최근까지 근력 개선 또는 만성 근육 손실을 유발하는 질병을 치료하기 위해 많은 연구가 있어왔지만, 지금까지 근력 개선 유도를 위해 대표적으로 사용되는 약물은 단백질 동화 스테로이드(AAS)계열의 steroidal androgen들이 주로 이용되어 왔다. AAS는 다양한 근육 및 체중 손실성 치료를 목적으로 처방되고 있으나, 장기간 또는 과다한 복용은 피부질환, 생식 및 내분비 기능 저하, 심혈관 질환 등 다양한 부작용을 초래한다고 잘 알려져 있다. 따라서, 상대적으로 부작용이 적은 근육 신생을 촉진하는 천연물을 탐색하는 연구는 매우 중요하다. 현재까지 천연물의 처리로 인한 근육 신생의 촉진은 대표적으로 세가지 기전의 영향을 받으며, 그 기전으로는 근육조절인자(MRF)의 양성 조절, 단백질 합성 기전의 활성화 및 단백질 분해기전의 억제가 있다. 본 리뷰에서는, 근육 신생 촉진 효과가 보고된 Black ginseng, Plum 등과 같은 식물 추출물 및 Creatine, Catechin 및 여러 지방산 등 천연물 유래 단일물질에 대하여 소개하고, 더 나아가 현재까지 알려진 상기의 천연물질들의 처리에 의한 근육 신생 촉진 기전에 대하여 기전별로 요약하고자 한다.
Carcinoembryonic antigen (CEA)는 다양한 종양에서 발현되는 자가 항원으로 면역치료에서 강력한 표지 인자이며 면역치료를 위한 표적 종양항원으로 널리 알려져 있다. 그러나 수지상세포 단독 치료는 동물모델에서 종양의 발생을 억제하는 데 효과가 있지만 이미 확립된 종양을 제거하는 데는 한계가 있다. 본 연구에서는 항종양 면역 효과를 증가시키기 위하여 화학치료제인 cyclophosphamide (CYP)와 종양 특이 면역치료법인 수지상세포 백신의 병합치료 효과를 CEA를 발현하는 마우스 종양 모델에서 검증하였다. 종양세포 주입 후 2일 소종양군과 10일 대종양군에서 CYP의 항종양 효과를 비교한 결과, 소종양군에서는 100 mg/kg에서 뚜렷한 종양 성장의 억제 효과가 관찰되었지만 대종양군에서는 약한 억제 효과가 관찰되어 본 연구에서는 대종양군을 병합치료의 적합한 모델로 설정하였다. CYP 와 수지상세포 백신의 병합치료(화학면역치료) 시 종양항원 특이 면역반응이 증가되었을 뿐만 아니라 상승적인 항종양 효과가 나타났다. 또한 CYP 치료에서 나타나는 체중 감소 및 조절 T세포와 골수유래 억제세포의 증가에 의한 면역억제는 화학면역치료에 의해 개선되었다. 항원 특이 면역치료를 병합한 화학면역치료가 화학치료의 부작용을 감소시키고 항종양 효과를 증가시킬 수 있는 치료 전략이 될 수 있을 것이다.
Objectives : This study was conducted to evaluate the acute toxicity and safety of Gumiganghwal-tang (Jiuweiqianghou-tang) in Sprague-Dawley rats though the current regulatory guideline. Methods : The preliminary study showed that the single oral administration of Gumiganghwal-tang(Jiuweiqianghou-tang) did not induce any toxic effect at a dose level of 2000 mg/kg. Based on the results, 2000 mg/kg was selected as the limited dose. In this study, 10 rats of each sex were randomly assigned to two groups of 5 rats each and were administrated singly by gavage at dose levels of 0 and 2000 mg/kg. Mortalities, clinical signs, and body weight changes were monitored for the 15-day period following administration. At the end of observation period, all animals were sacrificed and complete gross postmortem examinations were performed. Results : Throughout the study period, no treatment-related deaths were observed. There were no adverse effects on clinical signs, body weight, and gross findings at all treatment groups. Conclusions : These results showed that the single oral adminstration of Gumiganghwal-tang(Jiuweiqianghou-tang) did not cause any toxic effect at the dose levels of 2000 mg/kg in rats. In conclusion, the $LD_{50}$ of Gumiganghwal-tang (Jiuweiqianghou-tang) was considered to be over 2000 mg/kg body for both sexes.
