매화꽃봉오리의 추출용매별 항산화 효과 (Antioxidant Activities of Plunus mume flower buds Extract by Various Solvents)

  • This study investigated the antioxidant activities of distilled water, ethanol and methanol extracts of Plunus mume flower buds (PFB). The various solvent extracts of PFB were evaluated for their total polyphenol, flavonoid, reducing power and free radical scavenging activities by FRAP and DPPH analysis. The ethanol extract of PFB contained significantly higher amounts of total polyphenols (145 mg GAE/g) and flavonoids (25.43 mg QE/g) than methanol (132 and 25.42) and distilled water (113.6 and 18.04). Among solvent extracts of PFB, the ethanol extract showed the highest antioxidant activities. The 100% ethanol extract of PFB contained significantly higher amounts of total polyphenols and flavonoids than 70% and 50% ethanol extracts. Moreover, the 100% ethanol extract of PFB showed high efficacy in DPPH radical scavenging activity and in collagenase inhibition activity. This results suggest that 100% ethanol extract of PFB has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from PFB for usage of cosmeceutical and/or food industries.

Screening of Potent Anti-dementia Acetylcholinesterase Inhibitor-containing Edible Mushroom Pholiota adiposa and the Optimal Extraction Conditions for the Acetylcholinesterase Inhibitor

  • To develop a new anti-dementia acetylcholinesterase (AChE) inhibitor from edible mushrooms, AChE inhibitory activities were determined on water and ethanol extracts of various edible mushrooms from oriental medicine markets and agriculture markets. As a result, the 70% ethanol extract from Pholiota adiposa fruiting body had the highest AChE inhibitory activity of 30.6, and its water extract also had an AChE inhibitory activity of 23.8%. Therefore, we finally selected P. adiposa as a potent anti-dementia AChE inhibitor-containing mushroom. The AChE inhibitor of P. adiposa was maximally extracted when its fruiting body was treated with water for 3hr at $70^{\circ}C$ and 70% ethanol for 12 hr at $70^{\circ}C$, respectively.

율피의 생리활성 몇 미백효과를 이용한 화장품신소재에 관한 연구 (A Study on the Application of New Cosmetic Materials of Whitening Effect and the Physiological Activities of Chestnut Inner Shell)

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

동충하초의 항산화작용 및 아질산염 소거작용 (Antioxidative and Nitrite Scavenging Activities of Cordyceps militaris Extracts)

  • 인공재배한 번데기동충하초의 균사체와 자실체를 각각 물과 70% 에탄올로서 추출하여 전자공여 능과 아질산염 소거능을 조사하였다. 균사체 추출물의 전자공여능은 300∼1,000 ppm의 물추출물에서 37∼47%였으나, 에탄올추출물에서 57∼70%로서 에탄을 추출물이 우수하였다. 자실체 추출물의 전자공여능은 300∼1,000 ppm에서 물추출물과 에탄올추출물 모두 19∼48%로서 균사체 추출물에 비하여 낮은 편이었다. 아질산염 소거능은 pH 1.2, 3.0, 4.2, 6.0에서 측정한 길과, pH 1.2에서 가장 높았으며 PH 증가에 따라 감소하였다. 균사체 추출물의 아질산염 소거능은 pH 1.2에서 1,000 ppm의 물추출물은 23%, 에탄올추출물은 37%로서 에탄을 추출물이 우수하였다. 자실체 추출물의 경우에는 pH 1.2, 1,000 ppm 농도에서 물추출물은 12%, 에탄올추출물은 37%의 아질산염 소거능으로서 에탄올추출물이 우수하였다 전자공여능과 아질산염 소거능에서 균사체 추출물이 우수하였으며 용매 별로는 에탄올 추출물이 물추출물보다 우수하였다.

추출용매에 따른 흑양파의 항산화 활성 비교 (Comparison of Antioxidant Activities of Black Onion Extracts)

