키토산 유도체인 Sulfated N-acetyl Chitosan의 종양전이 억제효과 (Inhibitory Effects of Tumor Metastasis by Chitosan Derivative, of Sulfated N-acetyl Chitosan)

  • 키토산 유도체 인 sulfated N→acetyl chitosan을 합성하여 이 화합물을 쥐에 이식한 B16/BL6 mela noma에 대하여 폐암전이의 억제효과를 조사하였다. 키토산의 황산유도체는 13C-n.m.r.에 의하여 확인한 결과 3, 6, O-disulfate 엄을 알 수 있었다. SuI fated N-acetyl chitosan을 100mg/kg을 투여하였을 때 B16/BL6의 melanoma cells을 가장 효과적 으로 억제하였고 종양 무게의 증식도 대조군에 비해 억제되었다. 폐암의 전이에 있어서 B16/BL6에 sulfated N-acetyl chitosan을 LV. V로 주사하였을 때 B16/BL6의 melanoma의 전이 세포의 수는 자발적인 전이에 있어서도 용량 의존형(20 ~ 1OOmg/kg) 에 따라 줄어들었다. B16/BL6을 접종한 후 sulfated N-acetyl chitosan을 I.V.로 투여했을 때 전이세 포의 수는 현저히 감소하였다. Sulfated N-acetyl chitosan은 laminin의 종양세포에의 세포접착 능력을 부분적으로 억제하였다. 이와 같은 결과는 chitosan의 유도체인 sulfated N-acetyl chitosan이 세포접착 능력이 실험적으로 자발적인 암전이 모델에 효과적으로 작용하였다.

Preparation and validation of Chitosan-phthalocyanine complex - absorber of mutagens and carcinogens -

  • Phthalocyanine, a water soluble porphyrin derivative and dye, is known to inhibit the mutagenic and carcinogenic actions of compounds having polycyclic structures, e.g. heterocyclic amines. There is evidence that this adsorbent effect shows by a complex formation between the porphyrin-like structure of phthalocyanine and the planar molecular surfaces of theses compounds. That phthalocyanine can form an insoluble material when mixed with chitosan, a polyglucosamine, and that the solid chitosan-phthalocyanine, named Eco-Blue, thus prepared can efficiently adsorb polycyclic mutagenic compounds. The adsorption was experimented by UV/VIS spectrometry. The adsorbent effects of mutagens and carcinogens was identified by Gas chromatography (GC) and Ames Test. The adsorbed polycyclic mutagens were elutable with buffer, but only to small extents. Chitosan-phthalocyanine may be expected to be useful as an adsorbent against polycyclic mutagens and carcinogens.

PVA/Chitosan 블랜드필름의 효소와 pH 변화에 따른 분해효과 (Effects of Degradation according to Enzyme and pH changes of PVA/chitosan blend films)

  • Polyvinyl alcohol[PVA] is useful for the production of water-soluble packaging, paper, textile sizes. PVA and Chitosan are known as biodegradable polymers. PVA/chitosan blend films were prepared by solution blends method in the weight ratio of chitosan for the purpose of useful biodegradable films. Thermal and mechanical properties of PVA/chitosan blend films such as DSC, impact strength, tensile strength and morphology by SEM were determined. As a result, The ratio of 10.0wt% PVA/chitosan blend films were similar to PVA. Blend films were completely degraded pH 4.0 better than 7.0, 10.0 in the buffer solution. Also, Blend films were rapidly degraded enzyme(${\beta}-glucosidase$) solution better than pH solution by Enzymolysis.

Chitosan/whey Protein (CWP) Edible Films Efficiency for Controlling Mould Growth and on Microbiological, Chemical and Sensory Properties During Storage of Göbek Kashar Cheese

  • The objective of present study was to evaluate the effects of the application of chitosan and chitosan/whey protein on the chemical, microbial and organoleptic properties of Göbek Kashar cheese during ripening time (on 3rd, 30th, 60th and 90th d). Difference in microbiological and chemical changes between samples was found to be significant (p<0.05) during ripening period. Cheese samples with edible coating had statistically lower mould counts compared to the uncoated samples. Furthermore the highest and lowest mould counts were determined in control (4.20 Log CFU/g) and other samples (<1 Log CFU/g) at 60th and 90th d of storage. All samples exhibited higher levels of water soluble nitrogen and ripening index at the end of storage process. At the end of 90 day storage period, no signicant dierences in salt and fat values were observed among the cheeses studied. The edible coatings had a beneficial effect on the sensory quality of cheese samples. In the result of sensory analysis, while cheese C and the chitosan coated cheese samples were more preferred by the panellists, the chitosan/whey protein film-coated cheese samples received the lowest scores. This study shows coating suggests could be used to improve the quality of cheese during ripening time.

