임신부의 비타민 $B_{6}$ 섭취와 모체와 제대혈 및 태반 조직의 비타민 $B_{6}$농도 (Maternal Vitamin $B_{6}$ Intake and Vitamin $B_{6}$ Level in Maternal, Umbilical Cord Plasma and Placenta)

  • 본 연구는 정상만기분만 임신부 (30명)와 그들의 신생아를 대상으로 임신부와 비타민$B_{6}$ 섭취양상과 모체와 신생아 제대혈장 및 태반조직내의 비타민$B_{6}$ 농도를 측정하고 이들 농도와 임신결과와의 상관성을 조사하며, 모체에서 태아로 비타민$B_{6}$가 이동될 때, 태반의 역할을 규명하고자 하였다. 임신부의 평균 연령과 임신 전 체중 및 신장은 각각 25.9세, 53.8kg 및 161 cm 였으며, 임신 전 체중 및 신장은 각각 25.9세, 53.8 kg 및 161cm 였으며, 임신 전 BMI는 20.7kg/$m^2$로 정상 범위였다. 수축기와 이완기 평균 혈압은 122.5 mmHg 와 75.2mmHg의 결과로 안정적인 수준이었으며 헤모글로빈 농도와 헤마토크리트치는 12.1g/dl와 36.1%로 양호하였다. 연구대상자 중 93.3%에 해당하는 임신부가 영양보충제를 복용했으며 이 중 85.7%가 엽산과 철분이 포함된 빈혈 치료제를 이용한 것으로 조사되었다. 영양보충제의 평균 복용기간은 16.3주였다. 임신부가 1일 평균 열량 섭취량은 2189.5kcal로 권장량의 93.2%였으며, 단백질은 79.3g(113.3%)으로 권장량을 상회하였다. 비타민 B$_{6}$의 평균 섭취량은 권장량은 91.4%인 1.7mg이었고, 비타민 $B_{6}$의 평균 섭취량은 권장량은 91.4%인 1.7mg이었고, 비타민 $B_{6}$ 영양밀도와 단백질 g 당 섭취량은 각각 0.8와 0.02mg으로 양호한 결과였다. 그러나, 비타민$B_{6}$의 주된 공급식품이 곡류 및 전분류 (50%)와 야채 및 과일류 (33%)로 식물성 식품이 대부분이었다. 임신 말 모체 혈장과 신생아 제대혈장의 PLP 농도는 각각 16.7$\pm$4.1 nmol/l와 61.3$\pm$19.8 nmol/l로 제대혈장의 PLP 농도가 유의적으로 높았으며, 태반조직의 PLP 농도는 898.6$\pm$159.2ng/g으로 혈장의 PLP 농도에 비해 매우 높은 값을 보였다. 모체와 태반조직 및 신생아 제대혈장간 PLP 농도의 상관성을 조사한 결과, 모체 혈장과 태반 조직간 (p<0.0001), 태반 조직과 신생아 제대혈장간 (p<0.05), 모체 혈 장과 제대혈장간(p<0.05) 유의적인 양의 상관관계가 있었다.

  • P. shermanii IFO 1239을 이용 vitamin $B_{12}$를 생산할 때 산소의 영향은 매우 큼 것으로 나타났는데 배양 전반부는 혐기적 조건이, 후반부는 호기적 조건이 vitamin $B_{12}$ 생성증진에 효과적임이 밝혀졌다. 본 연구를 통하여 배양 48시간 동안은 복합배지상에서 혐기적으로 배양하다가 48시간 이후부터는 합성배지로 바꾸고 호기적 조건으로 배양해주는 방법이 vitamin $B_{12}$ 의 생성을 증가시켰다. 이런 배양방법을 이용하여 탄소원과 유기산이 vitamin $B_{12}$ 생성에 미치는 영향을 알아본 결과 glucose, fructose, lactose가 좋은 탄소원이었으며 유기산 중에서는 succinate 와 malate가 각각 34.9%와 18.7%의 vitamin $B_{12}$생성을 촉진시켰다.

