  • This experiment was designed to evaluate effects of Dandelion (Taraxzcum coreanum) and Dandelion fermented probiotics medium on the growth performance and meat quality in broiler chicks. A total of 150 "Ross" broilers,1-day old, were assigned to 5 treatments in a completely randomized design. There were 5 replications per treatment and 6 broilers in each replication for 5 weeks. The dietary treatments included a control (Dandelion and Dandelion fermentation was not added), antibiotic (0.05 chlortetracycline was added), 1.0% Dandelion supplementation and 0.5 and 1.0% Dandelion fermented probiotics, respectively. There was no significant difference in final body weight, weight gain, feed intake, and feed conversion ratio of broilers fed diets containing antibiotics, 1.0% Dandelion supplementation and 0.5 and 1.0% Dandelion fermented probiotics. The total cholesterol, HDL and LDL in plasma and meat cholesterol content was not affected by 0.5 and 1.0% Dandelion fermented probiotics and 1.0% Dandelion supplementation (P>0.05). The oleic acid content was significantly increased in 0.5% Dandelion fermented probiotics compared to that of the control (P<0.05). As a result, Dandelion and Dandelion fermented probiotics result in the influence on decreasing cholesterol in blood, particularly when adding probiotics, oleic acid that is the important factor in taste testing of meat increases thus the quality and taste of the chicken meat could be improved with the effect.

  • In the current study, the mesenchymal stem cells (MSCs) isolated and propagated from the human umbilical cord blood (UCB) were tested for their capabilities of differentiation into chondrocytes in vitro. The mesenchymal progenitor cells (MPCs) collected from UCB were cultured in a low glucose DMEM medium with 10% FBS, L-glutamine and antibiotics. The human MSC colonies were positively stained by PAS reaction. When the immunophenotypes of surface antigens on the MSCs were analyzed by fluorescence-activated cell sorter (FACS) analysis, these cells expressed positively MSC-related antigens of CD 29, CD44, CD 90 and CD105, whereas they did not express antigens of CD14, CD31, CD34, CD45, CD133 and HLA-DR. Following induction these MSCs into chondrocytes in the chondrogenic differentiation medium for 3 weeks or more, the cells were stained positively with safranin O. We clearly confirmed that human MSCs were successfully differentiated into chondrocytes by RT-PCR and immunofluorescent stain of type-II collagen protein. These data also indicate that the isolation, proliferation and differentiation of the hUCB-derived MSCs in vitro can be used for elucidating the mechanisms involved in chondrogenesis. Moreover this differentiation technique can be applied to developing cell-based tissue regeneration or repair damaged tissues.

  • This study was carried out to optimize the fertigation method using fermented swine liquid manure for the growth of two western vegetables, broccoli and celery. Plants were grown in a rain-shelter house and fertilized with a range of dilutions(efflux 5 dilution=Ef. 5, efflux 10 dilution=Ef. 10, efflux 25 dilution=Ef. 25, and efflux 50 dilution=Ef. 50) of the liquid manure or with conventional application of N : P$_2$O$\_$5/ : K$_2$O = 200 : 70 : 500kg/ha for broccoli, 250 : 210 : 240 kg/ha for celery as controls. After harvest, soil pH and K content decreased after using a high concentration of the liquid manure, Ef. 5, than after treatment with weaker concentrations at Ef. 25 and Ef. 50. On the other hand, soil electrical conductivity, content of P$_2$O$\_$5/, organic matter, total nitrogen, and NO$_3$-N at Ef. 5 increased as concentration of swine liquid manure increased. After harvest, available P$_2$O$\_$5/ in plant tissue did not differ significantly between any of the treatments. In broccoli, the lower concentration (Ef. 50) of swine liquid manure increased flowering over the other treatments, perhaps because the level of absorption into the plants is higher with lower concentration. The amounts of K and Ca in plant tissue were greatest after Ef. 25 and Ef. 50 treatments. Plant growth was best at Ef. 50 in broccoli, head height, head width, and head weight were the best with Ef. 25 and Ef. 50 treatments after harvest. In celery, leaf length was greater after Ef. 25 and Ef. 50 treatments than any other treatments. Total yield of celery of Ef. 25 and Ef. 50 treatments was twice that of conventional cultivation. On the other hand, yield severely decreased after application of high-concentration treatment at Ef. 5. In conclusion, fertigation of swine liquid manure, diluted in the range of Ef. 25 to Ef. 50, could improve yield and quality in broccoli and celery.

