Isolation of Angiotensin Converting Enzyme Inhibitors from Pig Blood (돼지혈액으로부터 항고혈압물질의 분리)

  • Angiotensin converting enzyme (ACE) inhibitors were isolated and purified from pig blood plasma. Pig blood plasma was obtained after removing blood cells by centrfugation, followed by the addition of anticoagulant to whole pig blood. To precipitate plasma proteins, pig blood plasma was treated with 4% trichloroacetic acid (TCA) as a final concentration. ACE inhibitors were isolated from plasma protein hydrolysates and TCA supernatant, using ultrafiltration, gel permeation chromatography, and reverse-phase high pressure liquid chromatography. ACE inhibitors isolated from plasma hydrolysates and TCA supernatant had $IC_{50}$ values of $23\;{\mu}M$ and $2\;{\mu}M$, pentapeptide, respectively.

Revision of the modified Blood Stasis Questionnaire II (어혈 진단 설문지 개정 연구)

  • Objectives : The objective of this study was to revise the modified Blood Stasis Questionnaire II. Methods : This revision focused on refining the Korean literature expression of Blood Stasis Questionnaire II consisting of 30-question questionnaire. Seven external experts and five researchers of Korean Institute of Oriental Medicine reviewed the questionnaire and its protocol, while the addition or deletion of questions and changes in scoring method were not dealt with in this revision. Results : Among thirty questions, four questions were corrected to appropriate expressions. In case of eight questions, explanations in Korean or Chinese were added. Thirteen questions in the phrase were changed in sentence form to unify the whole questionnaire. Conclusions : This study introduces the revised version of the modified Blood Stasis Questionnaire II. It is expected that clinical demand of this questionnaire will increase and it will be used vigorously in blood stasis research.

Studies on the Safety of Recombinant Bovine Somatotropin in Dairy Cow : Effects of ${\gamma}$BST on Hematologic and Blood Chemical Values in Dairy Cow (${\gamma}$BST의 젖소에 대한 안전성 연구 II. 성장호르몬이 젖소의 혈액상 및 혈액화학치에 미치는 영향)

  • Effects of recombinant bovine somatotropln(${\gamma}$BST) on hematologie and blood chemical values were investigated in twenty-five multiparous Holstein dairy cows. Recombinant BST was administered by two different routes ; intramusculary(12.5mg and 25mg/day) and subcutaneously(500mg and 750mg) in sustained-release vehicle every 2 weeks beginning 4 weeks postpartum and continuing for 7 months. Whole blood and serum samples were collected 0, 1, 2, 3, 5, and 7 months after beginning of treatments from control and ${\gamma}$BST-administered groups. Hematologic values including RBC, PCV, HB, MCH, MCHC, WBC and differential counts of treatment groups receiving ${\gamma}$BST were similiar to those of control group. Blood chemical values observed were total protein, albumin, A/G ratio, glucose, cholesterol, Ca, Pi, Ca/pi ratio, total bilirubin, creatinine, BUN, alkaline phosphatase, lactate dehydrogenase, alanine aminotransferase and aspartate aminotransferase. There were no significant differences in blood chemical values of cows administered with ${\gamma}$BST from those of control. Although some blood chemical values were fluctuated at a certain observation period, they were remained within the normal physiological ranges. It is concluded from the observations of these experiments that the dose and dosage froms of ${\gamma}$BST employed in this work might not affect hematologic and blood chemical values in dairy cows under the normal sanitary condition and adequate nutritional balance.

Study on the Precision in Determinations of Lead and Zinc in the Whole Blood (혈중 연 및 아연 분석의 정도에 관한 연구)

  • Analyses of lead and zinc were made by means of standard addition method using atomic absorption spectrophotometer(Baird Ltd., Model A5100) with flameless method for lead and flame method for zinc. The blood samples used were merely diluted with triton x-100, because it was simple, rapid and minimal risk of contamination. Mean recovery rate for lead added to the blood ranged from 97.7 to 101.3% with coefficient of variation ranging from 1.9 to 10.7%, and that for the added zinc ranged from 99.0 to 102.2% with coefficient of variation ranging from 2.1 to 9.1%. In repeated measurements of zinc in the blood, good reproducibility and interindividual variation were proved(p<0.01). In comparison of the lead and zinc concentrations in the blood determined by the standard addition method and standard method, there were good correlations between 2 sets of data (r=0.9731 for lead and r=0.9785 for zinc), although lead levels were estimated higher by the former method(p<0.01) and zinc levels by the latter method(p<0.01). It can be concluded that lead and zinc levels in blood standard addition method is reliable for determination of lead and zinc in the blood with good accuracy and reproducibility.

