Effects of Propolis on 808 nm Laser Induced Wounds

  • Wound healing is a complex and highly organized biological response to injury that results in the loss of tissue integrity. Our particular interest was propolis, traditional used as an antimicrobial and an anti-inflammatory agent. The purpose of this study was to see the effects of propolis on healing of the laser induced wounds and the level of collagen formation. 808 nm laser (20 J) was irradiated on the back of rats. Irradiated wounds were divided into gel control and 6 experimental groups (3 and 5% of different three kinds of propolis). Gel type of propolis (supplied by Seoul Propolis Co., Daejeon, Korea) in water based were applied on the laser wound once daily for 14 days. One fourth of rats were sacrificed on $3^{rd}$, $7^{th}$, $10^{th}$, and $14^{th}$ day and each wound was evaluated for degree of wound healing and the level of collagen formation. Healing of wound was evaluated by measuring and comparing the width and depth of the wounds. The levels of collagen were increased in 3 and 5% gel groups comparing to control on $3^{rd}$ day and the collagen level were increased more on $7^{th}$ day. Wounds of the experimental groups showed better healing in width comparing to the healing of the control. The results of this study demonstrated that propolis accelerated healing of wounds induced by laser irradiation and good collagen formation.

창상치유시 상피조직에 관한 조직화학적 관찰 (Histochemical Observation on the Epithelial Tissue in the Wound Healing)

  • The healing response may very with the tissue, the site and the degree of wound. the author observed histochemically the epithelial regeneration in the har palate wound of healthy male albino rats, varying in age from 120 to 150 days, and weighing about 100 gm. The deep wounds were made antero-posterior linealy by surgical knife to the depth of bone level. They were sacrified by ether anesthesia on 1, 2, 4, 7, 10 and 14 days after wounding. the staining methods used were Mcmanus' PAS reaction, Mowry's modification of the Hale reaction employing Muller's colloidal iron reagent, alloxan-Schiff reaction and hematoxylin-eosin stain. The results were as follows : 1. In the wound healing of hard palate, the epithelium had marked PAS positive reaction in the granular and the prickle cell layers on the from 2nd to 7th day. 2. Alloxan-Schiff reactions of regenerated epithelium were slightly increased on 7th day.

Wound-Induced Hair Follicle Neogenesis as a Promising Approach for Hair Regeneration

  • The mammalian skin contains hair follicles, which are epidermal appendages that undergo periodic cycles and exhibit mini-organ features, such as discrete stem cell compartments and different cellular components. Wound-induced hair follicle neogenesis (WIHN) is the remarkable ability to regenerate hair follicles after large-scale wounding and occurs in several adult mammals. WIHN is comparable to embryonic hair follicle development in its processes. Researchers are beginning to identify the stem cells that, in response to wounding, develop into neogenic hair follicles, as well as to understand the functions of immune cells, mesenchymal cells, and several signaling pathways that are essential for this process. WIHN represents a promising therapeutic approach to the reprogramming of cellular states for promoting hair follicle regeneration and preventing scar formation. In the scope of this review, we investigate the contribution of several cell types and molecular mechanisms to WIHN.

흉고직경별 약제 수간주입구멍의 크기에 따른 주입구멍의 치유도 - 포스팜 수간주입구멍에 대한 반응 - (The Size of Injection Wound, Tree Diameter, and Injection Wound Healing of a Tree - Response to Pospam Injection Wound -)

  • 천공기로 소나무와 밤나무에 구멍을 뚫고 살충제인 포스팜을 수간주입한 후에 주입공 주변의 변화를 조사하였다. 수간주입은 9월에 실시하였으며, 주입공은 지름 1cm와 0.5cm를 비교하였다. 왕성한 수세의 나무들 중에서 공시목을 선발하였으며, 소나무는 흉고직경 10, 20, 30cm, 밤나무는 10, 15, 20cm이었다. 처리 이듬해 6월의 결과조사에서, 외관상으로 부후의 징후를 보이는 나무는 없었으나, 수피를 벗겨낸 목질부에서는 뚜렷한 변색이 일어나고 있었다. 변색의 정도는 수간주입공의 크기에 비례하여, 1cm 주입공에서의 변색부가 0.5cm 주입공에서의 변색부보다 더 컸다. 그러나, 공시목의 종류나 크기는 변색부의 크기와는 관계가 없는 것으로 나타났다. 일부 공시목에서 주입공을 기점으로하여 목질부가 길이방향으로 갈라져 있는 현상이 나타났는데, 그 길이는 소나무에서 보다는 밤나무에서, 그리고 0.5cm 주입공에서 보다는 1cm 주입공에서 더 길었다. 갈라진 부분에서는 새로운 유조직이 자라나와 갈라진 부분을 메워가고 있었다. 흡수식 수간주입 자체가 나무에 미치는 피해는 밤나무에서, 그리고, 주입공의 크기가 1cm일 때 더 컸다.

