팥에 발생하는 바이러스 분리 동정 (Identification of Virus from Azuki Bean Plant)

  • 팥 바이러스병에 대한 기초자료를 얻고자 병징의 유형과 생육 단계별 감량정도 및 수량에 미치는 영향을 조사하고 병징 유형별 바이러스를 분리 동정한 결과는 다음과 같다. 1. 팥에 발생되는 바이러스병의 병징은 크게 mosaic, yellow mosaic 및 severe mosaic의 3가지 군으로 분류되었으며 병징별 분포는 mosaic>severe mosaic>yellow mosaic 순이었다. 2. 성숙기까지 매개충 차단 재배시의 이병률은 1.5%(방임구 20.7%)로서 생육 후기에 감량될수록 이병률이 낮았으며 10a당 수량도 171kg으로서 방임구에 비하여 45% 증수되었다. 3. 지표식물 검정 결과 mosaic 유형은 CMV, yellow mosaic 유형은 AMV, severe mosaic 유형은 AzMV의 기주범위와 유사하였다. 4. 항혈청에 의한 각 병징별 반응결과 yellow mosaic 유형의 시요는 AMV 항 혈청과 mosaic 유형의 시요는 CMV의 항혈청과 침강선이 형성되어 각각 AMV 및 CMV의 한 계통으로 판정되었다. 5. 병징 유형별 전자현미경 관찰결과 yellow mosaic유형은 18~58$\times$18nm의 타원형 입자가 다수 관찰되었고 mosaic 증상의 시요에서는 직경 30nm의 구형 입자, severe mosaic병징의 시요에서는 730$\times$12nm의 사상형 입자와 봉입자가 관찰되었다.

영세농가 콩 재배지의 Soybean mosaic virus, Soybean yellow mottle mosaic virus 및 Soybean yellow common mosaic virus 병 발생 조사 (Occurrence of Three Major Soybean Viruses, Soybean mosaic virus, Soybean yellow mottle mosaic virus and Soybean yellow common mosaic virus Revealed by a Nationwide Survey of Subsistence Farming Soybean Fields)

  • Soybean yellow mottle mosaic virus(SYMMV)와 Soybean yellow common mosaic virus(SYCMV) 병 발생은 최근에 우리나라에 보고되었지만 두 바이러스 병의 전국적인 분포에 대한 조사가 이루어지지 않아 본 연구팀은 2012년에 영세 농가를 중심으로 전국적인 조사를 실시하였다. 전국에 걸쳐 채집한 682점의 시료를 SYMMV, SYCMV, 그리고 Soybean mosaic virus(SMV) 특이 프라이머를 이용하여 진단한 결과, SMV 102점, SYMMV 116점, SYCMV는 17점이 PCR 양성으로 나타났다. SYMMV와 SYCMV의 복합감염은 검출되지 않았으나, SMV와 SYMMV 혹은 SMV와 SYCMV 복합감염은 각각 1건과 5건이 확인되었다. 처음으로 실시한 영세농가의 콩 바이러스 병 조사를 근거로 추정할 때, 바이러스의 발생은 자가 채종으로 인한 종자전염으로부터 발생되었을 것으로 추정된다. 따라서 바이러스 병의 예방을 위해서는 감염종자 관리 및 무병종자 보급이 무엇보다 필요함을 알 수 있다.

Evaluation and Verification of Barley Genotypes with Known Genes for Resistance to Barley yellow mosaic virus and Barley mild mosaic virus Under Field Conditions in South Korea

