Cloning and Characterization of a Multidomain GH10 Xylanase from Paenibacillus sp. DG-22

  • The xynC gene, which encodes high molecular weight xylanase from Paenibacillus sp. DG-22, was cloned and expressed in Escherichia coli, and its nucleotide sequence was determined. The xynC gene comprised a 4,419bp open reading frame encoding 1,472 amino acid residues, including a 27 amino acid signal sequence. Sequence analysis indicated that XynC is a multidomain enzyme composed of two family 4_9 carbohydrate-binding modules (CBMs), a catalytic domain of family 10 glycosyl hydrolases, a family 9 CBM, and three S-layer homologous domains. Recombinant XynC was purified to homogeneity by heat treatment, followed by Avicel affinity chromatography. SDS-PAGE and zymogram analysis of the purified enzyme identified three active truncated xylanase species. Protein sequencing of these truncated proteins showed that all had identical N-terminal sequences. In the protein characterization, recombinant XynC exhibited optimal activity at pH 6.5 and $65^{\circ}C$ and remained stable at neutral to alkaline pH (pH 6.0-10.0). The xylanase activity of recombinant XynC was strongly inhibited by 1 mM $Cu^{2+}$ and $Hg^{2+}$, whereas it was noticeably enhanced by 10 mM dithiothreitol. The enzyme exhibited strong activity towards xylans, including beechwood xylan and arabinoxylan, whereas it showed no cellulase activity. The hydrolyzed product patterns of birchwood xylan and xylooligosaccharides by thin-layer chromatography confirmed XynC as an endoxylanase.

Characterization of the Wild-Type and Truncated Forms of a Neutral GH10 Xylanase from Coprinus cinereus: Roles of C-Terminal Basic Amino Acid-Rich Extension in Its SDS Resistance, Thermostability, and Activity

  • A neutral xylanase (CcXyn) was identified from Coprinus cinereus. It has a single GH10 catalytic domain with a basic amino acid-rich extension (PVRRK) at the C-terminus. In this study, the wild-type (CcXyn) and C-terminus-truncated xylanase ($CcXyn-{\Delta}5C$) were heterologously expressed in Pichia pastoris and their characteristics were comparatively analyzed with aims to examine the effect of this extension on the enzyme function. The circular dichorism analysis indicated that both enzymes in general had a similar structure, but $CcXyn-{\Delta}5C$ contained less ${\alpha}-helices$ (42.9%) and more random coil contents (35.5%) than CcXyn (47.0% and 32.8%, respectively). Both enzymes had the same pH (7.0) and temperature ($45^{\circ}C$) optima, and similar substrate specificity on different xylans. They all hydrolyzed beechwood xylan primarily to xylobiose and xylotriose. The amounts of xylobiose and xylotriose accounted for 91.5% and 92.2% (w/w) of total xylooligosaccharides (XOS) generated from beechwood by CcXyn and $CcXyn-{\Delta}5C$, respectively. However, truncation of the C-terminal 5-amino-acids extension significantly improved the thermostability, SDS resistance, and pH stability at pH 6.0-9.0. Furthermore, $CcXyn-{\Delta}5C$ exhibited a much lower $K_m$ value than CcXyn (0.27 mg/ml vs 0.83 mg/ml), and therefore, the catalytic efficiency of $CcXyn-{\Delta}5C$ was 2.4-times higher than that of CcXyn. These properties make $CcXyn-{\Delta}5C$ a good model for the structure-function study of $({\alpha}/{\beta})_8$-barrel-folded enzymes and a promising candidate for various applications, especially in the detergent industry and XOS production.

