Inhibitory Effect of Lactobacillus plantarum K11 on the Adhesion of Escherichia coli O157 to Caco-2 Cells

  • Inhibitory effect of Escherichia coli O157 adhered to Caco-2 cells by the cells of Lactobacillus plantarum K11 and the cell-free culture supernatant (CFCS) and bacteriocin prepared from this strain was investigated. As the cell counts of viable L. plantarum K11 previously adhered to Caco-2 were increased, the rate of adhesion and adherent cell counts of E. coli O157 was lower. However, because the heated L. plantarum K11 rarely have the adhesion ability to Caco-2, the adhesion rate and adherent cell counts of E. coli O157 were high. In addition, the inhibitory effects of E. coli O157 adhesion by the CFCS and bacteriocin of L. plantarum K11 were dose-dependent manner. Therefore, the inhibition of adhesion of E. coli O157 to Caco-2 may result from the antimicrobial substances such as lactic acid and bacteriocin. Moreover the inhibitory activity of adhesion by the heated bacteriocin for 30 min at 100oC was similar to activity of non-treated bacteriocin, but the activity was disappeared by treatment with protease.

  • 동결건조법이 Nocardia mediterranei의 세포막 손상에 미치는 영향을 3H-thymidine 표지에 의한 DNA 유출 방법과 전자현미경으로 조사하였으며, 재수화 과정이 생존도에 미치는 영향을 연구하였다. 그 결과 N.mediterranei의 동결건조시 세포벽과 세포막의 손상이 세포 사멸의 원인으로 판명되었다.

Effect of Glutaraldehyde Treatment on Stability of Permeabilized Ochrobactrum anthropi SY509 in Nitrate Removal

  • For practical application, the stability of permeabilized Ochrobactrum anthropi SY509 needs to be increased, as its half-life of enzymatic denitrification is only 90 days. As the cells become viable after permeabilization treatment, this can cause decreased activity in a long-term operation and induce breakage of the immobilization matrix. However, the organic solvent concentration causing zero cell viability was 50%, which is too high for industrial application. Thus, whole-cell immobilization using glutaraldehyde was performed, and 0.1% (v/v) glutaraldehyde was determined as the optimum concentration to maintain activity and increase the half-life. It was also found that 0.1% (v/v) glutaraldehyde reacted with 41.9% of the total amine residues on the surface of the cells during the treatment. As a result, the half-life of the permeabilized cells was increased from 90 to 210 days by glutaraldehyde treatment after permeabilization, and no cell viability was detected.

  • TIM-1 (also known as KIM-1 and HAVcr-1) is a type I transmembrane glycoprotein member of the TIM family that may play important roles in innate and adaptive immune responses. The overexpression of proteins associated with membrane proteins is a major obstacle to overcome in studies of membrane protein structures and functions. In this study, we successfully coupled the overexpression of the TIM-1 protein with a C-terminal enhanced green fluorescent protein (GFP) tag in Escherichia coli. To the best of our knowledge, this report is the first to describe the overexpression of human TIM-1 in E. coli. The purified TIM-1-EGFP fusion protein recognized and bound directly to apoptotic cells and did not to bind to viable cells. Furthermore, we confirmed that the interactions of TIM-1-EGFP with apoptotic cells were blocked by TIM-1-Fc fusion proteins. This fusion protein represents a readily obtainable source of biologically active TIM-1 that may prove useful in future studies of human TIM-1.

Thymol and eugenol in essential oils enhance phage endolysin LysECP26-mediated cell wall disruption of Escherichia coli O157:H7

  • To enhance phage endolysin-mediated cell wall disruption of Escherichia coli O157:H7, the cells were co-treated with aromatic compounds, namely thymol or eugenol, found in essential oils and endolysin LysECP26. Interestingly, the minimal inhibitory concentrations of LysECP26 was four times lower when used in combination with either of the two compounds than when it was used alone. This synergistic activity was also confirmed by viable cell counting. Within 1 h of LysECP26 and eugenol or thymol co-treatment to the cells, there was a 2.3 or 3.8 log CFU/mL reductions, respectively. Additionally, field emission scanning electron microscopy showed cell wall disruption and severe morphological alterations of the cells in case of the combination treatments. Therefore, endolysin and thymol or eugenol co-treatment can help in developing efficient bio-control strategies against gram-negative pathogen E. coli O157:H7.

