Enrichment of Vitamins $D_3$, K and Iron in Eggs of Laying Hens

  • An experiment was conducted to produce eggs enriched with vitamins $D_3$, K and iron in eggs. Six hundred 97-wk-old ISA Brown force molted hens were allocated to completely randomized block arrangement of six dietary treatments: T1; control (C), T2; C+4,000 IU vitamin $D_3$+2.5 mg vitamin K+100 ppm Fe, T3; C+8,000 IU vitamin $D_3$+5.0 mg vitamin K+100 ppm Fe, T4; C+12,000 IU vitamin $D_3$+7.5 mg vitamin K+100 ppm Fe, T5; C+16,000 IU vitamin $D_3$+10.0 mg vitamin K+100 ppm Fe, T6; C+20,000 IU vitamin $D_3$+12.5 mg vitamin K+100 ppm Fe. Fe was supplemented with Fe-methionine. Each treatment consisted of five replicates of ten cages with two birds per cage. Egg production and egg weight were highest in T2 and incidence of soft and broken egg was highest in T6. Haugh unit was not different among treatments although it tended to be increased as dietary vitamins $D_3$ and K increased. Eggshell strength was not different among treatment. Concentrations of vitamin $D_3$ and K in egg yolk increased and plateaued approximately 20 days after feeding supplemented diets. The level of these vitamins peaked at 12,000 IU/kg vitamin $D_3$ and 7.5 mg/kg vitamin K supplementation and then decreased at the higher than these supplementation levels. The peak concentrations of vitamin $D_3$ and vitamin K were 4.6 times and 4.8 times greater than the control, respectively. Supplementary Fe also increased Fe content in egg yolk. It is concluded that vitamin $D_3$ and K in eggs can be effectively enriched by proper supplementation time and level of these vitamins.

Protective Effects of Vitamin C against Genomic DNA Damage Caused by Genotoxicants (유전독성물질의 유전체 손상 작용에 대한 Vitamin C의 방호효과)

  • Although it is popularly believed that vitamin C protects cells from various genotoxicants, the degrees and mechanisms of itsprotective actions are not fully understood. In this study, vitamin C's protective effects against various genotoxicants were quantified, together with subsequent analyses on the mechanisms of these protective effects. Comet assay was employed to measure the degree of DNA damage in Chinese hamster ovary cells (CHO-K1) exposed to five genotoxicants, $H_2O_2$, $HgCl_2$, N-methyl-N-nitro-N-nitrosoguanidine (MNNG), 4-nitroquinoline-1-oxide (4NQO), and UV-irradiation. In cases cells were treated with $H_2O_2$, $HgCl_2$, and 4NQO together with vitamin C, the damage to DNA decreased to the level of the control group. In cases of UV-irradiation, the protective effect of vitamin C appeared, but did not reach the control levels. Interestingly, vitamin C did not have protective effects against the genotoxicity of MNNG. The degrees of DNA damage of cells treated with vitamin C prior to exposure togenotoxicants were 28~49% lower than those of cells treated with vitamin C after being exposed to genotoxicants. In conclusion, vitamin C had strong antioxidanteffects against genotoxicants by being a primary antioxidant blocking genotoxicity reaching the cells, rather than being a secondary antioxidant acting on post-exposure DNA repair processes. However, vitamin C's protective effects appearto be limited, as there are genotoxicants, such as MNNG, whosegenotoxicityis not affected by vitamin C. Therefore, the results of this study warrant furtherstudies on toxic mechanisms of genotoxicants and their interactions with protective mechanisms of vitamin C.

