3,9-Diferuloyl-6-oxopterocarpen (Tensolin-F$^\circledR$): a Novel Anti-wrinkle Agent for Cosmeceuticals (새로운 주름개선 소재: 3,9-diferuloyl-6-oxopterocarpen (Tensolin-F$^\circledR$)의 개발과 화장품 응용)

  • As a novel anti-wrinkle agent, 3,9-diferuloyl-6-oxopterocarpen (Tensolin-F$^{(R)}$) has been synthesized and its anti-aging effects have been investigated. In the present study, to investigate the relationship between aging and Tensolin-F$^{(R)}$, we examined its effect on scavenging activities of radicals and reactive oxygen species (ROS), in vitro inhibition activity of matrix-metalloproteinase (MMP) and expression of UVA-induced MMPs in human dermal fibroblasts (HDF). Tensolin-F$^{(R)}$ was found to show activities of scavenging radicals and ROS with the $IC_{50}$/ values of 0.2 mM and 0.95 mM against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and superoxide radicals, respectively, in the xanthine/xanthine oxidase system. Fluorometric assays for the proteolytic activities of MMP-l (collagenase) were performed using fluorescent collagen substrates. Tensolin-F$^{(R)}$ inhibited the activities of MMP-l in a dose-dependent manner and the $IC_{50}$/ values calculated from semi-log plots were 0.025 mM. Also, UVA induced MMP-1 expression was reduced 85% by treatment with Tensolin-F$^{(R)}$ at 0.8 uM, which was reduced dose-dependent manner. The results of clinical study showed that 4.8 mM Tensolin-F$^{(R)}$ treated group reduced wrinkle significantly compared with placebo treated group (P 〈 0.05). Taken together, these result suggest that Tensolin-F$^{(R)}$ act as an anti-wrinkle agent by taking effects to antioxidation and reducing UVA-induced MMP-l production.-l production.

Enzyme Inhibition Activities of Ethanol Extracts from Germinating Rough Rice (Oryza sativar L.) (발아기간에 따른 벼(Oryza sativa L.)의 부위별 효소저해활성)

  • This study investigated the enzyme inhibitory effects of ethanol extracts from the different parts of rough rice (Oryza sativar L.) from its germination period. Rough rice was germinated at $37^{\circ}C$ for 6 days, then separated into hull+sprout and brown rice. ${\alpha}$-Glucosidase inhibitory activity had the highest value (39.38%) in hull+sprout extracts after 5 days of germination. ${\alpha}$-Amylase and DPP-4 inhibitory activity had the highest values (75.32% and 47.77%, respectively) in hull+sprout extracts after germination for 5 days. ACE inhibitory activity of hull+ sprout extracts increased from 43.16% at the beginning to 58.60% at 5 days, while brown rice extracts increased this activity from 0.88% at the beginning to 14.50% at 4 days. The xanthine oxidase inhibitory activity of hull+ sprout extracts increased from 62.02% at the beginning to 64.49% at one day, and then decreased. Lipase inhibitory activity had its highest value (55.17%) in hull+sprout extracts after germination for 5 days. These results indicate that the optimal germination period for increasing enzyme inhibitory activities may be 5 days, and that hull+sprout extracts have a higher enzyme inhibition activity than brown rice.

Qualities and Antioxidant Activity of Lactic Acid Fermented-Potato Juice (젖산 발효 감자주스의 품질 특성 및 항산화 활성)

  • This study was conducted to investigate the chemical properties and functionality of probiotic potato juice fermented by Lactobacillus casei. Free sugar content (especially glucose) of potatoes decreased by fermentation, but organic acid contents increased by fermentation. Although the free amino acid content of Superior juice significantly decreased after fermentation, Haryeong significantly increased after fermentation. ${\gamma}$-Aminobutyric acid, a functional amino acid, was detected at high levels in all samples and slightly decreased with fermentation, but not significantly. The total polyphenol content of potato juice showed insignificant changes in all samples by fermentation. The hydroxyl radical scavenging activity of all samples was more than 90%, and most of the activity was maintained after fermentation. The nitrite scavenging ability of all samples greatly decreased with fermentation; however a SOD-like activity slightly increased with fermentation, except for Haryeong. There was a significant xanthine oxidase inhibitory effect in fresh potato juice (more than 45%) and a low loss by fermentation. From our results, most of the chemical properties and functionality of potato juice are maintained after fermentation, although free sugar content and nitrite scavenging activity decline. Thus probiotic potato juice fermented by lactic acid could be used as a functional beverage.