Apoptosis and Cell Cycle Arrest in Two Human Breast Cancer Cell Lines by Dieckol Isolated from Ecklonia cava

  • Purpose: Dieckol, a phlorotannin compound isolated from Ecklonia cava, has been reported to have antioxidant, antiviral, anti-inflammatory, and anticancer properties. The purpose of this study was to investigate its anticancer effects on human breast cancer cell lines. Methods: In this study, the viability of two human breast cancer cell lines SK-BR-3 and MCF-7 was investigated after dieckol treatment using a WST-1 assay. Apoptosis and cell cycle distribution were assayed via Annexin V-fluorescein isothiocyanate and propidium iodide staining followed by flow cytometric analysis. Immunoblotting analysis was also performed using Bax/Bcl-2 to determine whether the dieckol-induced apoptosis was mediated by the intrinsic apoptotic pathway. Results: In a dose dependent manner, dieckol reduced the number of viable cells and increased the number of apoptotic cells. The effect of dieckol on the cell cycle distribution was analyzed using flow cytometry. Dieckol treatment significantly increased the percentage of MCF-7 and SK-BR-3 in the G2/M phase. Immunoblot analysis revealed that 24 hours of dieckol exposure increased the Bax/Bcl-2 ratio. Conclusion: Dieckol induced cytotoxicity in MCF-7 and SK-BR-3 human breast cancer cells inducing apoptosis and cell cycle arrest. Therefore, it is suggested that dieckol may be a potential therapeutic agent for breast cancer.

A Robotic System for Transferring Tobacco Seedlings

  • Germinatin and early growth of tobacco seedlings in trays containing many cells is increasing in popularity . Since 100 % germination is not likely , a major problem is to locate and replace the content of those cells which contain either no seedling or a stunted seedling with a plug containing a viable seedling. Empty cells and seedlings of poor quality take up valuable space in a greenhouse. They may also cause difficulty when transplanting seedlings into the field. Robotic technology, including the implementation of computer vision, appears to be an attractive alternative to the use of manual labor for accomplishing this task. Operating AGBOT, short for Agricultural ROBOT, involved four steps : (1) capturing the image, (2) processing the image, (3) moving the manipulator, (4) working the gripper. This research seedlings within a cell-grown environment. the configuration of the cell-grown seedling environment dictated the design of a Cartesian robot suitable for working ov r a flat plane. Experiments of AGBOT performance in transferring large seedlings produced trays which were more than 98% survived one week after transfer. In general , the system generated much better than expected.

수생산물의 생산과 관리에 관한 기초연구 : ELISA법을 이용한 Edwardssiella tarda의 직접 검출 (Study on the production and management of aquatic animals : direct detection of Edwardsiella tarda using an enzyme linked immunosorbent assay)

  • 어류의 감염 조직으로부터 edwardsiellosis의 원인균인 Edwardsiella tarda를 whole cell 자체로 직접 검출할 수 있는 solid phase ELISA법을 연구하였다. A. hydrophila ATCC7966, V. anguillarum HUFP5001, Y. ruckeri 11-4, E. ictaluri 및 Streptococcus sp. NG8206 등의 어병세균에 대해 ELISA법으로 실시한 교차반응 분석에서 A. hydrophila ATCC7966 균주와 V. anguillarum HUFP5001 균주가 E. tarda Edk-2에 대한 토끼 항혈청에 대해 높은 교차반응을 나타내었으나, 항혈청을 A. hydrophila ATCC7966 FKC로 흡착시킴으로써 교차반응을 제거할 수가 있었다. 그러나, 응집항체가 측정 결과와는 달리, ELISA 분석에서는 E. tarda 분리 균주간의 교차반응이 매우 높은 것으로 나타났다. Tissue homogenate내에 있는 항원을 검출함에 있어, 조직내의 지질이나 단백질 성분이 함께 분석용 plate에 coating되어 감도가 훨씬 감소하므로 ELISA법의 적용을 위해서는 감염 조직의 homogenate를 PBS에 100배 이상 희석한 후 진단을 실시해야 하는 것으로 나타났다. Tissue homogenate내에 있는 생균을 항원으로 하여 직접 검출할 때에는 검출한계가 $1{\times}10^3$ cells/ml로 나타나 FKC 항원의 사용에 비하여 더 증가된 감도를 보여주었다. 결론적으로 본 ELISA법은 양식장에서 발생한 edwardsiellosis를 진단함에 있어서 특이적이고 신속하며 민감한 방법으로 확인되었다.

