Genetic Analysis of Wheat for Plant Height by RNA-seq Analysis of Wheat Cultivars 'Keumkang' and 'Komac 5'

  • One of the most widely grown food crops in the world, wheat, is increasing more lodged since for increased rains and winds caused by abnormal climate. During the Green Revolution, shorter wheat cultivars were bred using many Rht genes to increase lodging resistance. However, since only some Rht genes were used for breeding shorter wheat, it may have had a limited impact on wheat breeding and reduced genetic diversity. Therefore, it is essential to search for genes that have breeding potential and affect dwarfism in order to increase the genetic diversity of dwarf characteristics in wheat. In this study, we performed the RNA-seq between 'Keumkang' and 'Komac 5' ('Keumkang' mutant) to analyze the difference in plant height. Differentially expressed genes (DEGs) analysis and Gene function annotation were performed using 265,365,558 mapped reads. Cluster set analysis was performed to compress and select candidate gene DEGs affecting plant height, stem and internode. Gene expression analysis was performed in order to identify the functions of the selected genes by condensing the results of the DEG analysis into a cluster set analysis. This analysis of these plant height-related genes could help reduce plant height, improve lodging resistance, and increase wheat yield. Its application to wheat breeding will also affect the increased genetic diversity of wheat dwarfism.

국내 밀 43개 품종에 대한 아그로박테리움 형질전환 효율성 검정 (Comparison of Agrobacterium-mediated Transformation Efficiency in 43 Korean Wheat Cultivars)

  • 본 연구는 국내 밀 43 품종에 대한 A. tumefaciens 형질 전환 효율을 검정하기 위해 GUS staining 분석을 수행한 것으로서 대부분의 밀 형질전환 연구가 특정 품종에 국한되어 있기 때문에 국내 장려 밀 품종에 대한 형질전환 효율에 영향을 미치는 조건에 대한 검정이 필요하다. 국내 밀 품종 중 32개에서 1개 이상의 조직에 염색된 신호가 관찰되었으며 4개 품종에서는 염색된 신호가 관찰되지 않았고 6개 품종에서 과도한 A. tumefaciens 성장이 관찰되었고, 7개 품종은 미성숙배의 크기 등의 이유로 GUS staining 분석이 불가능하여 추가 분석에서 제외하였다. 국내 밀 품종별 A. tumefaciens 감염 효율 비교 결과, 백립계 8개 품종 및 적립계 28개 품종에서는 평균적으로 유의한 차이가 없었다. 특히 조농밀, 조품밀, 새올밀, 조중밀, 새금강밀 등 적립계 품종에서 90% 이상의 높은 감염 효율이 관찰되었고, 전체 품종 중 22개는 50% 이상의 효율을 나타내었다. 본 연구를 통하여 조농밀 및 새올밀은 미성숙배에서 전체 조직에서 강한 GUS 발현으로 형질전환에 적합한 품종으로 나타났으며 금강 품종은 백립계에서 상대적으로 높은 발현을 보여 A. tumefaciens을 이용한 형질전환에 적합한 것으로 확인되었다. 추가적인 연구를 통해 품종의 조직배양 효율을 검정하고, 안정적인 GUS 발현 효율을 조사하는 것이 필요하며, 이를 통해 밀 형질전환에 이용 가능한 품종을 더욱 정밀하게 선발할 수 있을 것으로 판단된다. 해당 연구 결과는 국내 밀품종의 형질전환 효율을 높이기 위한 기초 자료로 활용 가능할 것이다.

Environmental Impacts of Korean and CIMMYT Wheat Lines on Protein Characteristics and Bread Making Quality

  • This study was conducted to compare the protein characteristics, dough rheology and bread loaf volume of Korean wheat cultivars and CIMMYT lines produced in diverse environments and to determine the genetic and environmental effects on bread making quality. Protein characteristics, including protein content and SDS-sedimentation volume, mixing properties during dough development and bread loaf volume were primarily influenced by the environment. Wheat cultivated in Jinju exhibited higher SDS-sedimentation volume based on constant protein weight and bread loaf volume than those in Suwon and Iksan. SDS-sedimentation volume based on constant protein weight, mixing time of mixograph and mixing tolerance of mixograph were positively correlated with bread volume. Korean wheat cultivars showed different allelic variations of $Glu-1$ and $Glu-3$ compared to CIMMYT wheat lines. Alchanmil, Keumkangmil and Tapdongmil could be suitable for bread making because these cultivars exhibited a 10 point $Glu-1$ score. However, Korean wheat cultivars should be introduced specific alleles in $Glu-3$ loci, including $Glu-A3b$ or $d$ and $Glu-B3b$, $d$, $f$ or $g$, to improve gluten strength related to increase bread loaf volume.

