식물성 식품에 존재하는 Flavonoids의 항산화 활성 (Antioxidant Activity of Flavonoids in Plant Origin Food)

  • Effective synthetic antioxidants such as butylated hydroxyanisole(BHA) and butylated hydroxytoluene(BHT) have been widely used in the food industry, but they are suspected to be toxic and carcinogenic effects. Therefore, the development of safely available natural antioxidants such as ascorbic acid, ${\alpha}$-tocopherol, ${\beta}$-carotene, flavonoids and selenium is essential. In particular, flavonoids, 2-phenyl-benzo-${\alpha}$-pyrones, are polyphenolic compounds that occur ubiquitously in food of plant origin. flavonoids occur in foods generally as O-glycosides with sugars bound usually at the C\ulcorner position. And variations in their heterocyclic ring gibes rise to flavones, flavonols, flavanones, flavanols, catechins, anthocyanidins, chalcone and isoflavones. Vegetables, fruits, and beverages are the main dietary sources of the flavonols, primarily as quercetin, kaempferol, and myricetin and the corresponding flavones, apigenin and luteolin. These flavonoids have biological activity such as antioxidant, anti-inflammatory, antithrombotic, antimutagenic, anticarcimogenic antiallergic and antimicrobial activity effects in vitro and in vivo. Flavonoids posses strong antioxidant activities acting as oxygen radicals scavenger, metal chelators and enzyme inhibitor. The antioxidant activity of flavonoids is determined by their molecular structure and more specially, by the position and degree of hydroxylation of the ring structure. All flavonoids with the 3`, 4`-dihydroxy(ortho-dihydroxy) posses marked antioxidant activity. And antioxidant activity increases with the number of hydroxyl groups substituted on the A-and B-rings. There is as yet no certainty about the effect of the presence of a double bond between C\ulcorner and C\ulcorner on the antioxidant activity of flavonoids.

Type II Isopentenyl Diphosphate Isomerase로서 Synechocystis sp. PCC6803의 sll1556의 작용 특성 (Functional Characterization of sll1556 of Synechocystis sp. PCC6803 as Type II Isopentenyl Diphosphate Isomerase)

  • Synechocystis sp. PCC6803의 type II Isopentenyl diphosphate isomerase gene(sll1556, Syidi2)의 특성을 살펴보기 위하여 ${\Delta}idi$인 E. coli $DH5{\alpha}$를 제작하고, 이 균주에서 cloning하고 발현시켰다. 라이코펜 합성 유전자들(crtE, crtB, and crtI)과 mevalonate pathway 유전자들(MvK1, MvK2, Mvd)를 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주를 mevalonate가 함유된 LB 배지에서 배양하면 mevalonate pathway 유전자들을 함유한 E. coli $DH5{\alpha}$균주는 mevalonate에 의해 생성된 isopentenyl diphosphate의 독성에 의해 매우 느린 성장을 보였다. 라이코펜 합성유전자들과 mevalonate 합성유전자들을 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주에 Syidi2를 도입한 결과, mevalonate가 함유된 LB배지에서 균체의 성장이 완전히 회복되었으며, 라이코펜이 합성되었음을 나타내는 붉은 균락이 형성되었다. 이에 따라, SyIdi1과 ECidi를 도입하여 비교한 결과, 라이코펜 합성 유전자들과 mevalonate pathway 유전자들을 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주 자체와 SyIdi1을 도입한 라이코펜 합성 유전자들과 mevalonate pathway 유전자들을 함유한 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주는 IPP의 독성에 의해 성장이 매우 느렸으나, SyIdi2, RSidi, HPidi, 및 ECidi를 함유하고 있는 ${\Delta}idi$ E. coli $DH5{\alpha}$ 균주는 균체의 성장과 라이코펜의 합성을 완전히 회복하였으며, 그중 가장 우수한 라이코펜 생합성 결과를 나타낸 것은 pSUP-LYCSyIdi2로서 균체당 라이코펜 생성량이 control 대비 2.8배이었다.

