Cloning and Characterization of ${\alpha}-Glucosidase$ Gene from Thermophilic Bacillus sp. DG0303

  • An ${\alpha}-glucosidase$ gene (aglA) from thermophilic Bacillus sp. DG0303 was cloned, sequenced, and expressed in Escherichia coli. The aglA was localized to the 2.1-kb PvuI-XmnI region within the 5.9-kb DNA insert of the gybrid plasmid pAG1. The gene consisted of an open reading frame of 1,686 bp with an unusual GTG initiation codon and TGA termination codon. The amino acid sequence deduced from the nucleotide sequence predicted a protein of 562 amino acid residues with a M, of 66,551 dalton. A comparative amino acid sequence analysis revealed that DG0303 ${\alpha}-glucosidase$ is related to bacillary oligo-1, 6-glucosidases. The Bacillus sp. DG0303 ${\alpha}-glucosidase$ showed a high sequence identity (36-59%) to the B. flavocaldarius, B. cereus, and B. thermoglucosidasius oligo-1, 6-glucosidases. The number of prolines in theses four ${\alpha}-glucosidases. was observed to increase with increasing thermostability of these enzymes. The cloned ${\alpha}-glucosidase was purified from E. coli $DH5{\alpha}$ bearing pAG1 and characterized. The recombinant enzyme was identical with the native enzyme in its optimum pH and in its molecular mass, estimated by sodium dodecy1 sulfate-polyacrylamide gel electrophoresis. The temperature optimum of the cloned ${\alpha}-glucosidase$ was lower than that of the native enzyme.

Polyopes lancifolia Extract, a Potent α-Glucosidase Inhibitor, Alleviates Postprandial Hyperglycemia in Diabetic Mice

  • This study was designed to investigate the inhibitory effects of Polyopes lancifolia extract (PLE) on ${\alpha}$-glucosidase activity, ${\alpha}$-amylase activitiy, and postprandial hyperglycemia in streptozotocin (STZ)-induced diabetic mice. The results of this study revealed a marked inhibitory effect of PLE on ${\alpha}$-glucosidase and ${\alpha}$-amylase activities. The $IC_{50}s$ of PLE against ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.20 mg/mL and 0.35 mg/mL, respectively. PLE was a more effective inhibitor of ${\alpha}$-glucosidase and ${\alpha}$-amylase activities than acarbose, the positive control. The postprandial blood glucose levels of STZ-induced diabetic mice were significantly lower in the PLE treated group than in the control group. Moreover, PLE administration was associated with a decreased area under the curve for the glucose response in diabetic mice. These results indicate that PLE may be a potent inhibitor of ${\alpha}$-glucosidase and ${\alpha}$-amylase activities and may suppress postprandial hyperglycemia.

자생 양치식물 9종의 성엽 및 근경 추출물의 α-glucosidase 억제 활성 (Alpha-glucosidase Inhibition Activity of Methanol Extracts Obtained from Nine Pteridophyte Species Native to Korea)

  • 본 연구는 자생 양치식물 9종의 성엽과 근경을 재료로 ${\alpha}$-glucosidase 억제활성을 분석하여 천연 ${\alpha}$-glucosidase 저해제로서 개발 가능한 식물 소재를 선발하기 위하여 수행하였다. 수확된 성엽과 근경은 수세 후 동결건조 하였으며, 건조시료를 분쇄하여 100% 메탄올 용매로 30분 동안 초음파 추출을 하였고, 추출 후 감압여과 하여 ${\alpha}$-glucosidase 억제활성을 측정하였다. 양성 대조구로는 acarbose를 사용하였다. 추출물 $50{\mu}L$에 0.7 unit ${\alpha}$-glucosidase 효소액 $100{\mu}L$를 넣고 혼합하여 $37^{\circ}C$에서 10분간 반응시킨 후, 1.5 mM ${\rho}$-NPG 기질용액을 $50{\mu}L$ 넣고 $37^{\circ}C$에서 20분간 반응시켰다. 1 M $Na_2CO_3$ 1 mL로 반응을 정지시키고 405 nm에서 흡광도를 측정하였다. 회귀분석을 이용하여 0.7 unit ${\alpha}$-glucosidase 용액의 활성을 50%를 억제하는데 필요한 시료의 농도($IC_{50}$ 값)를 구하였다. 양치식물 9종의 ${\alpha}$-glucosidase 억제활성은 성엽($IC_{50}=14.00{\sim}913.33{\mu}g{\cdot}mL^{-1}$)과 근경 추출물($IC_{50}=12.93{\sim}205.84{\mu}g{\cdot}mL^{-1}$)에서 공히 acarbose($IC_{50}=1413.70{\mu}g{\cdot}mL^{-1}$)에 비해 높았다. 양치식물의 추출물은 acarbose에 비해 성엽은 1.55~100.98배, 근경은 6.87~109.33배 높은 것으로 나타났다. 특히 성엽에서는 석위의 억제활성이, 근경에서는 꿩고비의 ${\alpha}$-glucosidase 억제활성이 가장 높았다. ${\alpha}$-Glucosidase 활성의 50%를 억제하기 위한 성엽과 근경의 필요 생체량은 공히 꿩고비(각 0.35, 0.27 mg)에서 가장 적은 것으로 나타났다. 성엽의 경우는 석위의 $IC_{50}$ 값이 가장 높았으나 가용성 고형분의 함량이 꿩고비에 비해 2.4배 낮아, 오히려 꿩고비의 경제성이 더 높은 것을 알 수 있었다. 본 연구의 결과 꿩고비는 적은 생체량으로도 높은 ${\alpha}$-glucosidase 억제활성을 나타내기 때문에 경제적인 천연 ${\alpha}$-glucosidase 저해제 소재로써 개발 가치가 매우 높은 것을 알 수 있었다.

