Sweet potato is grown extensively from tropical to temperate regions and is an important food crop worldwide. In this study, we established detection methods for 17 major sweet potato viruses using single and multiplex RT-PCR assays. To investigate the current incidence of viral diseases, we collected 154 samples of various sweet potato cultivars showing virus-like symptoms from 40 fields in 10 Korean regions, and analyzed them by RT-PCR using specific primers for each of the 17 viruses. Of the 17 possible viruses, we detected eight in our samples. Sweet potato feathery mottle virus (SPFMV) and sweet potato virus C (SPVC) were most commonly detected, infecting approximately 87% and 85% of samples, respectively. Furthermore, Sweet potato symptomless virus 1 (SPSMV-1), Sweet potato virus G (SPVG), Sweet potato leaf curl virus (SPLCV), Sweet potato virus 2 ( SPV2), Sweet potato chlorotic fleck virus (SPCFV), and Sweet potato latent virus (SPLV) were detected in 67%, 58%, 47%, 41%, 31%, and 20% of samples, respectively. This study presents the first documented occurrence of four viruses (SPVC, SPV2, SPCFV, and SPSMV-1) in Korea. Based on the results of our survey, we developed multiplex RT-PCR assays for simple and simultaneous detection of the eight sweet potato viruses we recorded.
Viral diseases have been a major limiting factor threating sustainable potato (Solanum tuberosum L.) production in Pakistan. Surveys were conducted to serologically quantify the incidence of RNA viruses infecting potato; Potato virus X (PVX), Potato virus Y (PVY), Potato virus S (PVS), Potato virus A (PVA), Potato virus M (PVM) and Potato leaf roll virus (PLRV) in two major potato cultivars (Desiree and Cardinal). The results suggest the prevalence of multiple viruses in all surveyed areas with PVY, PVS and PVX dominantly widespread with infection levels of up to 50% in some regions. Co-infections were detected with the highest incidence (15.5%) for PVX and PVS. Additionally the data showed a positive correlation between co-infecting viruses with significant increase in absorbance value (virus titre) for at least one of the virus in an infected plant and suggested a synergistic interaction. To test this hypothesis, glasshouse grown potato plants were challenged with multiple viruses and analyzed for systemic infections and symptomology studies. The results obtained conclude that multiple viral infections dramatically increase disease epidemics as compared to single infection and an effective resistance strategy in targeting multiple RNA viruses is required to save potato crop.

This study was performed to investigate the incidence of Hosta uirus X (HVX), a Potexvirus, from cultivated hosta ornamental plants in Korea and to ascertain seed transmission of the virus from infected parent plant to progeny ones for breeding program of hosta plants. Infection rate of HVX in cultivated hostas was 25.6 % (11 out of 43 collected samples contained HVX) based on Western blot and RT-PCR detection methods. Most of HVX-infected hostas showed visible systemic leaf symptoms (mosaic, mottle, curling, stunting or combinations). Variability of HVX was confirmed by sequences of coat protein gene of individual isolates from different hostas. HVX was seed-transmitted on Hosta 'Blue Cadet'. The virus was detected from seeds, and sprouts and seedlings from the virus-contaminated seed sources. Over 7.5 % of seeds were HVX-contaminated surveyed in this study, Our data suggest that HVX can be transmitted by seed source, and indexing of the virus should be done for breeding program of Hosta.
Among more than 30 viruses infecting strawberry, aphid-transmitted viruses including Strawberry mild yellow edge virus (SMYEV), Strawberry mottle virus (SMoV), Strawberry crinkle virus (SCV), and Strawberry vein banding virus (SVBV) have been considered as the most economically important viruses of strawberry in the world. To determine the incidence of these four viruses in major Korean strawberry cultivars, field surveys were conducted in major production areas during 2018-2019. In our surveys, SMYEV and SMoV were detected with low infection rates of 0.7% and 1.3%, respectively, whereas SCV and SVBV were not detected. No obvious symptoms were observed in the strawberry plants infected by SMYEV or SMoV. Since no sequences of SMoV Korean isolates have been reported, we initially determined nucleotide sequence of the 3' untranslated region (UTR) of seven SMoV isolates obtained during the surveys. The 3' UTR sequences (782 nt) of seven Korean isolates were phylogenetically compared with those of the previously reported SMoV isolates. Phylogenetic analysis revealed that most Korean isolates are closely related to Canadian isolates and only little evolutionary differentiation occurred among the Koreans isolates. This might be due to the low incidence of SMoV in strawberry in Korea.
