Derivation of Dynamic Characteristic Values for Multi-degree-of-freedom Frame Structures based on Frequency Response Function(FRF) (주파수응답함수 기반 다자유도 골조 구조물의 동특성치 도출 및 구조모델링 적용 )

  • In the seismic design of structures, seismic forces are calculated based on structural models and analysis. In order to accurately address the dynamic characteristics of the actual structure in the structural model, calibration based on actual measurements is required. In this study, a 4-story frame test specimen was manufactured to simulate frame building, accelerometers were attached at each floor, and 1-axis shaking table test was performed. The natural period of the specimen was similar to that of the actual 4 story frame building, and the columns were designed to behave with double-curvature having the infinite stiffness of the horizontal members. To investigate the effects seismic waves characteristics, historical and artificial excitations with various frequencies and acceleration magnitudes were applied. The natural frequencies, damping ratios, and mode shapes were obtained using frequency response functions obtained from dynamic response signals, and the mode vector deviations according to the input seismic waves were verified using the Mode assurance criterion (MAC). In addition, the damping ratios obtained from the vibration tests were applied to the structural model, and the method with refined dynamic characteristics was validated by comparing the analysis results with the experimental data.

  • The outlook for Korea's consumer price inflation rate has a profound impact not only on the Bank of Korea's operation of the inflation target system but also on the overall economy, including the bond market and private consumption and investment. This study presents the prediction results of consumer price inflation in Korea for the next three years. To this end, first, model selection is performed based on the out-of-sample predictive power of autoregressive distributed lag (ADL) models, AR models, small-scale vector autoregressive (VAR) models, and large-scale VAR models. Since there are many potential predictors of inflation, a Bayesian variable selection technique was introduced for 12 macro variables, and a precise tuning process was performed to improve predictive power. In the case of the VAR model, the Minnesota prior distribution was applied to solve the dimensional curse problem. Looking at the results of long-term and short-term out-of-sample predictions for the last five years, the ADL model was generally superior to other competing models in both point and distribution prediction. As a result of forecasting through the combination of predictions from the above models, the inflation rate is expected to maintain the current level of around 2% until the second half of 2022, and is expected to drop to around 1% from the first half of 2023.

An Empirical Study on Bank Capital Channel and Risk-Taking Channel for Monetary Policy (통화정책의 은행자본경로와 위험추구경로에 대한 실증분석)

  • This study empirically analyzes whether bank capital channel and risk-taking channel for monetary policy work for domestic banks in South Korea by analyzing the impact of the expansionary monetary policy on the rate spread between deposit and loan, capital ratio, and loan amount. For the empirical analysis, the Uhlig (2005)'s sign-restricted SVAR(Structural Vector Auto-Regression) model is used. The empirical results are as follows: the bank's interest rate margin increases, the capital ratio improves, risk-weighted asset ratio increases, and the amount of loans increases in response to expansionary monetary shock. This empirical results confirm that bank capital channel and risk-taking channel work in domestic banks, similar to the previous research results. The implications of this study are as follows. Although the expansionary monetary policy has the effect of improving the bank's financial soundness and profitability in the short term as bank capital channel works, it could negatively affect the soundness of banks by encouraging banks to pursue risk in the long run as risk-taking channel works. It is necessary to note that the capital ratio according to the BIS minimum capital requirement of individual banks may cause an illusion in supervising the soundness of the bank. So, the bank's aggressive lending expansion may lead to an inherent weakness in the event of a crisis. Since the financial authority may have an illusion about the bank's financial soundness if the low interest rate persists, the authority needs to be actively interested in stress tests and concentration risk management in the pillar 2 of the BIS capital accord. In addition, since system risk may increase, it is necessary to conduct regular stress tests or preemptive monitoring of assets concentration risk.

