An Investigation of Natural Lawn Condition on School Grounds and the Possibility of Revegetation on It's Surroundings (잔디운동장 훼손 현황과 풀을 이용한 도시 학교 운동장 주변의 녹화 가능성)

  • This study was carried out to investigate the deterioration rate and conditions of natural lawn play grounds from 71 schools in Daejeon metropolitan city. Also, we investigated the grasses and wild flowers of 33 families and 101 species on five additional school grounds. One year after planting in 71 schools from 2002 to 2004, we found out that the school lawn grounds coverage were deteriorated up to 51% in elementary schools, 76% in middle schools, and 42% in high schools. There are 45 species on the two school grounds located in mountain areas, 20 species on the three grounds located in downtown areas. There are 20 species in Compositae family, 17 species in Gramineae, 7 species in Polygonaceae, and 7 species in Leguminosae, 6 species in Caryophyllaceae, five species in Cruciferae, and 4 species in Cyperaceae.

A Study for Use of Wild Rhododendron mucronulatum for. albiflorum as Landscape plant (야생 흰진달래의 조경식생화를 위한 연구)

  • Rhododendron mucronulatum for. albiflorum, native species is a shrub that has white flowers on May to June, and rare species endangered by people's rash digging or cutting. But its physiological ecological characteristics and propagation method are not being known at all. Therefore, this study was executed to utilize this species as the planting material for landscaping by analysing its habitat environment and growth form, and also experimenting its seed and vegetative propagation, and it field culture and utilization. The results are as follows; 1. The elevation, gradient and direction of this species were 295-1,350m, 10-36$^{\circ}$, northwest respectively. It was found that the species is shade-liking plant that grows under forest cover of average 51.33%. 2. The soil pH and water content of its habitat were 5.4, 25.41% respectively. The organic matter content was 6.29% that was higher than 3.2%, the average organic matter content of forest soil in Korea. 3. Representative plant community within which this species was living was Quercus mongolica community, and its main neighboring species were Lindea obtusiloba, Fraxinus sieboldiana, Rhus trichocarpa, Rhododendron Schlippenbachii, Rododendron mucronulatum. 4. The leaf length and width of this species were 39.18mm, 12.60mm respectively. This result showed that generally its leaf size was larger than that of R. micranthum, R. yedoense var. poukhanense and R. mucronulatum var. ciliatum and smaller than that of R. mucronulatum and R. schlippenbachii. 5. The whole size of its pollen was, as 59${\times}$61$\mu\textrm{m}$, the largest of plants of Rhododendron family including R. mucronulatum and R. mucronulatum var. ciliatum. 6. The result of seed germination experiment at intervals of 5$^{\circ}C$ from 15$^{\circ}C$ to 30$^{\circ}C$ presented the highest germination rate of 94.7% at 20$^{\circ}C$ numerically, but high percent germination at all temperature levels without significant difference. And the seed of this plant proved to be sun-liking seed at requiring dormancy in germination. 7. Through seed germination experiment by treatment of growth regulators such as GA. Thiourea and Kinetin under dark condition, it was found that the effect of GA treatment on germination increase and acceleration was the highest. 8. In greenwood cutting, rooted rate by treatment of various concentration of IBA and NAA on clay and vermiculite bed was not wholly high, but 100ppm plots of both IBA and NAA of clay bed showed relatively good rooted rate. 9. As result of field culture experiment for finding out optimum growth temperature and light intensity, growth conditions such as height, number of leaves, fresh weight and chlorophyll contents were the best at night/day temperature of 20/25$^{\circ}C$ and under 1/2sun. Also, the photosynthetic rate was the highest at 25$^{\circ}C$. Accordingly, it was found that optimum temperature and light intensity for growth of this plant are 25$^{\circ}C$ (day temperature), 50% of natural light respectively.