갈근에서 분리한 화합물의 $\alpha$-glucosidase와 $\alpha$-amylase 활성 저해 효과 ($\alpha$-Glucosidase and $\alpha$-Amylase Inhibitory Activity of Compounds from Roots Extract of Pueraria thunbergiana)

  • Compounds of isolated from roots extract of Pueraria thunbergiana were tested their inhibitory effects on $\alpha$-glucosidase and $\alpha$-amylase. Inhibitory activity of methylene chloride (MC) fraction and ethyl acetate (EA) fraction against $\alpha$-glucosidase showed more than 60% at a concentration of $500{\mu}g/m{\ell}$. Among the nine compounds tested on $\alpha$-glucosidase, biochanin A, (-)-tuberosin and calycosin from MC fraction and daidzein from EA fraction were stronger inhibitors than acarbose ($IC_{50}=530{\mu}g/m{\ell}$), and their $IC_{50}$ were 9, 144, 328 and $20{\mu}g/m{\ell}$, respectively. Biochanin A and (-)-tuberosin also inhibited $\alpha$-amylase activity as like as acarbose $IC_{50}=20.5{\mu}g/m{\ell}$), and their $IC_{50}$ were 22 and $348{\mu}g/m{\ell}$, respectively. Although daidzein was already known $\alpha$-glucosidase inhibitory effects, it was newly evaluated that biochanin A and (-)-tuberosin inhibited $\alpha$-glucosidase as well as $\alpha$-amylase, and that calycosin did $\alpha$-glucosidase.

Inhibitory Effect of Buthus martensi Karsch Extracts on ${\alpha}$-Glucosidase Enzyme

  • While searching for ${\alpha}$-glucosidase inhibitors, the active compound was found in a methanol extract of Burthus martensi Kirsch. The separation of the active compound was performed using various chromatography methods and the physico-chemical properties of the purified compound were characterized. The compound showed very potent inhibitory activity against ${\alpha}-glucosidase$ with an $IC_{50}$ value of $5.3\;{\mu}g/ml$. Lineweaver-Burk plot indicated that its inhibition of ${\alpha}-glucosidase$ was competitive.

Expression of Thermostable $\alpha$-Glucosidase from Thermus caldophilus GK24 in Recombinant Saccharomyces cerevisiae

  • A gene (GenBank AF096282) coding for a $\alpha$-glucosidase (TcaAG, EC 3.2.1.20) from Thermus caldophilus GK24 was expressed in Saccharomyces cerevisiae, a generally recognized as safe (GRAS) host. The thermostable $\alpha$-glucosidase was produced inside of the GRAS host at 0.04 unit/mg-dry cell by the constitutively expressing ADH1 promoter and at 1.2 unit/mg-dry cell by the inductively expressing GALl0 promoter, respectively. No $\alpha$-glucosidase activities were found in the medium when the MF-alpha signal sequence from S. cerevisiae or $\alpha$-amylase signal sequence from Aspergillus oryzae were fused before the $\alpha$-glucosidase gene for the secretion.

효모 Pichia burtonii Y257-7에 의한 ${\alpha}$-Glucosidase 저해제의 생산 및 식후 혈당 상승 억제 효과 (Production and Anti-hyperglycemic Effects of ${\alpha}$-Glucosidase Inhibitor from Yeast, Pichia burtonii Y257-7)

