고온성(高溫性) 효모(酵母)에 의(依)한 Ethanol 발효(醱酵) (Studies on the Thermophilic Yeast for Ethanol Fermentation)

  • This experiment carried out to obtain the thermophilic yeast, suitable for ethanol fermentation, and two usable strain were isolated. And microbial characteristics of these strains were investigated, and ethanol fermentation tested. The results obtained were as follows; 1. The selected yeasts were identified D-71 with Saccharomyces cerevisiae, and J-515 with Saccharomyces fermentati. 2. The strains D-71 and J-515 were showed the highest ethanol fermentation activity in the crushed corn mash of high concentration at $35^{\circ}C$, and showed the slightly lower at $40^{\circ}C$ than in the case of $35^{\circ}C$. 3. The strains D-71 and J-515 were showed the very higher ethanol fermentation activity than that of compared strain at $35^{\circ}C$ and $40^{\circ}C$, and at these temperature, fermentation period was a little bit of short. 4. On fermentation test using D-71 and J-515, the residual total sugar in the mash was very lower at $35^{\circ}C$.

Antioxidant Activity of Indigenous Trametes Species in Korea

  • The fungal genus Trametes is globally distributed and comprises various wood-decay species, including the well-known medicinal mushroom Trametes versicolor, a popular remedy in traditional Asian medicine. Trametes species produce antioxidants, which have a wide range of health benefits. The pressent study evaluated seven indigenous Trametes species from Korea, which were cultivated in three different media (dextrose-yeast extract, DY; malt extract-yeast extract, MY; malt extract broth, MEB) and tested for antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assays. We found that the medium consumption rate did not significantly differ between the media and among the strains (72-76%). However, the T. versicolor strains had a relatively low consumption rate (14-65%). The 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) (ABTS) tests demonstrated that culture filtrates of T. cf. junipericola, T. orientalis, T. suaveolens, and T. versicolor possessed antioxidant activity against damage from free radicals. In particular, T. cf. junipericola (DY) and T. versicolor (MY) had >80% activity in the DPPH and ABTS assays, compared with that of the positive control (ascorbic acid). Thus, our study identified promising candidates with substantial antioxidant activity among the indigenous strains of Trametes spp. from Korea.

맥주의 발효과정에서 효모와 발효온도가 황화수소의 발생에 미치는 영향 (Effects of Yeast Strains and Fermentation Temperatures in Production of Hydrogen Sulfide During Beer Fermentation)

  • Ale과 lager 효모를 이용하여 맥주발효 실험한 결과, lager 효모는 발효가 활발하게 일어나는 발효 초기에 많은 양의 황화수소를 발생하였으며 ale 효모보다 더욱 많은 양의 황화수소가 발생되었다. 효모의 증식이 활발한 발효 초기에 많은 양의 황화수소가 만들어지는 것을 보아 효모의 증식과 황화수소의 생성 사이에는 상관관계가 있음을 확인하였다. 온도 차이에 따른 실험 결과에서, lager 효모의 경우에는 발효 온도가 높은 것이 발효율이 더 높아 발효 시간을 줄일 수 있는 장점은 있으나, 많은 양의 황화수소가 발생되는 것을 알 수 있었다. 또한 APL은 $15^{\circ}C$에서 발효시켰을 때, 발효 끝 부분에서 많은 양의 황화수소가 발생되었는데, 이것은 맥주에 좋지 않은 냄새의 원인이 되므로 장시간의 숙성기간을 필요로 한다는 것을 의미한다. EA의 경우에는 낮은 온도에서 오히려 더 많은 양의 황화수소가 생성이 되었으나, HA의 경우에는 lager 효모에서와 같이 높은 온도에서 더 많은 양의 황화수소가 생성되었는데, 이는 각 효모마다 $H_2S$를 생성하기에 적합한 온도가 있음을 의미한다.