우리나라 성인 여성의 항산화비타민 섭취량과 비만의 연관성 : 2007 ~ 2016년 국민 건강영양조사 자료를 이용하여 (Association between antioxidant vitamin intake and obesity among Korean women: using the Korea National Health and Nutrition Examination Survey 2007 ~ 2016)

  • 본 연구에서는 2007 ~ 2016년 국민건강영양조사의 검진 조사와 식품섭취조사를 완료한 만 19세 이상의 성인 여성 30,425명을 대상으로 항산화비타민 섭취량과 비만 및 복부비만의 연관성을 분석하였다. 체질량지수가 $25kg/m^2$ 이상이거나 허리둘레가 85 cm 이상인 대상자를 각각 비만과 복부비만으로 판정하였고, 개인별 일일 항산화비타민 섭취량은 24시간 회상법을 이용한 식사자료와 한국인 상용식품의 항산화비타민 함량 데이터베이스를 연계하여 추정하였다. 항산화비타민에는 카로티노이드 6종, 레티놀, 비타민 A (레티놀 활성 당량), 비타민 C, 토코페롤 4종, 비타민 E (${\alpha}$-토코페롤 당량)이 포함되었다. 정상군과 비만군의 열량 1,000 kcal당 일일 항산화비타민 섭취량의 평균을 비교한 결과, 비만군은 ${\alpha}$-카로틴, 레티놀, 비타민 E, ${\alpha}$-토코페롤, ${\gamma}$-토코페롤을 정상군보다 적게 섭취한 것으로 나타났다. 항산화비타민 섭취수준에 따라 대상자를 삼분위로 구분했을 때, 라이코펜을 가장 많이 섭취하는 군은 가장 적게 섭취하는 군에 비해 비만 (OR = 0.89, 95% CI: 0.83-0.96)과 복부비만 (OR = 0.88, 95% CI: 0.81-0.95) 유병률이 유의하게 낮았다. 총 카로티노이드, ${\alpha}$-카로틴, 비타민 A, ${\gamma}$-토코페롤 섭취량 또한 복부비만의 오즈비와 음의 연관성을 나타냈다. 식품군별 항산화비타민 섭취량을 비교했을 때, 비만군은 정상군에 비해 난류, 우유류, 조미료류, 곡류를 통한 항산화비타민 섭취량이 적었다. 추후 전향적 설계를 이용한 후속연구를 수행하여 항산화비타민 섭취량과 비만 간의 인과성을 확인할 필요가 있을 것으로 사료된다.

한국인 다소비 섭취 식품의 방사선 조사후 항산화 영양소와 비타민 $B_1$의 변화 (Changes in Antioxident Nutrients and Vitamin $B_1$ Contents of Gamma Irradiated Foods Consumed most Frequently in Korea)

  • Food irradiation has been steadily increased in many countries concomitantly with increasing international trades and concerns about naturally occurring harmful contaminants of food. Although, irradiation provides an excellent safeguard for the consumer by destroying harmful bacteria, it would be necessary to investigate nutritional adequacy of irradiated foods with a special emphasis on such foods as red pepper powders, cold-water fishes (mackerel), sesame seeds that are widely consumed in Korea. These food items are of special concern due to the fact that most of them are imported and that several nutrients contained in them are sensitively reacting to irradiation. We observed changes in the antioxidant nutrients contents, such as vitamin A, $\beta$-carotene, vitamin C and total phenol, and also vitamin B$_1$ in the foods that were irradiated at different dose levels. The nutrients contents in red pepper were respectively 98.6% and 92.3% for total phenol, and 97.8% and 94.5% for $\beta$-carotene, right after irradiation at the dose of 10 and 20 kGy. The vitamin C contents decreased with irradiation doses up to 76.4% (10 kGy) and 68.5% (20 kGy) and B$_1$ contents decreased to 54.9% and 80.7% of non irradiated red pepper. When the mackerel was irradiated at the applied dose of 3 and 7 kGy, total phenol concentration of the irradiated fish was respectively 91.3% and 94.2% of non irradiated fish. The amounts of vitamin A were 82.6% (3 kGy) and 78.0% (7 kGy) of unirradiated sample and vitamin B$_1$ contents reduced to 30.5% and 51.5%. $\alpha$-Tocopherol content was 33.9% of the non-irradiated one when it was irradiated at 7 kGy. Vitamin A and ${\gamma}$-tocopherol contents in sesame seeds linearly decreased with the applied doses (5 and 10 kGy) Vitamin A contents decreased up to 87.8% (5 kGy) and 82.9% (10 kGy) and that of ${\gamma}$-tocopherol were 72.4% and 66.5% of the unirradiated sample.