Effect of Vitamin C Supplementation on the Performance and Eggshell Quality of Layers (비타민 C 첨가가-산란계의 생산성과 난각품질에 미치는 영향)

  • An experiment was conducted to determine the effects of vitamin C supplementation on the performance and egg quality of layers. A total of 800 ISA Brown layers of 71 weeks old were assigned to one of the following diets containing 0, 3, 10, 50 or 100 ppm LG-vitamin C, 100ppm free ascorbic acid, 100 ppm Rovimix stay C-35 or 50 ppm LG-vitamin C supplemented with 100 ppm Zn methionine chelate. The feeding trial was conducted for flour weeks during the period of May to June when the average ambient temperature was $23.8^{\circ}C$ (5 :00 PM). Each treatment was replicated five times with 20 birds housed in 2 birds cage units. Vitamin C supplementation did not improve the overall performances (egg Production and egg Quality). Birds fed 100 ppm of LG vitamin C had the lowest egg Production while birds fed 100 ppm of Rovimix-35 had the lowest egg weight among birds fed different sources of vitamin C. Although supplementation of vitamin C over 50 ppm increased soft and broken egg Production, supplementation of 10 ppm LG vitamin C showed lowest soft and broken egg Production among all treatments. E99she11 thickness was highest in the treatment of 50 ppm LG vitamin C. In conclusion, supplementation of vitamin C at the level of 3 ~ 100 ppm did not significantly influence the laying performance and eggshell quality of laying hens under normal ambient(20 ~$26^{\circ}C$ ) condition.

A Study on the Variation of Vitamin C Content in Cooked Spinach by the Cookery Method. (시금치나물의 조리방법에 따른 Vitamin C 함량변화에 관한 연구)

  • The effect of the temperature and the time of scalding, and the addition of various seasoning on the remaining % of total Vitamin C contained in Spinach (Spinacia oleracea) was studied and discussed by means of chemical kinetics. The quantitative measurements of total Vitamin C were made by 2,4-dinitrophenyl hydrazine (DNPH) method with Clinical Electrophotometer (Fisher). The sample spinach contains 39.88mg. % of total vitamin C on the average, and the ratio of oxidized and reduced forms of Vitamin C is 1 : 4.7. When the sample spinach was scalded, the kinetics of the decrease of the remaining total vitamin C. % was observed to follow the first order reactions regardless of the temperatures applied (i,e. 70$^{o}$ , 80$^{o}$ , 90$^{o}$ and 100$^{o}$ ) with half-life range of 2.10 - 1.47 minutes. In the case of the addition of various seasonings, the addition of various seasonings, the kinetics remaining Vitamin C % showed to be the zero order reactions regardless of the kinds of seasonings and storage temperatures. With the addition of seasonings, the stability of Vitamin C was found to be increased in the order table salt+sesame oil+vinegar+soybean sauce, without seasoning, table salt+sesame oil, table salt+sesame oil+soybean sauce.

Analysis of Factors to Influence Requirements of Vitamins E and Vitamin C in Young and Healthy Men and Women (건강하고 젊은 남녀의 비타민 E와 비타민 C 요구량에 영향을 미치는 요인 분석)

  • Antioxidants such as vitamin C and E may play a preventive role in the development of cancer and coronary heart disease. The status of vitamins C and E may be affected by lifestyle habits such as smoking , drinking, and exercise. These habits can modify the dietary requirements of vitamin C and vitamin E. the purpose of this study was to determine whether Korean young healthy men and women consume vitamins C and E in sufficient quantities relative to their lifestyle habits. Among the participants in this study, 52% of the men and none of the women were smokers. ; 84% of all subjects drank alcohol more than once a week ; and the men exercised more often than the women. The concentrations of serum total , HDL-, and LDL- cholesterol were higher in the women than in the men, but the serum triglyceride concentrations were higher in the men than in the women. The men consumed less satuated fat than the women (p<0.05) . The daily intakes of vitamin C for the men and the women were 47.1mg and 65.6mg, respectively. On the other hand , the daily vitamin E intake was higher in the men (11.8mg) than women(6.9mg). The serum $\alpha$-tocopherol concentrations of all subjects were in a normal range, and in no subjects were they below the minimum value of ranges. However, about 19% of male subjects and 10% of female subjects showed deficient status, although the mean serum vitamin C levels were normal . Lifestyle habits fo the sort mentioned above have little influence on the serum vitamin C and $\alpha$-tocopherol concentrations. The serum $\alpha$-tocopherol concentration had a positive correlation with total fat and alcohol consumption. The serum vitamin C concentration was positively associated with regular exercise, but it was negatively correlated with the number of cigarettes smoked. Meanwhile, the serum lipid persoxide concentration , the indirect index of oxidative stress, was influenced by certain variable such as body mass index , the number of cigarettes smoked , alcohol consumption, energy expenditure, vitamin C intake, and serum ${\gamma}$-tocopherol concentration. Serum lipid peroxide concentration was positively associated with body mass index, the number of cigarette smoked , serum triglyceride , and HDL-cholesterol concentration. In conclusion , the vitamin E requirements of the subjects were met by the Recommended Daily Allowance (RDA) regardless of lifestyle habits. However, serum vitamin C concentrations showed individual variation and was below the normal ranges. Smoking and exercise influenced serum vitamin C concentration. Therefore, a reevaluation of the requirements of vitamin C relative to lifestyle habits is necessary.