Evaluation of Physiological Activities of Cnidium officinale Makino Extracts with Different Solvents (추출용매에 따른 천궁(Cnidium officinale Makino) 추출물의 생리활성 평가)

  • The objective of this study was to investigate the physiological activities of Cnidium officinale Makino extracts on extraction solvents. The yield of ethanol extract, 87.54%, was higher than that of the hot water extract(83.06%) and of the methanol extract(78.32%). In the inhibitory activity of xanthine oxidase, the ethanol extracts and methanol from Cnidium officinale Makino appeared to show significantly higher activity than hot water extract at $1,000{\mu}g/mL$. The DPPH radical scavenging activities of the ethanol and the methanol extracts at $50-2,000{\mu}g/mL$ were 19.96-89.01% and 19.41-88.21%, respectively. The SOD-like activities of all the extracts improved with an increase in the treatment concentration. Both collagenase and elastase inhibitory activities were shown higher from the ethanol and methanol extract compared to that of hot water extract. The tyrosinase inhibitory activity of ethanol extract, 12.04-73.85%($50-2,000{\mu}g/mL$), was higher than that of the other extracts. Taken together, these data suggest that Cnidium officinale Makino extract is effective in whitening and anti-wrinkle effect, thus it might strongly be considered as potential functional cosmetic components.

Study on the Anti-oxidant Effect of Pinus rigida Mill. inner Bark Extracts (리기다소나무(Pinus rigida Mill.) 내수피 추출물의 항산화효과에 관한 연구)

  • The aim of the study was to assess the cosmeceutical activity of Pinus rigida Mill. and it is possible that can be used as a cosmetic ingredient for application of cosmetic industries. The concentration of total phenolic compound of P. rigida water soluble fraction and P. rigida ethyl acetate soluble fraction showed 47 mg/L and 601 mg/L respectively. In the result of DPPH scavenging radical activity, P. rigida ethyl acetate soluble fraction showed 86% and it was similar to BHA effect at 10 ppm concentration. Xanthine oxidase inhibition of P. rigida water soluble fraction and P. rigida ethyl acetate soluble fraction were 76.3% and 80.5% at 500 ppm, respectively. In the result of tyrosinase inhibition effect related to skin-whitening, P. rigida water and ethyl acetate soluble fraction showed 42%, 10.9% at a 1,000 ppm. In these results, P. rigida has a great potential as a cosmeceutical ingredient with a natural anti-oxidant source.

A Study on the Application of New Cosmetic Materials of Whitening Effect and the Physiological Activities of Chestnut Inner Shell (율피의 생리활성 몇 미백효과를 이용한 화장품신소재에 관한 연구)

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

Antioxidant Effects and Melanin Inhibitory Effect of Natural Pimpinella komarovii Extracts in Jeju Island (제주도 자생 노루참나물 (Pimpinella komarovii) 추출물의 항산화 효과 및 멜라닌 억제 효과)