  • Soil-borne barley yellow mosaic disease caused by Barley yellow mosaic virus (BaYMV) or Barley mild mosaic virus (BaMMV) gives a serious threat to the winter barley cultivated in the southern regions in Korea. It is important to develop resistant varieties for stable and high-yield production. The objectives of this study were to evaluate 22 genotypes of exotic barley germplasms carrying the resistance genes rym1 through rym12, with the exception of rym10, and to determine the genes that confer resistance to BaYMV or BaMMV in Korea. Using the traditional visual scoring of symptoms at 4 locations over 3 years, average disease rate values differed (P < 0.001) among the genotypes. ELISA test revealed the presence of both BaYMV and BaMMV in all of the field sites but Jinju and significantly different rates of infection among genotypes and years. Barley genotypes differed in how virus quantities and pathogen-induced symptoms were correlated, especially in response to BaYMV. Disease incidence was affected by the climatic conditions present during the early growing stage before overwintering. A Chinese landrace, 'Mokusekko 3', carrying rym1 and rym5 was comparatively resistant at all locations studied. The barley genotypes carrying either rym6 or rym9 were susceptible to the viral strains. The genotypes carrying rym5 were resistant in Jinju and Milyang but susceptible in Iksan and Naju. The resistance genes rym2 and rym3 were effective in local strains and would be potent contributors to disease resistance.

한국산 글라디올러스에 발생하는 바이러스 (The Viruses in Gladiolus hybridus cultivated in Korea 1. Bean Yellow Mosaic Virus and Clover Yellow Vein Virus)

  • Gladioli (Gladiolus hybridus) showing flower colour breaking, leaf mosaics, necrotic fleck, and dwarfing or lack of visible symptoms were collected from gladioli growing areas in Taegu and Kyungpook province, Korea. The two viruses isolated from the naturally infected gladioli were identified as ban yellow mosaic virus (BYMV) and clover yellow vein virus (CIYVV) by their host range, immunosorbent electron microscopy (ISEM), enzyme-linked immunosorbent assay (ELISA), direct tissue blotting immunoassay (DTBIA) and intracellural symptoms. In ultrathin sections of BYMV and CIYVV infected tissues, laminated aggregate-type inclusions, cytopalsmic bodies and nuclear inclusions as well as filamentous virus particles were observed in the cytoplasm of parenchyma cells. By DTBIA and ISEM, BYMV was detected in all tested gladiolus plants showing severe or mild mosaic symptoms, whereas CIYVV were mainly detected from those of mild mosaic symptoms. BYMV is the most prevalent in commercial gladioli and present major production problems. Detection sensitivity of BYMV and CIYVV in crude sap of infected gladiolus leaves by ISEM was about twice compared with ELISA. In a comparison of ELISA, ISEM, DTBIA, BYMV was detected in same degree by DTBIA in samples where sap extracts were positive in both ELISA and ISEM. DTBIA provides a specific, rapid, and simple tool for large-scale diagnosis of BYMV.

용담(Gentiana spp.)에서 분리한 3종의 바이러스 (Three Viruses Isolated form Gentiana spp. in Korea)

  • Gentian plants (Gentiana spp.) showing yellow ringspot, mosaic, necrotic fleck and malformation were collected from their growing areas in Taegu, Kyungpook province and Alpine Agricultural Experiment Station, Korea. Three viruses isolated from the naturally infected gentian were identified as broad bean wilt virus (BBWV), cucumber mosaic virus (CMV) and clover yellow vein virus identified as broad bean wilt virus (CIYVV) by their host range, immunosorbent electron microscopy (ISEM) and electron microscopy. Electron microscopic examination of negatively stained preparations showed that BBWV and CMV are spherical particles of 28 nm and 30 nm in diameter, and CIYVV is filamentous particles of ca. 780 nm in length. By ISEM, BBWV was detected mainly in gentian showing yellow ringspot and mottle, CIYVV in necrotic fleck leaf of gentian and CMV in narrow and distorted mosaic leaf of gentian. BBWV and CMV are the most prevalent in the cultivated gentian. In ultrathin sections of BBWV infected tissues, large aggregates of crystalline array of virus particles and vesicular body were found in the cytoplasm and vacuole of mesophyll cells. In case of CIYVV, pinwheel- and laminated aggregate-type inclusions as well as filamentous virus particles were observed in the cytoplasm of mesophyll cells.

