Effects of Dietary Garlic Extracts on Whole Body Amino Acid and Fatty Acid Composition, Muscle Free Amino Acid Profiles and Blood Plasma Changes in Juvenile Sterlet Sturgeon, Acipenser ruthenus

  • A series of studies were carried out to investigate the supplemental effects of dietary garlic extracts (GE) on whole body amino acids, whole body and muscle free amino acids, fatty acid composition and blood plasma changes in 6 month old juvenile sterlet sturgeon (Acipenser ruthenus). In the first experiment, fish with an average body weight of 59.6 g were randomly allotted to each of 10 tanks (two groups of five replicates, 20 fish/tank) and fed diets with (0.5%) or without (control) GE respectively, at the level of 2% of fish body weight per day for 5 wks. Whole body amino acid composition between the GE and control groups were not different (p>0.05). Among free amino acids in muscle, L-glutamic acid, L-alanine, L-valine, L-leucine and L-phenylalanine were significantly (p<0.05) higher in GE than in control. However, total whole body free amino acids were significantly lower in GE than in control (p<0.05). GE group showed higher EPA (C22:6n3) and DHA (C22:5n3) in their whole body than the other group (p<0.05). In the second experiment, the effects of dietary garlic extracts on blood plasma changes were investigated using 6 month old juvenile sterlet sturgeon averaging 56.5 g. Fish were randomly allotted to each of 2 tanks (300 fish/tank) and fed diets with (0.5%) or without (control) GE respectively, at the rate of 2% of body weight per day for 23 d. At the end of the feeding trial, blood was taken from the tail vein (n = 5, per group) at 1, 12, and 24 h after feeding, respectively. Blood plasma glucose, insulin and the other serological characteristics were also measured to assess postprandial status of the fish. Plasma glucose concentrations (mg/dl) between two groups (GE vs control) were significantly (p< 0.05) different at 1 (50.8 vs 62.4) and 24 h (57.6 vs 73.6) after feeding, respectively, while no significant difference (p>0.05) were noticed at 12 h (74.6 vs 73.0). Plasma insulin concentrations (${\mu}IU$/ml) between the two groups were significantly (p<0.05) different at 1 (10.56 vs 5.06) and 24 h (32.56 vs 2.96) after feeding. The present results suggested that dietary garlic extracts could increase dietary glucose utilization through the insulin secretion, which result in improved fish body quality and feed utilization by juvenile sterlet sturgeon.

  • Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease, a chronic debilitating disease affecting ruminants worldwide. In the present study, we aimed to determine the major gene networks and pathways underlying the immune response to MAP infection using whole-blood cells, as well as provide the potential transcriptional markers for identifying the status of MAP infection. We analyzed the transcriptional profiles of whole-blood cells of cattle identified and grouped according to the presence of MAP-specific antibodies and the MAP shed by them. The grouping was based on the results obtained by ELISA and PCR analyses as follows: i) Test1 group: MAP-negative results obtained by ELISA and positive results obtained by PCR; ii) Test2 group: MAP-positive results obtained by ELISA and negative results obtained by PCR; iii) Test3 group: MAP-positive results obtained by ELISA and positive results obtained by PCR; iv) uninfected control: MAP-negative results obtained both by ELISA and PCR analysis. The results showed down-regulated production and metabolism of reactive oxygen species in the Test1 group, activation of pathways related to the host-defense response against MAP (LXR/RXR activation and complement system) in the Test2 and Test3 groups, and anti-inflammatory response (activation of IL-10 signaling pathway) only in the Test3 group. Our data indicate a balanced response that serves the immune-limiting mechanism while the host-defense responses are progressing.