액체배양에서의 홍국균의 생장, 색소 및 monacolin K 생산 특성 (Characteristics of Growth, Pigment and Monacolin K Production by Monascus strains in Liquid Culture)

  • The hyphal growth, production of color pigments and monacolin K by Monascus strains were investigated in liquid medium. Thirty five different strains were collected and cultured in potato dextrose yeast extract broth (PDYB), potato dextrose broth (PDB) and malt extract broth (MEB) medea at $25^{\circ}C$ for 7 days. The growth rates of most of strains were highest in PDYB medium. Growth rate as well as pigment production were influenced by suspension conditions of mycelia during liquid cultivation. Most of strains producing monacolin K corresponded to strains producing red pigment highly and showing more pH changes of liquid media. Monacolin K produced from strains was detected in culture broth as well as mycelia. Any citrinin was not detected in monacolin K producing strains. These results imply that the selection of the strains producing red pigment highly and showing more pH changes in liquid cultivation could be applied for primary screening of Monascus strains for preparation of red mold rice.

Optimal Conditions for Propagation in Bottom and Top Brewing Yeast Strains

  • The method of yeast propagation has an influence on yeast physiology, fermentation ability, flocculation rate, and taste stability of beer. In order to find optimal conditions for propagation, several parameters were investigated in combinations. The bottom brewing yeast grown at $10^{\circ}C$ indicated that a higher flocculation capacity during the $1^{st}$ fermentation. However, the taste stability and the aroma profile were not affected by parameters of propagation investigated. The beer quality was rather affected by storage duration. In addition, a correlation between tasting and chemiluminescence was found at the beer, which was produced using bottom brewing yeast. The propagation at $10-25^{\circ}C$ with addition of zinc ion indicated the best condition to improve fermentation ability, flocculation rate, and filterability for bottom brewing yeast, whereas the propagation at $30^{\circ}C$ with addition of zinc ion showed the best condition to increase fermentation ability for top brewing yeasts.

유류분해 미생물의 특성 및 제제화 가능성 평가

  • The various microbial tests were performed to determine bioremediation agent capacity for eight strains isolated from the oil contaminated regions. Two tests for isolated strains were conducted such as cell hydrophobicity and emulsifying activity. The biodegradation of SHM (saturated hydrocarbon mixture) and AHM (aromatic hydrocarbon mixture) with the strains also was carried out. The strains having higher cell hydrophobicity and emulsifying activity degraded petroleum oil effectively. The degradation capacity for SHM was represented more than 90% in YS-7 and WLH-1 of isolated strains, and KH3-2 were capable of degrading AHM. Especially, WLH-1 as yeast was shown more than two or three times in the degradation capacity of automobile engine lubricants and the biomonitoring results of contaminated soil for residual oil degrading test showed that the hydrocarbon biodegradation was increased in the second treatment by this strain.

효모에 따른 약주의 품질특성 1. 분리균주의 동정 및 휘발성 향기성분 (Characterization of Yakju Prepared with Yeasts from Fruits 1. Volatile Components in Yakju during Fermentation)

  • This study was carried out to obtain the basic information for improving the flavor quality of yakju. Three kinds of yakju were prepared with different yeast strains to investigate the effects of those strains on flavor components. A total of 23 strains were isolated from fruits such as apple, pear, persimmon and citron and two strains were excellent in producing ethanol and flavors. They were identified as Saccharomyces cerevisiae S 2 and Saccharomyces cerevisiae S 6 from morphological cultural test and physiological quality. Yakju A, B and C were prepared with S 2, S 6 and Saccharomyces cerevisiae IFO 1950, respectively. Flavor components of yakju were analyzed by gas chromatography and mass spectr ometry. A total of 57 peaks were detected and 13 compound were identified. They were 4 alcohol, 2 esters, 7 acids and miscellaneous compounds.