Nested PCR 기법을 이용한 토양으로부터 Barley yellow mosaic virus 검출 (Detection of Barley yellow mosaic virus from Soil Using Nested PCR)

  • 2단계의 nested PCR 방법을 이용하여 보리 및 벼 재배 토양에서 Barley yellow mosaic virus (BaYMV)를 검출하였다. BaYMV 분절 RNA1 외피단백질 영역의 특이 프라이머로 1차 PCR을 하고 내부서열로부터 작성된 프라이머로 2차 PCR을 실시하여 확보된 372 bp의 PCR 산물이 BaYMV 외피단백질 영역과 98%-100% 염기서열이 일치하여 BaYMV를 검출할 수 있음을 확인하였다. 이 결과는 토양으로부터 BaYMV 검출에 관한 최초의 보고이며 토양전염성 바이러스의 정확한 진단과 예찰에 적용될 수 있을 것으로 생각한다.

흰 토끼풀에서 분리한 강낭콩황반모자이크 바이러스의 동정 (Identication of Bean Yellow Mosaic Virus from Trifolium repens)

  • 흰토끼풀에 황색모자이크병징을 나타내는 이병주를 채집하여 병즙액의 지표식물반응과 물리적 성질조사, 풍매전염시험, 바이러스입자의 형태관찰 그리고 항혈청반응 등의 방법으로 병원바이러스의 동정을 시도한 결과 강낭콩황반모자이크바이러스(BYMV)로 밝혀졌다. 공시병원바이러스의 지표식물반응은 명아주에 전체국부반응, 잠두의 접종엽에 국부반응, 줄기와 상위엽에 각각 양저증상과 모자이크반응을 보였으며, 강남콩에는 엽맥양저, 황색모자이크 또는 축엽이 되고 순이 고사하였다. 또한 완두, 녹두, 팥과 red clover에는 모자이크 반응을 나타냈다. 바이러스입자의 형태 및 크기는 740nm정도의 사상입자이며, 복숭아혹진딧물에 의하여 전염이 가능하였다. 바이러스병즙액의 생리적 성질은 내열성이 $60\~65^{\circ}C$, 내희석성이 $10^{-3}\;-\;10^{-4}$, 내보존성이 3일이었다. 순화된 공시바이러스는 일본 식물바이러스 연구소로부터 분양받은 BYMV항혈청과의 혼합침강반응결과 양성반응을 보였다.

RP-PCR을 이용한 보리누른모자이크바이러스 (BaYMV)와 보리마일드모자이크바이러스(BaMMV)의 외피단백질 유전자 검정 및 해석 (Analysis and Detection of Coast Protein Gene of Barley Yellow Mosaic Virus and Barley Mield Mosaic Virus by RT-PCR)

  • Using the reverse transcription polymerase chain reaction (RT-PCR), a rapid and sensitive assay method for the detection and identification of barley yellow mosaic virus (BaYMV) and barley mild mosaic virus (BaMMV) was adapted. Two units of primers from each virus were selected and used for the determination of two different viruses. PCR fragments of BaYMV (ca. 0.9kb) and BaMMV (ca. 0.8kb) were obtained from the designed method for the assay of BaYMV and BaMMV coat protein. PT-PCR fragments were cloned using vector pT7 Blue and the sequences of the selected clones were analyzed. coat protein of BaYMV and that of BaMMV consisted of 297 amino acids (891 nucleotides) and 251 amino acids (753 nucleotides), respectively. The snalysis of coat protein genes from these two viruses showed that 45.6% of nucleotides sequence ad 34.9% of amino acid in BaYMV were homologous to those in BaMMV.

Movement of Zucchini yellow mosaic vims Involved in Symptom Severity on Zucchini Squash

  • Zucchini squash (Cucurbita pepo cv. Black Beauty) plants infected with A strain of Zucchini yellow mosaic virus (ZYMV-A) isolated from a hollyhock plant showed systemically severe mosaic symptom, similar to previously established Cu strain of ZYMV. However, initial symptom of squash infected by ZYMV-A strain was generally more severe than those infected by ZYMV-Cu. Using leaf-detachment assay, examination of kinetics of accumulation of the coat protein (CP) in systemic loaves of squash plants showed that CPs of ZYMV-A appeared earlier than those of ZYMV-Cu. However, both ZYMV-A and ZYMV-Cu showed similar kinetics of CP accumulation 7 days post-inoculation. These results indicate that different rates and initial severity of systemic symptom development were due to differences in the rate of movement rather than vims replication.