  • This study was purposed on revealing the connections between blood types and brain functions or correlation character among Junior and Highschool students. Observed records are from those kids who volunteered for KRIJUS(Korea Research Institute of Jungshin Science)'s brain wave measurement from September 2005 to December 2008. The whole 2208 Junior and Highschool studentskids group consists of junior 628 female, 1002 male and high 213 female, 365 male. Blood types are as follows A>B>O>AB dominately ordered. Second, blood type had no relations with corelation character. Third, blood type had no relations with brain function rate. Thus, it would not be such a wise decision to tell one's character depending on his/her blood type.

  • A 3D human ventricular model is proposed to simulate an integrative analysis of heart physiology and blood hemodynamics. This consists of the models of electrophysiology of human cells, electric wave propagation of tissue, heart solid mechanics, and 3D blood hemodynamics. The 3D geometry of human heart is discretized to a finite element mesh for the simulation of electric wave propagation and mechanics of heart. In cellular level, excitations by action potential are simulated using the existing human model. Then the contraction mechanics of a whole cell is incorporated to the excitation model. The excitation propagation to ventricular cells are transiently computed in the 3D cardiac tissue using a mono-domain method of electric wave propagation in cardiac tissue. Blood hemodynamics in heart is also considered and incorporated with muscle contraction. We use a PISO type finite element method to simulate the blood hemodynmaics in the human ventricular model.

Blood Viscosity Measurements Using a Pressure-Scanning Capillary Viscometer

  • A previously designed capillary viscometer with measuring differential pressure was modified to measure the viscosity of non-Newtonian fluids including unadulterated blood continuously over numerous shear rates in a single measurement. Because of unavoidable experimental noise and a limited number of data, the previous capillary viscometer experienced an inaccuracy and could not directly determine a viscosity without an iterative calculation. However, in the present measurement there are numerous data available near the point of interest so that the numeric value of the derivative, d(In Q)/d(In Q$\sub$w/), is no longer sensitive to the method of differentiation. In addition, relatively low and wide shear rate viscosity measurements were possible because of the present precision pressure-scanning method with respect to time. For aqueous polymer solutions, excellent agreement was found between the results from the pressure-scanning capillary viscometer and those from a commercially available rotating viscometer. In addition, the pressure-scanning capillary viscometer measured the viscosity of unadulterated whole blood without adding any anticoagulants.

  • Recently, the high blood pressure is appeared the most frequent disease of a man's the whole sickness. But, to treat this disease is uncertain and produces an adverse reaction of a medicine therapy. Also, a patients are burden high price for doctor's treatment. Therefore, we studied effectively and financially to execute the treatment of the high blood pressure using the Chinese Medicine Theorem that recently arousing a great interest of the people. The main theories of this paper are Blood Vessel Theory and Acupuncture. The composed circuits are low-frequency generating circuit, charging-discharging circuit. The former will substitute a needle that use in the acupuncture therapy method and the latter will charge and discharge the bioelectric action potential of a patient in Ear Acupuncture Point. Also, all circuits will be controlled a micro-computer circuit.