Objectives: This study was designed to investigate the effect of Gami-cheongpyesagan-tang extract (GCST) on high fat diet-induced obesity in rats. Methods: The mice were divided into six groups; normal diet control, high fat diet control (HFD), HFD+GCST administrated group (100, 200, and 400 mg/kg) and olistat-admistrated group. Obesity was induced by high fat diet (45%) for 7 weeks in mice, and GCST was administrated orally every day for 7 weeks. The body weight, food intake, and serological markers such as total cholesterol, triglyceride, lipid contents, leptin, adiponectin and glutamic oxaloacetic transaminase/glutamic pyruvic transaminase were measured in mice. The mRNA expression of obese-associating genes such as sterol regulatory element-binding protein (SREBP)-1c, fatty acid synthase (FAS), stearoyl-CaP desaturase (SCD-1), peroxisome proliferator-activated receptor $(PPAR)-{\alpha}$, COA oxidase (ACO), and carnitine palmitoyltransferase ($CPT-1{\alpha}$) was analyzed by reverse transcription polymerase chain reaction. Results: The administration of GCST at 400 mg/kg, significantly reduced the increase of body weight and food intake as well as food efficiency compared to HFD group. GCST decreased the serum levels of triglyceride, total cholesterol, low-density lipoprotein-cholesterol, leptin in HFD control group and inhibited lipid accumulation in liver and adipose tissues, but did not increase high-density lipoprotein-cholesterol. In the liver tissues of GCST administrated HFD group, the mRNA levels of SREBP-1c, FAS and SCD-1 were decreased and the mRNA levels of $PPAR-{\alpha}$, ACO, and $CPT-1{\alpha}$ were increased. Conclusions: These results indicate that GCST could improve high fat diet induced obesity through inhibiting the hyperlipidemia in fatty Liver. It suggest that GCST may be used clinically for declining the accumultion of body fat with hyperlipidemia.
Background: Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease associated with multiple metabolic disorders. The medicinal plant Curcuma longa L. is widely distributed in Asia and has been used to treat a spectrum diseases in clinical practice. To date, there are inadequate reports of the effects of C. longa 50% EtOH extract (CE) on NAFLD. Therefore, in this study, we evaluate the CE on an NAFLD animal and elucidate the mechanism of action. Methods and Results: C57BL/6J mice fed a methionine-choline deficient diet (MCD) were treated with CE or milk thistle, and changes in inflammation and stetosis were assessed. Experimental animals were divided into six group (n = 10); Normal, MCD, MCD + CE 50 mg/kg/day (CE 50), MCD + CE 100 mg/kg/day (CE 100), MCD + CE 150 mg/kg/day (CE 150), and the Control, MCD + Milk thistle 150 mg/kg/day (MT 150). Body weight, liver weight, liver function, and histological changes were assessed in experimental animals. Quantitative real-time polymerase chain reaction and western blot analyses were performed on samples collected after 4 weeks of treatment. We observed that CE administration improved MCD-diet-induced lipid accumulation, and triglyceride (TG) and total cholesterol (TC) levels in serum. Treatment with CE also decreased hepatic lipogenesis through modulation of the sterol regulatory element binding protein-1 (SREBP-1), CCAAT-enhancer binding protein α (C/EBPα), fatty acid synthase (FAS), and peroxisome proliferator-activated receptor γ (PPARγ) expresion. In addition, the use of CE increased adenosine monophosphate-activated protein kinase (AMPK) phosphorylation and inhibited the up-regulation of toll-like receptor (TLR)-2 and TLR-4 signaling and the production of inflammatory mediators. Conclusions: In this report, we observed that CE regulated lipid accumulation in an MCD dietinduced NAFLD model by decreasing lipogenesis. These data suggeste that CE could effectively protect mice against MCD-induced NAFLD, by inhibiting the TLR-2 and TLR-4 signaling cascades.