Asymbiotic germination and seedling growth of Calanthe striata f. sieboldii Decne. ex Regel (금새우난초(Calanthe striata f. sieboldii Decne. ex Regel)종자의 비공생 발아 및 신초증식)

  • Calanthe striata f. sieboldii Decne. ex Regel is a terrestrial orchid with beautiful flowers arranged in racemose inflorescences. This species is threatened due to over-collection and loss of suitable habitats. Asymbiotic germination is useful in the conservation efforts to re-establish plants in the wild, and for commercial propagation. In this study, we investigate the effects of sodium hypochlorite (NaOCl), three types of culture media (Phytomax Orchid Maintenance - POM, Seed Germination Maintenance - SGM, and Murashige and Skoog 1962 - MS), and plant growth regulators on embryo swelling, protocorm formation, and embryo diameter of C. striata f. sieboldii. Treatment with 1% NaOCl for 30 min greatly enhanced embryo swelling (28.3%), embryo diameter ($205.8{\mu}m$), and embryo protocorm formation (54.8%) compared to seeds without NaOCl treatment (embryo swelling 8.5%, embryo diameter $14.6{\mu}m$, and protocorm formation 13.4%) on POM medium. Protocorm formation on POM medium supplemented with 1.0 mg/L N6-benzyladenine (BA) (95.6%) was better than the control (54.5%). Additionally, the effects of activated charcoal (AC) and sucrose on seedling growth in in vitro culture were examined. The protocorm converted into healthy plants with well-developed shoot primordia on the POM medium with AC and sucrose. The most suitable conditions for seedling growth after 10 weeks of culture were the POM medium with AC or sucrose. These results show effective asymbiotic seed germination and in vitro seedling growth of C. striata f. sieboldii.

Genetic variation and population structure of Asarum misandrum (Aristolochiaceae) in Korea (각시족도리풀(Asarum misandrum)의 유전적 다양성 및 집단 구조)

  • Genetic variation in Asaum misandrum, a woodland herb in Korea, was investigated based on allozyme analysis with starch gel electrophoresis. All of previously reported populations in Korea were sampled and seven loci from six enzymes were analyzed. Overall genetic variation of A. misandrum population showed considerably high levels of genetic variation within the species (A = 2.05, P = 71.4, $H_E$ = 0.294). A positive $F_{IS}$ value of A. misandrum indicated overall deficiency of heterozygotes, and a low $F_{ST}$ value (0.112) meant very little differentiation among populations. Factors contributing to the high levels of genetic diversity found within populations of A. misandrum include population maintenance via wide distribution range from Korea to Japan and primarily outcrossing breeding system. Although it showed moderate genetic diversity level, most habitats of the species were scattered and discontinuous. Besides, low numbers of individuals were found in the most habitats and individuals are collected frequently from the wild due to the unique shape of the flowers as well as the rarity of the species. Thus, there is a need to set up a reasonable conservation strategies including the maintenance mechanism of genetic diversity of A. misandrum.

Optimization of a Virus-Induced Gene Silencing System with Soybean yellow common mosaic virus for Gene Function Studies in Soybeans

  • Virus-induced gene silencing (VIGS) is an effective tool for the study of soybean gene function. Successful VIGS depends on the interaction between virus spread and plant growth, which can be influenced by environmental conditions. Recently, we developed a new VIGS system derived from the Soybean yellow common mosaic virus (SYCMV). Here, we investigated several environmental and developmental factors to improve the efficiency of a SYCMV-based VIGS system to optimize the functional analysis of the soybean. Following SYCMV: Glycine max-phytoene desaturase (GmPDS) infiltration, we investigated the effect of photoperiod, inoculation time, concentration of Agrobacterium inoculm, and growth temperature on VIGS efficiency. In addition, the relative expression of GmPDS between non-silenced and silenced plants was measured by qRT-PCR. We found that gene silencing efficiency was highest at a photoperiod of 16/8 h (light/dark) at a growth temperature of approximately $27^{\circ}C$ following syringe infiltration to unrolled unifoliolate leaves in cotyledon stage with a final SYCMV:GmPDS optimal density $(OD)_{600}$ of 2.0. Using this optimized protocol, we achieved high efficiency of GmPDS-silencing in various soybean germplasms including cultivated and wild soybeans. We also confirmed that VIGS occurred in the entire plant, including the root, stem, leaves, and flowers, and could transmit GmPDS to other soybean germplasms via mechanical inoculation. This optimized protocol using a SYCMV-based VIGS system in the soybean should provide a fast and effective method to elucidate gene functions and for use in large-scale screening experiments.