Distribution of Abused Drugs in 275 Alcohol-positive Blood Samples of Korean Driver (음주운전자 275명 혈액 중 마약류 및 남용약물의 분석)

  • Even though driving under the influence of drug (DUID) is a worldwide problem, we, Korea has no regulation system yet except for alcohol, and there are little cases reported related to DUID. In order to investigate the type of abused drugs for drivers in Korea, we tried to analyze controlled and non-controlled drugs in alcohol-positive blood samples. 275 whole bloods, which were positive for alcohol on the roadside test, were collected from the police for two months ($Nov.{\sim}Dec.$ 2006). The analytical strategy was constituted of three steps: First, alcohol in blood samples were confirmed and quantified by gas chromatography. Second, controlled drugs were screened by $Evidence_{investigator}\;^{TM}$ (Randox, U.K.) as preliminary test. It was based on immunoassay by biochip array analyzer. Nine groups of drug abuse were screened: amphetamines, methamphetamines, cannabis, cocaine, opiates, barbiturates, methadone, benzodiazepines I (oxazepam) & II (lorazepam). Finally, confirmation of these drugs was performed by GC-MS. Blood samples were extracted by solid-phase extraction by $RapidTrace^{TM}$ (Zymark, U.S.A.). After trimethylsilyl (TMS) derivatization, eluates were analyzed to GC-MS. Total 49 drugs were investigated in this study including controlled drugs, antidepressants, 1st generation antihistamines, dextromethorphan, nalbuphine, ketamine, etc. For rapid detection, we developed the automated identification system. It was made up a new software, "DrugMan", modified Chemstation data analysis menu and newly developed macro modules. A series of peak selection, identification and reporting of the results were performed automatically by this system. Concentrations of alcohol in 275 blood samples were ranged from 0.011 to 0.249% (average, 0.119%). Among 149 blood samples, just six samples (4.0%) were showed positive results to the immunoassay: one methamphetamine and five benzodiazepines group I. By GC-MS confirmation, only benzodiazepines were detected and methamphetamine was not detected from immunoassay positive blood sample. Besides these drugs, 5 chlorpheniramines, dextromethorphan, diazepam, doxylamine, ibuprofen, lidocaine and topiramate were also detected in whole bloods by GC-MS. Conclusively, the frequency of drug abuse for Korean drivers was relatively low. There was none case which illegal drug was detected. However these results were limited to alcohol positive blood samples, so it is necessary to analyze more samples including alcohol negative blood.

Effect of Total Mixed Ration with Fermented Feed on Ruminal In vitro Fermentation, Growth Performance and Blood Characteristics of Hanwoo Steers

  • In this study, two experiments were conducted to evaluate the total mixed ration with fermented feed (TMRF) and total mixed ration (TMR) by rumen in vitro fermentation and their effects on the growth performance and blood characteristics of Hanwoo steers. In experiment 1, three Hanwoo steers ($600{\pm}47$ kg), each permanently fitted with a ruminal cannula were used. In this experiment, three diets designated as T1, TMRF (18.4% fermented feed, tall fescue, mammoth wild rye forage and whole crop barley); T2, TMRF (17.7% fermented feed, rice straw and whole crop barley); and T3, TMR (rice straw, whole crop barley and probiotics, but no fermented feed), which were subjected to rumen in vitro fermentation for 48 h. The results demonstrated that DM disappearance rate gradually increased with advancing fermentation time, but T1 and T2 were higher than the T3 (p<0.05) from 3 h to 12 h, but insignificant (p>0.05) at 24 and 48 h. None of the specific VFAs were affected except for acetic and non volatile lactic acids, which were produced more in T2 than in T1 and T3 at 24 h and 48 h of incubation. A/P was lower in T1 and T2 than inT3 at 24 h (p<0.05) and 48 h (p>0.05) of incubation. These results confirmed that TMRF-related treatment shows a superior performance to that of TMR during the ruminal fermentation period. In experiment 2, the three diets in experiment 1 plus 1 more control diet (concentrates, probiotics and 2% rice straw of body weight) were fed to the 48 Hanwoo steers ($160{\pm}10$ kg) for a period of 168 d. The results demonstrated that the daily and total live weight gain and feed efficiency were higher (p<0.05) in the TMRF and TMR groups than in the control group. SGOT, SGPT and BUN (p<0.05) were reduced in TMRF relative to the control and TMR groups by 168 d which confirmed that TMRF shows better blood profiles than the TMR and control groups. Overall, these results appear to show that TMRF has better in vitro ruminal characteristics than those of TMR; growth performance and blood profiles were also found to be superior in TMRF than in the TMR and control groups. Thus, our findings suggest that TMRF-based feed supplies are favorable for Hanwoo cattle.