Absorption Behavior in the Body of Chitosan Oligosaccharide according to Molecular Weight; An In vitro and In vivo Study

  • Chitosan has a wide range of applications in biomedical materials as well as in dietary supplements. Chitosan oligosaccharide with free-amine group (COFa) is an improvement over traditional chitosan that lacks the usual impurities and materials detrimental to the body. Based on a previous study of water soluble chitosan (WSC, chitosan lactate), we investigated the molecular weight (Mw) - dependent absorption phenomena of COFa in vitro and in vivo with various Mws. The absorption of CO Fa was significantly influenced by its molecular weight. As Mw increases, the absorption decreases. The absorption profiles for 5 K COFa (Mw=5 kDa) were observed to be more than 10 times higher than those of high molecular weight chitosan (100 K HWSC Mw=100 kDa) in both in vitro and in vivo transport experiments. Furthermore, the in vitro transport experiment suggested that transcellular transport of the COFa (Mw <10 kDa) through Caco-2 cell layer could occur with a negligible cytotoxic effect. The COFas showed a cytotoxic effect on Caco-2 cells that was dependent on dose and Mw. COFa could be transported transcellularly through the Caco-2 cell layer.

  • Chitosan is derived from chitin by deacetylation in the presence of alkali, which is a copolymer consisting of $\beta$-(1longrightarrow4)-2-acetamido-D-glucose and $\beta$-(1longrightarrow4)-2-amino-D-glucose units with the latter usually exceeding 80% (Arvanitoyannis et al., 1998). Chitosan has been developed as new physiological material since it possesses antibacterial activity, hypocholesterolemic activity and antihypertensive action. However, even though chitosan has very strong functional properties in many areas, its high molecular weight and high viscosity may restrict the use in vivo. In addition, there is little doubt that such properties will influence absorption in the human intestine. Recently, studies on chitosan have attracted interest for converted chitosan to oligosaccharide, because the oligosaccharide possesses not only water-soluble property but also versatile functional properties such as antitumor activity, immune-enhancing effects, enhancement of protective effects against infection with some pathogens in mice, antifungal activity, calcium absorption accelerating effect (Jeon et al., 1999) and antimicrobial activity. There is, however, little information on the toxicity of chitosan oligosaccharide. (omitted)

키토산 전처리가 감즙염색 면직물의 염색성과 항균성에 미치는 효과 (The Effects of Chitosan Pretreatment on the Dyeabilities and Antibacterial Activities of Persimmon Juice-Dyed Cotton Fabrics)

  • Environmentally and human compatible chitosan were pretreated on cotton fabrics which were then dyed with 100% persimmon juice. The chitosan concentration was 1% and the chitosan types were high molecular weight chitosan (1980cps), low molecular weight chitosan (18첸), chitosan oligomer and water soluble chitosan. The properties of the fabric surfaces, the dyeabilities, the color fastnesses, the antibacterial activities, the strengths, the elongations and the drape stiffnesses were evaluated. The properties of the chitosanpretreated, persimmon juice-dyed cotton fabrics (CLP) were compared to those of the untreated (CN), chitosan treated (CL) and persimmon juice-dyed fabrics (CP). The results were as follows. The fibers extruded from the surface of CN decreased on CP. The air between the fibers within CN were substituted by chitosan solution or persimmon juiceand decreased within CLP according to SEM observations. The effects of chitosan treatment, the chitosan molecular weights and the degrees of deacetylation of chitosan on the dyeabilities of the persimmon juice-dyed cotton fabric were not distinct. The curing after chitosan padding improved the dyeabilities of CLP compare to noncuring. The strengths of CP decreased and those of CL increased, compared to those of CN. The strengths of CLP were greater than those of CP. The elogations of CP and CL were greater than those of CN. The strengths and elongations of CLP were greater than those of CN. The chitosan treatments improved the strengths but not the elongations. The drape stiffnesses of CL, CP and CLP were greater than those of CN. The antibacterial activites of chitosan pretreated, persimmon juice-dyed cotton fabrics against Staphylococcus aureus were increased by more than 98% by persimmon juice.