  • 1. A. ficuum의 genomic library 검색을 통해 Axe 유전자를 포함하고 있는 5.0 kb의 XbaI DNA 절편을 cloning 했다. Cloning 된 절편의 부분 염기서열 결정 결과 약 1.4 kb의 AXE coding 부위를 확인했으며, cDNA cloning과 그 염기서열의 결정을 통해 AXE coding 부위 내에는 두 개의 intron 이 존재함이 확인되었다. 2. AXE coding 부위의 아미노산 잔기 서열 검색 결과 A. awamori의 AXE와 약 92%의 상동성과 95%의 유사성이 있음이 확인 되었다. 3. 약 900 kb의 AXE의 cDNA를 yeast의 YEp352 vector의 GAL1 promoter의 전사 방향으로 cloning한 후 발현시킨 결과 형질전환체에서 acetyl esterase 활성을 확인했으며, 활성도는 숙주균주에 비해 약 4-5배의 높은 OD unit로 나타내는 것을 확인할 수 있었다.

Thermostable Xylanase from Marasmius sp.: Purification and Characterization

  • We have screened 766 strains of fungi from the BIOTEC Culture Collection (BCC) for xylanases working in extreme pH and/or high temperature conditions, the so-called extreme xylanases. From a total number of 32 strains producing extreme xylanases, the strain BCC7928, identified by using the internal transcribed spacer (ITS) sequence of rRNA to be a Marasmius sp., was chosen for further characterization because of its high xylanolytic activity at temperature as high as $90^{\circ}C$. The crude enzyme possessed high thermostability and pH stability. Purification of this xylanase was carried out using an anion exchanger followed by hydrophobic interaction chromatography, yielding the enzyme with >90% homogeneity. The molecular mass of the enzyme was approximately 40 kDa. The purified enzyme retained broad working pH range of 4-8 and optimal temperature of $90^{\circ}C$. When using xylan from birchwood as substrate, it exhibits $K_m$ and $V_{max}$ values of $2.6{\pm}0.6\;mg/ml$ and $428{\pm}26\;U/mg$, respectively. The enzyme rapidly hydrolysed xylans from birchwood, beechwood, and exhibited lower activity on xylan from wheatbran, or celluloses from carboxymethylcellulose and Avicel. The purified enzyme was highly stable at temperature ranges from 50 to $70^{\circ}C$. It retained 84% of its maximal activity after incubation in standard buffer containing 1% xylan substrate at $70^{\circ}C$ for 3 h. This thermostable xylanase should therefore be useful for several industrial applications, such as agricultural, food and biofuel.

Purification and Characterization of Two Endoxylanases from an Alkaliphilic Bacillus halodurans C-1

  • Two endoxylanases from an alkaliphilic bacterium, Bacillus halodurans C-1, were purified 3.8- and 7.9- fold with specific activities of 9.4 and 19.8U/mg protein, respectively. The molecular masses of both purified enzymes were 23 and 47 kDa, respectively, and 23 kDa xylanase I (Xyl I) exhibited an optimum pH at 7.0, whereas 47 kDa xylanase II (Xyl II) showed a broad pH range of 5.0 to 9.0. The temperature optima of both xylanases were $60^{\circ}C\;and\;70^{\circ}C$, respectively. Both were stable in the pH range of 6.0 to 9.0 and 5.0 to 10.0, respectively, and they were stable up to $60^{\circ}C\;and\;70^{\circ}C$, respectively. The $K_m\;and\;V_{max}$ of Xyl I were 4.33mg/ml and $63.5{\mu}mol/min/mg$, respectively, whereas Xyl II had a $K_m$ value of 0.30 mg/ml and $V_{max}$ of $210{\mu}mol/min/mg$. Both xylanases hydrolyzed xylans from birchwood, oat spelt, and larchwood. However, they showed different modes of action; a series of xylooligosaccharides larger than xylotriose were released as the major products by Xyl I, whereas xylobiose and xylotriose were the main products by Xyl II. The maximum synergistic action of the two enzymes on hydrolysis of xylan was 2.16 with the ratio of Xyl I to Xyl II at 1:9.