  • 흑양파를 ethanol, methanol 및 물로 각각의 추출물을 제조하고, 이 추출물의 항산화활성을 비교 검토함으로서 흑양파 추출물을 이용한 다양한 가공식품의 제조에 필요한 기초자료를 제공하고자 하였다. 흑양파 물 추출물이 28.56%로 수율이 가장 높았으며, 총 페놀도 13.5 mg/g로 가장 많았다. MTT assay 이용한 Raw 264.7의 세포독성에서 물 추출물이 가장 양호한 세포보호 효과를 보였다. 흑양파 ethanol 추출물과 물 추출물에서 유의적으로 높은 환원력을 나타내었고, 나머지 추출물에서는 차이가 없었다. 흑양파 추출물의 농도 증가에 따라 DPPH 라디칼소거능이 증가하였으며, 추출물의 농도가 $1000{\mu}g/mL$일 때 양파 ethanol 추출물은 23.29%, 흑양파 물 추출물은 65.99%로 흑양파가 양파에 비하여 항산화력이 매우 높았다. ABTS 저해활성은 DPPH에 의한 라디칼 소거능과 마찬가지로 추출물의 농도에 의존적으로 상승하였다. 이상의 결과를 종합하면 흑양파 ethanol 이나 methanol 추출물보다는 물 추출물이 우수한 생리활성 효과를 나타내므로 흑양파 물 추출물을 새로운 식품소재로 이용할 수 있을 것으로 판단된다.

차조기(Perilla frutescens var. acuta kudo) 잎의 물과 에탄올 추출물의 항산화 활성 (Antioxidant Activities of Extract with Water and Ethanol of Perilla frutescens var. acuta kudo Leaf)

  • 본 연구에서는 차조기의 30, 50, 70, 95% 에탄올 및 물추출물과 분획물의 전자 공여능, 지질산화 억제능, SOD 유사활성 및 xanthin oxidase 저해 활성을 측정하였다. 추출물의 전자 공여능은 $18{\sim}30%$ 정도로 낮았으나, 분획물에서는 95%과 70% 에탄올 추출물의 chloroform 분획이 50% 수준의 활성을 보였고, ethyl acetate 분획에서는 모든 추출용매에서 75% 이상의 활성을 보였다. 특히, 70% 에탄올 추출물의 ethyl acetate 분획물이 93.4%로 가장 높은 활성을 보였으며 $RC_{50}$에 요구되는 농도는 200.5 ppm이었다. 저장 온도 $40^{\circ}C$에서 linoleic acid의 자동산화를 가장 효과적으로 억제하는 것으로 나타난 추출물은 70% 에탄올 이었다. 분획물의 경우, 95% 에탄추출물은 hexane과 ethyl acetate 가용성분이 그 외 에탄올 추출과 물추출은 chloroform과 ethyl acetate 가용성분이 가장 높은 억제효과를 나타내었다. SOD 유사활성은 에탄올 추출물이 $28{\sim}32%$, 물추출물이 30.3%이었으며, 70% 에탄올 추출물이 32.4%로 가장 높았다. 분획물에서는 70% 에탄올 추출물의 ethyl acetate 가용분이 38.8%로 활성이 가장 높았으며 $IC_{50}$에 요구되는 농도는 1549.0ppm이었다. Xanthine oxidase 저해 활성은 에탄올추출물이 $36{\sim}41.2%$이었으며, 물추출물이 46.9%로 추출물중 가장 높았다. 분획물에서는 물추출의 ethyl acetate 가용분이 1,000 ppm 농도에서 52%의 활성을 보여 가장 높았다.

Antioxidant Activities and Phenolic Compounds Composition of Extracts from Mulberry (Morus alba L.) Fruit

  • The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity

감잎의 물 및 에탄올 추출물이 한국인 위암 세포주에 미치는 항암효과 (Anticancer Effect of Persimmon Leaf Extracts on Korean Gastric Cancer Cell)

