제주지역에 서식하고 있는 블루베리 (Vaccinium corymbosum L.)로부터 수용성 추출물을 효과적으로 제조하기 위하여 5가지 종류의 탄수화물 분해효소 (AMG, Celluclast, Termamyl, Ultraflo, Viscozyme)와 또 다른 5종의 단백질 분해효소 (Alcalase, Flavourzyme, Kojizyme, Neutrase, Protamex)를 이용하여 효소적 추출을 시도하였다. 이들 효소를 이용하여 제조된 여러 가지 블루베리 효소 추출물에 대하여 1,1-diphenyl 2-picrylhydrazyl (DPPH), 활성산소종 (ROS), 일산화질소 $(NO{\cdot})$ 등의 소거 활성, 금속 킬레이트결합능, 그리고 지질 과산화 저해능과 같은 항산화 효과를 검토하였다. 모든 효소 추출물의 페놀함량은 $517.85\~597.96\;mg/100 g$ 건조시료이었으며, 항산화 효과에 대한 평가에서 특히 DPPH 및 $NO{\cdot}$ 소거활성, 그리고 금속 킬레이트 결합능 등이 우수한 것으로 나타났다. Viscozyme 추출물은 DPPH 소거활성 $(0.046{\pm}0.002\;mg/mL)$이, 그리고 AMG 추출물은 $NO{\cdot}$ 소거활성$(0.339{\pm}0.011\;mg/mL)$ 및 지질 과산화 억제활성 $(0.28{\pm}0.01\;mg/mL)$이 각각 우수하였다. 과산화 수소 소거활성에 있어서는 블루베리 효소 추출물들이 천연 항산화제인 a-토코페롤보다 상대적으로 높았다. 이러한 결과로 볼 때 블루베리는 항산화 효과가 우수한 화합물들을 함유하고 있을 것으로 판단되며, 향후 천연 항산화 자원으로서 이용이 가능할 것으로 사료된다.
In this study, silkworms fed Cudrania tricuspidata leaves were powdered, and hydrolytic enzymes, including viscozyme, papain, and flavourzyme, were added to verify the functionality of the different mixtures. In the general component analysis, the C. tricuspidata silkworm (CS) group exhibited higher crude protein and ash content than did the other groups, and the enzyme-treated groups exhibited higher carbohydrate content than the CS group. The DPPH and ABTS radical scavenging activities were significantly higher in the CS treated with viscozyme (CSV) and the CS treated with viscozyme/flavourzyme (CSVF) than in the other groups, with the CSV group showing the highest reducing power. ACE inhibitory activity was significantly higher in the CS treated with visocozyme/papain (CSVP) than in the CS group. In conclusion, rather than using powdered silkworms fed C. tricuspidata leaves, it would be more effective to use hydrolysates from C. tricuspidata silkworms as raw materials for functional foods.
This study aimed to investigate the enzymatic hydrolysis of mannitol using Viscozyme$^{(R)}$ L, Celluclast$^{(R)}$ 1.5 L, Saczyme$^{(R)}$, Novozym$^{(R)}$, Fungamyl$^{(R)}$ 800 L, Driselase$^{(R)}$ Basidiomycetes sp., and Alginate Lyase, and to optimize of reaction conditions for production of reducing sugar. Response surface methodology (RSM) based on central composite rotatable design was used to study effects of the independent variables such as enzyme (1-9% v/w), reaction time (10-30 h), pH (3.0-7.0) and reaction temperature ($30-70^{\circ}C$) on production of reducing sugar from mannitol. The coefficient of determination ($R^2$) of $Y_1$ (yield of reducing sugar by Viscozyme$^{(R)}$ L) and $Y_3$ (yield of reducing sugar by Saczyme$^{(R)}$) for the dependent variable regression equation was analyzed as 0.985 and 0.814. And the p-value of $Y_1$ and $Y_3$ showing 0.000 and 0.001 within 1% (p < 0.01), respectively, was very significant. The optimum conditions for production of reducing sugar with Viscozyme$^{(R)}$ L were 9.0 % (v/w) amount of enzyme, 30.0 hours of reaction time, pH 4.5 and $30.0^{\circ}C$ of reaction temperature, and those with Saczyme$^{(R)}$ were 9.0% (v/w) of amount of enzyme dosage, 30.0 h of reaction time, pH 7.0 and $30.0^{\circ}C$ of reaction temperature, consequently, the predicted reducing sugar yields were 22.5 and 27.9 mg/g-mannitol, respectively.