  • Cytochrome P450 enzymes have been intensively investigated in hepatic tissues and several mammalian cell lines. Compared to most studies about cytochrome P450 isozymes in liver in vivo and hepatic, cell lines in vitro, the study of cytochrome P450IA1 in human breast cancer cells could be very important to understand the mechanism of the regulation of CYPIA1 gene expression and cell growth. MCF-7 human breast cancer cells are well characterized to study estrogen and antiestrogen action due to the fact that they contain high level of estrogen receptor and have biological markers characterized. And also MCF-7 cells express high level of arylhydrocarbon hydroxylase activity and human cytochrome P450IA1 cDNA was cloned from MCF-7 cells. Ah receptor was characterized in many breast cancer cell lines and polycyclic aromatic hydrocarbon such as 3-MC induced the expression of CYPIA1 gene and cytochrome P450- dependent monooxygenase activity. We undertook a study to examine the effect of estrogens and other chemicals on the regulation of human CYPIA1 gene expression in MCF-7 cells via RTPCR analysis, that might help us to understand the mechanism of the regulation of CYPIA1 gene expression and MCF-7 cell growth. Expression vector containing the functional 5'-regulatory region of human CYPIA1 fused to the CAT reporter gene was transfected into estrogen receptor positive MCF-T cells or estrogen receptor negative MDA-MB-231 cells. After these cells were treated with various chemicals, RTPCR was carried out to measure both CYPIA1 mRNA and CAT mRNA levels. 1nM 3-MC increased in both P450 and CAT mRNA levels over those of control by two folds in MCF-7 cells but does not in MDA-MB-231 cells. Estrogen or tamoxifen or retinoic acid or chrysin decreased in both P450 and CAT mRNA levels that were induced by 3-MC in MCF-7 when each chemical was administered with 3-MC concomitantly. These results suggested that the level of CYPIA1 gene expression is modulated with estrogen-related molecules and make it possible to speculate that ER is related to CYPIA1 gene expression and cell growth in breast cancer cells. Molecular Authentication and Phylogenetic Relationship of Bupleurum Species by the rDNA-ITS Sequences (rDNA-ITS 염기서열 분석을 통한 시호 종 감별용 유전자 마커 개발 및 유연관계 분석)

  • Objectives : Bupleuri Radix (Siho) is prescribed as the root of different Bupleurum species on the pharmarcopoeia in Korea and China. Moreover, other species and varieties of the genus Bupleurum have been also distributed on the herbal market as Bupleuri Radix. However, due to the morphological similarity and frequent occurrence of intermediate forms, the correct identification of this radix is very difficult. To develop a reliable method for correct identification and improving the quality standards of official Bupleuri Radix, we analyzed sequences of the ribosomal RNA gene and internal transcribed spacer (rDNA-ITS) region. Methods : PCR amplification of rDNA-ITS region was performed using ITS1 and ITS4 primer from 6 Bupleurum species and 1 variety, B. falcatum L. (Siho), an improved breed of B. falcatum L. (Samdo-Siho), B. chinense DC. (Buk-Siho), B. scorzonerifolium Willd. (Nam-Siho), B. longiadiatum Turcz. (Gae-Siho), B. euphorbiodes Nakai (Deungdae-Siho) and B. latissimum Nakai (Seom-Siho), and nucleotide sequence was determined after sub-cloning into the pGEM-Teasy vector. Authentic marker nucleotides were estimated by the analysis of ClastalW using entire rDNA-ITS sequence of three samples per species. Results : In comparative analysis of the rDNA-ITS sequences, we found specific nucleotides to distinguish Korean (B. falcatum L. and its variety) and Chinese official species (B. chinense DC. and B. scorzonerifolium Willd.) from others at positions 411 and 447, and positions 89, 101, 415 and 599, respectively. Futhermore, we also found nucleotide indels (insertion and/or deletion) and substitutions to identify each of different Bupleurum species, 2 positions for B. falcatum L. and its variety, 6 positions for B. chinense DC., 49 positions for B. scorzonerifolium Willd., 8 positions for B. euphorbioides Nakai, 7 positions for B. longiradiatum Nakai and 9 positions for B. latissimum Nakai. These sequence differences at corresponding positions are avaliable nucleotide markers to determine the botanical origins of Bupleuri Radix. Moreover, we confirmed the phylogenetic relationship of B. latissimum Nakai, a Korean endemic speices, among Bupleurum species based on the rDNA-ITS sequence. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by the providing of definitive information that can identify each plant species and distinguish it from unauthentic adulterant Bupleurum species.

