백하수오 알칼로이드 성분의 혈관이안 효능 (Vasodilatory Effect of the Alkaloid Component from the Roots of Cynanchum wifordi Hemsley)

  • Natural products are one of the useful source of cardiovascular drugs, in particular, when they have antioxidant activity. Gagaminine, an alkaloid isolated from the roots of Cynanchum wilfordi Hemsley, has been reported to potently inhibit the aldehyde oxidase activity ({TEX}$IC_{50}${/TEX}=0.8$\mu$M) and reduce lipid peroxidation. However, the effect of gagaminine on vascular smooth muscle has not yet been investigated. In the present study, we examined whether gagaminine relaxes vascular smooth muscle by isometric tension study. In order to observe its relaxation effect on the arteries, conductivel vessel (rat thoracic aorta) and resistance vessel (pig coronary artery) were purposely used. Results indicated that gagaminine relaxed in a concentration-dependent manner $\alpha$-adrenoceptor agonist, phenylephrine (PE)-induced contraction of rat aorta. Pretreatment with gagaminine inhibited PE-induced contraction, noncompetitively. {TEX}$Ca^{2+}${/TEX}-induced contraction was significantly diminished by gagaminine. In pig coronary artery, gagaminine relaxed thromboxane receptor (U 46619)-mediated contraction in dose-dependent manner. Pretreatment with gagaminine also reduced the maximum contraction induced by KCl. These observations strongly suggest that agagminnine relaxes vascular smooth muscle, irrespective of both resistance and conductive artery. We demonstrate that gagaminine, a potent natural antioxidant, has a significant vasodilatory effect and its action mechanism van be ascribed at least in part to {TEX}$Ca^{2+}${/TEX} antagonistic action as evidenced by inhibition {TEX}$Ca^{2+}${/TEX}-induced contraction (rat aorta) and KCl-induced contraction (porcine artery). Furthermore, neither $\alpha$ -adrenoceptor nor thromboxane receptor seems responsible for the relaxation of gagaminine.

Glucosamine increases vascular contraction through activation of RhoA/Rho kinase pathway in isolated rat aorta

  • Diabetes is a well-known independent risk factor for vascular disease. However, its underlying mechanism remains unclear. It has been reported that increased influx of the hexosamine biosynthesis pathway (HBP) induces O-GlcNAcylation of proteins, leading to insulin resistance. In this study, we determined whether or not O-GlcNAc modification of proteins could increase vessel contraction. Using an endothelium-denuded aortic ring, we observed that glucosamine induced OGlcNAcylation of proteins and augmented vessel contraction stimulated by U46619, a thromboxane $A_2$ agonist, via augmentation of the phosphorylation of MLC20$MLC_{20}$, MYPT1(Thr855), and CPI17, but not phenylephrine. Pretreatment with OGT inhibitor significantly ameliorated glucosamine-induced vessel constriction. Glucosamine treatment also increased RhoA activity, which was also attenuated by OGT inhibitor. In conclusion, glucosamine, a product of glucose influx via the HBP in a diabetic state, increases vascular contraction, at least in part, through activation of the RhoA/Rho kinase pathway, which may be due to O-GlcNAcylation.

  • As an attempt to develop new functional health beverage by using medicinal herb, Artemisia capillaris, we investigated the effect of scopoletin in Artemisia capillaris on vessel and regional cerebral blood flow (rCBF) of rats ingesting health drink prepared with Artemisia capillaris extracts and various ingredients. Artemisia capillaris extract decreased the vessel contraction and increased rCBF significantly. The extracts were grouped by heat temperature and mixed ratio and tested their respective characteristics. Then each condition was combined and produced the most effective one. The drink produced consisted of Artemisia capillaris extract 42%, honey 9.8%, citric acid 0.035%, cyclodextrin 1.47% and water. Brix, pH and acidity of the product were 9.2, 4.4 and 0.04%, respectively, This drink scored to have highest level on overall acceptance by the sensory evaluation. The above results showed that development of such functional beverage using Artemisia capillaris can be used as a functional material improving blood circulation in beverage industry.

鼻出血에 應用되는 艾柏飮의 效能에 關한 實驗的 硏究 (An Experimental Study on the Effects of Aebaegeum Applicable to the Treatment of Epistaxis)

  • Aebaegeum has been widely used in the treatment of epistaxis, as based on Oriental Medical 1iteratures. In order to investigate experimentally the clinical effects of Aebaegeum, hemostatic time, plasma recalcification time and prothrombin time of hypoprothrombinemia induced by warfarin injection, action on isolated ileum, action on blood vessel, action on blood pressure and respiration and action on heart were observed. The result of the studies were obtained as follows: 1. Hemostatic time of vein ruptured mice was significantly shortened. 2. Plasma recalification time and prothrombin time of hypoprothrombinemic rats induced by warfarin injection were significantly shortened. 3. Spontaneous mobilities in the isolated ileum of mice were significantly suppressed, and contraction by acetylcholine chloride and barium chloride were inhibited. 4. Dilatation of blood vessel and downing of blood pressure of rabbit were noted. 5. Contraction of heart in the original position and isolated heart of frog were significantly shortened. According to the above result, it is expected that Aebaegeum can be widely applicable to the treatment of epistaxis.

