국내산 야생9차를 발효정도에 따라 몇 단계의 발효차를 제조하여 일반세균, 젖산균 및 효모 총 8균주에 대한 항균활성을 검색하고, 항균활성 물질의 미생물에 대한 최소저해농도, 열 및 pH에 대한 안정성, 에탄올추출물의 용매분획별 항균 활성을 분석한 결과는 다음과 같다. 국내산 야생녹차의 물추출물과 에탄올추출물의 최소저 해농도는 그람양성균에서는 B. subtilis가 0.2 mg/mL로 가장 낮았고, 그람음성균에서는 P fuorescens가 0.3∼0.5 mg/ mL로 가장 낮았으며, 젖산균과 효모는 1 mg/mL이하의 농도에서는 항균활성이 나타나지 않았다. 발효가 많이 진행된 차일수록 불발효차인 녹차에 비하여 항균활성은 점점 감소하였으며, 에탄올추출물이 물추출물보다 항균효과가 더 높았다. 차 항균활성 물질의 열 및 pH 안정성 은 50∼121$^{\circ}C$ 및 pH 3∼11까지 각 균주의 생육 저해환의 크기가 대조구와 비슷하게 나타나 녹차, 반발효차, 발효차 모두 열과 pH에 안정한 것으로 확인되었다. 차 에탄올추출물을 용매 분획한 결과 물분획물에서 생육억제효과가 가장 크게 나타났고, 그 다음으로는 에틸아세 테이트 분회물, 에테르 분획물의 순으로 항균활성이 높았으며, 헥산 분획물에서는 항균활성을 보이지 않았다. 각 분획물의 항균활성은 발효가 많이 진행된 차일수록 생육 저해환의 크기가 적었으며, 발효도가 높은 국내산 강발효차와 중국산 홍차의 경우 에테르 분획물에서는 항균활성이 나타나지 않았다.
본 연구에서는 한방재료인 감초에서 항균력을 가지는 유효물질을 얻기 위해 추출 조건을 최적화한 후 최적 추출물의 항균 활성과 안정성을 조사하였다. 추출용매를 선정한 후 용매의 농도, 추출 온도, 추출 시간 등을 결정하였다. 감초를 서로 다른 극성을 가진 물, 메탄올, 에탄올, 아세톤, 에틸 아세테이트, 헥산 등의 용매로 추출한 후 항균 활성을 조사함으로써 에탄올을 최적 추출 용매로 선정할 수 있었다. 에탄올 농도별로 추출하였을 때는 무수에탄올일 때 항균 활성이 가장 높았으나 95% 농도와는 유의한 차를 보이지 않았으므로 비용을 고려하여 추출 용매는 95% 에탄올로 선택하였다. 95% 에탄올로 감초를 추출할 때 상온에서 12시간 추출하는 것이 바람직한 것으로 조사되었다. 이후 14종의 Gram(+) 세균, Gram(-) 세균, 효모 등을 이용하여 추출물의 항균활성을 조사하였다. 여러 미생물에 대한 최소 억제 농도(Minimal inhibitory concentration, MIC)는 Gram(-) 세균은 $250{\mu}g/mL$, Gram(+) 세균은 $25{\mu}g/mL$ 이였고 효모는 $50-500{\mu}g/mL$으로 조사 되어, Gram(+) 세균에 대한 탁월한 항균활성을 가졌다. 추출물을 열과 pH 처리한 후 항균활성이 감소하지 않고 잘 유지되는 것으로 보아 상기의 추출물은 열과 pH에는 비교적 안정하다고 판단된다. 감초 에탄올 추출물은 여러 미생물에 대한 항균활성을 가지며 열과 pH 에 대해서도 안정하므로 식품의 천연보존료로서 사용이 가능하리라 사료된다.