This study was performed to determine the effects of dietary fat sources, i.e., beef tallow, soybean oil, olive oil and coconut oil (each 3% in feed), on the growth performance, meat quality and gene expression in growing-finishing pigs. A total of 72 crossbred pigs (Landrace${\times}$Large White${\times}$Duroc) were used at $71{\pm}1$ kg body weight (about 130 d of age) in 24 pens ($320{\times}150$ cm) in a confined pig house (three pigs per pen) with six replicate pens per treatment. The growing diet was given for periods of $14{\pm}3$ d and the finishing diet was given for periods of $28{\pm}3$ d. The fat type had no significant effect either on growth performance or on chemical composition or on meat quality in growing-finishing pigs. Dietary fat type affected fatty acid composition, with higher levels of unsaturated fatty acids (UFAs) and monounsaturated fatty acids (MUFAs) in the olive oil group. Microarray analysis in the Longissimus dorsi identified 6 genes, related to insulin signaling pathway, that were differentially expressed among the different feed groups. Real time-PCR was conducted on the six genes in the longissimus dorsi muscle (LM). In particular, the genes encoding the protein kinase, cAMP-dependent, regulatory, type II, alpha (PRKAR2A) and the catalytic subunit of protein phosphatase 1, beta isoform (PPP1CB) showed the highest expression level in the olive oil group (respectively, p<0.05, p<0.001). The results of this study indicate that the type of dietary fat affects fatty acid composition and insulin signaling-related gene expression in the LM of pigs.
본 연구는 갈근과 지황 복합물과 유산소 운동이 난소절제로 갱년기를 유도한 흰 쥐에서 나타나는 지질 및 당대사 이상에 미치는 영향을 확인하였다. 난소절제 후 8주간 AIN-76A 식이를 급여한 결과 체중증가량, 식이섭취량, 체 지방, 혈장 LDL-C, 공복혈당 증가를 통해 갱년기 동물 모델이 유도되었음을 확인할 수 있었다. 갈근과 지황 복합물과 유산소 운동은 서로 다른 기전으로 에스트로겐 분비 저하로 나타나는 비만 및 대사이상을 정상화하는 유익한 결과를 나타냈다. 지방 조직에서 확인된 관련 기전으로는 갈근과 지황 복합물로 인한 아디포넥틴 유전자 발현 증가 및 렙틴저항 억제, 유산소 운동으로 인한 아디포넥틴 및 인슐린 수용체 유전자 발현 증가, 갈근과 지황 복합물 및 유산소 운동의 병행으로 인한 아디포넥틴 유전자 발현 증가 상승효과가 관찰되었다. 이상의 결과로 갈근과 지황 복합물과 유산소 운동의 병행은 폐경기 에스트로겐 결핍으로 인한 대사이상을 개선하는 효과가 있을 것으로 기대한다. 향후에는 폐경기 유도 동물모델의 개선과 함께 단백질 및 신호전달분자 수준에서 추가적인 기전연구가 필요할 것으로 사료된다.