국산밀을 이용한 white layer cake의 제조 적성 (White Layer Cake-Making Properties of Korean Wheat Cultivars)

  • 8품종의 국산밀에 대한 이화학적 특성 및 white layer cake 제조 특성을 조사하였다. 국산밀의 제분율은 $62.5{\sim}71.8%$의 분포로 품종간의 차이가 큰 것으로 나타났다. 국산밀가루의 단백질 함량은 $7.70{\sim}10.58%$의 범위였으며, 회분 함량는 $0.51{\sim}0.71%$의 범위로 수입밀 박력분에 비해 다소 높았다. 국산밀가루의 mixograph 반죽특성에서 우리밀, 알찬밀, 올그루밀의 반죽 물성이 매우 약해 cake의 제조에 적합하였고 은파밀, 그루밀은 단백질 함량에 비해 안전성이 다소 떨어졌으며, 금강밀과 탑동밀가루는 반죽의 안정성이 높은 경질밀의 특성을 보여 cake의 제조에 부적합함을 보여주었다. 국산밀 cake반죽의 pH와 비중은 각각 $6.71{\sim}7.08,\;0.73{\sim}0.76\;g/cc$의 범위로 박력분 반죽과의 차이가 크지 않은 것으로 나타났다. White layer cake의 부피, 비체적은 각각 $837.5{\sim}952.5\;cc,\;2.16{\sim}2.40\;cc/g$의 범위로 알찬밀이 가장 높은 값을 주었다. 국산밀 cake은 symmetry와 uniformity에서 박력분 제조 cake 보다 약간 떨어지는 경향을 주었다. 국산밀 cake의 crumb 색도는 박력분 보다 다소 어둡고 황색도가 높은 것으로 나타났다. 국산밀 cake의 텍스쳐는 품종에 따라 차이를 보였으며 알찬밀, 우리밀에서 경도가 낮아 부드러운 것으로 나타났다.

Analysis of Genetic Diversity of Korean Wheat Cultivars Using Microsatellite DNA Polymorphisms

  • Genetic background and phylogenetic relationships among 20 Korean wheat cultivars were assessed using microsatellites after amplifying with 13 SSR primer pairs. Average allele number per primer pair was 3.36. Genetic similarities for every pair of cultivars ranged from 0.42 to 0.97, with 0.69 of overall average. Korean cultivars were divided into two major groups based on microsatellite DNA polymorphisms. Group I consisted of relatively old cultivars developed until 1970s, and group II contained the recent cultivars developed during 1980s and 1990s. Amongst old elite cultivars/lines, ‘Yukseung 3’, ‘Norin 12’ and ‘Norin 72’ contributed most to the genetic background of cultivars belonging to group I, and ‘Norin 4’, ‘Norin 12’, ‘Norin 43’ and ‘Norin 72’ to group II, respectively. The phylogenetic relationship of Korean wheat cultivars was in accordance with the genealogical data of each cultivar. The genetic background of each cultivar was assessed from the point of breeding and germplasm management such as variety identification and duplicated accessions for assisting in developing a system for the registration of new variety based on the molecular characterization in future.

Relationships between Genetic Diversity and Fusarium Toxin Profiles of Winter Wheat Cultivars

  • Fusarium head blight is one of the most important and most common diseases of winter wheat. In order to better understanding this disease and to assess the correlations between different factors, 30 cultivars of this cereal were evaluated in a two-year period. Fusarium head blight resistance was evaluated and the concentration of trichothecene mycotoxins was analysed. Grain samples originated from plants inoculated with Fusarium culmorum and naturally infected with Fusarium species. The genetic distance between the tested cultivars was determined and data were analysed using multivariate data analysis methods. Genetic dissimilarity of wheat cultivars ranged between 0.06 and 0.78. They were grouped into three distinct groups after cluster analysis of genetic distance. Wheat cultivars differed in resistance to spike and kernel infection and in resistance to spread of Fusarium within a spike (type II). Only B trichothecenes (deoxynivalenol, 3-acetyldeoxynivalenol and nivalenol) produced by F. culmorum in grain samples from inoculated plots were present. In control samples trichothecenes of groups A (H-2 toxin, T-2 toxin, T-2 tetraol, T-2 triol, scirpentriol, diacetoxyscirpenol) and B were detected. On the basis of Fusarium head blight assessment and analysis of trichothecene concentration in the grain relationships between morphological characters, Fusarium head blight resistance and mycotoxins in grain of wheat cultivars were examined. The results were used to create of matrices of distance between cultivars - for trichothecene concentration in inoculated and naturally infected grain as well as for FHB resistance Correlations between genetic distance versus resistance/mycotoxin profiles were calculated using the Mantel test. A highly significant correlation between genetic distance and mycotoxin distance was found for the samples inoculated with Fusarium culmorum. Significant but weak relationships were found between genetic distance matrix and FHB resistance or trichothecene concentration in naturally infected grain matrices.