A Novel Approach to Investigating Protein/Protein Interactions and Their Functions by TAP-Tagged Yeast Strains and its Application to Examine Yeast Transcription Machinery

  • Tandem affinity purification (TAP) method combined with LC-MS/MS is the most accurate and reliable way to study the interaction of proteins or proteomics in a genome-wide scale. For the first time, we used a TAP-tag as a mutagenic tool to disrupt protein interactions at the specific site. Although lots of commonly used mutational tools exist to study functions of a gene, such as deletional mutations and site-directed mutagenesis, each method has its own demerit. To test the usefulness of a TAP-tag as a mutagenic tool, we applied a TAP-tag to RNA polymerase II, which is the key enzyme of gene expression and is controlled by hundreds of transcription factors even to transcribe a gene. Our experiment is based on the hypothesis that there will be interrupted interactions between Pol II and transcription factors owing to the TAP-tag attached at the C-terminus of each subunit of Pol II, and the abnormality caused by interrupted protein interactions can be observed by measuring a cell-cycle of each yeast strain. From ten different TAP-tagged strains, Rpb7- and Rpb12-TAP-tagged strains show severe defects in growth rate and morphology. Without a heterodimer of Rpb4/Rpb7, only the ten subunits Pol II can conduct transcription normally, and there is no previously known function of Rpb7. The observed defect of the Rpb7-TAP-tagged strain shows that Rpb7 forms a complex with other proteins or compounds and the interruption of the interaction can interfere with the normal cell cycle and morphology of the cell and nucleus. This is a novel attempt to use a TAP-tag as a proteomic tool to study protein interactions.

충남 예산군 신암면 일대 과수원의 과일과 꽃들로부터 효모의 분리 및 분포 특성 (Isolation and Diversity of Yeasts from Fruits and Flowers of Orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, Korea)

  • 충청남도 예산군 신암면 일대 과수원들의 과일과 꽃에서 효모들을 분리한 후 이들의 26S rDNA의 D1/D2 부위의 염기서열을 결정한 다음 NCBI의 BLAST 데이터베이스에 등록되어 있는 효모들과의 분자생물학적 유연관계를 비교, 분석하여 동정하였다. 2013년 4월부터 10월 사이의 과일 19점으로부터 모두 25종의 효모들을 48균주 분리하였고 Pichia kluyveri, Hanseniaspora uvarum, Rhodotorula mucilaginosa, Torulaspora delbrueckii 등 4종의 효모 등이 많이 분포하고 있었다. 과수원 주위의 꽃 39점에서 48종의 효모 등을 108균주 분리하였고 이 중 Microstroma juglandis와 Pseudozyma aphidis가 가장 많이 분리되었다.

Bioconversion of Ginsenosides from Red Ginseng Extract Using Candida allociferrii JNO301 Isolated from Meju

  • Red ginseng (Panax ginseng), a Korean traditional medicinal plant, contains a variety of ginsenosides as major functional components. It is necessary to remove sugar moieties from the major ginsenosides, which have a lower absorption rate into the intestine, to obtain the aglycone form. To screen for microorganisms showing bioconversion activity for ginsenosides from red ginseng, 50 yeast strains were isolated from Korean traditional meju (a starter culture made with soybean and wheat flour for the fermentation of soybean paste). Twenty strains in which a black zone formed around the colony on esculin-yeast malt agar plates were screened first, and among them 5 strains having high ${\beta}$-glucosidase activity on p-nitrophenyl-${\beta}$-D-glucopyranoside as a substrate were then selected. Strain JNO301 was finally chosen as a bioconverting strain in this study on the basis of its high bioconversion activity for red ginseng extract as determined by thin-layer chromatography (TLC) analysis. The selected bioconversion strain was identified as Candida allociferrii JNO301 based on the nucleotide sequence analysis of the 18S rRNA gene. The optimum temperature and pH for the cell growth were $20{\sim}30^{\circ}C$ and pH 5~8, respectively. TLC analysis confirmed that C. allociferrii JNO301 converted ginsenoside Rb1 into Rd and then into F2, Rb2 into compound O, Rc into compound Mc1, and Rf into Rh1. Quantitative analysis using high-performance liquid chromatography showed that bioconversion of red ginseng extract resulted in an increase of 2.73, 3.32, 33.87, 16, and 5.48 fold in the concentration of Rd, F2, compound O, compound Mc1, and Rh1, respectively.
