Objectives : In order to investigate the possibility of Sorbus commixta Heal. stem (SC) as a natural material, antioxidant activities of the hot water and ethanol extracts were examined. Methods : The samples of SC were pulverized, and fractions were extracted repeatedly three times from hot water and 70% ethanol at room temperature for 2 hours. The antioxidant activities were analyzed from total polyphenol, flavonoid contents, DPPH, ABTS, hydroxyl radical, $Fe2^+$ chelating, and nitrite scavenging activity. Results : Total polyphenol contents were significantly higher in ethanol extract group ($504.39{\mu}g/m{\ell}$) than in hot water extract group ($364.64{\mu}g/m{\ell}$). Total flavonoid contents were also significantly higher in ethanol extract group ($160.09{\mu}g/m{\ell}$) than in hot water extract group ($124.59{\mu}g/m{\ell}$). DPPH, ABTS, $Fe2^+$ chelating were slightly higher in ethanol extract gorups than in hot water extract groups, and increased in a dose-dependent manner. The hydroxyl radical scavenging activity (18.42~23.61%) of ethanol extract groups were shown to be approximately twice higher than that (7.63~10.37%) of hot water extract groups at $12.5{\sim}50{\mu}g/m{\ell}$ concentration. Nitrite scavenging activities of both ethanol and hot-water extract groups were shown to be higher in a dose dependent manner at the concentration of $12.5{\sim}50{\mu}g/m{\ell}$ at pH 3.0 than at pH 1.2, and ethanol extract groups (86.55~96.64%) had higher activity than the hot water extract groups (42.59~92.63%), which was higher than that of the control group antioxidant BHT (72.96~80.11%). Conclusions : The extracts of SC displayed antioxidant activities which suggested a natural material can be developed to functional material.
The thinning Green ball apple was extracted using water and ethanol and a phenolic concentration of thinning Green ball apple was $50-200{\mu}g/mL$. The water and ethanol extracts of thinning Green ball apple showed 94.69% and 92.24% 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity and 100.30% and 99.16% 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity at phenolic concentration of $200{\mu}g/mL$, respectively. The water and ethanol extracts of thinning Green ball apple showed antioxidant protection factor of 1.76 antioxidant protection factor and 1.76 antioxidant protection factor, respectively. The water and ethanol extracts showed 101.46% and 99.64% anti-oxidative effect on thiobarbituric acid reactive substances at phenolic concentration of $200{\mu}g/mL$. Hence, the water and ethanol extracts of thinning Green ball apple can be considered a potential anti-oxidant. The water and ethanol extracts showed 33.28% and 32.14% hyaluronidase inhibition, respectively, at phenolic concentration of $150{\mu}g/mL$. The water and ethanol extracts showed 47.33% and 40.92% elastase inhibition and 46.19% and 65.58% collagenase inhibition at phenolic concentration of $200{\mu}g/mL$, respectively. About these experiments, thinning Green ball apple was found to exhibit anti-oxidation activity as well as hyaluronidase, elastase and collagenase inhibitory activities. Therefore, thinning Green ball apple can be considered a potential source for functional food.
국내산 재배 생약류 28종 중 열수 추출물에서 항산화력을 나타낸 것으로 조사된 작약, 목단, 황금, 두충, 시호 그리고 산수유의 열수 추출물 6종을 냉동건조한 후 70% ethanol로 용해하여 ethanol 가용성 획분(ESF)과 ethanol 불용성 획득(EIF)으로 분획하였다. 이 분획물들을 60% linoleic acid에 3,000ppm씩 가한 후 35$^{\circ}C$의 항온기에서 저장하면서 BHA 첨가구와의 산화 안정성을 비교하였다. 이 결과 작약가 항금의 ESF는 유도기간이 각각 12일과 9일인데 반해 BHA 첨가구는 9일로 나타나 산화 안정성이 더 높거나 같은 것으로 나타났다. 한편 6종의 생약류의 ESF는 EIF보다 linoleic acid에 대한 항산화 효과가 모두 높은 것으로 나타났다.
Physiological activities of Korean-grown ginger (KG) and Chinese-grown ginger (CG) extracts were examined. Ginger was extracted with water, and 50 and 100% ethanol, and then nitrite-scavenging activity (NSA), superoxide dismutase (SOD)-like activity, and electron-donating ability (EDA) of extracts were investigated. NSA at pH 1.2 was the most effective in 50% ethanol extracts of both origins. SOD-like activities of water and 50% ethanol extracts of both samples were 8.66-35.95% lower than those of 1 and 0.1% L-ascorbate solutions. SOD-like activity of KG extracts was higher than that of CG extracts, and water extracts of samples were the highest. EDA of KG extract was higher (22.23-86.95%) than that of CG extract, while both sample extracts showed lower EDA than those of 1 and 0.1% L-ascorbate solutions.
Red ginseng extracts were prepared with various concentrations of ethanol. Extract yields were examined and saponins in the extracts were identified and determined by TLC and HPLC, respectively. Yields of the extracts, $19.7{\sim]50.3%$, were the highest in water extract and showed significant decrease with the increase of ethanol concentration used for extraction. Contrary to the extract yields, saponin yields from red ginseng were conspicuously increased with the increase of ethanol concentration and were $3.47{\sim}5.13%$ of crude saponins and $1.28{\sim}1.93%$ of six major ginsenosides. Saponin contents in the red ginseng extracts were $6.9{\sim}24.2%$, of crude saponin and $2.57{\sim}9.22%$, of six major ginsenosides.
Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+ radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.
Objectives : This study was performed to investigate the antioxidant activity of water and ethanol extracts from Sasa borealis leaves, stems and roots. Methods : Sasa borealis leaves, stems and roots extract were prepared using water and ethanol. The total polyphenol and total flavonoid contents were analyzed. 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity, SOD like activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), hydroxyl radical scavenging activity, and Nitrite scavenging activity assays were carried out to determine the antioxidant activities. Results : The antioxidant activities of the Sasa borealis appeared higher in ethanol extract than water extracts. Total polyphenol and total flavonoids contents in ethanol extracts of leaves were $24.6mg/m{\ell}$ and $14.3mg/m{\ell}$, respectively, which were much higher than those of any other parts. SOD like activity was 70% ethanol extract of the leaves was highest with 15.68%. Electron donating ability was 70% ethanol extract of the leaves had the highest 59.07%. It exhibited high electron donating ability than BHT(45.68%). Nitrite scavenging activity of 70% ethanol extract was higher than the water extract at pH 2.5 and pH 4.2. Nitrite scavenging activity was 70% ethanol extract of the leaves was the highest 75.2%. Hydroxyl radical scavenging activity was 70% ethanol extract of the leaves was highest with 16.16%, showed very low activity than BHT(61.56%). Conclusions : These results suggest that 70% ethanol extracts from Sasa borealis leaves, exhibited higher antioxidant activities than those of root and stem, and can be potentially used as proper natural antioxidants.
The effects of the water extracts from thyme(TM) and tarragon(TG) on shelf-life and quality of kimchi were investigated by measuring the changes in pH, acidity, number of total microorganisms, number of Lactobacillii and Leuconostoc during fermentation at 1$0^{\circ}C$, and were tested for antimicrobial activities against Lactobacillus plantarum and Leuconostoc mesenteroides. TM and TG were extracted with water, ethyl ether, ethyl acetate and ethanol. Water, ethyl ether, ethyl acetate and ethanol extracts of TM showed antimicrobial activities against Lactobacillus plantarum and did not observed against Leuconostoc mesenteroides. On the other hand, water, ethyl acetate and ethanol extracts of TG showed antimicrobial activities against Leuconostoc mesenteroides and did not observed against Lactobacillus plantarum. The decrease of pH and the increase of acidity showed lower in kimchi prepared by adding water extracts from TM than in products from TG. The number of total microorganisms were also detected less in the kimchi prepared by adding water extracts from TM. And, the properties of barkless of kimchi measured instrumentally were higher for kimchi prepared by adding water extracts from TM, also maintaining good crispness. The optimal addition amounts of both TM and TG for good overall and spicy taste of kimchi were 0.03%. The results suggested the possible use of the extracts of TM and TG can be successfully used for the quality and extension of shelf-life of kimchi.
The present study was to investigate the antioxidant activity in ethanol and water extracts of root and stem of Sargassum coreanum. Antioxidant activities were evaluated by total polyphenol contents, DPPH radical scavenging activity, chelating effect, reducing power, and rancimat method. Total polyphenol contents of ethanol and water extracts were 32.79 mg/g and 15.55 mg/g, respectively. Ethanol extract showed higher DPPH radical scavenging activity than water extract and similar activity to BHT. Reducing power of extracts was increased in a concentration-dependent manner and ethanol extract had more reducing power than water extract. Ethanol and water extracts have little chelating effect at all concentrations. Antioxidant index (AI) of ethanol extract measured by Rancimat was higher than that of water extract, but their AI was lower than that of BHT. These results indicate that ethanol extract of S. coreanum root and stem has more potent antioxidant activity than water extract through DPPH radical scavenging and reducing power, and could potentially be used as a good source of natural antioxidants.
본 연구는 다양한 생약재를 사용하여 생약재 복합물을 구성하고, 복합물의 이화학적 특성과 항산화 활성에 대하여 조사하였다. 생약재 복합물의 추출 수율은 물 추출물이 35.00%, 에탄올 추출물 25.33%이었으며, 분획물 중에서는 ethyl acetate층의 수율이 물 및 80% 에탄올 추출물에서 각각 0.64% 및 3.76%였다. 생약재 복합물의 물 및 80% 에탄올 추출물의 총플라보노이드 함량은 각각 2.34% 및 2.42%이었고 총페놀성 화합물 함량은 5.04% 및 4.56%이었으며, 용매별 분획물 중 ethyl acetate층의 총플라보노이드 함량이 가장 높았고 총페놀성 화합물 함량 또한 ethyl acetate층에서 가장 높았다. 추출물에 가장 많이 함유된 성분은 salicylic acid와 ${\rho}$-coumaric acid이었다. 물 추출물과 80% 에탄올 추출물의 전자공여능은 각각 43.32% 및 41.32%이었으며, 각각의 용매 분획물의 전자공여능은 총페놀성 화합물 함량의 패턴과 유사하였다. 아질산염 소거능은 물 및 80% 에탄올 추출물에서 9.68% 및 8.94%였으며, 용매별 분획물의 아질산염 소거능은 전자공여능과 유사하였다.