Evaluation of Korean Noodle Quality of Korean Winter Wheat over Years and Locations

  • Noodle texture parameters of Korean style wet and dry noodles and relationships between noodle quality and flour characteristics were evaluated for two years, 1997 and 1998, and at two locations, Suwon and Deokso, using Korean winter wheat cultivars and lines. No significant difference for chewiness was found between cultivars over locations. Noodles made from flours from 1997 showed significantly higher chewiness than those from 1998. Chewiness of cooked noodles showed positively significant correlations with protein content and SDS sedimentation volume and negatively significant correlations with starch peak viscosity and flour swelling volume. Korean winter wheat cultivars, except for Gobunmil, Keumkangmil Tapdongmil, Suwon 265 and Suwon 280, showed chewiness of cooked noodles similar to commercial flours used for noodle making in Korea and Japan.

Effect of High Temperature on Grain Characteristics and Quality during the Grain Filling Period

  • Global warming has significant effects on the growth and development of wheat and can cause a reduction in grain yield and quality. Grain quality is a major factor determining the end-use quality of flour and a reduction in quality can result economic losses. Therefore, it is necessary to study the physiological characteristic of wheat to understand its response to temperature elevation, which can aid in the development of strategies to mitigate the negative effects of high temperature and sustain wheat production. This study investigated the effects of elevated temperature on grain characteristics and quality during the grain filling period of two Korean bread wheat cultivars Baekkang and Jokyoung. These two bread wheat cultivars were subjected to an increasing temperature conditions regime; T0 (control), T1 (T0+1℃), T2 (T0+2℃) and T3 (T0+3℃). The results showed that high temperature, particularly in T3 condition, caused a significant decrease in the number of grains per spike and grain yield compared to the T0 condition. The physical properties, such as grain weight and hardness, as well as chemical properties, such as starch, protein, gluten content and SDSS, which affect the quality of wheat, were changed by high temperature during the grain filling period. The grain weight and hardness increased, while the grain size not affected by high temperature. On the other hand, amylose content decreased, whereas protein, gluten content and SDSS increased in T3 condition. In this study, high temperature within 3℃ of the optimal growth temperature of wheat, quantity properties decreased while quality-related prosperities increased. To better understand the how this affects the grain's morphology and quality, further molecular and physiological studies are necessary.

Association of Puroindolines Genotypes and Grain Properties, Milling Performances and Physical Properties of Flour in Korean Wheats

  • Puroindoline alleles, grain characteristics, milling performances and physical properties of flour of 22 Korean wheat cultivars were evaluated to determine the influence of puroindolines genotypes on grain and flour characteristics and to provide useful information for improving milling and end-use quality in Korean wheat breeding programs. Nine Korean wheat cultivars carried with Pina-D1a/Pinb-D1a, 11 cultivars had Pina-D1a/Pinb-D1b and 2 cultivars were Pina-D1b/Pinb-D1a. Korean wheats carrying with Pina-D1a/Pinb-D1a genotypes showed lower test weight and thousand kernel weight, area and roundness of grain and hardness index in grain characteristics, lower flour yield and higher proportion of break flour in milling performances and lower average particle size of flour, ash and damaged starch content, water retention capacity, yellowness-blueness and higher lightness of flour than wheats with Pina-D1a/Pinb-D1b or Pina-D1b/Pina-D1a genotypes. Pina-D1a/Pinb-D1b genotypes showed lower 1000-kernel weight, grain area, higher average of particle size of flour, higher ash and damaged starch content than Pina-D1b/Pina-D1a genotypes. There was no difference in hardness index of grain, milling performances, flour color between Pina-D1a/Pinb-D1b and Pina-D1b/Pina-D1a genotypes. These results could present the information to improve milling quality and physical properties of flour in Korean wheat breeding programs.

Evaluation of agronomic traits variability in wheat germplasm

  • Wheat is one of the world's most important crop species. Recently, abnormal climate changes directly influence on the significant reduction of wheat productivity in the world. This threaten allow breeders to find new genetic resources. Wheat is one of the widely grown crops in the world. Individual cultivars / germplasm is adapted in that region where the climate is unique to each other. Therefore, introducing new genetic resources which was good in one place may better perform in another region. In this study, we evaluated germplasm in Korean environment and measured numerous agro-morphological characteristics. Information that are provided by the National Agrobiodiversity Center (Jeonju, Korea) and National Plant Germplasm System (Aberdeen, USA) were included in the analysis. Cluster analysis was performed using the unweight pair-group method of averages. The results of PCA indicated principal discriminatory characteristics of wheat landraces and varieties. Significant differences indicated high variability among the quantitative traits. Cluster analysis results showed that the groups were divided by geological climate condition. The preliminary evaluation of germplasms in Korean environment would help to develop wheat cultivars via providing useful genetic traits that are resided in alien germplasms.