Accelerated Wound Healing by ]Recombinant Human Basic Fibroblast Growth Factor in Healing-impaired Animal Models

  • The stimulatory effect of recombinant human basic fibroblast growth factor (bFGF) on wound healing was evaluated in healing-impaired animal models. Full-thickness wounds were made in prednisolone-treated mice, streptozotocin (STZ)-induced diabetic rats and mitomycin C (MMC)-treated rats. Saline or bFGF at a dose of 1, 5, or $25\mu\textrm{g}$ per wound was applied to the open wound once a day for three to five days. The degree of wound healing was assessed using wound size and histological parameters such as degree of epidermal and dermal regeneration. Local application of bFGF accelerated wound closure significantly in a dose-dependent manner in all healing-impaired wounds (p<0.05). The wound healing effect of bFGF was further confirmed by histological examination in MMC-treated rats. Epidermal and dermal regeneration were enhanced in bFGF-treated wounds with a dose-related response. Dermal regeneration parameters such as collagen matrix formation and angiogenesis were significantly increased in $5\mu\textrm{g}$, or $\25mu\textrm{g}$ of bFGF-treated wounds when compared to saline-treated wounds (p<0.05). pectin immunostaining on day 8 for vascular endothelium showed an increased number of neovessels in bFGF-treated wounds. These results suggest that topical application of bFGF has beneficial effects on wound healing by angiogenesis and granulation tissue formation in healing-impaired wounds.

Overexpression of AtCAF1, CCR4-associated factor 1 homologue in Arabidopsis thaliana, negatively regulates wounding-mediated disease resistance

  • The CCR4-CAF1-NOT complex-mediated degradation of mRNA is a fundamental aspect of gene regulation in eukaryotes. We herein examined the role of AtCAF1 in the innate immune and wound responses of plants. Our results showed that overexpression of AtCAF1 significantly downregulated the transcript level of EFR but not FLS2 and BRI1, as well as abolished up-regulated expression pattern of EFR in response to wounding. Consistently, Agrobacteriummediated transient expression of GUS was highly enhanced in the transgenic plants overexpressing AtCAF. Furthermore, JA responsive genes were down-regulated by overexpression of AtCAF, causing the transgenic plants overexpressing AtCAF more susceptible to necrotrophic fungal pathogen, Botrytis cinerea. These results suggest that The CCR4-CAF1-NOT complex-mediated degradation of mRNA negatively regulates wounding-mediated disease resistance in Arabidopsis thaliana.

Effect of Pycnogenol on Skin Wound Healing

  • This study was carried out to investigate the effects of pycnogenol (PYC) on the cutaneous wound healing of the mice. The wounds were extracted on days 1, 3, 5, and 7 post-injury for histomorphometrical analysis including wound area, infiltrating inflammatory cells, wound contracture including collagen deposition. As the result, the wound area of PYC-treated group was larger than the control group on days 1 to 7. Inflammatory cells in the PYC-treated wounds were decreased at day 1 compared to the control wound tissue. From day 3 to 7, there was no significant difference between the control and the PYC-treated skin wounds. Though the degree of contraction in the PYC-treated group was lower than that of the control group from days 1 to 5, but appeared significantly higher on day 7. Compared to the control group, collagen accumulation in the PYC-treated group was higher than that of the control group from days 5 to 7. From this result, it may support the possibility that PYC would be useful agent for early inflammatory response and matrix remodeling phase of the skin wounds.

Wound-State Monitoring for Burn Patients Using E-Nose/SPME System

  • Array-based gas sensors now offer the potential of a robust analytical approach to odor measurement for medical use. We are developing a fast reliable method for detection of microbial infection by monitoring the headspace from the infected wound. In this paper, we present initial results obtained from wound-state monitoring for burn patients using an electronic nose incorporating an automated solid-phase microextraction (SPME) desorption system to enable the system to be used for clinical validation. SPME preconcentration is used for sampling of the headspace air and the response of the sensor module to variable concentrations of volatiles emitted from SPME fiber is evaluated. Gas chromatography-mass spectrometry studies prove that living bacteria, the typical infectious agents in clinical practice, can be distinguished from each other by means of a limited set of key volatile products. Principal component analysis results give the first indication that infected patients may be distinguished from uninfected patients. Microbial laboratory analysis using clinical samples verifies the performance of the system.

A novel excisional wound pain model for evaluation of analgesics in rats

  • Background: Management of pain from open wounds is a growing unmet healthcare need. However, the models available to study pain from wounds or to develop analgesics for the patients suffering from them have primarily relied on incisional models. Here, we present the first characterized and validated model of open wound pain. Methods: Unilateral full-skin excisional punch biopsy wounds on rat hind paws were evaluated for evoked pain using withdrawal responses to mechanical and thermal stimulation, and spontaneous pain was measured using hind paw weight distribution and guarding behavior. Evaluations were done before wounding (baseline) and 2-96 hours post-wounding. The model was validated by testing the effects of buprenorphine and carprofen. Results: Pain responses to all tests increased within 2 hours post-wounding and were sustained for at least 4 days. Buprenorphine caused a reversal of all four pain responses at 1 and 4 hours post-treatment compared to 0.9% saline (P < 0.001). Carprofen decreased the pain response to thermal stimulation at 1 (P ≤ 0.049) and 4 hours (P < 0.011) post-treatment compared to 0.9% saline, but not to mechanical stimulation. Conclusions: This is the first well-characterized and validated model of pain from open wounds and will allow study of the pathophysiology of pain in open wounds and the development of wound-specific analgesics.