Quercetin의 지질대사 개선 효과에 대한 작용기전을 확인하기 위해 C57BL/6J mouse를 사용하여 실험을 수행하였다. 고콜레스테롤혈증을 유도하기 위해 6주간 1% 콜레스테롤과 0.5% cholic acid를 함유하는 고콜레스테롤 식이를 급여하였으며, quercetin은 0.05%와 0.1%의 수준으로 고콜레스테롤 식이에 추가하여 같은 기간 동안 제공하였다. Quercetin은 혈청과 간의 중성지방 및 콜레스테롤 수준을 용량 의존적으로 감소하는 것으로 나타났다. 고콜레스테롤 식이를 섭취한 쥐의 간에서 지방 합성을 촉진하는 SREBP-1c, ACC1 및 FAS 유전자 발현이 quercetin 섭취에 의해 억제되는 것을 확인하였다. Quercetin은 간세포 내에서 에너지 대사를 조절하는 AMPK 활성을 증가시켰다. 이에 반해 암세포 증식을 촉진하고 지방간에서 높게 발현되는 miR-21 발현은 quercetin 섭취에 의해 감소되었다. 본 연구의 결과는 quercetin이 고콜레스테롤 식이 섭취 쥐에서 혈청과 간의 지질 수준을 낮추는 지질대사 개선 효과가 있으며, 이러한 효과의 일부는 간 내 지방합성 유전자 (SREBP-1c, ACC1 및 FAS) 발현, AMPK 활성 및 miR-21 조절을 통해 매개된다는 것을 시사한다.
본 연구에서는 정향과 마가목 복합물 (SS)의 항비만 효과를 알아보기 위해 실험을 진행하였다. SS 투여는 3T3-L1 세포 내 TG와 TC가 유의적으로 감소하는 효과를 나타냈으며, 지질 합성 관련 유전자와 지방산 산화 관련 유전자 발현을 조절하는 효과를 보여주었다. 비만이 유도된 C57BL/6 mice에서 SS 투여는 혈청 내 leptin 호르몬 수치를 감소시켰으며, AMPK/ACC/SREBP-1 경로를 경유하여 TG 합성을 억제하였다. 또한, 조직병리학적 분석을 통해 지질 축적과 지방세포의 크기가 감소된 것을 확인하였다. 따라서 SS는 비만의 예방과 치료를 위한 잠재력을 갖춘 소재로 사료된다.
본 연구에서는 유전성 비만 마우스를 이용하여 아프리칸 망고 추출물의 항비만 효과를 알아보고자 하였다. 기존의 연구 내용을 아프리칸 망고 추출물의 섭취 농도를 식이의 1%와 2%로 정하고, AIN93G diet에 아프리칸 망고 추출물 분말을 1%와 2% 섞어 diet를 제작하였다. 실험 식이를 8주간 제공한 후 실험 종료 시 실험군들의 체중 변화는 ob/ob군에 비해 유의적으로 감소하였다. 실험동물의 간 및 지방조직의 무게를 측정한 결과 간의 무게와 지방조직의 무게가 ob/ob군에 비해 유의적으로 감소하였다. 실험동물의 혈청 지질 함량은 TG, TC, LDL-콜레스테롤이 ob/ob군에 비해 감소하였으나 유의성은 나타나지 않았고, HDL-콜레스테롤은 고농도 섭취 시 ob/ob군에 비해 유의적으로 증가하였다. Adiponectin 유전자의 발현은 ob/ob군이 가장 낮았고, 실험식이 섭취군들은 ob/ob군에 비해 유의적으로 증가하였다. Adipogenic enzyme과 transcription factor인 FAS, LPL, PPAR-${\gamma}$, C/EBP, SREBP 유전자의 발현은 ob/ob군이 가장 높게 나타났고, 실험식이 섭취군들은 ob/ob군에 비해 유의적으로 낮았다. 전반적으로 본 실험에서는 아프리칸 망고 추출물이 leptin 유전자가 결핍된 비만 동물 모델에서 체중 감소 및 혈청 지질 함량을 개선시켜 주는 효과가 있고, adipogenic enzyme과 transcription factor 조절에 유익한 효과가 나타났다. 이러한 결과로부터 아프리칸 망고 추출물이 항비만 효과를 가진 천연 기능성 식품의 개발에 있어 기초자료로 활용할 수 있을 것으로 기대할 수 있다.
