Effects of Enrichment Planting with Five Native Species and Different Plantation Treatments on Seedling Growth Characteristics at Logged-over Forest in Lao PDR (라오스 개벌림에서 다섯가지 자생수종과 각기 다른 조림처리의 Enrichment Planting이 묘목의 생장특성에 미치는 효과)

  • Enrichment planting is commonly used for increasing the density of tree species in secondary forests and one of forest rehabilitation programs in Lao PDR. This study aimed to determine the performance of five native species in enrichment planting using different canopy opening treatments, and to examine the suitable species and silviculture techniques applicable to a logged-over forest in Lao PDR. Results of the study showed that only crown height had significant difference among species and planting treatments. The survival rate and crown diameter were significantly different by species, but not by planting treatments. However, root collar diameter (RCD) and height of seedlings showed significant differences among planting treatments. Among the study species, Hopea odorata and Dalbergia cochinchinensis showed significantly higher survival rate and height growth, indicating their suitability in enrichment planting for rehabilitation of forest in Lao PDR. The results also suggested that 2 m width line planting stimulated RCD growth, but wider line planting, such as gap planting, promoted height growth of seedlings.

Characterization of Fusarium solani Causing Fusarium Root Rot of Lisianthus in Korea (꽃도라지 뿌리썩음병을 일으키는 Fusarium solani 의 특성)

  • Lisianthus (Eustoma grandiflorum) is a flowering ornamental plant used widely in Korea. In 2015, wilting, damping-off, stunting, and root rot symptoms were observed in lisianthus plants of Yeoju and Gimhae, Korea. Affected seedlings appeared yellow and showed poor development of root systems in the field and in nursery boxes. Furthermore, affected plants were yellow, stunted, and died at approximately 2-3 months after transplanting. Fusarium species were consistently isolated from the basal stems of diseased plants. Nine isolates were identified as Fusarium solani based on morphological characteristics. Macroconidia of isolates were relatively wide, straight-to-slightly curved, and microconidia formed in false heads on long monophialides. Abundant chlamydospores were produced at the middle or tips of hyphae. To confirm this identification, a molecular analysis of the translation elongation factor 1 alpha (TEF) and RNA polymerase II subunit (RPB2) genes was conducted. The sequences of TEF and RPB2 showed 99.2-99.9% and 98.0-98.1% similarity, respectively, to those of reference F. solani strains in NCBI GenBank. Pathogenicity was tested using root dipping inoculation of healthy lisianthus seedlings. Symptoms were observed within 7 days of inoculation only in inoculated plants. This is the first report of F. solani causing Fusarium root rot on lisianthus in Korea.

The Variation of Population of Pinus koraiensis Sieb. et Zucc. in Mt. Jiri (지리산(智異山) 잣나무집단(集團)의 변이(變異)에 관(關)한 연구(硏究))

  • These studies were carried out to know the variation of morphological tree forms, needles, cotyledons, seed characters and top height growth of seedlings of Pinus koraiensis S. et Z. by populations in Mt. Jiri. The results are summarized as follows: 1. It was observed that the characteristics of length, needles of serration density of needles, percentage of full seeds length, of seeds, thickness, of seeds length of cotyledons, width of cotyledon, numbers of the stomata line in cotyledons and the top height growth of seedlings were significantly different in individual trees and populations. 2. The characteristics of branch angle specific gravity of wood and resin canal numbers of cotyledons were significantly different in populations. 3. It was that the characteristics of numbers of stomata line, width, of seeds weight, of seeds serration density of cotyledons, numbers of cotyledons were significantly different in individuals. 4. The resin canals of needles showed three principle resin canals in individual trees and population, and the coefficients of variation were not shown in individual trees and populations. Stem forms showed to be a straight form with all populations. 5. The characteristics of cotyledons had generally higher tendency than the characteristics of needles in comparison with coefficience of variation.

Effects of Supply Methods of Nutrient Solution on Growth of Grafted Cactus Gymnocalycium mihanovichii var. friedrichii Grown Hydroponically (접목선인장 비모란 수경재배시 배양액의 공급방법이 생육에 미치는 영향)

  • This experiment was conducted to elucidate the proper suppling frequency of nutrient solution for grafted cactus Gymnocalycium mihanovichii var. friedrichii grown hydroponically without medium. Grafted cactus seedlings were planted onto the cultivation bed without medium, using labor-saving tray. The treatments like 1, 3, 5 and 7 times of nutrient solution supply per day and continuous soaking of plant root in the nutrient solution during the daytime were tested in summer and winter season. The growth of grafted cactus was worst in the treatment of one time supply of nutrient solution per day, and there were not significant difference in growth of grafted cactus among other treatments both in summer and winter season. 17.6% of grafted cactus seedlings failed to rooting in the treatment of one time supply of nutrient solution per day in winter season. The proper suppling frequency of nutrient solution, for the grafted cactus Gymnocalycium mihanovichii var. friedrichii grown hydroponically without medium, was three times supply of nutrient solution per day both in summer and winter seasons considering growth and rooting of plants.