  • This study was performed to investigate the in vitro and in vivo anticancer effects of persimmon leaf extracts on human gastric cancer cells. In vitro anticancer effects of persimmon leaf extracts (water extract at 8$0^{\circ}C$ for 3 hours, water extract at room temperature for 48 hours, 50% ethanol extract at 8$0^{\circ}C$ for 3 hours, 50% ethanol extract at room temperature for 48 hours, 75% ethanol extract at 8$0^{\circ}C$ for 3 hours and 75% ethanol extract at room temperature for 48 hours) on SNU16 (Korean gastric cancer cell) were investigated by MTT assay. Persimmon leaf extracts exhibited strong in vitro anticancer effects. We found that the higher the ethanol content of the solvent, the stronger the in vitro anticancer effects. Extraction yields, contents of flavonoids, vitamin A, vitamin C and vitamin E were measured. We found that the higher the ethanol content of the solvent, the higher the extraction yields and the contents of flavonoids, vitamin A and vitamin E. Among persimmon leaf extracts, 75% ethanol 8$0^{\circ}C$ extract showed the highest extraction yield, the highest contents of flavonoids, vitamin A and vitamin E and exhibitied the strongest in vitro anticancer effect on SNU16. Therefore, 75% ethanol 8$0^{\circ}C$ extract was chosen as the material to investigate in vivo anticancer effects. In vivo anticancer effect of persimmon leaf 75% ethanol 8$0^{\circ}C$ extract was investigated in SNU16 transplanted nude mice. Twenty five female nude mice (BALB/c) were blocked into five groups according to body weight and raised for 4 weeks with diets containing 4% (w/w), 8% (w/w) persimmon leaf 75% ethanol 8$0^{\circ}C$ extract, with IT (intratumoral) injection treatment with 1.65 mg/100 $\mu$1, 3.3 mg/100 $\mu$1 concentration every other day 3 weeks after SNU16 was transplanted. Persimmon 75% ethanol 8$0^{\circ}C$ extract significantly lowered tumor weight and tumor volume in SNU16 transplanted nude mice. Tumor weight and tumor volume in all experimental groups were significantly lower than those in the control group. Helper T cell (CD4) levels of mice injected with 3.3 mg/100 $\mu$1 extract significantly increased. Cytotoxic T cell (CD8) levels in all experimental groups significantly increased and helper/cytotoxic T cell ratios in all experimental groups significantly decreased. Natural killer cell and MHC class II molecule in all experimental groups significantly increased. In conclusion, persimmon leaf 75% ethanol 8$0^{\circ}C$ extract exhibited strong in vitro and in vivo anticancer effects against SNU16 cells and it increased cytotoxic T cell, natural killer cell and MHC classII molecule in experimental groups in SNU16 transplanted nude mice.

야관문 추출물의 생리 활성 (Physiological activities of Lespedeza cuneata extracts)

  • 본 연구에서는 유용성분을 함유하고 있는 야관문을 기능성 소재로 활용하고자 열수 및 50% 에탄올을 사용하여 추출물을 제조한 후 야관문 추출물의 생리 활성을 조사하였다. 추출수율은 50% 에탄올 추출물에서 17.60%로 열수추출물 12.60%보다 높은 수율을 나타내었다. 총 페놀 및 플라보노이드 함량은 50% 에탄올 추출물에서 각각 242.26 mg/g 및 160.73 mg/g으로 높은 함량을 나타내었다. DPPH radical 소거활성은 열수추출물에서 92.07% 및 50% 에탄올 추출물에서 96.38%로 나타나 50% 에탄올 추출조건의 항산화 활성이 높았다. Superoxide radical 소거활성 및 tyrosinase 저해활성은 50% 에탄올 추출물 $1,000{\mu}g/mL$에서 각각 94.46% 및 63.31%로 높은 활성을 나타내었다. 아질산염 소거활성은 50% 에탄올 추출시 높게 나타났으며, pH에 따라 산성에 가까울수록 아질산염 소거활성이 높게 나타났다. Nitric oxide 생성량은 50% 에탄올 추출 $250{\sim}1,000{\mu}g/mL$에서 $7.15{\sim}20.61{\mu}M$로 유의적으로 높은 활성을 나타내었으며 시료 농도에 따라 농도 의존적으로 증가하였다. 인간 폐암세포주(A549), 인간 자궁경부암 세포주(HeLa), 인간 간암세포주(Hep3B) 및 마우스 복수암 세포주(sarcoma180)에 대한 암세포 생육 저해활성을 측정한 결과, 열수 추출물 및 50% 에탄올 추출물 모두 암세포생육 저해활성을 보여주었으며, 특히 HeLa에서 상대적으로 높은 항암활성을 나타내었다. 따라서, 야관문 50% 에탄올 추출물은 향후 기능성 식품 소재 개발에 있어 산업적으로 활용 가능할 것으로 기대된다.

동백(Camellia japonica L.)유박 추출물의 항균활성 (Antimicrobial Activity of Defatted Camellia(Camellia japonica L.) Seeds Extract)

  • To developed natural food preservatives, ethanol and water extracts were prepared from the defatted camellia(Camellia japonica L.) seeds. Antimicrobial activities were examined against 14 microorganisms which were food borne pathogens and/or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract showed antimicrobial activities against several microorganisms tested, but not on lactic acid bactria. Especially, minimum inhibitory concentrations(MIC) for yeasts were as low as 1mg/ml. Water extract exhibited antimicrobial activities for the yeasts tested, but almost bacteria were not observed. The ethanol extract was fractionated in the order of hexane, chloroform, ethylaetate and butanol fractions to test antimicrobial activity. The strongest activity for the tested yeasts were found in the butanol fractions, but bacteria were chloroform fraction. Hansenula anomala treated with ethanol were observed by scanning electron micrograph, and the results exhibited morphological changes, including the irregularly contracted cell surface and expanded ellipsoidal shape.
