The effect of baicalin in a mouse model of retinopathy of prematurity

  • Baicalin is a flavonoid derived from the dried root of Scutellaria baicalensis. In this study, oxygen-induced retinopathy was used to characterize the anti-angiogenic properties of baicalin in mice. Pups were exposed to a hyperbaric oxygen environment to induce retinal angiogenesis and were subjected to intraperitoneal injection of baicalin. Avascular area, neovascular tufts, and neovascular lumens were quantified from digital images. Compared to the vehicle, baicalin clearly reduced the central avascular zone and the number of neovascular tufts and lumens. High-dose baicalin (10 mg/kg) significantly reduced the expression of matrix metalloproteinase-2 (MMP-2), MMP-9, angiotensin II, and vascular endothelial growth factor (VEGF). These results show that baicalin is a powerful antiangiogenic compound that attenuates new vessel formation in the retina after systemic administration, and is a candidate substance for therapeutic inhibition of retinal angiogenesis. Antifungal Activities of Isothiazoline/Cabamate based Organic Antifungal Agent Activated-Cement Mortars (AACM)

  • Antifungal agents are used to impart antibacterial or bactericidal properties to commodities and various articles used in industries and can be classified into two broad groups i.e organic and inorganic. Inorganic antifungal agents comprise of Ag, Zn, or Cu, etc. These elements tend to exhibit high level of antifungal activities, non-uniform dispersion in substrates, and have poor properties in expensive and cheap adhesiveness. In this study, the organic antifungal agent was used for the purpose of investigating the antifungal activity of antifungal agent activated-cement mortar (AACM) on the aspergilus niger of various fungus which can be easily discovered in the interiors and exteriors of buildings. In addition, an experiment on the basic physical properties of AACM such as compressive and flexural strength was carried out. The conclusion of this investigation revealed that a dosage increase of antifungal agent exhibits a high inhibitory effect on the aspergilus niger, and although there is a slight decrease in the strength of AACM, the strength of AACM was almost equal to that of inactivated cement mortar.

Antagonism against Helicobacter Pylori and Proteolysis of Lactobacillus Helveticus CU631 and Strain Identification

  • The antagonistic activities of 30 strains of lactobacilli against Helicobacter pylori were determined and Lactobacillus helveticus CU631 has been selected as the strain which possesses the strongest inhibitory effect in the disc diffusion assay showing inhibition zone diameter of $10{\pm}1.5mm$, whereas those of L. plantarum and L. fermentum have been shown to be $4.0{\pm}0.6mm$. H. pylori G88016 revealed the highest vacuolating toxin producing activity among the 8 strains, the inhibitory activity of L. helveticus CU631 in vacuolating toxin producing activity of H. pylori manifested in the co-culture of two strains and in the 5:5 mixture of supernatant of the two strains. Both L. helveticus CU631 and cell free culture supernatant had a strong inhibitory activities in urease and cytotoxin producing activities of H. pylori NCTC11637 and CJH12. An accelerated proteolytic activity of water soluble peptides by L. helveticus CU631 during the refrigeration storage has been manifested in the cream cheese. DNA seqences of 16S-23S ribosomal RNA spacer region showed typical pattern among the various strains of L. helveticus, which could be used in the identification of L. helveticus CU 631.

Gentamicin/CTMA/Montmorillonite as Slow-Released Antibacterial Agent

  • This paper presents the characteristics of gentamicin-loaded into cetyl trimethyl ammonium intercalated montmorillonite (GtM/CTMA/Mt) as a hybrid composite for a slow-released antibacterial delivery systems. The work describes the successful immobilization of gentamicin into the interlayers of surfactant-modified montmorillonite. Physicochemical characterization of the material is carried out by means of X-ray diffraction, scanning electron microscopy, transmission electron microscopy, and Fourier transform infrared spectroscopy. The kinetics of the gentamicin release is investigated by in vitro study and analyzed based on UV-Vis spectrometry. In addition, antibacterial study is performed towards Klebsiella pneumoniae Staphylococcus aureus, Escherichia coli, and Streptococcus pyogenes. The results show that the gentamicin loading into CTMA/Mt increases the effectiveness of the antibacterial activity, as shown by the higher inhibition zone for all tested bacteria, compared to gentamicin as a positive control. The kinetics study suggests that the gentamicin release obeys the modified Korsmeyer-Peppas model. The physicochemical study and activity test demonstrate the feasibility of the GtM/CTMA/Mt for practical applications.

  • A new type of macrolides, lincosamides and streptogramin B antibiotics (MLS) resistance, showing the characteristic phenotype growing within the inhibition zone area around the clindamycin disk, was identified among clinically isolated Staphylococcus aureus. We named the phenotype as MLS resistance with foggy-D shape (fMLS). The average frequency of MLS isolates was 9%. All of fMLS isolates have only erm(A) for the resistance determinant. The growth pattern of the challenged MLS isolate looks intermediate phase between the patterns of inducible MLS resistance and constitutive MLS resistance.