  • Following the study on the fermentation conditions influencing the production of vitamin $B_{12}$ by Propionibacterium shermanii(Korean J Biotechnol. Bioeng. 7,126-131, 1992), the effects of some factors supplemented in the medium on the production of vitamin $B_{12}$ were studied. Maximum production of vitamin $B_{12}$ was observed when $Co^{+2}$ was supplemented at the concentration of 2-4 ppm in the fermentation medium. Increase of the supplemented $Co^{+2}$ to 12 ppm did not inhibit the growth of the organism, but it accelerated the lysis of the organism. In the literature, peptone was reported to activate the biosynthesis of vitamin $B_{12}$. Examination of the effect of peptone on the growth and the production of vitamin $B_{12}$ showed that at early stage more vitamin$B_{12}$ was observed in the supplemented medium, but no difference was observed in the later stage of fermentation. Examination of the time for addition and the amount of 5,6-dimethylbenzimidazole, a precursor known to influence the production of vitamin $B_{12}$, showed that a maximum yield of vitamin $B_{12}$ was observed when 15 mg/L was added to the fermentation medium after 2 days' incubation. The effect was comparable with the increase of the production of vitamin $B_{12}$ when the fermentation condition was changed to aerobic condition after 2 days' culture under anaerobic condition.

노인의 혈중 호모시스테인, 엽산, 비타민 $B_{12}$ 수준 및 영양소 섭취 상태와 신경인지기능과의 관련성 (Relationship among Plasma Homocysteine, Folate, Vitamin $B_{12}$ and Nutrient Intake and Neurocognitive Function in the Elderly)

  • This study examined the relationship among plasma homocysteine, folate, and vitamin $B_{12}$ levels and neurocognitive function in 118 community-dwelling elderly subjects (mean age, $75.1{\pm}6.7$ years). The Mini-Mental State Examination (MMSE-KC) was used to screen and assess neurocognitive function in the participants. Dietary intake data including the use of dietary supplements were obtained using the 24-hour recall method by well-trained interviewers. Plasma folate and vitamin $B_{12}$ concentrations were analyzed by radioimmunoassay, and homocysteine was assessed by a high performance liquid chromatography-fluorescence method. The proportions of participants with suboptimal levels of plasma folate (< 3 ng/mL), vitamin $B_{12}$ (< 221 pmol/mL), and homocysteine (> $15{\mu}mol/L$) were 16.1%, 5.9%, and 21.2%, respectively. A multiple regression analysis showed that plasma homocysteine was negatively associated with plasma folate and vitamin $B_{12}$ levels. The MMSE-KC test scores were significantly associated with plasma homocysteine and folate, but not with vitamin $B_{12}$, after adjusting for age, gender, body mass index, living with spouse, education, current smoking, energy intake, and chronic diseases such as hypertension, diabetes, thyroid disease, dyslipidemia, stroke, and cardiovascular disease. A general linear model adjusted for covariates revealed that MMSE-KC test scores increased from the lowest to the highest quartiles of vitamin $B_1$, vitamin $B_2$, vitamin $B_6$, vitamin $B_{12}$, and vitamin C intake (p for trend = 0.012, 0.039, 0.014, 0.046, 0.026, respectively). These results indicate that the problem of folate inadequacy and hyperhomocysteinemia are highly prevalent among community-dwelling elderly people and that dietary intake of the B vitamins and vitamin C is positively associated with cognitive function scores.

  • 비타민은 동물과 어류에 있어 성장, 발육, 및 대사기능에 비교적 적은 양으로 요구되는 필수 미량원소이다. 이들 미세 영양분이 결핍됨으로써 식욕 감퇴에서부터 심한 조직 변형등의 증상들을 일으킨다. 수용성 vitamin B6는 비교적 적은양이 요구되며, 주로 보조 효소 기능을 가지고 있다. 비타민 B6라는 명칭은 유사한 대사 기능들을 가진 화학적으로 관련된 pyridoxine, pyridoxamine, pyridoxal들을 의미한다. 비타민 B6는 비 반추 포유류, 조류와 어류의 음식물을 통해 섭취되는 성분이며, 비타민 B6 화합물들은 식물과 미생물 등에 의해 합성된다. 대사적으로 활성형인 비타민 B6 보조효소는 pyridoxal-5-phsphate(PLP)이며, decarboxylases, aminotransferases, sulfhydrases, tryptophanases를 포함한 아미노산 대사에 관여하는 여러 효소들(PLP-dependant)에 coenzyme으로 작용한다. 비타민 B6 요구량이 육식 동물보다 고단백질을 섭취하는 어류에서 더 높다. B6는 탄수화물과 지질의 대사에도 역시 관여하며 heme과 serotonin의 합성에 필수적이다. 어류에서 결핍증(현상)은 빨리 나타나는데, 이러한 증세로는 신경계 분열, 경련, 유영 부조화, 피부병변, 부종, 안구 돌출, 근 긴장성 경련 등이 포함된다. 비타민 B6는 pyridoxal kinase와 pyridoxine(pyridoxamine) oxidase의 촉매 반응에 의해 생체내 활성형인 PLP로 된다. PLP는 PLP-의존성 효소에서 보조 효소로 필수적인 역할을 하는 관계로 이 논문에서는 비타민 B6 생합성 및 대사와 비타민 B6 의존성 효소(aminotransferase)의 특성과 작용기작에 대한 효소학적 연구를 중점으로 이들 enzyme들의 구조와 기능에 대한 최근 연구 동향을 살펴보고자 한다.