  • Investigated ware the decomposition of mulch paper, weed occurrence, and rice growth and yield in transplanting rice culture with paper mulching in 1996 and 1997. The mulch paper were developed from domestic old corrugated container. In the experiment of 1996 where 25 percent of nitrogen fertilizer was applied as tillering fertilizer at 15 days after transplanting, it took about 28 to 40 days for the mulch paper to reach 50 percent decomposition according to types of mulch paper and fertilizer amount. The decomposition was faster in 18kg N/10a fertilization plot than in 12kg N/10a fertilization plot, and slower in the mulch paper treated with polyamidepolyamine epichlorohydrin resin(PPE) for the reinforcement of wet strength than in the mulch paper without PPE. In the experiment of 1997 where nitrogen fertilizer was not applied at tillering stage and instead 70 percent of nitrogen fertilizer was supplied as basal fertilizer, it took more than 42 days for the mulch paper(PPF treated) to reach 50 percent decomposition. Paper mulching was found to be very effective in controlling the paddy weeds at early stage of rice growth, but the efficacy, of weed control decreased especially in the mulch paper without PPE at later stage of rite growth due to the occurrence of Ludwigia prostrata in 1996. Few weeds occurred even in non-mulched plot without herbicide in 1997, rending difficulties in evaluating the efficacy of mulch paper in weed control, However, it could be deduced from the durability of mulch paper that paper mulching would have had sufficient efficacy of weed control. Rice yield was only 185kg/10a in the plot without mulching and weed control, but 657kg/10a and 771kg/10a in the paper(PPE treated) mulching plot with 12kgN/10a and 18kgN/10a, respectively, in 1996. The paper mulching plot showed rice yield similar to the weed control plot with herbicide in 1997. It would be concluded that paper mulching could ensure effective weed control and as high yield as the conventional rice cultivation with herbicidal weed control if the corresponding amount of basal nitrogen fertilizer is increased instead of omitting the nitrogen fertilizer at tillering stage.

  • Ultraviolet (UV) induces photo aging, erythema, sunburn, photo-toxicity, photo-allergy, and skin tumor, To investigate photo-protective effects of AmorePacific Beauty-03 (APB-03; mixture of red ginseng extract powder and soybean extract powder) on UV-induced damaged skins, 40 SKH hairless female mice were orally administered APB-03 or saline five times a week and irradiated with UV three times per week far up to 12 weeks. Visible skin changes and skin damage in dermis and epidermis by replica image analysis and histological analysis. In APB-03-treated group, better skin, negative replica appearance and less wrinkle formation were observed compared to the UV control group. These results demonstrate oral administration of APB-03 have photo-protective effects on UV-damaged hairless mouse skin.

  • This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.

  • The study was conducted to develope a rapid method for the detection of Yersinia enterocolitica in spring water and vegetables via multiplex polymerase chain reaction (PCR) technique using ail, yst, uirF and subgenus-specific Y16S primers. Specificity and sensitivity of multiplex PCR and application of best primers for the detection of Y. enterocolitica from spring water and vegetables were investigeted. Y. enterocolitica ATCC 27729 strains gave 356 bP and 200 bp (Y16S) and 134 bp (yst) bands. but Y. enterocolitica ATCC 9610 and ATCC 23715 strains gave 200 bp and 134 bp bands. In the meanwhile, non-pathogenic Yersinia species, such as Y. frederikseni, Y. inter-media, Y. kristenseni and Y. pseudotuberculosis gave only single 200 bp band, and other bacteria including Escherichia coli O157:H7 ATCC 25392, Shigella dysenteri. Staphylococcu aureus ATCC 25923 and Listeria mo-nocytogenes ATCC 19111 did not show any bands. Among primers, yst and Y16S primer showed the best sensitivity. Seven CFU/mL Y. enterocolitica cells could be detected with yst and Y16S primers and the sensitivity was significantly improved by the further 2nd PCR after 38 cycles of first PCR amplication. Spring water, cabbage and mushroom were inoculated with Y. enterocolitica to determine the sensitivity of multiplex-PCR for the rapid detection of Y. enterocolitica. Multiplex-PCR assay could detect 7 or 70 cells in spring water and vegetables using whole cell lysate with repeating PCR amplication.