Surveys on Disease Occurrence in Major Horticultural Crops in Kangwon Alpine Areas (강원도 고랭지 주요 원예작물의 병해 발생 상황)

  • These surveys were conducted to check the occurrence of disease in various horticultural crops in alpine areas, especially Daekwallyong areas, Pyongchang-Gun, and Hyeongseong-Gun in Kangwon province. TuMV on Chinese cabbage was one of the most serious diseases, especially in 1994 and 1997. The incidence of soft rot and clubroot has been increased gradually. Brittle root rot on Chinese cabbage was significantly decreased. Soft rot, gray mold, downy mildew, powdery mildew, bottom rot and Alternaria leaf spot were the common diseases on most vegetable crops. Gray mold (Botrytis cinerea) on celery, cercospora leaf spot (Cercospora sp.) on melon, powdery mildew (Erysiphe cichoracearum) on lettuce, and clubroot (Plasmodiophora brassicae) on parsley are newly found in Korea. The most common and predominant diseases were viruses, especially CMV, TMV, TuMV, BBMV, and gray mold, wilts, and cercospora leaf spot on many flowers in alpine areas. Fusarium oxysporum f. sp. eustomae causing wilting on lisianthus (Eustoma grandiflorum), Turnip mosaic virus causing mosaic and color breaking on stock, Cercospora spp. causing cercospora leaf spot on various wild lily, Cladosporum echinulatum causing leaf spot on carnation, and phytoplasma causing witches' broom on statice (Limonium sinuatum) and blazing star (Liatris spp.) were newly found during these surveys in Korea.

A Functional Analysis of OsCPK11, a Calcium-dependent Protein Kinase (CDPK) Gene in Rice (벼의 칼슘-의존성 단백질 카이네즈 유전자인 OsCPK11의 기능적 분석)

  • CDPKs have pivotal roles in plant $Ca^{2+}$-mediated transduction signaling. A total of 29 CDPK genes have been identified in rice (Oryza sativa L.), but their key functions have not been completely noted. This study focused on the OsCPK11 gene, which has not been studied, to determine its functional characteristics. A study of tissue-specific expressions revealed that the OsCPK11 gene is expressed in young leaves, mature leaves and flowers of rice. An expression of the gene was also confirmed in gibberellin-treated aleurone layers of rice. Regarding the phenotypic characteristics of Tos17-inserted OsCPK11 mutants, the heights of the mutants were not distinguishable from the heights of wild type plants, but the number of caryopses and the caryopses' weights were significantly statistically different. In addition, many grains of the mutants had white belly materials in their endosperm. The cDNA of the OsCPK11 was cloned, and an OsCPK11 protein of about 60.5 kD was obtained by using a GST affinity chromatography and an SDS-PAGE. An analysis of the amino-acid sequence of the protein indicated that the OsCPK11 protein has the structural characteristics of typical CDPKs. The results provided useful information about the functions of the OsCPK11 gene and further noted the roles CDPKs have in $Ca^{2+}$-mediated signaling in plants.

Effect of Seeding Rate (Sheep fescue 70% + Wildflowers 30%) on the Growth Characteristics, Seasonal Anthesis Distribution and Botanical Composition in Wildflower Pastures (쉽 페스큐(70%)와 야생화(30%)의 파종비율이 야생화 초지의 생육특성, 계절개화분포 및 식생변화에 미치는 영향)