Production and Location of Xylanolytic Enzymes in Alkaliphilic Bacillus sp. K-1

  • The production and location of xylanolytic enzymes in alkaliphilic Bacillus sp. K-1, isolated from the wastewater treatment plant of the pulp and paper industry, was studied. When grown in alkaline xylan medium, the bacteria produced xylanolytic enzymes such as xylanase, $\beta$-xylosidase, arabinofuranosidase, and acetyl esterase. Two types of xylanases (23 and 45 kDa) were found to be extracellular, but another type of xylanase (35 and/or 40 kDa) was detected as pellet-bound that was eluted with 2% triethylamine from the residual xylan of the culture. The xylanases were different in their molecular weight and xylan-binding ability. Arabinofuranosidase and $\beta$-xylosidase were found to be intracellular and extracellular, respectively, and acetyl esterase was found to be extracellular. The extracellular xylanolytic enzymes effectively hydrolyzed insoluble xylan, lignocellulosic materials, and xylans in kraft pulps.

양송이 수확 후 배지로부터 분리한 Bacillus subtilis AB-55가 생산하는 xylanase의 특성 (Xylanase properties of Bacillus subtilis AB-55 isolated from waste mushroom bed of Agaricus bisporus)

  • A bacterium AB-55, isolated from waste mushroom bed of Agaricus bisporus in Sukseong-myeon, Buyeo-gun, Chungcheongnam-do, Korea, was screened onto xylan agar congo-red plate by the xylanolysis method and was used to produce an xylanase in shaker buffle flask cultures containing oat spelt xylans. The phylogenetic analysis using 16S rRNA gene sequence data showed that the strain AB-55 had the highest homology (99.0%) with Bacillus subtilis and it was named as Bacillus subtilis AB-55. A xylanase was purified by ammonium sulfate precipitation (50~80%), gel filtration on sephacryl S-300, and ion exchange chromatography on DEAE sepharose FF. The molecular weight of the xylanase was estimated as 44 kDa by SDS-PAGE. Optimal pH and temperature for the xylanase activity was pH 7 and $50^{\circ}C$, respectively. N-terminal amino acid sequence of the enzyme was identified as Ser-Ala-Val-Lys-His-Gly-Ala-Ile-Val-Phe. The substrate specificity of the enzyme exhibited that it hydrolyzed efficiently oat spelt xylan as well as beechwood xylan, but showed no activity against Avicel and carboxymethyl clellulose (CMC). The enzyme activity was enhanced by $Fe^{2+}$ and $Mn^{2+}$ whereas was entirely inhibited by $Hg^+$.

  • Xylan의 효소적 가수분해는 고부가가치 기능성 물질 또는 바이오에너지 생산을 위한 발효성 당을 얻는 가장 유용한 방법 중 하나이다. endo-${\beta}$-Xylanase는 xylan 주사슬 내부의 ${\beta}$-1,4-결합을 가수분해하여 xylobiose, xylotriose를 포함한 다양한 XOS를 생산하는 핵심 효소이다. 이들 효소 중에서 glucuronoxylanase GH30은 methylglucuronic acid가 측쇄에 수식된 xylan에 특이적으로 작용한다. 본 연구에서는 Paenibacillus amylolyticus KCTC 3005에서 유래한 2종의 xylan 가수분해효소(PaXN_10과 PaGuXN_30) 유전자를 클로닝하고, Escherichia coli에서 각각 발현시켰다. PaXN_10 (38.7 kDa)은 ${\beta}$-xylanase GH10 계열, PaGuXN_30 (58.5 kDa)은 glucuronoxylanase GH30에 해당하는 효소이며, $50^{\circ}C$와 pH 7.0에서 최대 활성을 나타내었다. 가수분해 특성 연구를 통해 P. amylolyticus가 목질계 glucuronoxylan을 분해하는 효소 시스템을 제안하였다. 세포 외로 분비되는 PaGuXN_30은 glucuroxylan을 가수분해하여 methylglucuronic acid 측쇄를 가지는 다양한 aldouronic acid mixtures를 생성하며, 이러한 분해산물은 세포 내로 이동하여 PaXN_GH10에 의해 xylose, xylobiose와 같은 저분자 XOS로 분해되어 세포 내 대사경로에 이용될 수 있다. 또한 이들 효소의 가수분해특성을 이용하여 다양한 탄수화물 소재 생산이 가능할 것으로 기대한다.

