Development of Organic Fertilizer based on the Cow Dung -II. Studies on Rapid Fermentation (우분(牛糞)의 유기질비료화(有機質肥料化) 연구(硏究) -II. 속성부숙방법(速成腐熟方法))

  • To manufacture a good organic fertilizer through rapid composting process, cow dung was mixed with bulking materials such as rice straws, shredded bark, wood chips, and saw dust. The mixing ratio of the cow dung and bulking material was two to one on volume basis and moisture content was adjusted to 60 to 70 percent, C/N ratio 25 to 30 and aeration forced with suction during the aerated pile fermentation. Aerated pile was established outdoors and curing pile was in vinylhouse and the experiment was conducted for 3 years from '88 to '90. After pilling, temperature in aerated pile was reached to the maximum level at 7 to 9 days and the temperatures of cow dung + shredded bark, cow dung + wood chips, and cow dung + rice straws were higher than that of cow dung, and cow dung + saw dust. Total carbon, and C/N ratio in aerated pile fermentation period were increased in the middle stage and then they were decreased with the process of days, and nutrient contents in curing pile period had the same tendency as in the aerated pile.

  • This study was conducted to manufacture a good qualified organic fertilizer with cow dung through rapid composting process, and the proposal guideline of the degree of maturity could be estimated with the measurement of final product. It included total nitrogen content of above 2% on an oven-dry material basis, C/N ratio of below 20, CEC of more than about 60 me/100g, ratio of carbon in reducing sugar to the total carbon of below 35%, and temperature in pile of above $60^{\circ}C$. The total nitrogen content, the C/N ratio, and the Paper Chromatographic method couldn't be the guideline to evalute the maturity of cow dung compost. CEC was increased in increased fermentation and it was high in the high fermented temperature plots which were cow dung+ shredded bark in 1988, cow+dung+wood chips in 1989, and cow dung+rice straws in 1990. The ratio of carbon in reducing sugar to total carbon in 1990 was lower in cow dung+saw dust than cow dung+rice straws that was the highest temperature in pile. Generally cow dung was mixed well with saw dust and thus the total carbon of the product was high. The measurement of the temperature in pile seems to be a indirect guideline of maturity.

Morphological Characteristics of Decomposition and Browning of Oak Sawdust Medium for Ground Bed Cultivation of Lentinula edodes (표고 지면재배용 참나무 톱밥배지의 분해와 갈변의 형태적 특성)

  • This study investigated the internal and external morphological characteristics of decomposition and browning of oak sawdust medium for ground bed cultivation of Lentinula edodes. Within fifty days after L. edodes inoculation, surface hyphae on the bed browned. In 110 days, the fungal hyphae occupied and decomposed wood fibers, vessels and parenchymatous cells from the inside as white profuse hyphal mass was amorphously dissolving the saw dust particles from the outer surface. Most of the white hyphal bed surface became cleanly brown, however, some colony surface became blackened and slimy with contaminating bacteria, hyphae and spores. The brown layer was ca. 0.34 mm thick with highly dense and white hyphal mass beneath, whereas the blackened layer was ca. 1.17 mm thick with shrunken hyphae and less decomposed sawdust particles beneath. The surface hardness of the brown surface was ca. $0.73kgf/cm^2$, soft and resilient, while that of the blackened was ca. $0.91kgf/cm^2$, hard and nonresilient. By 150 days Lentinula edodes mushrooms fruited only on the brown surface and not on the blackened medium.

