Extract of water-soluble polysaccharide (CFWx), showing inhibiting activity on ${\alpha}-glucosidase$, was prepared from the fruiting bodies of Cordyceps militaris by hot-water extraction, and ethanol precipitation. Chemical characteristics of CFWx were as follows: carbohydrate content 30% including 16% of uronic acid; 51% protein content; monosaccharide composition, Man:Glu:Gal (30:43:27); molecular weight $3-5{\times}10^4$. CFWx was further purified by ion-exchange, gel-permeation, and affinity chromatography and $CFWx-AH-{\alpha}$ fraction was isolated. Fundamental structure of $CFWx-AH-{\alpha}$ was deduced as ${\alpha}-(1{\to}4$)-D-glucan with ${\alpha}-(1{\to}3$)- and/or ${\alpha}-(1{\to}6$)-D-glycosidic side chains based on methylation analysis.

This study was conducted to investigate effects of dietary pigment sources on shell color of juvenile abalone, Haliotis discus hannai. Three replicate groups of the abalone, average weight 173 mg, were fed the diets containing various pigment sources such as Porphyra powder, Spirulina, yeast astaxanthin and paprika extract for 16 weeks. Survival and weight gain were not affected by dietary pigment sources (P>0.05). Shell color of abalone fed diets containing Porphyra powder and Spirulina was approached to yellow-red and orange which are similar to shell color of wild abalone. However, shell color of abalone fed the diets containing yeast astaxanthin and paprika extract were similar to that of control diet showing bright green. Porphyra powder and Spirulina contain not only fat-soluble pigment such as chlorophyll and carotenoids but also water-soluble pigment such as phycoerythrin and phycocyanin. These results would be useful information to change shell color of abalone in aquaculture.
In the present study, we investigated whether Houttuynia cordata Thumb (Saururaceae; HC) extracts have an anti-inflammatory effect in human monocytic THP-1 cells and human eosinophilic EoL-1 cells. The dried and powdered whole plants of HC were extracted with 80% EtOH. The combined extract (HC-1) was concentrated under reduced pressure. The residue was diluted with water, and then successively partitioned with n-hexane, EtOAc, and BuOH to produce the n-hexane (HC-2), EtOAc (HC-3), BuOH (HC-4), and the water-soluble fractions (HC-5), respectively. HC extracts have no cytotoxicity on THP-1 cells and EoL-1 cells at a high concentration of $10\;{\mu}g/ml$ for 24 h, except HC-2 extract ($10\;{\mu}g/ml$). Interleukin-6, Interleukin-8 and Monocyte chemoattractant protein-1 in THP-1 cells were increased after the treatment with the extract from house dust mite or LPS. The increase of cytokine production was strongly suppressed by HC-3 extract, in comparision with other extracts. HC-3 also had inhibitory effect on Interleukin-6 production increased by mite extract and LPS in EoL-1 cells. However, HC-3 extract increased Interleukin-8 production induced by mite extract and LPS in EoL-1 cells. These results suggest that HC extracts may be used as useful agents for treating allergic disorders such as asthma and atopic dermatitis.

This study was carried out to investigate the effect of Cinnamomum camphora on melanogenesis. The MeOH extract of Cinnamomum camphora inhibited mushroom tyrosinase activity in dose-dependent manner. Moreover, it significantly suppressed the melanin production in melan-a cells at the concentration of $100{\mu}/m{\ell}$. The MeOH extract was partitioned with ethyl acetate, n-butanol and water. Among them, the BuOH soluble fraction exhibited significant inhibitory effect on mushroom tyrosinase. In addition, the BuOH soluble fraction reduced the melanin production in melan-a cells. But, the BuOH soluble fraction had less inhibition effects on melan-a cell originated tyrosinase. So, it was performed western blotting for melanogenic proteins (tyrosinase, tyrosinase-related protein (TRP-2)) using melan-a cells. The BuOH soluble fraction inhibited the protein expression of tyrosinase at the concentration of $100{\mu}/m{\ell}$. The results suggested that the BuOH soluble fraction of C. camphora might be a potent inhibitor of melanin biosynthesis in melan-a cells.
BACKGROUND/OBJECTIVES: Several medicinal properties of Smilax china L. have been studied including antioxidant, anti-inflammatory, and anti-cancer effects. However, the antiobesity activity and mechanism by which the water-soluble fraction of this plant mediates its effects are not clear. In the present study, we investigated the lipolytic actions of the water-soluble fraction of Smilax china L. leaf ethanol extract (wsSCLE) in 3T3-L1 adipocytes. MATERIALS/METHODS: The wsSCLE was identified by measuring the total polyphenol and flavonoid content. The wsSCLE was evaluated for its effects on cell viability, lipid accumulation, glycerol, and cyclic adenosine monophosphate (cAMP) contents. In addition, western blot analysis was used to evaluate the effects on protein kinase A (PKA), PKA substrates (PKAs), and hormone-sensitive lipase (HSL). For the lipid accumulation assay, 3T3-L1 adipocytes were treated with different doses of wsSCLE for 9 days starting 2 days post-confluence. In other cell experiments, mature 3T3-L1 adipocytes were treated for 24 h with wsSCLE. RESULTS: Results showed that treatment with wsSCLE at 0.05, 0.1, and 0.25 mg/mL had no effect on cell morphology and viability. Without evidence of toxicity, wsSCLE treatment decreased lipid accumulation compared with the untreated adipocyte controls as shown by the lower absorbance of Oil Red O stain. The wsSCLE significantly induced glycerol release and cAMP production in mature 3T3-L1 cells. Furthermore, protein levels of phosphorylated PKA, PKAs, and HSL significantly increased following wsSCLE treatment. CONCLUSION: These results demonstrate that the potential antiobesity activity of wsSCLE is at least in part due to the stimulation of cAMP-PKA-HSL signaling. In addition, the wsSCLE-stimulated lipolysis induced by the signaling is mediated via activation of the ${\beta}$-adrenergic receptor.
