Jeollabuk-do, Korea. All of them grew in 50% glucose-containing yeast extract-peptone-dextrose (YPD) broth and Pichia holstii SY20-2 was also halophile, growing in 20% NaCl-containing YPD broth. All of them, except Cryptococcus tephrensis SY26-1, were assimilated to glucose. Cell-free extract from Kazachstania servazzii SY14-3 showed the highest 98.6% of ${\alpha}$-glucosidase inhibitory activity and maximal production of the ${\alpha}$-glucosidase inhibitor was obtained with 24h incubation at $30^{\circ}C$. The antihypertensive angiotensin I-converting enzyme inhibitory activity of the unrecorded yeasts were showed 58.6-80.4% in their supernatants.

Comparison of Biochemical Composition and Antimicrobial Activity of Southern-Type Garlic Grown in the Eastern and Western Regions of Jeju (제주 동부와 서부지역 남도마늘의 생화학적 성분 및 항균활성 비교)

  • Components and physiological activity of crops are strongly affected by the natural environments of the growth regions. In this study, we carried out soil analysis and determined the allicin, alliin, total pyruvate, total phenol and flavonoid, and reducing sugar contents of garlic from garlic-cultivating regions in the eastern and western portions of Jeju Province. Significantly higher contents of macronutrients were found in the soil and garlic from the eastern region of Jeju compared to western region, but micronutrients were higher in the western region than eastern region. In addition, the alliin and total phenolic contents were higher in the western region than in the eastern region. Also, allicin, total pyruvate, total flavonoid content, and ${\alpha}$-glucosidase inhibitory activity of the eastern region samples were higher than those of the western region in Jeju. This study promotes our understanding of the different components of garlic according to the cultivation regions of Jeju.

Antioxidant Activity and Inhibition Activity against α-Glucosidase and PTP1B of Chrysanthemum indicum L. Extract and Fractions (감국(Chrysanthemum indicum L.) 추출물과 분획물의 항산화 및 α-Glucosidase와 PTP1B 저해 활성)

  • In this study, we evaluated the polyphenol content, flavonoid content, antioxidant and biological of 70% EtOH extract and its fractions (dichloromethane, ethyl acetate and water) of the plant Chrysanthemum indicum L. (CI) belonging to family chrysanthemum. antioxidant activity was determined 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS), and superoxide scavenging activity. The biological activity tests included measurement of ${\alpha}$-glucosidase and PTP1B. The results show that content of polyphenol and flavonoid in 70% EtOH of CI 30.20 mg and 31.58 mg, respectively. Among the different samples, ethyl acetate (EA) fraction showed the highest level of polyphenol (87.87 mg) and flavonoid (65.12 mg). The DPPH, ABTS and superoxide dismutase(SOD) antioxidant assay of EA fraction showed 93.84%, 93.38% and 84.68% inhibition respectively. The EA fraction also showed the highest antioxidant activity compared to 70% EtOH extract and other fractions. EA fraction ($20{\mu}g/mL$) showed the highest 30.29% and 99.24%, respectively compared to other samples. The results of this study show that the EA fraction contained more antioxidant and antidiabetic components than 70% EtOH extract and other fractions. In conclusion, we believe that CI may be useful natural antioxidant and functional material.

ACE, α-Glucosidase and Cancer Cell Growth Inhibitory Activities of Extracts and Fractions from Marine Microalgae, Nannochloropsis oculata (해양 미세조류 Nannochloropsis oculata 추출.분획물의 ACE, α-glucosidase 및 암세포 저해 활성)

  • Extracts of the marine microalgae Nannochloropsis oculata were obtained using 80% methanol (MeOH) and water. The 80% MeOH extract was further fractionated with n-hexane, chloroform, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water to isolate the active fraction. Seven samples were prepared and their angiotensin converting enzyme (ACE), $\alpha$-glucosidase, and cancer cell growth inhibitory activities in vitro were determined. The most profound ACE inhibitory activity was observed in the chloroform fraction, while the others had moderate effects. By contrast, greater $\alpha$-glucosidase inhibitory activity was found in the EtOAc fraction, n-hexane fraction, and water extract of N. oculata. The antiproliferative effects of the extracts and fractions against HL-60, U937, CT-26, and SK-Hep1 cancer cells were also determined. The n-BuOH fraction had the strongest antiproliferative effects on CT-26 cells in a time-dependant manner (P<0.05). These results suggest that the extracts and fractions from N. oculata could be used as a potential functional food or as pharmaceutical ingredients.

Antioxidant abilities and physiological properties of dried Haw extracts prepared using different drying methods (건조 방법에 따른 산사과 추출물의 항산화 및 생리활성)

  • This study was performed to determine the effects of drying methods (HD, hot air drying; FD, freeze-drying) on the antioxidant and physiological abilities of Haw (Crataegus pinnatifida Bunge). Powder color values of dried Haw, L and b, were higher for HD, while the chroma values were higher for FD. The total polyphenol, and flavonoid contents of HD Haw and FD Haw were found to be $9.29{\pm}0.50mg\;GAE/mL$ and $15.48{\pm}0.38mg\;GAE/mL$, and $9.41{\pm}0.26mg\;RHE/mL$ and $26.46{\pm}0.34mg\;RHE/mL$, respectively. DPPH radical scavenging abilities at 100 mg/mL concentration were higher for FD (64.90%) than HD (28.66%), as were the reducing power, ABTS radical scavenging ability, and nitrite scavenging ability. However, the HMG-COA reductase of HD (74.67%) was higher than that of FD (72.10%). The ${\alpha}-glucosidase$ inhibitory abilities of HD and FD at a concentration of 0.1 mg/mL were 24.69% and 97.38%, respectively. These results indicate that Haw is a potential functional material and that freeze-drying Haw is better than hot air drying.