  • This study was carried out in the Chungnam National University grassland experimental field from October, 2007 to December, 2009 in order to find out the growth characteristics of the wildflowers, the seasonal anthesis distribution of the wildflowers, and the botanical composition of the wildflowers which were altogether composed of 70% sheep fescue and 30% wildflowers. The experimental species contained 34 species in total to which sheep fescue (fundamental turfgrass), 4 native wildflowers, and 29 introduced wildflowers belonged. At the time of the anthesis of the wildflowers, the average length was 29.9 cm one year later and 38.4 cm two years later. They bloomed out into nine colors, but into one or three colors during August, September, and October. The blooming season leant toward May and June and from August there was lots of difficulty in the wildflower blooming and the maintenance of their consistency. Especially, at the age of two (2009) the color, seasonal distribution, and consistency emerged as problems owing to the reduction of annual wildflowers after winterization. The annual ration of wildflowers, sheep fescue, and weeds came to 25%, 61%, and 14% each one year later (2008) and 22%, 65%, and 13% each two years later (2009). Based on this result, cultivating wildflower grassland in the ratio of 70% (sheep fescue) to 30% (wildflowers) is the proper method not only for maintaining green space by sheep fescue, but also for commanding a fine view of wild flowers.

Planting Patterns and Landscape Redevelopement of Pilam Seowon in Jangseong-Gun (장성 필암서원(筆巖書院)의 식재현황과 정비방안)

  • This study, as a purpose of base study for landscape redevelopement of Seowon, pointed out problems of planting patterns in each sector and suggested an alternative through analyzing old literature, complete enumeration survey in each sector and taking aerial images. the conclusions drawn from this study are as follows. A Pilam Seowon(historic site no 242) is located in a Pyungya-Sanrok(plain and mountain) district in Jangsung-gun and has Junhak-Humyo(Study room in the front and Shirne in the back) type. 23 taxonomic group of arbor, 6 taxonomic group of shrub and 5 taxonomic group of flowers are planted high-densitily in limited flat surface. It can be classified into being planted by family and being planted through landscape redevelopement in 1980s. Korean traditional trees are planted in this area like Pinus densiflora, Juniperus chinensis L., Thuja orientalis L., Zelkova serrato Makino, Sophora japonica L., Lagerstroemia indica, Prunus mume include Ginkgo biloba which is old big tree and is registered in Jangsung-gun as protected trees. The tea tree in this area was introduced from wild tea plantation in Yonhwa moutain and was planted. From now on, we have to pay consideration a form management of planted trees in the historical area, rearrangement of plant density due to over planting, removal of alien trees like Pinus bungeana and Canna generalisa, prevention a dwarfishness of main area due to over planting in outside facilities, recovery a alienation among each planting area, and planting concept from plants drawings on wall of buildings can be suggested. The implications of this case study is that introduction of over planting can make more problems like losing sense of place. Therefore, we must do a actual state survey on traditional landscape area like Seowon from preservation point of view.

A Study on Decanting of Old Wine : Focused on Fortified Wine (올드 와인의 디캔팅 연구 : 강화 와인을 중심으로)

  • This study was tested on Ratafia Champagne Trouillard 1947 of old fortified wine and analyzed differences from existing wines. Old fortified wine in Champagne, France and blanding is Pinot Noir, Chardonnay and Pinot Meunier. Alcohol level is 18% and test date is Feb. 15-21, 2019(six days of decanting period/15 p.m. on the last tasting day). Tester is composed of one FICB grand commander one KOV Finland commander. The wine opening was tested for two blades after wire removal and the decanting time was applied to the calculation formula of 2019(this year)-1947(vintage year)/12=6 days set in this study. Aroma smelled like cherries, fruits, soy sauce and licorice and bouquet was identified in five stages. The first stage was presented with the smell of pot, the second stage was light coffee, the third stage smell of fruit and flowers, the fourth stage smell of wild honey and the fifth stage smell of refined brandy. Then, the test was analyzed in seven stages. This study has the following implications: First, the new concept of old wine was applied to fortified wine. Specific computational formulas for the decanting period were derived. The decanting presented five steps of aromas and bouquet. Wine testing has been expanded from the previous five to seven levels. A new taste of Champagne old fortified wine was analyzed.