생체재료로서의 고순도 수용성 키토산 올리고당의 제조와 특성 (Preparation and Characterization of Highly Pured Water-soluble Chitosan Oligosaccharides as Biomaterials)

  • 본 연구에서는 유전자 전달체로 응용하기 위하여 다양한 분자량의 고순도 수용성 키토산 올리고당을 한외여과막을 이용하여 분급화 하였다. 분급화한 고순도 수용성 키토산 올리고당의 평균분자량과 분포를 측정하였고, 매우 좁은 분포를 갖는 것을 확인하였다. 고순도 수용성 키토산 올리고당의 분자량은 한외여과막에 따라 1에서 10 kDa의 범위를 가진다는 것을 젤크로마토그래피 측정을 통하여 확인하였다. 분급화된 키토산의 구조는 $^1H$ NMR과 FTIR을 통하여 구조를 확인하였고, UV를 통하여 탈아세틸화도를 측정한 결과 90% 이상의 높은 탈아세틸화도를 갖는다. 본 연구를 통하여 제조된 고순도 키토산 올리고당은 세포독성이 없고 아주 좁은 분자량 분포와 높은 순도를 갖고 있음을 확인하였다.

분리 대두단백질과 염화칼슘을 첨가하여 제조한 수용성 키토산 필름의 특성 (Effect of Soy Protein Isolate and Calcium Chloride on the Properties of Water Soluble Chitosan Film)

  • 수용성 키토산 필름 용액에 분리 대두단백질(SPI)과 $CaCl_2$ 첨가가 키토산 필름의 물성에 미치는 영향을 알아보기 위해 이들을 첨가하여 제조한 필름의 색도, puncture strength, 인장강도와 신장률, 수증기투과도를 측정하였다. 필름의 puncture strength는 0.15% $CaCl_2$를 첨가하여 제조한 키토산 필름이 $458.8\;g_f$로 대조구의 $174.5\;g_f$보다 약 2.5배 증가된 가장 높은 값을 나타냈다. 인장강도의 경우 대조구가 0.06 MPa인 반면에 0.05% $CaCl_2$를 첨가한 키토산 필름은 1.58 MPa로 가장 높은 값을 보였고 SPI를 2.5% 첨가한 필름은 0.73 MPa로 나타났다. 필름의 신장률은 대조구가 12.25%인 반면 0.15% $CaCl_2$를 첨가한 필름은 44.80%로 $CaCl_2$의 양이 증가함에 따라 커지는 경향을 보였으며, 수증기투과도는 $CaCl_2$를 첨가한 필름이 SPI를 첨가한 필름보다 더 낮은 값을 보였다.

Effect of the Fish Oil Fortified Chitosan Edible Film on Microbiological, Chemical Composition and Sensory Properties of Göbek Kashar Cheese during Ripening Time

  • Objective of the present study is to investigate the effect of coated edible films with chitosan solutions enriched with essential oil (EO) on the chemical, microbial and sensory properties of Kashar cheese during ripening time. Generally, no differences were found in total aerobic mesophilic bacteria, streptococci and lactoccocci counts among cheeses but these microorganism counts increased during 60 and 90 d storage especially in C1 (uncoated sample) as compared with coated samples. Antimicrobial effectiveness of the films against moulds was measured on 30, 60, and 90 d of storage. In addition of fish EO into chitosan edible films samples were showed to affect significantly decreased the moulds (p<0.05) as 1.15 Log CFU/g in C4 (with fish oil (1% w/v) fortified chitosan film) on the 90th d, while in C1 as 3.89 Log CFU/g on the 90th d of ripening. Compared to other cheese samples, C2 (coated with chitosan film) and C4 coated cheese samples revealed higher levels of water-soluble nitrogen and ripening index at the end of storage. C2 coated cheese samples were preferred more by the panellists while C4 coated cheese samples received the lowest scores.