양파 과육부에 함유되어 있는 유용 성분인 quercetin과 그 배당체를 다양한 착즙 및 추출방법에 따른 수율을 측정, 분석하였으며 그 결과를 요약하면 다음과 같다. 양파과육 의 quercetin과 배당체의 최적 추출 조건으로는 50$^{\circ}C$에서 60% methanol를 용매로 사용했을 때 15분의 추출시간인 것으로 나왔으며 용매와 시료의 비율이 0.8 mL/g일 때 추출 수율이 가장 좋았다. 초음파를 60분 동안 처리 시 대조군에 비하여 2.06배의 추출 증가 효과를 볼 수 있었으며 microwave는 60초 조사 시에 최대의 추출 증가 효과를 나타내어 microwave를 처리하지 않은 시료와 비교하여 quercetin과 배당체가 2.14배 많이 추출되었다. 효소 처리 시 cellulase 용액과 시료의 비율이 0.5 mL/g일 때 quercetin과 배당체의 수율이 가장 높았으며 1.65 배 더 많이 추출되었고 viscozyme과 시료의 비율이 0.5 mL/g일 때 quercetin과 배당체의 수율이 가장 높았으며 추출증가율은 2.29 배인 것으로 나왔다. 가장 많은 quercetin과 배당체의 추출 수율을 보인 방법은 viscozyme과 시료의 비율이 0.5 mL/g일 때의 0.070${\pm}$0.002 mg/g인 것으로 나타났다.
The ginsenoside Rh2 has strong anti-cancer, anti-inflammatory, and anti-diabetic effects. However, the application of ginsenoside Rh2 is restricted because of the small amounts found in Korean white and red ginsengs. To enhance the production of ginsenoside Rh2-MIX (comprising 20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3 as a 10-g unit) with high specificity, yield, and purity, a new combination of enzymatic conversion using the commercial enzyme Viscozyme L followed by acid treatment was developed. Viscozyme L treatment at pH 5.0 and $50^{\circ}C$ was used initially to transform the major ginsenosides Rb1, Rb2, Rc, and Rd into ginsenoside F2, followed by acid-heat treatment using citric acid 2% (w/v) at pH 2.0 and $121^{\circ}C$ for 15 min. Scale-up production in a 10-L jar fermenter, using 60 g of the protopanaxadiol-type ginsenoside mixture from ginseng roots, produced 24 g of ginsenoside Rh2-MIX. Using 2 g of Rh2-MIX, 131 mg of 20(S)-Rh2, 58 mg of 20(R)-Rh2, 47 mg of Rk2, and 26 mg of Rh3 were obtained at over 98% chromatographic purity. Then, the anti-cancer effect of the four purified ginsenosides was investigated on B16F10, MDA-MB-231, and HuH-7 cell lines. As a result, these four rare ginsenosides markedly inhibited the growth of the cancer cell lines. These results suggested that rare ginsenoside Rh2-MIX could be exploited to prepare an anti-cancer supplement in the functional food and pharmaceutical industries.
Ferulic acid is a phenolic compound that serves as a major biosynthetic precursor of vanillin in higher plants. We investigated the ability of the 3 commercial enzymes - Ultraflo L, Viscozyme L, and ${\alpha}-Amylase$ - to induce the release ferulic acid from the Ipomoea batatas L. (sweet potato) stem. The rate of release for ferulic acid was optimal when Ultraflo L (1.0%) was used compared with the other enzymes, whereas Viscozyme L was most effective for the release of vanillic acid and vanillin. Thus, these enzymes may be useful for the large-scale production of ferulic acid and other phenolic compounds from sweet potato stem.
In this study, the production of total reducing sugar from macro green-algae Enteromorpha intestinalis by enzymatic hydrolysis was investigated. As a result of enzymatic hydrolysis using 13 kind commercial enzymes, the highest yield of 8.75% was obtained from Viscozyme L, which is multi-enzyme complex such as cellulase, arabanase, beta-glucanase, hemicellulase and xylanase. As a control, only 0.33% and 0.27% yield were obtained from 1% sulfuric acid and 0.05 M citrate buffer (pH 4.8), respectively. In the case of enzyme mixture, the mixture of $Viscozyme^{(R)}$ L and $Cellic^{(R)}$ CTec2 (1:1) was presented the highest yield of 10.67%. Finally, the 14.99% yield was obtained at 36 hr under the condition of 10% biomass and 30% enzyme mixture.