Development of SNP Molecular Marker for Red-fleshed Color Identification of Peach Genetic Resources (복숭아 유전자원의 적색 과육 판별 SNP 분자표지 개발)

  • Various colors of fruit skin and flesh are the most popular commercial criteria for peach classification. In order to breed new red-fleshed peach cultivar, many cross seedlings and generations should be maintained. Therefore it is necessary to develop early selection markers to screen seedlings with target traits to increase breeding efficiency. For the comparison of transcription profiles in peach cultivars differing in flesh color expression, two cDNA libraries were constructed. Differences in gene expression between red-fleshed peach cultivar, 'Josanghyeoldo' and white-fleshed peach cultivar, 'Mibaekdo' were analyzed by next-generation sequencing (NGS). Expressed sequence tag (EST) of clones from the two cultivars were selected for nucleotide sequence determination and homology searches. Putative single nucleotide polymorphisms (SNP) were screened from peach EST contigs by high resolution melting (HRM) analysis displayed specific difference between 8 red-fleshed peach cultivars and 24 white-fleshed peach cultivars. All 72 pairs of SNPs were discriminated and the HRM profiles of amplicons were established. In the study reported here, the development of SNP markers for distinguishing between red and white fleshed peach cultivars by HRM analysis offers the opportunity to use DNA markers. This SNP marker could be useful for peach marker assisted breeding and provide a good reference for relevant research on molecular mechanisms of color variation in peach cultivars.

Optimization of In vitro Cultures for Production of Seedling and Rootstock of Rehmannia glutinosa(Gaertn.) DC. (지황 배양묘 및 종근 생산을 위한 기원검증 및 최적기내배양조건 확립)

  • Rehmannia glutinosa(Gaertn.) DC. is a herbaceous perennial plant and belonging to the Scrophulariaceae and used as roots for medicinal part and purpose. R. glutinosa is and usually used for fresh rehmannia root or prepared rehmannia root. However, it is very difficult to propagate using the seeds because of lack germination so it is propagated using the vegetative method as the rootstock. Currently, propagation and harvesting using the rootstock of R. glutinosa has difficulties about production of the high quality and quantity in R. glutinosa because of root rot disease. To optimize in vitro cultures and to improve the rootstock and seedling of R. glutinosa after morphological and genetical determination, 5 plant culture media (MS, DJ, LS, QL, and WPM) were used in this study then WPM was selected for better growth, for multiplication condition(WPM + IAA 1.0 mg/L + IBA 0.5 mg/L), and for root enlargement condition(WPM + NAA 0.1 mg/L) of R. glutinosa. Based on these results, in vitro seedlings of R. glutinosa were transferred to soil for acclimation with environment adaptation and shown the positive effects about root enlargement and root formation. Therefore, it can be used for high quality of R. glutinosa production and production of the rootstock based on propagation using in vitro seedlings of R. glutinosa.

Current Status and Perspective of Smart Vegetable Seedling Production Technology in the Republic of Korea (국내 스마트 채소 육묘 기술 개발 현황 및 전망)

  • In this study, we summarized the definition of smart vegetable seedling production technology, analysis of smart seedling production system, a hardware and software configuration model for smart seedling production system, research and development trends in smart seedling production system, and proposed future research and development plans for smart seedling production technology. Smart vegetable seedling production is a data-based seedling production, management, and distribution system that utilizes 4th Industrial Revolution technology to improve seedling productivity and quality. The production of vegetable seedlings using smart seedling production technology can be efficiently managed by collecting, analyzing, and managing information on seedlings, environment, and tasks at each stage of production by linking with the smart seedling integrated management system. However, there is still a lack of standardization of seedling standards and quality for each vegetable crop to establish smart seeding production technology, as well as development of smart seedling production element technology, which requires national wide R&D support.