채소류의 수확 후 원재료 및 데침 처리에 의한 비타민 B5 및 B6 함량 변화 (Vitamin B5 and B6 Contents in Fresh Materials and after Parboiling Treatment in Harvested Vegetables)

  • 국내 다소비 채소류 39종에 대해 원재료 및 데침 조리 시 함유된 수용성 비타민 $B_5$$B_6$의 함량변화를 자외부 흡광 검출기(HPLC/DAD) 및 형광검출기(HPLC/FLD)로 분리 검출하였다. 각 성분의 분석법을 밸리데이션하여 신뢰도 높은 분석법임을 확인한 후 조제분유를 이용하여 내부 분석품질을 관리하였고, 비타민 $B_6$의 경우에는 국제 정도 관리를 통하여 정밀한 분석 수행능력을 평가 받았다. 검증된 분석법으로 채소류 39종의 원재료 및 데침 조리 시 비타민 $B_5$$B_6$ 함량 변화를 분석한 결과, 비타민 $B_5$의 경우 근채류에서는 마늘, 엽채류에서는 포항초, 경채류는 두릅, 과채류의 숙과용 호박이 각각의 분류에서 가장 높은 함량을 나타내었으며, 그 중 경채류인 두릅($2.462mg{\cdot}100g^{-1}$)이 가장 높은 함량을 나타내었다. 비타민 $B_6$의 경우에는 근채류에서 마늘, 엽채류에서는 근대, 경채류는 마늘쫑, 과채류의 적색토마토가 각각의 분류에서 가장 높은 함량을 나타내었고, 그 중 엽채류의 근대($0.127mg{\cdot}100g^{-1}$)가 가장 높은 함량으로 평가되었다. 대부분의 채소류에서 데침 처리 시 원재료에 비해 $B_5$$B_6$의 함량이 감소하거나 검출되지 않는 양상을 나타내었으며, 특히 비타민 $B_5$는 두릅, 비타민 $B_6$는 노란감자 및 근대에서 각각 약 20배 및 4배 대폭 감소하는 경향을 나타내었다. 이 결과는 채소작물의 이용 및 가공, 국민 식생활 정보 및 학교 조리 급식 등 영양적 부분과 국민 건강을 위한 중요한 기초자료로 이용될 수 있을 것이다.

Analysis of Vitamin $B_{12}$ in the Korean Representative Foods and Dietary Intake Assessment for Koreans

  • This study was conducted to provide a vitamin $B_{12}$ database for the representative Korean food items and to assess the dietary intake assessment of vitamin $B_{12}$ for Koreans. The vitamin $B_{12}$ content of 106 foods had been determined by high performance liquid chromatography (HPLC) using column switching method. Rich sources of vitamin $B_{12}$ were meats, milk, and egg ($0.3-3.4\;{\mu}g/100g$). Vegetables and fruits contained vitamin $B_{12}$ below limit of detection (LOD). The major food sources for vitamin $B_{12}$ intake were milk (72.0%), meats (22.3%), egg (3.6%), and fishes & shellfishes (2.1%). Mean vitamin $B_{12}$ intake of Koreans was $3.16\;{\mu}g$/person/day. The proportion of population with intake below estimate average requirement (EAR) and above recommended intake (RI) of vitamin $B_{12}$ was 60.7 and 36.5%, respectively. The vitamin $B_{12}$ intake level of young children with 1-2 years which was 834.6% of RI while the intake level of the older adults 50 years and older was only 70.0% of RI. Also, there were regional differences between urban and rural area. The population with intake below RI was larger than that with intake above RI in Korea.

미숙아를 출산한 산모의 임신시 영양소 섭취실태에 관한 연구 (I) (A Study on Nutrient Intake During Pregnancy of Women of Premature Delivery (I))