Beneficial Role of Ginseng Saponin on Hemodynamic Functions of Porcine Blood Vessel

  • The previous reports have showed that ginseng saponins, which are the active ingredients of Panax ginseng, cause the relaxation of artery that are contracted due to a various of hormones or potassium ($K^+$). Recently, we also showed that ginsenosides differentially regulate channel activity. The purpose of this study was to examine whether ginseng saponins affect contraction induced by $K^+$, serotonin (5-HT), or acetylcholine (Ach) in porcine coronary vessel. Treatment with concentrations of ginseng saponins caused a relaxation of 25 mM KCl-induced porcine coronary artery contraction. Also, ginseng saponin induced a significant dose-dependent relaxation of $3\;{\mu}M$ 5-HT-induced porcine coronary artery with the endothelium. In the porcine artery with the endothelium, ginseng saponins induced a relaxation by $3\;{\mu}M$ 5-HT in a concentration-dependent pattern. Ginseng saponins induced relaxation of both 25 mM KCl- and $3\;{\mu}M$ 5-HT-induced coronary artery contraction in the absence and presence of the endothelium. In contrast, treatment with $100\;{\mu}g/mL$ ginseng saponin did not induce relaxation in coronary artery contraction induced by Ach ($0.01\;{\mu}M$ to $30\;{\mu}M$) in the presence of the endothelium, but did cause significant relaxation of coronary artery contractions by Ach ($0.01\;{\mu}M$ to $30\;{\mu}M$) in the absence of the endothelium. These findings indicate that ginseng saponin (> $100\;{\mu}g/mL$) significantly inhibits porcine coronary artery contractions caused by $K^+$, 5-HT, and Ach. Therefore, in this study, we demonstrated that ginseng saponin may show beneficial roles on abnormal coronary contraction.

이산화염소의 활성산소 발생에 따른 화랑곡나방 심장박동 억제 효과 (Inhibitory Effect of Chlorine Dioxide Using Reactive Oxygen Species Against Heart Contraction of the Indianmeal Moth, Plodia interpunctella)

  • 곤충의 혈액순환은 심장 박동에 의해 도움을 받는다. 다양한 생리적 변화는 심장 박동 조절을 수반하게 된다. 심장박동에 대한 교란은 곤충의 생존을 위협하게 된다. 본 연구는 활성산소를 유발하여 살충력을 발휘하는 이산화염소가 혈액순환계에 미치는 영향을 심장박동을 통해 분석하였다. 화랑곡나방(Plodia interpunctella) 유충의 등핏줄은 몸의 윗면 중앙에 위치하고 후방으로 복부 10번째 마디에서 시작하여 전방으로 첫 번째 가슴 마디까지 연결된 관 구조를 나타냈다. 등핏줄의 수축과 이완은 주로 복부 3-10번째 마디에 위치한 등핏줄에서 일어났으며 이 부위에 5개의 심실이 관찰되었다. 심장박동빈도는 $25^{\circ}C$에서 분당 평균 118.6회의 수축 리듬을 보였다. 그러나 온도에 따라 심장박동빈도는 현격한 변화를 보였다. 혈강에 이산화염소를 다양한 농도로 투여한 경우 심장박동빈도는 약제 농도 증가에 따라 감소하였다. 이산화염소(100 ppm)을 훈증 처리할 경우 노출 시간의 경과에 따라 심장박동리듬이 현격하게 감소하였다. 이러한 이산화염소의 심장박동 억제효과는 활성산소 저해제인 비타민 E와 함께 주입할 경우 회복되는 현상을 나타냈다. 이상의 결과는 이산화염소가 화랑곡나방의 심장박동에 억제효과를 주었으며 이러한 억제효과는 이물질이 유발하는 활성산소에 기인된 것으로 해석된다.