Effects of Antioxidant Vitamins Supplementation on Antioxidative Status and Plasma Lipid Profile in Korean NIDDM Patients (항산화비타민 보충이 인슐린비의 의존형 당뇨병 환자에서 항산화상태에 혈액지방성분에 미치는 영향)

  • We investigated the effects of antioxidant vitamins supplementation on antioxidative status and plasma lipid profile in female Korean non-insulin diabetes mellitus(NIDDM) patients. Forty-five patients were groups by types of vitamin to take into three groups-Vitam in E group(400IU/day, n=15), Vitamin C group (1,000mg/day, n=15) and Vitamin E plus C group (400 IU plus 1,000mg/day). Supplementation period was 4 weeks. After vitamins supplementation, plasma vitamin E concentration significantly increased in vitamin E and vitamin E + C group, but plasma retinol concentration were not affected by vitamin E or vitamin C supplementation. And plasma levels of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS), indicator of lipid peroxidation and increased susceptibility of LDL towards lipid peroxidation, were significantly decreased in all three groups after vitamins supplementation. Also catalase activities in erythrocytes were significantly decreased after antioxidant vitamin supplementation in all subjects. And after vitamins supplementation, post prandial 2 hour glucose and total cholesterol levels was decreased in all subjects. And after vitamins supplementation, post prandial 2 hour glucose and total cholesterol levels was decreased in all patients, especially there was a significant difference in vitamin C, Vitamin E + C group. In this study, antioxidant vitamins supplementation might have a protective function against the free radical-mediated lipid peroxidation and decrease the plasma total cholesterol levels in Korean female NIDDM patients.

  • Modified radish Kimchi and radish-root juice were fermented under $N_2$ atmosphere at $22{\sim}23^{\circ}C$ and $37^{\circ}C$, respectively and their changes of vitamin C content. were determined by dinitrophenylhydrazine and indophenol methods. In the earlier period of the fermentation, vitamin C content decreased temperarily and then began to increase. When the flavor of Kimchi was most acceptable, vitamin C showed the maximum value which eras equivalent to or more than the initial, after which vitamin C content decreased gradually. The increase of vitamin C content seemed to come from the biosynthesis of the vitamin owing to the enzymatic action occurred in radish-root. The addition of galacturonic acid to Kimchi and to radish-root juice elevated the vitamin C content, which suggested galacturonic acid was a good substrate for the synthesis of vitamin C. The optimum pH of the Kimchi fermentation for the better vitamin C content ranged 4.0 to 4.5.

Antioxidant activity of vitamin C in iron-overloaded swine plasma (철분 투여한 돼지에서 비타민 C의 항산화 작용)