재조합 균주 Escherichia coli가 생산하는 Bacillus stearothermophilus Exo-xylanase의 정제 및 특성 (Purification and Characterization of Exo-xylanase from Escherichia coli Cells Harboring the Recombinant Plasmid pMGl)

  • Bacillus stearothermophilus exo-xylanase 유전자 DNA가 삽입된 재조합 plasmid pMG1을 가지고 있는 E.coli JM109 exo-xylanase 생산 최적 배양 조건, 생산 효소의 정제 및 정제 효소의 특성 등을 조사 연구하였다. 상기 재조합 E.coli 균주는 0.5 fructose, 0.5 yeast extract, 1.0 tryptone 및 1.0 sodium chloride가 함유된 배지에서 약 10시간 배양했을 때 최대량의 효소를 생산하였으며 생산효소의 94는 세포내에 존재하는 것으로 분석되었다. 생산 효소는 ammonium sulfate 분획, ion exchange chromatography 및 gel filtration 등의 과정을 거쳐 단일 단백질로 정제하였으며 정제 효소는 pH 6.0과 $45^{\circ}C$에서 가장 높은 효소 활성을 나타내었다.또한 1mM $Ca^{2+}$$Co^{2+}$ 이온의 첨가는 각각 약 25% 정도의 활성화 효과를 나타내는 반면, 본 효소의 pNPX에 대한 $K_{m}$은 2.75mM, pl값을 4.7, 그리고 분자량은 gel-filtration 법으로는 약 200,000dal., SDS-polyacrylamide gel 전기영동법으로는 약 66,000dal 으로 측정되어 세 개의 동일한 subunit로 구성된 효소 단백질인 것으로 추정되었다. 본 정제 효소는 xylobiose, xylotrioxe 및 xylotetraose 등의 xylo-oligosaccharide를 효과적으로 분해함은 물론이고, 분해율은 낮으나 birchwood xylan, larchwood xylan 및 oatspelt xylan 등의 xyland에도 작용, xylose 생산을 확인함으로써 본 효소는 그 예가 극히 드문 bacterial exo-xylanase인 것으로 분류되었다.

Deinococcus geothermalis의 Xylan 최적 분해조건 및 분해산물 분석 (Characteristics of Xylan Degradation and HPLC Analysis of Hydrolyzed Xylans by Deinococcus geothermalis)

  • 대표적인 중호열성 방사선저항성 미생물인 Deinococcus geothermalis에는 다른 Deinococcus 속과 비교해 당대사 작용에 관여하는 유전자가 풍부하게 존재하는 특징이 있다. 본 연구에서는 D. geothermalis를 이용하여 xylan의 최적 분해조건을 확인하였고, beechwood xylan, birchwood xylan 및 oat spelt xylan의 최종 분해산물을 HPLC를 이용하여 분석하였다. 기질의 종류에 따른 당화율을 비교한 결과 beechwood xylan, birchwood xylan 그리고 oat spelt xylan 순서로 당화율이 높게 나타났다. D. geothermalis를 이용한 xylan의 최적 분해조건인 $40^{\circ}C$, pH 8.0 그리고 마그네슘 이온을 첨가하였을 때 당화율이 7.5배 증가하였다. Beechwood xylan과 birchwood xylan의 최종 분해산물은 xylose, xylobios, xylotriose, xylotetraose, xylopentaose, 그리고 xylohexalose였으며, xylose의 함량이 가장 높았다. 또한 oat seplt xylan의 최종 분해산물은 xylose, xylopentaose 그리고 xylohexalose가 생성되었다. Xylan의 효율적인 당화를 위하여 전처리 방법으로서 방사선조사를 하였고, 방사선조사 후 D. geothermalis에 의한 beechwood xylan, birchwood xylan 그리고 oat spelt xylan의 당화율이 증가하였다. 본 연구를 통하여 D. geothermalis 및 방사선을 이용한 전처리 방법이 xylooligosaccharides를 생산하는데 유용함을 확인하였다.