  • In order to design and develop a product that can treat the fatty liver, natural complex food with all natural ingredients was developed and supplemented to rats with high fat diet to induce fatty liver. As a result, when the amount of natural complex food was increased in diet of subjects, the activities of the blood serum AST, ALT, ALP, 3-GT and LDH were decreased. The total protein concentration levels of the 30% and the 50% natural complex food groups did not show changes in respect to the control group, but the 100% natural complex food groups showed significant decrease (p<0.05). Likewise, the amount of blood serum albumin in the 30% and the 50% natural complex food groups did not show improvement, but the 100% natural complex food did showed significant changes (p<0.05). The amount of blood serum triglyceride decreased as the amount of natural complex food was increased. In order to investigate the appearances of the accumulated fat in the liver, the animals were dissected. Livers of the control group (no natural complex food) were appeared as a white color, which means serious fat accumulation. However, all the natural complex food groups (30,50 and 100% natural complex food) showed noticeable decrease of fat content. Even the histology showed that livers of the control group had expansion of the fat, but a11 the natural complex food groups had e decreased as the contents and continued to show destroyed fatty cells. By observing the biological numeric data, the physical appearance and the history of the fatty liver, it is highly expected that natural complex food is very effective in treating the liver damaged -by the to fat and the cholesterol.

  • We performed a randomized placebo-controlled trial to determine whether or not fermented red ginseng supplementation modulates blood glucose and insulin resistance in type 2 diabetic patients. A total of 38 patients were randomized to either a fermented red ginseng group or placebo group. The patients in the experimental or placebo group consumed 780 mg of fermented red ginseng or cellulose supplement per day for 12 weeks, respectively. Lifestyle factors and dietary intakes of the patients were not altered during the 12-weeks period. In the fermented red ginseng group after 12 weeks, the fasting blood glucose levels were significantly decreased ($136.29{\pm}16.45$ mg/dL to $127.71{\pm}17.74$ mg/dL) and $HbA_1c$ was also decreased. Especially, high HbA1c (HbA1c $\geq$8%, $8.45{\pm}0.56%$ to $7.82{\pm}0.53%$) was significantly decreased compared to low HbA1c (HbA1c <8%, $6.71{\pm}0.85%$ to $6.44{\pm}0.49%$) in the fermented red ginseng group. Serum low-density lipoprotein was slightly decreased in the fermented red ginseng group compared to the placebo group. Homeostasis model assessment-insulin resistance was significantly reduced in the fermented red ginseng group compared to the placebo group. These results suggest that fermented red ginseng supplementation could be helpful to reduce blood glucose by improving insulin resistance in type 2 diabetic patients.

  • This study was carried out to the structure of plant community from Sinseongam to Jungdaesa in Odaesan National Park, furthermore, it seeks to curate the basic data for planning of the Abies holophylla's forest management in Odaesan National Park. In order to identify the current ecological environment, this study explored the actual vegetation as primary research and set to twenty plots(i.e. $400m^2$) for analysing detailed structure of plant communities. The research methodology was qualitative analysis, therefore it used TWINSPAN and DCA analysis tools. Especially, TWINSPAN performed well in several comparisons of classification techniques, DCA is one of the ordination technique showed that the plant communities. The plant community was analysed classification and ordination by TWINSPAN and DCA, moreover it was analysed the structure of plant community such as importance percentage of woody species, DBH class distribution, the index of diversity and rate of sample tree growth. The main vegetation was A. holophylla-Quercus mongolica forest and Deciduous broad-leaved forest in the communities where located in low altitude and valley, whereas main vegetation where located in high altitude and slope was Q. mongolica forest. The research site's plant communities were classified four groups. In all of communities, A. holophylla was dominant species in main canopy layer, furthermore, the three communities (community I, II, III) are growing up next generation of A. holophylla excluding community IV. The communities (community I, II, III) can be sustained current status which dominates the A. holophylla communities, simultaneously, there might be expanded the Deciduous broad-leaved communities by Carpinus cordata, Betula schmidtii and so on. While, it showed that the community IV tended to be weaken the forces of A. holophylla, therefore the community IV can be transferred to C. cordata-Deciduous broad-leaved communities in the future. The age of sample trees was 79~128(i.e. A. holophylla), 75~87(i.e. Pinus koraiensis) and 190 years(i.e. Ulmus davidiana var. japonica). The index of Shannon's Species diversity (H') were ranged from 0.3889 to 1.3332 in the communities.