  • We investigated several biological activities using the ethanol extract and its fractions from Pimpinella komarovii leaves to evaluate the usefulness of its extract as a functional biomaterial. The ethanol extract showed antioxidant activities, such as DPPH scavenging activity $(IC_{50}=231.8{\mu}g/m{\ell})$. superoxide scavenging activity $(IC_{50}=23.6{\mu}g/m{\ell})$, and xanthine oxidase inhibitory activity $(IC_{50}=587.8{\mu}g/m{\ell})$. Its EtOAc fraction showed the strongest antioxidant activities among several fractions. The inhibitory effect of ethanol extract on tyrosinase activity was higher than water fraction. When $50{\mu}g/m{\ell}$ of EtOAc fraction was applied, the inhibition ratio of tyrosinase activity was much higher (42%) than that of melasolv. The EtOAc fraction also showed higher inhibitory effect on melanogenesis in Melan-a cells. The n-hexane and EtOAc fractions dose-dependently inhibited the NO production in a RAW 264.7 cells. These results suggest that extract of Pimpinella komarovii could be used as functional biomaterial in developing a skin whitening agent having the antioxidant activity.

Antioxidant Activity and Physiological Function of the Anomala albopilosa Extracts (청동풍탱이(Anomala albopilosa)추출물의 항산화성 및 생리기능)

  • We analyzed antioxidant and physiological activities to investigate the functional effects of ethanol extracts of Anomala albopilosa imago and A. albopilosa larva. In order to effectively screen for anti-inflammatory agents, we first investigated the antioxidant activities such as DPPH radical scavenging capacity, superoxide radical scavenging capacity, xanthine oxidase inhibitory activity, and nitric oxide scavenging capacity of the A. albopilosa extracts. By the screening system, we found that A. albopilosa extracts had antioxidant activity which increased with increments of the extract concentration. Moreover, we examined the inhibitory effect of the A. albopilosa extracts on the production of anti-inflammatory factors that the nitric oxide (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and prostaglandin $E_2\;(PGE_2)$ production activated with LPS ($1{\mu}g/mL$) in murine macrophage cell line RAW 264.7. A. albopilosa extracts potentially inhibited the iNOS and COX-2 in a dose-dependent manner. The inhibition of iNOS activity was correlated with the decrease in nitrite levels. Additionally, the $PEG_2$ production is markedly inhibited after a treatment with the A. albopilosa extracts.

Enhancement of the Antioxidant and Anticancer Activities of Berberis koreana Bark by Using a Low Temperature and High-Pressure Extraction Process (저온고압 추출공정을 이용한 매자나무 수피의 항산화 및 항암활성 증진)

  • This study was conducted to characterize the enhanced antioxidant and anticancer activities of Berberis koreana bark following a low temperature and high pressure extraction process. The results indicate that the B. koreana bark extracted as described showed a 93% increase in DPPH radical scavenging activity. Inhibition activity of xanthine oxidase was highest by this extraction process. In addition the growth of human lung cancer cells (A549), human stomach cancer cells (AGS), human breast cancer cells (MCF-7) and human liver cancer cells (Hep3B) were inhibited by 70.8%, 86.2%, 84.3% and 62.5% respectively. These data indicate that this low temperature and high pressure extraction technique results in the efficient extraction of bioactive compounds from rigid plant materials. This process could also be combined with other techniques to improve extraction yields and identify new biologically active substances from relatively hard plants.

Effect of Rheum undulatum Extract on Antioxidant Activity and Activity of Matrix Metalloproteinase-1 in Human Skin Fibroblasts (대황 추출물의 항산화 활성 및 MMP-1 저해 활성)

  • Rheum undulatum L. has been commonly used as a cure for hematemesis, dropsy, and haematuria in the Oriental medicine for a long time. The main constituents of R. undulatum are chrysophanol and emodin, which are an antioxidative substance that has an anthraquinone structure. In the present study, to develop a new anti-aging agent, we examined the antioxidant activity and the inhibitory effect of the R. undulatum extract on the synthesis of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The R. undulatum extract was found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide radicals in the xanthine/ xanthine oxidase system by a dose-dependent manner, respectively. UVA-induced MMP-1 expression was reduced about 79.5% by 1 ${\mu}g$/ml of the R. undulatum extract and also inhibited MMP-1 activity in a dose-dependent manner. In conclusion, it was observed that the R. undulatum extract has the antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity. Therefore, these results suggest that the R. undulatum extract can be developed as a new anti-aging component of cosmetics.