The incidence of Cherry necrotic rusty mottle virus (CNRMV) and Cherry green ring mottle virus (CGRMV) have recently been occurred in Korea, posing a problem for sweet cherry cultivation. Since infected trees have symptomless leaves or ring-like spots on the pericarp, it is difficult to identify a viral infection. In this study, the incidence of CNRMV and CGRMV in sweet cherry in Gyeongbuk province was surveyed using a newly developed duplex reverse transcriptase polymerase chain reaction (RT-PCR) method that can detect both viruses in a single reaction. CNRMV and CGRMV co-infection rates were 29.6%, 53.6%, and 17.6%, respectively, in samples collected from three different sites (Daegu, Gyeongju and Gyeongsan) in Gyeongbuk province during 2012 and 2013. This duplex RT-PCR method offers a simple, rapid, and effective way of identifying CNRMV and CGRMV simultaneously in sweet cherry trees, which can aid in the management of viral infections that could undermine yield.

Soybean mosaic virus (SMV) resistance of Korean recommended soybeans was evaluated naturally and by mechanical inoculation in Suwon. Based on the differential reaction of forty-four soybean genotypes tested to nine different SMV strains, soybeans were classified into twenty-four groups. Myeongjunamulkong and Ilpumgeom-jeongkong showed a high degree of resistance to nine SMV strains, having no symptom. The other cultivars produced various reactions according to inoculation of each SMV strain: symptomless, mosaic or systemic necrosis. Only five cultivars such as Kwangankong, Eunhakong, Tawonkong, Namhaekong, Sobaegnamulkong were totally susceptible to every strain. There was variation in disease incidence. Soybeans, having the highest levels of resistance to G5H and G7H in the greenhouse, showed the lowest levels of SMV incidence in the field of Suwon. Myeong-junamulkong, Ilpumgeomjeongkong, Soyangkong, Pungsannamulkong, Sodamkong, Jangmikong, Geomjeong-kong2, Pureunkong, Sinpaldalkong2, Duyoukong, and Geumgangkong were fairly resistant to SMV. And SMV incidence of Taekwangkong, Saealkong and Baegunkong was over 45% with symptom of bud necrosis. And soybeans, highly resistant to SMV in the field and the greenhouse, were mainly derived from Jangyeobkong and Hwang-keumkong resistant to G1-G7.
The incidence of virus disease on peppers was investigated at the 52 areas in the whole country in 2002, 2004 to 2006. Among the six viruses, Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Pepper mild mottle virus (PMMoV), Broad bean wilt virus 2 (BBWV2), Tobacco mild green mosaic virus (TMGMV) and Tomato spotted wilt virus (TSWV), infecting peppers in Korea, the incidence of CMV, PepMoV, PMMoV and BBWV2 was 29.4%, 13.6%, 14.3%, 25.6%, orderly. TMGMV and TSWV had the same low infection rate of 1.0%. The infection rate of CMV was higher as 53.3% and 34.2% in 2002 and 2004, but it was decreased to 18.2% and 11.9% in 2005 and 2006, respectively. The infection rate of BBWV2 was lower as 1.3% in 2002 and 1.8% in 2004, but it was increased abruptly to 41.3% in 2005 and 58.2% in 2006. For the types of mixed infections of pepper viruses, the incidence of CMV+PepMoV was 62.6% in 2002 and 50.0% in 2004, and that of CMV+BBWV2 was increased suddenly from 33.3% in 2005 to 83.2% in 2006. The triplex infection of CMV+BBWV2+PepMoV was 6.4% in average. CMV caused severe mosaic and BBWV2 induced ring spots, and the two mixed virions caused chlorosis on the leaves of red peppers. TSWV induced the typical symptoms of multiple ring spots on the leaves and fruits of red peppers.