마두령(馬兜鈴)이 혈관(血管) 평활근(平滑筋)에 미치는 영향(影響) (Effects of Fructus Aristolochiae on the Vascular Smooth Muscle)

  • Fructus Aristolochiae has been used in Korea for many centuries as a treatment for various disease.The purpose of the present study is to determine the effect of Fructus Aristolochiae on norepinephrine(NE) induced blood vessel contraction in rabbits. Rabbit(2 kg, male) were killed by $CO_2$ exposure and a segment (8-10mm) of each rabbit was cut into equal segments and mounted in a tissue bath. Contractile force was measured with force displacement transducers under 2-3 g loading tension. The dose of norepinephrine(NE) which evoked 50% of maximal response ($ED_{50}$) was obtained from cumulative dose response curves for NE ($10^{-6}{\sim}10^{-3}M$). Contractions evoked by NE ($ED_{50}$) were inhibited significantly by Fructus Aristolochiae in abdominal aorta and femoral artery. Fructus Aristolochiae inhibited the relaxation pretreated propranolol and L-NNA in femoral artery. But Fructus Aristolochiae did not effect the relaxation pretreated ODQ in femoral artery and abdominal aorta. These results indicate that Fructus Aristolochiae can relax NE induced contraction of rabbit blood vessel selectively, and that this relaxation relates to nitric oxide synthesis and sympathetic action.

수종 한약재가 혈관에 미치는 영향 (Effects of Several Herbs on the Blood Vessel)

  • Rhizoma Arisaematis, Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Folium Perillae, Radix Sophorae Subprostratae, Radixi, Radix Ledeboutriellae, Rhizoma Atractylodis, Herba Ephedrae, Radix Puerariae and Radi Aconitx Bupleuri have been used in Korea for many centuries as a treatment for various disease. Contractions evoked by NE $(ED_{50})$ were inhibited significantly by Radix Sophorae Subprostratae, Radix Aconiti and Herba Ephedrae in renal artery. These results indicate that each herb can relax NE induced contraction of rabbit and pig blood vessel selectively, and that this relaxation relates to Gui-Gyung(歸經).

Effects of Ginsenoside Rg3 Epimers on Swine Coronary Artery Contractions

  • The previous reports demonstrated that ginseng saponins, active ingredient of Panax ginseng, inhibited blood vessel contraction induced by various hormones or high $K^+$. Recently, we demonstrated that 20(R)- and 20(S)-ginsenoside $Rg_3$. regulate ion channel activities with differential manners. The aim of this study was to examine whether ginsenoside $Rg_3$ isomers also show differential effects on swine coronary artery contractionresponses induced by high $K^+$, serotonin (5-HT) or acetylcholine. Treatment of 20(S)- but not 20(R)-ginsenoside $Rg_3$ caused a concentration-dependent relaxation of coronary artery contracted by 25mM KCI. 20(S)- and 20(R)-ginsenoside $Rg_3$ induced significant relaxations of coronary artery contraction induced by 5-HT $(3{\mu}M)$ in the presence of endothelium with concentration-dependent manner and, also in the absence of endothelium only 20(S)-ginsenoside $Rg_3$ induced a strong Inhibition of coronary artery contraction induced by 5-HT in a concentration-dependent manner. 20(S)-ginsenoside $Rg_3$ caused relaxation of coronary artery in the absence and presence of endothelium. In contrast, treatment of 20(S)- and 20(R)-ginsenoside $Rg_3\;(100{\mu}M)$ did not show significant inhibition of coronary artery contraction induced by acetylcholine $(0.01\;to\;30{\mu}M)$ in the presence of endothelium, whereas both isomers caused significant inhibition of coronary artery contraction induced by acetylcholine $(0.01\;to\;30{\mu}M)$ in the absence of endothelium in a concentration-dependent manner. These findings indicate that 20(S)-or 20(R)-ginsenoside $Rg_3$ exhibits differential relaxation eff3cts of swine coronary artery contractions caused by high $K^+$, acetylcholine, and 5-HT treatment and that this differential vasorelaxing effects of ginsenoside $Rg_3$ isomers also might be dependent on endothelium.

합성제어기를 이용한 혈압 시뮬레이터의 구현 (Blood pressure simulator using hybrid controller)

  • A hybridized simulator for generating blood pressure waveform is proposed to study the remedy and/or evaluation of the conventional sphygmomanometer utilizing the oscillometric method which is widely applied. The blood pressure of a flowing fluid was controlled for the blood vessel's condition caused by a rhythmical and periodical contraction/relaxation because of the special excitatory and conductive system of the heart. In this study, a hybridized controller composed of the PI feedback controller and the feedforward controller. The inverse dynamics function is proposed to operating the control valve while the pressure is applied in an oil pressure tank. The proposed hybrid simulator reproducing the blood pressure waveform in an artificial blood vessel has kept the control performance consistent over all range. Based on these results, the proposed simulators could be applied to the development and compensation of the non invasive sphygmomanometer type as well as to study the characteristics of the blood pressure and blood vessel.