Enterococcus faecium MJ-14가 생산하는 박테리오신과 물리화학적 처리의 상승효과 (Synergistic Effect of Physico-chemical Treatment and Bacteriocin Produced by Enterococcus faecium MJ-14)

  • E.faecium MJ-14가 생산하는 박테리오신 30 BU/ML 처리에 의해 L. monocytogepes 균수는 0.6 log unit 감소 되었으나, 박테리오신 (30 BU/mL $65^{\circ}C$, 5분간 가열처리와 병용한 경우에는 약 2.7 log unit가 감소하였다. 박테리오신 30 BU/mL와 itric acid혹은 acetic acid $0.5\%$를 병용 처리한 경우에는 24시간 만에 초기균수가 각각 약 2 log unit와 3 log unit감소하였다. 박테리오신 30 BU/mL와 NaCl $8\%$를 병용 처리했을 때에는 박테리오신 단독 처리구보다 약 1 log unit더 감소되었다. 또한, 박테리오신 30 BU/mL단독 처리했을 때에 비해 sodium benzoate $500{\mu}g/mL$혹은 sodium lactate $1000{\mu}g/mL$와 각각 병용 처리하면 2 log unit가 더 감소되었으며, sodium nitrate $100{\mu}g/mL$와 potassium nitrate $100{\mu}g/mL$와의 병용 처리한 경우 박테리오신 단독 처리구에 비해 각각 3.5 log unit와 2 log unit이상 더 감소하였다. 한편, 탈지유에 E. faecium MJ-14의 박테리오신 300 BU/mL을 첨가한 경우 $4^{\circ}C$에서 24시간 만에 L. monocytogenes의 균수는 대조구에 비해 각각 약 1.5 log unit감소되었으며,돈육에 첨가한 경우 $-20^{\circ}C$에서 7일간 만에 대조구보다 약 2 log unit의 감소효과가 나타났다.

Mutans streptococci에 대한 polyphosphate의 항균효과 (ANTIBACTERIAL EFFECT OF POLYPHOSPHATES ON MUTANS STREPTOCOCCI)

  • 치아우식증의 원인균인 S. mutans GS5와 S. sobrinus 6715에 대한 polyP의 효과를 관찰하여 보다 안전하고 효과적인 치아우식증 예방을 위한 임상적용의 가능성을 고찰하고자 첫째, 다양한 사슬길이의 polyP를 첨가한 후 흡광도를 측정하여 MIC를 결정하고, 둘째, 실험균주를 흡광도 $0.3{\sim}0.5$까지 증식시킨 후 MIC 농도의 polyP를 첨가하여 흡광도의 변화를 측정함으로써 균주증식 후 성장 억제효과를 관찰하였으며, 셋째, 생균수 측정으로 polyP의 항균효과를 평가하였고, 넷째, 핵산유리의 정도로 polyP의 킬레이션 작용여부를 관찰하였으며, 다섯째, polyP의 비수용성 글루칸 합성능력을 관찰하였으며, 여섯째, 투과전자현미경으로 세포막과 세포질 내의 구조적 변화를 관찰하였다. 이상의 연구를 통하여, polyP의 살균작용이 S. mutans와 S. sobrinus에 대한 성장을 억제시키는 효과가 있는 것으로 가늠된다. 이와 같은 성장 억제효과는 polyP의 킬레이션에 의한 것이라기보다는 균주 세포의 구조적, 형태적 변화가 주된 요인이었던 것으로 판단된다.