The functional properties of Hericium erinaceus were analyzed. The crude protein content was 8.01%. free sugars were mainly composed of glucose(47.09 mg%) and fructose(34.65 mg%), but sucrose and maltose were not detected. The free amino acids were mainly glutamic acid(1,468.12 mg%), alanine(716.07 mg%) and threonine(643.95 mg%) in Hericium erinaceus. It doesn't difference between water and ethanol extract on soluble solid content. Comparing minerals of extracts from Hericium erinaceus, water extract showed higher contents than ethanol extract except for I This tendency is similar to superoxide scavening activity and electron donating activity. But phenolic compounds, ethanol extract was higher than water extract. In comparison of water and ethanol extract of Hericium erinaceus, as a whole water extract was excellent.
Hot water soluble extract from sporophyll of Undaria pinnatifida was examined for antioxidant activity using l,l-diphenyl-picrylhydrazyl (DPPH) and lipid peroxidation assay. The $IC_{50}$ value (7.6 mg/ml) of hot water extract from sporophyll of U. pinnatifida was more higher than ascorbic acid (0.025 mg/ml) and BHT (0.25 mg/ml). The hot water extract from sporoof U. pinnatifida was stable from pH 2 to pH 7 but decreased at alkali pH. The heat stability of hot water extract from sporophyll of U. pinnatifida was stable from $0^{\circ}C$ to $120^{\circ}C$. An effect on an antioxidant activity of hot water extract from sporophyll of U. pinnatifida was studied with various metal ions and EDTA. Antioxiactvity of hot water extract from sporophyll of U. pinnatifida was inhibited by EDTA but increased by adding $Cu^{2+}$, $Co^{2+}$ and $Mn^{2+}$, respectively.
Objectives : This study was carried out to evaluate whether the water extract from cause the cellular damage in HepG2 cell line. It was reported that Dictamnus dasycarpus Turcz(DDT) intake induce poisoning symptoms in human population. These symptoms was closely related to liver toxicity, however, mechanisms for liver toxicity caused by DDT have not been elucidated exactly. Here, hepatotoxicity caused by DDT was evaluated using HepG2 cell line. Methods : Water extract of DDT was treated into HepG2 cell with various doses such as 0, 0.1, 0.5, 1.0 and $5.0mg/m{\ell}$. In order to cell viability, both MTT and LDH assay were carried out. Also, apoptosis array kit was used to identify whether cell death caused by DDT is due to apoptosis or not. In addition, reactive oxygen species (ROS) was measured after treatment of water extract. Results : We found out significant changes in the apoptosis related factors of hepatocyte. The cell viability of HepG2 treated with DDT water extract was decreased in dose-dependent. Also most of the apoptosis related factors were significantly increased. We found out that Caspase 3, Cytochrome C and ROS had increased in dose-dependent. In addition, other apoptosis related factors Bcl 2 and Bax, which were also constant changes. However, there was no significance. Conclusions : These results suggest that water soluble extract of DDT is expected to have oral toxicity, including hepatocellular damage Therefore, it is suggested that DDT could cause various side effects and toxicity of clinical conditions.
Precipitation was formed during the preparation of decoction from a mixture of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine were identified in coprecipitated product and these components were the active ingredients of two herbal medicine. The coprecipitated product was very slightly soluble in water and sparingly soluble in ethanol. The stoichiometric ratio of baicalin and berberine was found to be 1:1. The lipid-water partition coefficients of coprecipitated product were increased more than baicalin and berberine in chloroform, but were decreased in other organic solvents. The content of baicalin and berberine in coprecipitated product, determined by HPLC, were 23.08% and 26.75%, but the content of active ingredients in supernatant were 0.66% and 0.26%, respectively. The dissolution profile of baicalin of coprecipitated product was increased more than extract of Scutellariae Radix in artificial gastric juice, but was decreased in artificial intestinal juice. The dissolution rate of berberine of coprecipitated product was lower than extract of Coptidis Rhizoma in artificial gastric juice and intestinal juice commonly.
The herb of Geranium sibirium L, a drug knwon as "Chui Sonni Poul" distributed widely, has been used as a folk-medicine for the treatment of diarrhea. The dried entire herb is boiled with methanol and methanol is distilled of from the filtered methanol extract under reduced pressure. Then the extract is boiled with water and filtered off. From the filtrate, the following substances are isolated and identified by treating with organic solvents as ether, ethyl acetate, and etc.: 1. Gallic acid: a colorless needle crystal which is soluble in alcohol and water. mp.235.deg. C, positive (dark blue) against the ferric chloride reagent. 0.76 percent of gallic acid is yielded from the herb. 2. Quercetin: a light yellow crystal. mp.194.deg. C. negative against the ferric chloride reagent. 3. Ellagic acid: a light yellow crystal which is insoluble in ehter and acetone and slightly soluble in alcohol. Positive (blue) against the ferric chloride reagent. The crystal obtained by recrystalisation from pyridine, does not melt by 360.deg. C. Represent a yield of 0.03 percent from the herb. 4. Crude tannin: a approximately 7.6 percent of crude tannin is yielded by treating with ethyl acetate. gallic acid and querceetin are yielded by hydrosis with dliute sulfuric acid.