Mechanisms of Platelet Adhesion on Elastic Polymer Surfaces: Protein Adsorption and Residence Effects

  • Platelet adhesion onto elastic polymeric biomaterials was tested in vitro by perfusing human whole blood at a shear rate of 100 sec$\^$-1/ for possible verification of mechanisms of initial platelet adhesion perfusion of blood on the polymeric substrates was performed after treatments either with or without pre-adsorption of 1% blood plasma, and either with or without residence of the protein-preadsorbed substrate in phosphate buffered solution. The surfaces employed were elastic polymers such as poly(ether urethane urea), poly(ether urethane), silicone urethane copolymer, silicone rubber and poly(ether urethane) with the anti-calcifying agent hydroxyethane bisphosphate. Each polymer surface treated was exposed in vitro to the dynamic, heparinized whole blood perfused for upto 6 min and the surface area of platelets initially adhered was measured by employing in situ epifluorescence video microscopy. The blood perfusion was performed on the surfaces treated at the following three different conditions: directly on the bare surfaces, after protein pre-adsorption and after residence in buffer for 3 days of the surfaces protein pre-adsorbed for 2 h. The effects of blood plasma pre-adsorption on the initial platelet adhesion was surface-dependent. The amount of the adsorbed fibrinogen and the surface coverage area of the adhered platelets were dependent on the surface conditions whether substrates were bare surfaces or protein pre-adsorbed ones. To test an effect of possible morphological (re)orientations of the adsorbed proteins on the initial platelet adhesion, the polymeric substrate pre-adsorbed with 1% blood plasma was immersed in phosphate buffered solution for 3 days and then exposed to physiological blood perfusion. The surface area of the platelets adhered on these surfaces was significantly different from that of the surfaces treated with protein pre-adsorption only. These results indicated that platelet adhesion was dependent on the surface property itself and pre-treatment conditions such as blood perfusion without any pre-adsorption of proteins, and blood perfusion either after protein pre-adsorption or after subsequent substrate residence in buffer of the substrate pre-adsorbed with proteins. Understanding of these results may guide for better designs of blood-contacting materials based on protein behaviors.

  • Intravascular oxygenation represents an attractive. alternative support modality for therapy originated with acute respiratory distress syndrome(ARDS). However. the clinical study concluded that more gas exchange was needed for intravascular oxygenation to be clinically effective in ARDS treatment. In this study, we tried to enhance gas exchange on the VIVLAD using microencapsulation of hemoglobin and perfluorocarbon emulsion(PFC emulsion). Blood gas measurements were performed by collecting blood samples from the arterial and venous sides of the circuit, and processing them in a blood/gas analyzer. The function of hemosome. blood/hemosome mixed solution. and blood/PFC emulsion mixed solution were tested by an oxygen dissociation curve using a blood/gas analyzer. As a result, it was shown that the oxygen transfer of hemosome and blood/hemosome mixed solution were higher than that of whole blood. Also. it showed that the carbon dioxide transfer of whole blood/PFC emulsion mixed solution was higher than that of others. Therefore, we determined that hemosome and PFC emulsion could increase oxygen transfer and carbon dioxide transfer. respectively.

  • This study was designed to provide basic strategies of nursing intervention in order to keep and obtain healthy, responsible and regular donors by investigating and analyzing relationships of health promotion behavior. self-efficacy and self-esteem of blood-donors. The subjects collected for the research were four hundred and sixty-eight individuals who participated in platelet donation at a university hospital apheresis unit in Seoul. The data for this study were collected between May and June, 2002 using questionnaires. The results were as follows. 1. The proportion of subjects who have not had previous experience of whole blood-donation was $13.7\%$, while the proportion of subjects who have had two times previous experiences was $16.2\%$ and over five times was $30.6\%$. 2. Mean score of health promotion behavior was 2.7 (range 1-4), self-efficacy was 3.8 (range 1-5) and self-esteem was 3.3 (range 1-4), which showed higher than middle. 3. There was positive significant relationship between the number of blood-donation and health promotion behavior as well as the number of blood donation and self-efficacy. 4. Positive significant relationship existed between the number of whole blood-donation and the number of apheresis blood-donation. Therefore, to increase recurrence of blood-donation. it is necessary to establish more effective and organized nursing intervention strategies that can promote the health promotion behavior, self-efficacy and self-esteem of blood-donors.