  • 본 연구에서는 우리나라 전통발효식품에서 분리한 효모들을 이용하여 새로운 항당뇨성 ${\alpha}$-glucosidase 저해물질을 개발하기 위하여, 먼저 48종의 효모들의 ${\alpha}$-glucosidase 저해활성을 측정한 결과, Pichia burtonii Y257-7의 무세포 추출물이 가장 높은 저해활성을 보여 시험균주로 최종 선발하였다. ${\alpha}$-glucosidase 저해제 최적 생산 조건을 검토한 결과, Pichia burtonii Y257-7를 $28^{\circ}C$에서 24시간 LB 배지(pH 6.0)에 배양하여 얻은 무세포 추출물에서 가장 높은 ${\alpha}$-glucosidase 저해활성을 보였다. 최적 생산조건에서 얻은 ${\alpha}$-glucosidase 저해물질을 $0.45{\mu}m$$0.2{\mu}m$ syringe filter unit로 여과한 후 Sephadex G-50 column chromatography와 연속 용매 추출을 실시하여 부분정제한 후 일반 쥐와 당뇨유발 쥐를 이용하여 항당뇨 효과를 조사하였다. 정상 쥐와 streptozotocin으로 당뇨를 유발시킨 당뇨유발 쥐에 Pichia burtonii Y257-7이 생산하는 ${\alpha}$-glucosidase 저해제의 부분정제물을 전분과 함께 500 mg/kg, 1000 mg/kg 농도로 경구 투여한 결과, 시판 혈당강하제인 acarbose보다는 효과가 낮았지만 농도 의존적으로 정상 쥐와 당뇨유발 쥐 모두에서 혈당 상승 억제 효과를 확인할 수 있었다.

  • 버섯을 이용하여 새로운 혈당상승 억제 물질을 개발하여 건강 기능성 식품 소재 산업에 응용하고자 느타리버섯, 노랑느타리버섯, 분홍느타리버섯, 새송이버섯과 잣버섯들의 ${\alpha}$-glucosidase 저해활성을 측정한 결과 잣버섯 자실체의 에탄올 추출물이 가장 높은 ${\alpha}$-glucosidase 저해활성을 보였다. 잣버섯 자실체 분말을 95% 에탄올로 $30^{\circ}C$에서 48시간 추출하였을 때 ${\alpha}$-glucosidase 저해물질이 가장 많이 추출되었고 이 ${\alpha}$-glucosidase 저해물질을 함유한 잣버섯 에탄올 추출물은 일반 쥐와 streptozotocin으로 당뇨를 유발시킨 당뇨 쥐에서 모두 농도 의존적으로 식후 혈당상승 억제 효과를 보였다.

Sargassum yezoense Extract Inhibits Carbohydrate Digestive Enzymes In Vitro and Alleviates Postprandial Hyperglycemia in Diabetic Mice.

  • In this study, we investigated whether Sargassum yezoense extract (SYE) could inhibit ${\alpha}-glucosidase$ and ${\alpha}-amylase$ activities, and alleviate postprandial hyperglycemia in streptozotocin (STZ)-induced diabetic mice. Freeze-dried S. yezoense was extracted with 80% ethanol and concentrated for use in this study. The hypoglycemic effect was determined by evaluating the inhibitory activities of SYE against ${\alpha}-glucosidase$ and ${\alpha}-amylase$ as well as its ability to decrease postprandial blood glucose levels. The half-maximal inhibitory concentrations of SYE against ${\alpha}-glucosidase$ and ${\alpha}-amylase$ were $0.078{\pm}0.004$ and $0.212{\pm}0.064mg/mL$, respectively. SYE was a more effective inhibitor of ${\alpha}-glucosidase$ and ${\alpha}-amylase$ activities than the positive control, acarbose. The increase in postprandial blood glucose levels was significantly alleviated in the SYE group compared with that in the control group of STZ-induced diabetic mice. Furthermore, the area under the curves significantly decreased with SYE administration in STZ-induced diabetic mice. These results suggest that SYE is a potent inhibitor of ${\alpha}-glucosidase$ and ${\alpha}-amylase$ activities and alleviates postprandial hyperglycemia caused by dietary carbohydrates.

Bacillus lentimorbus B-6 균주로부터 생산된 $\alpha$-Glucosidase 억제제의 생물 유화제로서의 특성 (Emulsifying Character of $\alpha$-Glucosidase Inhibitor Produced from Bacillus lentimorbus B-6)

  • Bioemulsifiers are those chemicals which are produced from microorganisms but which have both hydrophilic and hydrophobic groups. $\alpha$-Glucosidase inhibitor ($\alpha$-GI) produced from Bacillus lentimorbus B-6 (B-6) showed bioemulsifying activity. But $\beta$-glucosidase inhibitor produced from B-6 didn't show emulsifying activity. $\alpha$-GI was purified from supernatant of B-6 grown in minimal culture medium containing glucose and sodium glutamate by Sephadex G-100 column chromatography and isolated from $\beta$-GI by dialysis against water. Toluene was determined as the best substrate for emulsifying activity of $\alpha$-GI. $\alpha$-GI showed thermostability at $100^{\circ}C$ for 15 min, high salt tolerance up to 32% NaCl and wide range of pH-stability at pH $4\sim10$. Emulsifying character of $\alpha$-GI can be useful for the liposome formation for the treatment of diabetes mellitus.