  • Inadequate nutritional status of pregnancy can cause underweight and premature birth, undergrowth and deliverance of physically and mentally defected babyies. The purpose of this study is to provide guidelines for preventing preterm delivery in the aspect of nutritional factors. The nutrient intakes were compared between a preterm delivery group and a normal term delivery group to recognize risk factors of preterm delivery. The results obtained are summarized as follows. The pregnancy period was statistically longer in the normal term group (p<0.0001). Weight increase was statistically higher in the normal term group (p<0.0001). Calories (p<0.05), carbohydrates (p<0.0005), dietary fibers (p<0.0001), potassium (p<0.0005), vitamin $B_1$ (p<0.0005), vitamin $B_6$ (p<0.05), vitamin C (p<0.0001), and folic acid (p<0.05) intakes were statistically higher in the normal term group. Nutrient density of vitamin $B_1$ (p<0.05) and vitamin C (p<0.0001) in the normal term group was statistically higher. Nutrient adequacy ratio of zinc (p<0.05), vitamin $B_1$ (p<0.05) and folic acid (p<0.05) were statistically higher in the normal term group. Index of Nutritional Quality of vitamin $B_1$ (p<0.05) and vitamin C (p<0.0001) were statistically higher in the normal term group. In this study, the normal term delivery showed higher intakes of calories, carbohydrates, dietary fiber, crude fiber, potassium, vitamin $B_1$, vitamin $B_6$ vitamin C and folic acid than the preterm delivery group. Deficiencies in various nutrients may lead to preterm delivery, therefore, balanced nutrient intake is recommended to prevent preterm delivery.

관상동맥협착 환자의 혈장 총 Homocysteine, 엽산 및 비타민 $B_{12}$ 농도 (Plasma Total Homocysteine, Folate and Vitamin $B_{12}$ Concentrations in Patients with Coronary Stenosis)

  • Recently elevated plasma homocysteine concentration is considered an independent risk factor for atherosclerosis and thrombosis with coronary artery disease. Folate and vitamin $B_{12}$ are cofactors and closely related with metabolism of homocysteine. The purpose of this study is to evaluate the correlation between homocysteine and folate and vitamin $B_{12}$ in patients with ischemic heart disease. Twenty-six patients, in whom coronary angiographic finding revealed more than $50\%$ of stenosis at least in one coronary vessel were enrolled as the patient group, and thirty subjects, in whom angiographic finding revealed in not significant stenosis, but complained of chest pain, were selected as the control group. Fasting venous blood was obtained and measured the concentration of plasma total homocysteine, folate and vitamin $B_{12}$ by high performance liquid chromatography and fluorescence detection method. We examined the correlation between homocysteine and folate and/or vitamin $B_{12}$ in the control group and the patient group, respectively. Compared with the control group, the patient group had relatively higher plasma total homocysteine concentration ($10.7\pm4.2\;vs\;9.6\pm3.5$ umol/L), but showed no significant difference. Folate and vitamin $B_{12}$ concentration are low in the patient group, but showed no significant difference between patient and control group. Plasma total homocysteine concentration showed negative correlation with folate and vitamin $B_{12}$ in both the control group and the patient group, and showed significantly negative correlation in patient group {r = -0.550 (p < 0.01) vs r = -0.609 (p < 0.01)}. We knew that the plasma total homocysteine concentration were relatively elevated in patient group compared with the control group. Because plasma total homocysteine concentrations are closely negative correlated with folate and vitamin $B_{12}$ in the patient group, folate and vitamin $B_{12}$ supplement can lower the mortality and morbidity of ischemic heart disease. 저열량 생즙 식이가 성인의 혈청 페리틴 농도에 미치는 영향 (Effects of a Low-Calorie Raw Juice Diet on the Level of Serum Ferritin in Korean Adults)

  • This study examines the effects of a low-calorie raw juice diet on the level of serum ferritin in adults and analyzes nutrient intake from the diet. There were significant differences between juices; the highest calorie was provided by pear juice, highest crude protein, vitamin A, and vitamin B2 levels were from green Juice 1; and highest vitamin C and vitamin B1 levels were from fruit juices. The ratio of estimated energy requirements (EER) for the participants was 56.2% from the raw juice diet. The percentages of recommended intake (RI) from the raw juice diet of protein (57.9%), dietary fiber (19.1%), niacin (6.2%), calcium (0.1%), and magnesium (0.2%) were lower than 75%. However, those of RI of vitamin A, vitamin B1, vitamin B2, vitamin B6, and vitamin C were 1796.5%, 7481.7%, 1915.5%, 30858.7%, and 7500%, respectively, exceeding the tolerable upper intake level (UL) for vitamin A, vitamin B6, and vitamin C. There were significant decreases in weight, the body mass index (BMI), body fat mass, and skeletal muscle mass in males and females. After the diet program, serum iron and SOD (superoxide dismutase) showed significant decreases, whereas RBC, hemoglobin, hematocrit, and serum ferritin showed significant increases. There were negative correlations between serum ferritin and weight and between serum ferritin and skeletal muscle mass for all participants. There were negative correlations between serum ferritin and skeletal muscle mass for males and between serum ferritin and body fat mass for females. These results suggest that a raw juice diet can supplement a regular diet to prevent excess or deficient nutrient intake.