산불 관련 해양환경오염원들에 대한 해조류의 활력 측정 (Viability Assay of Seaweeds Responding to Mountain Fire-Related Pollutants)

  • Plant ash and soil drainage, derived by frequent mountain fires during winter, might cause biological contamination to seaweeds at seashore and river mouse area. To thalli of Ulva pertusa, maximum non-lethal concentration(MNLC), lethal concentration 50 $(LC_{50})$ and minimum lethal concentration (MLC) of pine needle ash were shown as 60, 350 and 550 mg/mL, respectively. The yellow loess and granite sand did not damage at concentrations of 20 and 200 mg/mL, respectively To thalli of Porphyra yezoensis, the MNLC, LC5O, MLC of pine needle ash were shown as 0.08, 0.4 and 1.0 mg/mL, respectively. Effects of yellow loess and granite sand were approximately 1/2 and 1/10 of the ash. To thalli of Undaria pinnatifida, the pine needle ash, yellow loess and granite sand did not damage at the concentration range of 20 to 40 mg/mL. Change of pigments $(chlorophyll\;\alpha,\;lutein,\;\beta-carotene,\;phycoerthrin)$ was also determined at the MNLC, $LC-{50}$ and MLC of pine needle ash. Among three seaweeds tested, P. yezoensis produced the most 2.7-fold of lutein and 2.3-fold of $\beta-carotene$ at $LC-{50}$ of the ash. Thus the P. yezoensis, appeared as a sensitive indicator, could be used as one of test organisms for determination of the biological effect of pollutants contaminated in marine environment.

Phaffia rhodozyma 세포파쇄액으로부터 항산화제 Astaxanthin의 미셀 형성을 통한 가용화 및 추출 (Solubillzation and Extraction Of Antioxidant Astaxanthin by Micelle Formation from Phaffia rhodozyma Cell Homogenate)

  • Astaxanthin (3,3'-dihydroxy-${\beta}$, ${\beta}$-carotene-4-4'-dione), a natural pigment of pink to red color, is widely distributed in nature particularly in the skin layer of salmonoids and the crust of shrimp, lobster, etc. Recently, it was produced from the yeast culture of Phaffia rhodozyma. Because of its high thermal stability and antioxidant functionality, its applications can be extended into food, cosmetics, and pharmaceutical ingredient beyond the traditional feed additive. Because of its very high lipophilicity, astaxanthin has been extracted traditionally by strong organic solvents such as chloroform, petroleum ether, acetone, etc. In this study, we developed a surfactant-based solubillization system for astaxanthin, and used it to extract astaxanthin from disrupted yeast cells. Among Tween 20, Triton X-100 and SDS, Tween 20 was identified as the most suitable surfactant in terms of extraction capacity and safety. The ethylene oxide group of Tween 20 was identified as the most significant factor to increase the HLB value that determined the extraction capacity. The effects of micelle formation condition, such as the molar ratio of astaxanthin and Tween 20, pH, and ionic strength were also investigated. pH and ionic strength showed no significant effects. The optimal molar ratio between astaxanthin and Tween 20 was 1 : 12. Antioxidant activity of astaxanthin was higher than ${\beta}$-carotene and ${\alpha}$-tocopherol. Astaxanthin in the crude extract from the yeast cell was more resistant to air and/or light degradation than pure astaxanthin, probably because of the presence of other carotenoids and lipids.

들깨 품질평가 현황과 전망 (Current Status and Prospects of Quality Evaluation in Perilla)

  • 1. 들깨는 용도면에서 볼 때 종실은 식용으로, 기름은 식용과 공업용, 깻잎은 채소로 또 깻잎의 정유성분은 향료로 이용될 수 있는 다용도 작물이다. 2. 종실의 단백질 함량은 18-28% 정도이고 단백질의 아미노산조성에 있어 글루타메이트(glutamate)함량은 계란보다 월등히 많다. 3. 종실의 기름 함량은 35~54%로서 기름중에는 리놀렌산(linolenic acid)이 60% 이상 함유되어 있으므로 생리적 기능성 식품으로 우수한 특성을 갖고 있으며 필수지방산의 공급원으로서도 중요하다. 4. 기름의 특성상 들기름은 고도의 불포화 지방산인 리놀렌산이 주성분이기 때문에 들기름을 짜서 상온에서 보관하게 되면 산패(oxidation)되기 쉬우므로 장기간 저장 이용하기 어렵다. 5. 깻잎은 비타민 C와 베타카로틴($\beta$-carotene) 함량이 많고 다른 미량성분도 풍부하게 함유되어 있으므로 들깨잎은 생식용 채소로서의 이용가치가 높다. 6. 깻잎의 정유(essential oil) 성분은 페릴라알데하이드(perilla aldehyde)가 주성분이며 이는 식품이나 식품첨가제로 쓰일 때 광범위한 항균작용(antimicrobial activity)을 나타낸다. 7 들깨나 자소잎의 붉은 색소중에는 안토시아닌(anthocynins)이 다량 함유되어 있으며 이는 식품색소로 널리 쓰인다.