Disease incidences of cucumber virus diseases in Jeonnam province were 52.5%, 16.1%, 35.2%, and 50.9% in 1999, 2000, 2001, and 2002, respectively. Rod- and flexuous rod-shaped virus particles were observed with the frequencies of 63.2% and 10.5%, respectively from the samples collected in 1999 under EM observation. Rod-shaped virus particles are considered as tobamovirus while flexuous rod shaped particles are considered as potyviruses. To further confirm their nature, total of 312 diseased virus samples were collected from 2000 to 2002, and tested by RT-PCR. Disease incidences of tobamoviruses including Cucumber green mottle mosaic virus and Kyuri green mottle mosaic virus were 48.7% and 3.8%, respectively while those of potyviruses including Zucchini yellow mosaic virus, Papaya ringspot virus, and Watermelon mosaic virus were 15.7%, 9.3%, and 5.1%, respectively. Interestingly, Cucumber mosaic virus was hardly detected. About 5.8% of tested samples were infected with more than one virus. Tobamovirus infection was consistently observed from September to December regardless of planting time, whereas infection of potyviruses was observed in many cucumber cultivating areas where it was planted in September and October.
The incidence of Broad bean wilt virus 2 (BBWV2) on red pepper was investigated using the samples obtained from 24 areas of 8 provinces in Korea. Two hundred and five samples (79%) out of 260 collected samples were found to be infected with BBWV2. While the single infection rate of BBWV2 was 21.5%, the co-infection rate of BBWV2 with Cucumber mosaic virus, Pepper mottle virus, Pepper mild mottle virus and/or Potato virus Y was 78.5%. To characterize the genetic diversity of BBWV2 Korean isolates, 7 isolates were fully sequenced and analyzed. Phylogenetic analyses revealed that BBWV2 isolates could be divided largely into two groups as Group I and Group II. Based on the partial sequence analyses, 153 selected BBWV2 isolates were subgrouped into GS-I (21.6%), GS-II (3.9%) and GS-III (56.9%). BBWV2 GS-III, which was predominant in Korea, appears to be a new combination between Group I RNA-1 and Group II RNA-2. Viral disease incidence of BBWV2 on red pepper was under 2% before 2004. However, the incidence was increased abruptly to 41.3% in 2005, 58.2% in 2006 and 79% in 2007. These rapid increases might be related with the emergence of new combinations between BBWV2 groups.
Incidences of major feline viral diseases provide basic information for preventing viral disease in cats. Despite the growing interest in feline viral diseases, sero-surveillances have been lacking. In this study, we analyzed the diagnoses of feline viral diseases and conducted a sero surveillance of feline panleukopenia virus (FPV), feline calicivirus (FCV), feline herpesvirus-1 (FHV-1), and feline infectious peritonitis virus (FIPV) in Korean cats. Of the 204 confirmed cases since 2015, the numbers of diagnoses for FPV, FIPV, FCV, feline influenza virus, and FHV-1 were 156, 32, 12, 3, and 1 case, respectively. In total, 200 sera, collected between 2019 and 2021, were screened for the presence of antibodies against FPV, 2 FCVs, FHV-1, and FIPV using a hemagglutination inhibition test and a virus-neutralizing assay (VNA). The overall seropositive rates in cats tested for FPV, the 2 FCVs, FHV-1, and FIPV were 92.5%. 42.0%, 37.0%, 52.0%, and 14.0%, respectively. A low correlation (r = 0.466) was detected between the VNA titers of 2 FCV strains. The highest incidence and seropositive rate of FPV reveal that FPV is circulating in Korean cats. The low r-value between 2 FCVs suggests that a new feline vaccine containing the 2 kinds of FCVs is required.