Ying blood(營血) flows within Blood Circulation means living body ingredient which thing reveals by circulation of the blood change phenomenon. Ying Qi(營氣) is Ying(營) in meaning which go on meridians, therefore meridians as wandering living body reaction route mean. This Ying Qi(營氣)'s physiology transform blood and grows up whole body and flows within Blood Circulation. It becomes most important ingredient of composition of blood, Wei Qi(衛氣) transforms and becomes nutrition constantly by running regeneration of material which transform in Jungcho(中焦) and spread by spreading action of upper chest exhaust in lower belly above the blader of apriority, Kidney-yang.

  • To analyze in-vivo blood flow characteristics in a chicken embryo, in-vivo experiment was carried out using micro-PIV technique. Because endothelial cells in blood vessels are subject to shear stress of blood flow, it is important to get velocity field information of the placental blood flow. Instantaneous velocity fields of an extraembryonic blood vessel using a high-speed camera and intravital microscope. The flow images of RBCs were obtained with a spatial resolution of $20\times20{\mu}m$ in the whole blood vessels. The mean velocity field data confirm that the blood flow does show non-Newtonian fluid characteristic. The blood in a branched vessel merged smoothly without any flow separation into the main blood vessel with the presence of a slight bump. This in-vivo micro-PIV measurement technique can be used as a powerful tool in various blood flow researches.

Radioprotective Effect of Propolis on the Blood Corpuscle of a Mouse by SEM after X-irradiation on the Whole Body (전신 X-선 조사 후 마우스의 대동맥 혈구에서 SEM을 이용한 프로폴리스의 방사선방어효과 연구)

  • After x-ray 5Gy radiation on the whole body of a mouse using a linear accelerator, its leucocyte, erythrocyte, and platelet were observed by SEM. Also, after injecting propolis into the abdominal cavity, the radio-protective effect of blood corpuscles was studied. The observation of micromorphology in blood corpuscles revealed that the number of leukocyte, erythrocyte, and thrombocyte decreased in the experimental group and the lump got together in blood corpuscles after 10 and 20 days. In RBC, crack or break on the surface and poikilosperocytes were observed. In the irradiation group, the size of leucocytes was smaller than that in control group and the number of villus at the verge substantially decreased. The blood corpuscles in the propolis group, however, had the similar results to control group.

  • The novelty of this study resides in the fabrication of stoichiometric and stress-reduced $Si_3N_4/SiO_2/Si_3N_4$ triple-layer membrane sieves. The membrane sieves were designed to be very flat and thin, mechanically stress-reduced, and stable in their electrical and chemical properties. All insulating materials are deposited stoichiometrically by a low-pressure chemical vapor deposition system. The membranes with a thickness of 0.4 ${\mu}m$ have pores with a diameter of about 1 ${\mu}m$. The device is fabricated on a 6" silicon wafer with the semiconductor processes. We utilized the membrane sieves for plasma separations from human whole blood. To enhance the separation ability of blood plasma, an agarose gel matrix was attached to the membrane sieves. We could separate about 1 ${\mu}L$ of blood plasma from 5 ${\mu}L$ of human whole blood. Our device can be used in the cell-based biosensors or analysis systems in analytical chemistry.

  • The spectra of whole blood EDTA samples from two people were generated using a CARY 5E (UV-VIS-NIR) spectrophotometer from 400 to 1000nm which contain visible and NIR region. Only the data between 400 and 800nm were used to analyze the components of blood. Using the same spectrophotometer, the spectra of Water, normal saline, plasma were generated These spectra were subtracted from each blood sample, and then the first derivative of each of the subtracted data was taken to minimize baseline variations and indicated the wavelength-shift of peak and valley. Normalization and division between two blood samples were used to correlate the quantity ratio of specific components with feature of spectra. Samples were controlled at $30^{\circ}C,\;37^{\circ}C$, ambient temperature.