호중구의 세포사멸은 자연적으로 일어나지만 여러 외부자극에 의한 신호의 전달에 의하여 증가하거나 지연된다. 본 연구에서는 항암, 항생제로 개발된 프레닐 페놀계인 ascochlorin의 유도체 중에서 백혈구 암의 세포사멸을 유도하는 4-O-methyl-ascochlorin (MAC)이 호중구의 자연 세포사멸 및 지연되는 세포사멸에 어떠한 영향을 미치는가와 그 작용기작을 연구하였다. 호중구의 세포사멸은 사람 말초 혈액으로부터 분리하여 세포 배양 시간에 따라 형태 변화, annexin-V/propidium iodide의 염색, 및 DNA 전기영동 등으로 조사하였다. MAC는 농도 및 시간 의존 형으로 호중구의 세포사멸을 증가시켰다. 그러나 granulocyte macrophage-colony stimulating factor나 lipopolysaccharide 등에 의한 세포사멸의 지연은 MAC에 의하여 부분적으로 억제되었다. MAC에 의한 세포사멸의 유도는 pancaspase, caspase-8 및 caspase-3 억제제인 zVAD-fmk. zIETD-fmk, 및 zDEVD-fmk에 의하여 억제되었으며 procaspase-8과 procaspase-3의 단백질 양도 MAC로 처리한 호중구에서 현저히 감소하였다. 미토콘드리아 막 투과성은 MAC에 의하여 현저히 감소하였으나 zVAD-fmk에 의하여 완전히 봉쇄되지 못하였다. 이들 결과 들은 MAC에 의한 호중구 세포사멸의 증가는 caspase-8 및 caspase-3의 활성을 통하여 일어나지만 미토콘드리아의 막성분에는 영향이 없다는 것을 제시하고 있다.
Acetyl-CoA carboxylase (ACC) catalyzes the first step in fatty acid biosynthesis: the synthesis of malonyl-CoA from acetyl-CoA. As essential regulators of fatty acid biosynthesis and metabolism, ACCs are regarded as therapeutic targets for the treatment of metabolic diseases such as obesity, In ACC, the biotinoyl domain performs a critical function by transferring an activated carboxyl group from the biotin carboxylase domain to the carboxyl transferase domain, followed by carboxyl transfer to malonyl-CoA. Despite the intensive research on this enzyme, only the bacterial and yeast ACC structures are currently available, To explore the mechanism of ACC holoenzyme function, we determined the structure of the biotinoyl domain of human ACC2 and analyze its characteristics using NMR spectroscopy. The 3D structure of the hACC2 biotinoyl domain has a similar folding topology to the previously determined domains from E. coli and P. Shermanii, however, the 'thumb' structure is absent in the hACC2 biotinoyl domain. Observations of the NMR signals upon the biotinylation indicate that the biotin group of hACC2 does not affect the structure of the biotinoyl domain, while the biotin group for E. coli ACC interacts directly with the thumb residues that are not present in the hACC2 structure. These results imply that, in the E. coli ACC reaction, the biotin moiety carrying the carboxyl group from BC to CT can pause at the thumb of the BCCP domain. The human biotinoyl domain, however, lacks the thumb structure and does not have additional non-covalent interactions with the biotin moiety; thus, the flexible motion of the biotinylated lysine residue must underlie the "swinging arm" motion. This study provides insight into the mechanism of ACC holoenzyme function and supports the "swinging arm" model in human ACCs.