Studies of Liming Effect on the Improvement of an Acid Sulphate Paddy Soil (특이산성답(特異酸性畓) 토양(土壤)의 개량(改良)을 위(爲)한 석회시용(石灰施用) 효과(?果)에 관(關)한 연구(硏究))

  • These studies were carried out for the elucidation of liming effect on the growth of rice seedlings and the chemical characteristics of an acid sulphate paddy that shows not only extremely high acidity of soil but also poor growth of rice plants, consequently low yield. Thus the liming effect on the changes of acidity, oxidation-reduction potential, and the contents of iron, aluminium, sulphate, and phosphorus fractions in the soil was investigated under the waterlogging and drying condition. The reclaimable or inhibitory effect of phosphorus, iron and aluminium on the growth of rice seedlings was also investigated under liming. The results are summarized as follows: 1. After liming, the pH of the acid sulphate subsoil decreased again on drying. 2. The oxidation-reduction potential reached a minimum after 5 days of flooding and greatly decreased on liming but increased after drying. 3. The contents of ferrous iron soluble in water-and Morgan's solution reached a maximum after 15 days of flooding and only the content of water soluble ferrous iron was greatly decreased. 4. The content of aluminium soluble in water-and Morgan's solution decreased by flooding and liming, and showed a tendency to increase on drying. 5. In the limed acid sulphate soil, the content of water soluble calcium showed a highly significant negative correlation with the content of sulphate and liming decreased sulphate content in the soil. 6. The contents of total phosphorus was 496.3 ppm in the acid sulphate topsoil and 387.5 ppm in the subsoil. The content of each phosphorus fraction was in the order of Fe-P>Occ. Fe-P>Ca-P>Occ. Al-P>Al-P and Fe-P content in the soil was the highest fraction among them. 7. Lime application increased greatly Ca-P and Al-P, and Occ. Fe-P and Occ. Al-P only slightly, but decreased Fe-P differently in each soil. 8. Effect of phosphorus on the dry matter yield of rice seedlings was great. The optimum amount of phosphorus to produce maximum dry matter yield of rice seedlings appeared to be 6.8% of maximum absorption (absorption coefficient) without liming and 10.0% with liming. 9. In rice seedlings liming increased the content and uptake of calcium and silica but decreased those of iron and aluminium. Phosphorus application increased the content and uptake of phosphorus and decreased iron while the application of iron and aluminium increased their contents and uptake but decreased those of phosphorus. 10. Liming greatly alleviated such toxicity of iron and aluminium. 11. When phosphorus was applied, the dry matter yield of rice seedlings showed highly significant positive correlations with uptake of phosphorus, calcium and silica each. When iron and aluminium were applied, dry matter yields indicated significant positive correlations with the contents or uptake of calcium and silica each, but significant negative correlations with the content or uptake of iron and aluminium. 12. Under the application of phosphorus and lime, dry matter yields showed significant positive correlations with pH and Morgan's extractable calcium each of the soil samples after harvest. Under the application of lime, iron and aluminium, dry matter yields showed significant positive correlations with pH, calcium and silica each, but negative correlations with iron and aluminium contents each of the soil samples after harvest.

  • The inheritance of isoenzymes in young coat tissues of trembling aspen (Populus tremuloides Michx.) was studied by electrophoretically analysing five parental clones and their full-sib progenies obtained by crossing one female clone to four male clones. The distal 2cm section of vigorous young roots of 70 seedlings per family were subjected to horizontal starch gel electrophoresis. The resulting gels were tested for activity of 7 enzyme systems. Evidences for the inheritance of isoenzymes observed indicated that the isoenzyme variants of every isoenzyme zone were under control of codominant alleles at a single locus. Chi-square test of joint segregation data of the two loci, 6-PGD-2 and PGI-2, indicated that the pair of loci was not linked.

Assessing weediness of herbicide tolerant genetically modified soybean

  • Imports of genetically modified (GM) soybeans (Glycine max) for food or feed consumption in Korea have been increasing. Although the cultivation of GM soybeans has not yet been allowed in Korea, the number of field tests for GM soybeans has also been rising. This study was conducted to investigate whether herbicide tolerant GM soybean can survive and persist in uncultivated environments when they escape from transportation routes or from isolated fields. Seeds of GM and non-GM soybeans and wild soybeans (Glycine soja) were buried in 2 and 15 cm soil depths and their viability was examined after 1, 2, 6, and 10 months. GM and non-GM soybean seeds completely lost their viability within six months of burial, whereas seeds of wild soybean maintained their viability during the study period. Seeds of soybean and wild soybeans that were sown on the soil surface germinated and grew to vegetative cotyledon stage. Seedlings of GM and non-GM soybean did not compete well with weeds, including Cerastium glomeratum, Alopecurus aequalis var. amurensis, Capsella bursa-pastoris, Conyza canadensis, Stellaria aquatica, and Erigeron annuus. Also, GM soybean did not survive through winter. However, wild soybeans competed well with the weeds and became dominant in August. Herbicide tolerant GM soybean is unlikely to persist under uncultivated environments and to become weeds.