한국산 동백종자 초임계 추출물의 피부 항산화 및 항노화 연구 (Anti-oxidant and Anti-aging Effect of Extracts from Seed of Camellia japonica L. by Supercritical Fluid Extraction)

  • Objectives : The aim of this study was to investigate the anti-oxidant and anti-aging effect of the seed of Camellia japonica L. extracted by supercritical CO2. Methods : The cell viability was performed by MTT assay. Nitric oxide (NO) production was performed by NO assay. The anti-oxidant effect was performed by beta-carotene bleaching assay and the intracellular proteome was analyzed expression of each 15 proteins by 2-D electrophoresis. Results : Fatty acid analysis of extract from Camellia japonica L. has shown oleic acid was 84%. And the antioxidant effect was about 186% compared with alpha-tocoperol(0.1%) by beta carotene bleaching assay. In 2D PAGE analysis, fifteen protein changes in five mechanisms which was collagen synthesis pathway, MMPs, ECM-cell interaction, cytokine, antioxidant enzymes were analyzed. Conclusions : On this study extract from seed of Camellia japonica L. by supercritical extraction could be used as a antioxidant and antiaging substance for the skin.

Changes in Free Amino Acid, Carotenoid, and Proline Content in Chinese Cabbage (Brassica rapa subsp. Pekinensis) in Response to Drought Stress

  • Chinese cabbage grown during autumn season is confronted with drought conditions for a certain period, especially during the early growth stage. In this study, we investigated the effects of drought stress on plant growth characteristics, as well as free amino acid, carotenoid, and proline in Chinese cabbage. Chinese cabbage seeds (Bulam Plus) were germinated, and all the seedlings were transplanted into plastic containers (28 cm diameter ${\times}$ 22 cm high) containing a commercial growth medium. The soil water content was measured and maintained at 10% for the drought-stressed plants and at 30% for the control plants, for three weeks. The results revealed that plant growth parameters were lower in the drought-stressed plants than in the control plants. The total free amino acid content tended to decrease in both drought-stressed and control plants with time. The total free amino acid content was found to be lower in the drought-stressed plants than in the control plants and the proline content was unaffected. Moreover, at three weeks after treatment, carotenoid content in drought stressed plants was significantly higher than that in the untreated plants. We believe that our study makes a significant contribution to the literature because the effects of drought stress on plant growth parameters, free amino acid, carotenoids, and proline accumulation in autumn growing cultivar of Chinese cabbage have not been widely studied in Korea, and our study provides valuable information in this regard, as Chinese cabbage is consumed throughout the year in Korea.

Biological effects of ginseng petroleum extract on hydrogen peroxide induced lipid peroxidation and oxidative DNA damage

  • Reactive oxygen species(ROS)에 의한 세포독성이나 DNA손상은 노화와 암에 밀접한 관련이 있다. 본연구에서는 ROS중 hydroxyl radical 생성에 관여되는 $H_2O$$_2$에 의해 유도되는 산화적 세포독성이나 DNA손상에 억제적으로 작용할 수 있는 천연물을 창출하기 위한 연구를 하였다. 인삼(Panax Ginseng)C.A. Meyer의 석유에텔의 추출물(GPE)과 일부분획성분(P2)에 대하여 in vitro에서 지질과산화억제효과 및 프리라디칼소거효과를 시험하고 CHL Cell에서의 $H_2O$$_2$ 유도 세포독성과 산화적 DNA손상에 미치는 영향을 연구하였다. 한편 이들 물질을 기존의 항산화제인 ascorbic acid, dl-$\alpha$-tocopheorl 및 $\beta$-carotene등과 비교하였다.