장미(Rosa sp. cv Paul's Scarlet) 현탁 배양체의 배지에서는 정상적인 성장 조건하에서 cationic peroxidase (POD)만 관찰되었으나, 효모 세포벽으로부터 추출한 숙주 특이성이 낮은 elicitor를 10 mg glucan/L의 농도로 투여하여 24시간 정도 배양하였을 때 기존의 POD와 뚜렷하게 구분되는 anionic POD가 유도됨을 관찰하였다. Elicitor에 의해 유도되는 prominent한 anionic POD (pI 6.1)와 대조구로서 pI 8.4의 cationic isozyme을 분리한 후 효소학적 성질을 비교하였다. Ammonium sulfate농축, ion exchange chromatography, gel filtration chroma tography를 통하여 약 200배로 농축된 두 효소를 분리하였으며, 이 두 효소의 몇몇 기질에 대한 kinetic parameter들은 매우 의미있는 차이를 보여 주었다. 그 중에서 ferulic acid에 대한 Km은 anionic POD의 경우 4.6 mM, cationic POD는 1.38 mM로 측정되었으며, $\textrm{H}_2\textrm{O}_2$에 대한 Km은 각각 anionic POD가 0.72 mM, cationic isozyme이 0.45 mM로 나타났다. 특히 NADH 산화 반응의 활성은 anionic POD가 cationic POD에 비하여 약 2배 높은 것으로 측정되었다. 0.1 mM coniferyl alcohol의 첨가에 의한 NADH 산화 반응의 억제는 anionic POD fraction에서 약 60% 정도 일어난 반면에 cationic POD fraction에서는 약 15% 정도 일어났다.
본 연구는 유지효모 R. toruloides를 활용하여 적색 파프리카 추출물의 중성지방 억제 효과를 확인하기 위하여 수행되었다. 대조구, 양성대조구, 0.02, 0.1 및 0.5% 적색 파프리카 70% 에탄올 추출물을 첨가한 실험구의 총 지질 함량은 각각 80.45±2.40, 86.61±11.58, 79.82±2.80, 85.30±0.91 및 92.40±12.40 mg/g으로 나타났으며, 유지효모의 중성지방 함량은 각각9.89±0.36, 7.58±1.11, 7.28±0.17, 6.68±0.35 및 5.23±0.67 mg/dL으로 나타났다. 대조구, 양성대조구, 0.02, 0.1 및 0.5% 적색 파프리카 70% 에탄올 추출물을 첨가한 실험구의 지방소적 형성을 관찰한 결과, 적색 파프리카 70% 에탄올 추출물의 첨가농도가 증가함에 따라 대조구에 비해 소량의 형광이 관찰되는 것으로 보아 유지효모 내 생성되는 지방소적이 감소됨을 확인하였다. 대조구, 양성대조구, 0.02, 0.1 및 0.5% 적색 파프리카 70% 에탄올 추출물을 첨가한 실험구의 세포수는 각각 1.90±0.16, 2.17±0.25, 1.85±0.11, 2.28±0.21 및 2.49±0.03×108 cell/mL으로 나타났으며, 유리지방산의 함량은 각각2.44±0.05, 2.71±0.07, 2.46±0.16, 2.52±0.10 및 2.98±0.14 mg/g으로 나타났다. 본 연구결과에서 적색 파프리카 70% 에탄올 추출물의 첨가농도에 따른 유지효모 내 중성지방 억제 효과, 지방소적의 감소, 세포수 및 유리지방산의 함량이 증가한 결과를 통해 적색 파프리카 70% 에탄올 추출물이 중성지방 억제 효과에 유의적인 영향을 미치는 것으로 확인되었으며, 추후 활성 물질의 규명에 관한 추가적인 연구가 필요할 것으로 사료된다.

The aim of this study was to investigate the effect of isolated lactic acid bacteria (LAB) on the quality of high moisture rye silage. Rye forage (Secale cereale L.) was harvested at the heading stage (27.3% of dry matter (DM)) and cut into approximately 3-5 cm lengths. Then, the forage divided into 4 treatments with different inoculants: 1) No additives (CON); 2) Lactobacillus brevis strain 100D8 at a 1.2 × 105 colony-forming unit (cfu)/g of fresh forage (LBR); 3) Leuconostoc holzapfelii strain 5H4 at a 1.0 × 105 cfu/g of fresh forage (LHO); and 4) Mixture of LBR and LHO (1:1 ratio) applied at a 1.0 × 105 cfu/g of fresh forage (MIX). About 3 kg of forage from each treatment was ensiled into a 20 L mini-bucket silo in quadruplicate for 100 days. After silo opening, silage was collected for analyses of chemical compositions, in vitro nutrient digestibilities, fermentation characteristics, and microbial enumerations. The CON silage had the highest concentrations of neutral detergent fiber and acid detergent fiber (p = 0.006; p = 0.008) and a lowest in vitro DM digestibility (p < 0.001). The pH was highest in CON silage, while lowest in LBR and MIX silages (p < 0.001). The concentrations of ammonia-N, lactate, and acetate were highest in LBR silage (p = 0.008; p < 0.001; p < 0.001). Propionate and butyrate concentrations were highest in CON silage (p = 0.004; p < 0.001). The LAB and yeast counts were higher in CON and LHO silages compare to LBR and MIX silages (p < 0.001). However, the mold did not detect in all treatments. Therefore, this study could conclude that L. brevis 100D8 and Leu. holzapfelii strain 5H4 can improve the digestibility and anti-fungal activity of high moisture rye silage.