본 연구에서는 미역을 이용하여 초고온 열산 가수분해, 효소 당화, 발효과정을 거쳐 아세톤, 부탄올, 에탄올을 생성하는 실험에 대해 진행하였다. 초고온 열산 가수분해에서의 최적 조건은 10%의 slurry, 270 mM의 황산, $160^{\circ}C$에서의 7.5분이었다. 초고온 열산 가수분해는 열처리 시간을 줄이고 적은 농도의 황산을 사용해도 더 많은 당과 적은 저해물질을 생성해 낸다는 장점이 있다. 효소 당화에서는 Viscozyme L (${\beta}-glucanase$, Novozymes)을 12 unit/ml으로 처리하는 것이 25.1 g/l로 가장 많은 단당을 생성했다. 발효에서는 C. acetobutylicum KCTC 1724이 비교적 낮은 pH 5.0에서 많은 아세톤, 부탄올, 에탄올을 생성하는 장점이 있었지만 mannitol을 모두 소비하지 못하는 단점이 있어 고농도의 mannitol 배지에 순치한 C. acetobutylicum KCTC 1724을 사용하여 발효를 진행하였다. 그 결과, 아세톤, 부탄올, 에탄올이 각각 0.99 g/l, 5.62 g/l, 2.44 g/l로 순치하지 않은 C. acetobutylicum KCTC 1724를 이용해 발효했을 때 보다 부탄올은 2.45 g/l, 에탄올은 1.10 g/l 증가했으며 수율($Y_{ABE}$)은 0.24에서 0.37로 증가했다.
해조류 중 홍조류인 꼬시래기(G. verrucosa)로부터 효모를 이용한 발효를 위해 열산가수분해, 효소당화 및 에탄올 발효수율 향상을 검토하고, 기존의 혼합당의 흡수효율을 높이기 위해 고농도 당 순치를 수행하였다. 열산가수분해는 200 mM 황산(H2SO4)을 이용하여 10% (w/v)의 꼬시래기(G. verrucosa)의 슬러리, 130℃의 온도에서 60분 동안 열산가수분해를 수행하였다. 또한 wild type 효모와 고농도 galactose에 순치(adaptive evolution)된 효모를 이용한 발효를 실시한 결과, wild type 효모의 경우 발효 144시간에 8.5 g/l 에탄올 발효로 에탄올수율계수 YEtOH = 0.19와 galactose에 순치된 효모의 경우 21.5 g/l 에탄올 발효로 에탄올수율계수 YEtOH = 0.50을 나타내었다. 이러한 연구결과는 해양 바이오매스인 해조류로부터 바이오 연료의 효율적인 생산방법을 제공할 수 있을 것으로 판단된다.
온주밀감 및 당유자 진피 추출물의 추출수율은 탄수화물분해효소인 viscozyme 추출물과 단백질분해효소인 kojizyme 추출물에서 높았으며, 당유자의 효소 추출물들이 온주밀감에 비하여 높게 나타내었다. 총 폴리페놀 함량은 탄수화물분해효소인 ultaflo와 단백질분해효소인 alcalase 및 flavourzyme에서 높았으며, 온주밀감 추출물들이 당 유자에 비하여 높게 나타내었다. 총 플라보노이드 함량은 탄수화물분해효소인 ultaflo와 단백질분해효소인 alcalase 및 물 추출물에서 높았다. DPPH radical 소거활성은 온주밀감에 비하여 당유자의 효소 추출물에서 매우 높게 나타났으며, 탄수화물분해효소인 viscozyme과 단백질분해효소인 kojizyme에서 가장 높았다. 특히 viscozyme 추출물은 86% 이상의 매우 높은 활성을 나타내었다. Hydrogen peroxide 소거활성은 추출물의 농도가 증가됨에 따라 약간씩 증가하였으나, 2.0 mg/mL에서는 온주밀감 진피의 kojizyme 추출물을 제외하고는 91% 이상의 매우 높은 활성을 나타내었다. Alkyl radical 소거활성은 추출물의 농도가 증가됨에 따라 급격히 증가하였으며, 모든 효소 추출물들은 0.5 mg/mL 이상에서 매우 높은 활성을 나타내었다. Hydroxyl radical 소거활성은 추출물의 농도가 증가됨에 따라 급격히 증가하였으며, 모든 효소 추출물들은 다른 활성산소종들의 활성에 비하여 상대적으로 낮은 활성을 나타내었다.