  • Iron deficient anemia in piglets could be overcome by supplementary iron. Overloaded iron induced peroxidation of cell membrane and increased malonaldehyde (MDA). Antioxidant activity of vitamin C has been studied in iron-overloaded swine plasma. Erythrocyte fragility, MDA, glutathione, vitamin A, and vitamin E were measured in swine plasma with or without iron (0~1mg/dl) and vitamin C (0~10mg/dl). Erythrocyte fragility increased from 8% to 45% in iron group and reduced from 57% to 43% in vitamin C group with dose dependant response. MDA was $0.94{\pm}0.05$ and $1.86{\pm}0.10$ nmol/ml in piglet and pig, respectively, and significantly high in pig (p<0.05). Iron increased MDA from $1.86{\pm}0.10$ to $9.46{\pm}0.04$ nmol/ml in pig, but not in piglet (p<0.05). Vitamin C reduced MDA from $9.46{\pm}0.04$ to $4.80{\pm}0.10$ nmol/ml in pig. Iron increased glutathione from $90.12{\pm}0.10$ to $108.52{\pm}5.29$ nmol/dl in pig, and vitamin C reduced glutathione from $108.52{\pm}5.29$ to $93.52{\pm}2.44$ nmol/dl (p<0.05). Vitamin A and E were $24.86{\pm}2.70$ to $138.29{\pm}6.70{\mu}g/dl$, respectively in iron group, and $35.76{\pm}0.60$ to $177.21{\pm}2.95{\mu}g/dl$, respectively in supplementary vitamin C group (p<0.05). These data indicated an antioxidant activity of vitamin C in iron-overloaded swine plasma.

Associations of dietary vitamin A and C intake with asthma, allergic rhinitis, and allergic respiratory diseases

  • BACKGROUND/OBJECTIVES: Asthma and allergic rhinitis (AR) are closely related and considered as allergic respiratory diseases (ARD), and their prevalence has recently increased. Data on the association of dietary antioxidant vitamin intake with asthma and AR in adults are limited. The present study aimed to investigate the associations of vitamin A and C intake with asthma, AR, and all cases of both diseases in young adults who participated in a cross-sectional national survey, with the use of high-sensitivity C-reactive protein (hs-CRP) level as an effect modifier. SUBJECTS/METHODS: This study included 6,293 male and female adults aged 20-49 years from the Korea National Health and Nutrition Examination Survey (KNHANES) conducted between 2016 and 2018. The questionnaire-based reports on asthma and AR diagnosis were used to determine outcome variables. Further, 24-h recall data on dietary vitamin A and C, carotene, and retinol intake were acquired. Logistic regression analysis was performed to calculate odds ratios (ORs) and 95% confidence interval (CI). RESULTS: Dietary vitamin C intake was inversely associated with asthma prevalence among participants with hs-CRP levels (≥ 1 mg/L); the OR of asthma prevalence was 0.27 (95% CI, 0.08-0.84) for participants with vitamin C consumption ≥ 75 mg/day compared with those consuming < 20 mg/day. Similar association analyses limiting to non-users of dietary supplements were performed to rule out the potential effects of supplement intake on the outcomes; results showed a stronger association. However, the association between vitamin C and asthma was not significant in participants with hs-CRP levels < 1 mg/L; the OR of asthma was 1.44 (95% CI, 0.66-3.16) for participants with vitamin C consumption ≥ 75 mg/day compared with those consuming < 20 mg/day. Vitamin C intake was not associated with AR. Moreover, there was no association between vitamin A intake and neither asthma nor AR. CONCLUSIONS: These findings suggest that higher vitamin C intake may play a potential role in reducing asthma prevalence. Nevertheless, further studies should be conducted to evaluate whether this association is causal.

Lipid Peroxidation and Antioxidant Vitamin Status in Oral Cavity and Oropharyngeal Cancer Patients

  • Background: This study was conducted to determine levels of lipid peroxidation and antioxidant vitamin status in patients with oral cavity and oropharyngeal cancer. Methods: The study group consisted of a total number of 80 subjects between the age 40-68 years, 40 with clinically and histopathologically proved cases of oral cavity and oropharyngeal cancer and 40 normal healthy, age and sex matched volunteers as controls. Levels of lipid peroxidation products as malondialdehyde (MDA) and antioxidant vitamins as vitamin A and vitamin C were estimated and compared between the two groups. Results: There was a statistical significant difference in the mean MDA, plasma vitamin A and vitamin C in the oral and oropharyngeal cancer patients compared with the healthy controls (p<0.0001). Conclusions: Lipid peroxidation (MDA) is higher and plasma antioxidant vitamins like vitamin A and vitamin C were lower in oral cavity and oropharyngeal cancer patients than healthy controls.