We studied the effects of Saccharomyces cerevisiae boulardii CNCM I-1079 (LSB) supplemented to lactating sows on reproductive traits and farrowing duration and to piglets from day 7 of life on post-weaning performance and IgG concentration. Ninety-six Landrace × Yorkshire sows started the trial 5 days before the expected farrowing date. Sows were distributed into 2 groups according to parity number and backfat thickness: control (CON: regular lactation diet) and LSB (CON + LSB at 2 × 109 colony forming units [CFU]/kg of feed). Seven days after birth, litters were randomly selected from each group and supplemented creep feed with or without LSB at 2 × 109 CFU/kg. At weaning, piglets from CON sows were shifted to a commercial farm and allocated to 14 pens in groups of 25 piglets/pen according to the creep feed supplemented during lactation. Piglets followed a 3-phase feeding program: creep, pre-starter and starter, with or without LSB at 2 × 109 CFU/kg LSB in creep and pre-starter, and 1 × 109 CFU/kg LSB in starter. The piglets were vaccinated against classical swine fever on days 41 and 72 of life. One day before each vaccination and at the end of the trial, blood samples were collected from 15 randomly selected piglets per treatment and assessed for total IgG. Supplemented sows with non-supplemented litters displayed the lowest backfat thickness loss during lactation (p < 0.05). The LSB supplementation shortened farrowing duration (p < 0.05) and increased feed intake (p < 0.05) during the first week of lactation. The LSB-fed piglets were heavier at the end of creep (p < 0.05), pre-starter (p < 0.05), and the trial (p < 0.05); grew faster during creep (p < 0.05), starter (p < 0.05), and overall (p < 0.05); and displayed an improved feed conversion ratio during creep (p < 0.05). Total IgG content was higher at days 40 (p < 0.05) and 71 (p < 0.05) in LSB-fed piglets. We conclude that supplementing sows with Saccharomyces cerevisiae boulardii CNCM I-1079 from late gestation until weaning shortens farrowing duration, increases feed intake, and minimizes backfat losses during lactation. When supplemented to piglet diet, post-weaning performance is improved. This improvement observed could be linked to a better immune status, as suggested by the higher IgG.
본 연구에서는 Bacillus amyloliquefaciens ISP-5 균주의 최적배지조성을 확인하기 위해 5종의 탄소원(glucose, fructose, sucrose, maltose, corn starch)과 5종의 질소원(yeast extract, peptone, tryptone, soytone, soy bean flour)을 이용하여 각각의 생육을 측정하였다. 그 결과 glucose와 soy bean flour를 각각 최적 탄소원과 질소원으로 확립하였으며 최적 배지를 이용하였을 때 상업용 배지(TSB)에 비해 약 2500배의 생육을 확인할 수 있었다. B. amyloliquefaciens ISP-5 배양액을 상추에 엽면 살포하였을 때 상추의 엽폭과 엽장은 대조군에 비해 8.6%, 12.9%의 증가하였으며, 생체중과 건물중은 대조군에 비해 각각 24.2%, 23.9% 증가하였다. 이러한 결과들을 종합해보았을 때 B. amyloloiquefaciens ISP-5 균주를 이용한 미생물 제제는 상추용 식물생장촉진제로써 활용할 수 있을 것이라 기대된다.