봉약침액(蜂藥鍼液)의 안정성(安定性) 연구(硏究) (A Study on the Stability of Diluted Bee Venom Solution)

  • Objective : The purpose of this study was to investigate the stability of bee venom according to the keeping method and period. Method : The author observed microbial contamination of bee venom in nutrient agar, broth, YPD agar and YPD media and antibacterial activity for S. aureus, E. coli manufactured 12, 6 and 3 months ago as the two type of room temperature and $4^{\circ}C$ cold storage. Results : 1. 1:3,000 and 1:4,000 diluted bee venom solution did not show microbial contamination both room temperature and cold storage within twelve months. 2. There was antibacterial activity of diluted bee venom for S. aureus in cold storage within twelve months and there was no antibacterial activity of diluted bee venom for S. aureus in twelve months, room temperature storage. 3. We could not observe the zone of inhibition around paper disc of all for E.coli. in 1:3,000, 1:30,000 and 1:3,000,000 diluted bee venom solution, respectively. According to results, we expect that diluted bee venom solution is stable both cold and room temperature storage within twelve months.

골프장 잔디의 Rhizoctonia 마름병에 대한 생물학적 방제 (Biological Control on Rhizoctonia Blight of Turfgrasses in Golf Courses)

  • This study was undertaken to find a new formulation of soil amendment, and selection of antogonists and to effectively control brown and large patch of turfgrasses caused by Rhizoctoniz solani AG1-1 and AG 2-2. Fourteen inorgainc chemicals (1%, w/w) were added individually in vitro, and some chemicals showed suppressiveness to R. solani. Alum suppressed effectively mycelial growth of R. solani in the range of 17 to 77% as compared with control. The four chemicals such as Al2(SO4)3, alum, CaO, and NH4NO3 were finally selected. Out of three organic compounds, composted pine bark (CPB) showed prominent suppressive effect as compared with milled alfalfa and pine leaves. After inoculation of R. solani isolates AG-1 and AG2-2 on the turf seedlings, water soaked lesions and blight symptoms were developed on the whole seedlings. According to inhibition zone method, mycelial growth of the fungus were greatly suppressed by culture filterates of the antagonists, Gliocladium virens (Gl1-) and Pseudomonas sp. (P713). CPB soil amendment mixed with antagonists (1% w/w) controlled not only brown and large patch of turfgrasses, but also promote the good growth of the seedlings. In addition, the controlling effect was maintained more than 30 days. Especially, the controlling effect of two antagonists was similar to Cㅖㅠ soil amendment with the antagonists and also stimulated a favorable growth of the seedlings. Therefore, its is expected that continuous control of Rhizoctonia blight of turfgrasses can be obtained in field by subsequent applications of the antagonists.

세파드록실의 방출에 미치는 폴리우레탄 매트릭스 제조방법의 영향 (The Effect of Fabrication Methods on the Release of Cefadroxil from a Polyurethane Matrix)

  • In order to evaluate the effect of fabrication methods on the controlled release of an antibiotic from a polymeric device, two types of polyurethane-cefadroxil matrix were prepared by the solvent casting method or the freeze drying method, using bovine serum albumin as a pore former. The amount of cefadroxil released from various formulations at $37^{\circ}C$ was measured by HPLC. The duration of antimicrobial activity of matrices against S. aureus was evaluated by measuring the diameters of the inhibition zone. The morphology of the matrices was investigated by scanning electron microscopy (SEM). Changing the fabrication method could alter the release rate of cefadroxil from the matrix. The matrix fabricated by the freeze drying method had more porous inner structure and showed higher release rate than that prepared by the solvent casting method. However, the duration of antimicrobial activity was shorter when the matrix was fabricated by the freeze drying method.

  • High Voltage Pulsed Current Stimulation(HVPCS) and Microcurrent Electrical Neuromuscular Stimulation(MENS) have been used to promote the healing of decubitus ulcer and surgical wounds. The benefits of HVPCS and MENS are thought to include an inhibitive effect on bacterial growth. The purpose of this study was to compare the inhibitive effect of two different electrical stimulation techniques growth in vitro. Using agarose-based media, the two bacterial species Staphylococcus aureus, Esherichia coli - which are commonly isolated from open wounds were incubated in an incubator for 24 hours following exposure to HVPCS(400 V, 120 pps, $70{\mu}s$) and MENS($100{\mu}A$, 0.3 Hz). We then measured the zone of inhibition around each electrode. Both HVPCS and MENS produced an inhibitive effect on bacterial growth in this vitro study. However MENS was more effective than HVPCS.

Properties of the Fusants of Lactobacillus acidophilus 88 and Lactobacillus casei subsp. casei KCTC 1121

  • Protoplast fusion between L. casei KCTC 1121 and L. acidophilus 88 was attempted to obtain improved strains. The fusants produced a bacteriocin against indicator strains, making a smaller inhibition zone compared to that of L. acidophilus 88. After culturing for 2 months on selective medium, the selected fusants were still stable without segregation. Fusants showed higher lipase activity compared to those of the two parent strains. Fusant No.4, 11, and 15 exhibited excellent lactic acid productivity. Fusant No.4 and 15 exhibited improved proteolysis ability compared to the two parent strains. Whereas L. casei possessed both ${\beta}-galactosidase$ and $phospho-{\beta}-galactosidase$ activities, and L. acidophilus 88 had only ${\beta}-galactosidase$ activity, the fusants had both the intermediate enzyme activities. Cell size of the fusants was greater than that of the parents.
