Development of Reverse Transcription Semi-nested PCR Primer Pairs for the Specific and Highly Sensitive Detection of Human Aichivirus A1

  • Human Aichivirus A1 (HuAiV-A1) is a waterborne human pathogenic virus classified as Picornaviridae and Kobuvirus. In this study, we developed a method that can detect about 35 minutes faster with the same detection sensitivity level than the previously reported HuAiV-A1 diagnostic RT-PCR primer. The RT-PCR primer sets developed in this study are capable of detecting HuAiV-A1 at a level of about 100 ag and formed 563 bp amplification product. In addition, the RT-nested PCR method was able to amplify 410 bp using the RT-PCR product as a template. The detection sensitivity of our method was 10 times higher than the method with the highest detection sensitivity to date. Therefore, the detection method of HuAiV-A1 developed in this study is expected to be used in the water environment in which a small amount of virus exists. Also, this detection method is expected to be used as HuAiV-A1 diagnostic technology in both clinical and non-clinical field.

  • This study was designed to evaluate the anti cariogenic effect of F in primers, bonding agents, composite resins or glass ionomer cements in enamel. Twenty-five extracted teeth were selected and a cavity was prepared on either the buccal or the lingual surface of each tooth. After pumicing and etching, the samples were divided into 5 groups. In group A, the samples were primed, bonded and filled with ART bond and Brilliant Enamel (Coltene, Switzerland). Group B composed of Optibond and Herculited XRV (Kerr, USA), group C composed of Syntac and Tetric(Vivadent, Lichtenstein), and group D composed of Scotch-bond Multipurpose and Z 100 (3M, YSA). In group E, the samples were filled with glass ionomer cement (Fuji II LC, Japan), All surfaces except the 2mm beyond the cavosurface margin of the sample were protected, and samples were then put into an acid buffer for 3 days to develop the initial caries. The samples were then sectioned through the filling body into thin wafers and then examined with a polarizing microscope under water imbibition. The fluoride in primer, bonding agent, or composite filling material did not prevent the initial caries in the enamel area adjacent to the filling body whereas the fluoride in the glass ionomer did prevent the initial stage caries.

Characterization of Modified Chloroprene Rubber by Nanosilica as a Primer (Modified Chloroprene Rubber를 이용한 Primer 제조 및 특성평가)

  • Water In this study, CR/silica nanocomposites were produced by dispersing nanosilica on chloroprene rubber (CR) to apply toluene-free primers for mobile devices. The properties of the modified chloroprene rubber using nanosilica was evaluated through FT-IR, SEM, EDS, Contact angle. The SEM images indicated that P-4 (4 phr) was the most homogenously dispersed. Pencil hardness measurements and Contact angle indicated that the hardness of the CR/silica nanocomposite and the hydrophobicity increased with increase in the silica content. The peel strength of P-4 (4phr) was the highest and the initial peel strength of P-4 sample (2.9 kgf/inch) was 50% higher than that of the P-0 sample.

Diagnostic Method for the Detection of JC Polyomavirus Using Loop-mediated Isothermal Amplification (등온증폭법을 이용한 고감도 JC polyomaviruses 진단법 개발)

  • JC polyomavirus (JCPyV) is a human pathogenic virus belonging to the family Polyomaviridae, a viral group containing dsDNA nucleic acid. A recent recommendation is to apply the presence of JCPyV as a fecal indicator for water contamination in environments like sewage, and techniques to monitor JCPyV in water are being proposed. To date, the conventional PCR system has been applied as a diagnostic method for detecting JCPyV. There is a need for a more rapid and sensitive JCPyV diagnostic detection method in clinical and environmental samples. In this study, we developed a loop-mediated isothermal amplification (LAMP) primer set for the detection of JCPyV. Our results indicate that the LAMP method using a specific primer set shows about 10-fold higher detection sensitivity than the conventional PCR system. The effectiveness of the LAMP method developed in this study has been validated by PCR product digestion using the HaeIII restriction enzyme. We, therefore, propose that the LAMP method using a specific primer set can be applied as a rapid and sensitive detection method for monitoring JCPyV in clinical and environmental samples.

  • The purpose of this study is to evaluate the adhesive properties of polyurethane mixed aqueous dispersions by omitting the primer, dealing with the preparation of skins for synthetic leather with excellent adhesion by omitting the pre-treatment process. The two-component mixed polyurethane water dispersion was prepared by synthesizing an ester-based polyurethane resin (PU-T) and a carbonate-based polyurethane resin (PU-C) to obtain the final resin. As a result of measuring the peel strength of the adhesive specimens omitting the pre-treatment agent, it was confirmed that the state adhesive strength (ethylene vinyl acetate (middle): $4.2kg_f/cm$ and rubber (outsole): $4.4kg_f/cm$) there was. This makes it possible to omit the pre-treatment process which has been indispensably used in the shoe manufacturing process, thereby reducing the process time and reducing the amount of volatile organic compounds (VOCs) generated in the pre-treatment product, resulting in environmentally advantageous.

  • Pre-construction primer (PCP), or shopprimer, have been applied to steel plates to control temporary corrosion during ship fabrication. For surface preparation at ship block stage, in common shipyard practices, welding beads, burnt and rusted areas shall be blasted or power tool cleaned and the contamination such as zinc salt shall be removed with blasting or power tool. Whereas, the sound film of PCP needs not to be removed or roughened as the paint having good compatibility with PCP is used for the first coat. In many cases, however, full blasting or sweep blasting on the sound PCP treated block assemblies was requested. There still has been argument about the legitimacy of this practice, thus, it is critical to evaluate the quality of the coating system applied on the sound PCP retained condition, comparing with the one applied on the full blasted or sweep blasted condition. In this study, two different epoxy systems for water ballast tank were applied on the surfaces with sound PCP condition, full blasted condition, and sweep blasted condition. Coating performances such as durability, anti-corrosion, cathodic disbondment resistance were evaluated. The test results clearly indicated that the sound film of PCP needed not to be removed or roughened as the paint having good compatibility with PCP based on inorganic zinc silicate.

  • Cast-in-place foamed concrete is a high porous material placed as base the layer of mortar screeds in the panel heating system, so the quality of mortar is affected by it. Therefore, this study is aims to investigate how the absorption characteristics of foamed concrete influences on mortar screeds according to the foamed ratio-62%, 67% and 72%- and the surface treatments of foamed concrete-water and acrylic emulsion primer spray, etc. The result of this study shows that water-spray in the surface of foamed concrete has a good effects to reduce mortar cracking.

  • Human Aichivirus (Aichivirus A; AiV-A) is a positive-sense single-strand RNA non-enveloped virus that has been detected worldwide in various water environments including sewage, river, surface, and ground over the past decade. To develop a method with excellent sensitivity and specificity for AiV-A diagnosis from water environments such as groundwater, a combination capable of reverse transcription (RT)-nested polymerase chain reaction (PCR) was developed based on existing reported and newly designed primers. A selective method was applied to evaluate domestic drinking groundwater samples. Thus, a procedure was devised to select and subsequently identify RT-nested PCR primer sets that can successfully detect and identify AiV-A from groundwater samples. The findings will contribute to developing a better monitoring system to detect AiV-A contamination in water environments such as groundwater.