Studies on the Hydrolysis of Wood with Dilute Sulphuric Acid Solution and Trichoderma viride Cellulase (묽은황산(黃酸) 및 Cellulase에 의(依)한 목재당화(木材糖化)에 관(關)한 연구(硏究))

  • 1. The study was conducted on the optimum condition of the treated substrate with dilute sulphuric acid solution and cellulase for saccharification. The wood (saw dust) of Alnus hirsuta Rupr. (10~15 years) was treated with 0.3%, 0.6%, 0.9%, 1.2%, 1.5%, $H_2SO_4$ solution at $1.5kg/cm^2$ for 15min., 30min., 45min., and 60min., followed by thermal treatment at $190^{\circ}C$ for 30min., and screening with 60 mesh sieve, after which to 0.5 grams of each sample was added 0.5ml cellulase solution, and 50ml 0.1M acetic acid buffer solution (pH 5.0), after incubating at $40^{\circ}C$ for 96hr. 2. The crude cellulase of Trichoderma viride Perx. ex. Fr. SANK 16374 was produced by the submerged culture process and produced in the culture fluid was salted out quantitatively by the use of ammonium sulfite. 3. Reducing sugar was determined by the 3.5-dinitrosalicylic acid (DNS) method. 4. The reducing sugar was increased with increase of the sulphuric acid concentration and saw dust was treated with 1.5% $H_2SO_4$ solution at $1.5kg/cm^2$ for 45min. showed the best saccharification (16.0%). The reducing sugar formation did not show statistically significant in 5% levels by thermal treatment time 45min. and 60min. 5. The substrate for cellulase which was treated with 0.9% $H_2SO_4$ solution at $1.5kg/cm^2$ for 60min. showed the best reducing sugar formation (23.6%). And did not show significant difference in 5% levels at 0.9%, 1.2%, and 1.5% $H_2SO_4$ solution.

  • Enzymatic hydrolysis of the substrate from Alnus hirsuta (Spach) Rupr (8-14years) was investigated using cellulase preparations of Trichoderma viride Pers. ex. Fr. SANK 16374 and conduced on the optimum reaction conditions of the cellulase on saccharification. The crude cellulase was produced by the submerged culture process and produced in the culture fluid was salted out quantitatively by the use of ammonium sulfate. The method of delignification from wood(Saw dust) was treated by the peracetic acid (PA) method. Reducing sugar was determined by the dinitrosalicylic acid (DNS) method. The results were summerized as follows; 1. The optimum pH of cellulase was 5.0 and the range of stability with respect to pH was generally from 4.0 to 6.0 2. The optimum temperature of cellulase was generally $40^{\circ}C$, but reducing sugar formation did not show significent differences at 5% levels in the reaction temperature from $40^{\circ}C$ to $50^{\circ}C$. 3. The redusing sugar were increased with increase of cellulase concentration. 4. The reducing sugar were decreased with increase of substrate concentration. 5. Fructose was a very good inhibitor of the enzyme from Trichoderma viride, but glucose inhibition was generally weak.

Investigation of the Utilization of Organic Materials and the Chemical Properties of Soil in the Organic Farms in Korea (국내 유기농재배지 유기물 시용실태 및 토양의 화학적 특성)

  • A survey of 31 organic farmers were conducted to investigate the actual conditions of organic matter application. The amounts of organic matter application in the fields were higher in order of fruit, vegetable and rice farm. The average was 50 ton/ha in vegetable farms. In the green vegetable farms saw dust and animal manure were mainly utilized to make compost. Rice straw, wood chip, and forest bushes were also used for composting. In the fruit vegetable farms materials relatively lower in nitrogen content such as rice straw and cattle manure were used in vegetative period and materials higher in nitrogen content such as oil cake and wild grass were used in reproductive phase. Nutrient balance investigated in the farm in Icheon region who produce lettuce, angelica, and kale continuously in one cropping year indicated surplus in three major nutrients. Nitrogen and phosphorous were in excess by 29 and 10 kg respectively in the organic rice farm in yang-pyoung region. While soil chemical properties in the organic farms are within the adequate range in open field, it is much higher than the limits in the greenhouse soils. Overall application of organic matter is in an oversupply state. This results suggested that the organic matter management should be based on the soil conditions for sustainable cultivation. Chemical composition of organic matters and soil test reports should be considered prior to the application of organic matter.