Phylogenetic Analysis of Cucurbit Chlorotic Yellows Virus from Melon in 2020 in Chungbuk, Korea (2020년 충북지역 멜론에서 발생한 Cucurbit Chlorotic Yellows Virus의 계통분석)

  • Cucurbit chlorotic yellows virus (CCYV) is a plant virus that causes damage to cucurbit crops such as watermelon and cucumber, and is transmitted by an insect vector known as the whitefly. Since CCYV was first detected on cucumber in Chungbuk in 2018, it has been reported in other areas including Gyeongsang in Korea. In 2020, we performed field surveys of yellowing diseases in the greenhouses growing melon and watermelon in Chungbuk (Jincheon and Eumseong). Reverse transcription-polymerase chain reaction analysis of 79 collected samples including melon, watermelon, and weeds resulted in detection of CCYV in 4 samples: Three samples were singly infected with CCYV and one samples was mixed infected with CCYV, Cucurbit aphid borne yellows virus, and Watermelon mosaic virus. The complete genome sequences of the four collected CCYV melon isolates (ES 1-ES 4) were determined and genetically compared with those of previously reported CCYV isolates retrieved from GenBank. Phylogenetic analyses of RNA 1 and 2 sequences revealed that four ES isolates were clustered in one group and closely related to the CCYV isolates from China. The analysis also revealed very low genetic diversity among the CCYV ES isolates. In general, CCYV isolates showed little genetic diversity, regardless of host or geographic origins. CCYV has the potential to pose a serious threat to melon, watermelon, and cucumber production in Korea. Further studies are needed to examine the pathogenicity and transmissibility of CCYV in weeds and other cucurbits including watermelon.

Development of a High-Performance Concrete Compressive-Strength Prediction Model Using an Ensemble Machine-Learning Method Based on Bagging and Stacking (배깅 및 스태킹 기반 앙상블 기계학습법을 이용한 고성능 콘크리트 압축강도 예측모델 개발)

  • Predicting the compressive strength of high-performance concrete (HPC) is challenging because of the use of additional cementitious materials; thus, the development of improved predictive models is essential. The purpose of this study was to develop an HPC compressive-strength prediction model using an ensemble machine-learning method of combined bagging and stacking techniques. The result is a new ensemble technique that integrates the existing ensemble methods of bagging and stacking to solve the problems of a single machine-learning model and improve the prediction performance of the model. The nonlinear regression, support vector machine, artificial neural network, and Gaussian process regression approaches were used as single machine-learning methods and bagging and stacking techniques as ensemble machine-learning methods. As a result, the model of the proposed method showed improved accuracy results compared with single machine-learning models, an individual bagging technique model, and a stacking technique model. This was confirmed through a comparison of four representative performance indicators, verifying the effectiveness of the method.

3D Film Image Inspection Based on the Width of Optimized Height of Histogram (히스토그램의 최적 높이의 폭에 기반한 3차원 필름 영상 검사)

  • In order to classify 3D film images as right or wrong, it is necessary to detect the pattern in a 3D film image. However, if the contrast of the pixels in the 3D film image is low, it is not easy to classify as the right and wrong 3D film images because the pattern in the image might not be clear. In this paper, we propose a method of classifying 3D film images as right or wrong by comparing the width at a specific frequency of each histogram after obtaining the histogram. Since, it is classified using the width of the histogram, the analysis process is not complicated. From the experiment, the histograms of right and wrong 3D film images were distinctly different, and the proposed algorithm reflects these features, and showed that all 3D film images were accurately classified at a specific frequency of the histogram. The performance of the proposed algorithm was verified to be the best through the comparison test with the other methods such as image subtraction, otsu thresholding, canny edge detection, morphological geodesic active contour, and support vector machines, and it was shown that excellent classification accuracy could be obtained without detecting the patterns in 3D film images.

  • With the recent development of IT technology, research and interest in various biosignal measuring devices is increasing. As an aging society is in full swing, research on the elderly population using IT-related technologies is continuously developing. This study is about the development of life pattern detection and fall detection algorithm, which is one of the medical service areas for the elderly, who are rapidly developing as they enter a super-aged society. This study consisted of a system using a 3-axis accelerometer and an electrocardiogram sensor, collected data, and then analyzed the data. It was confirmed that behavioral patterns could be classified from the actual research results. In order to evaluate the usefulness of the human activity monitoring system implemented in this study, experiments were performed under various conditions, such as changes in posture and walking speed, and signal magnitude range and signal vector magnitude parameters reflecting the acceleration of gravity of the human body and the degree of human activity. was extracted. And the possibility of discrimination according to the condition of the subject was examined by these parameter values.

Development of SCAR Markers for the Discrimination of Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma based on the RAPD (RAPD 분석을 통한 대황(大黃)과 종대황(種大黃) 감별용 SCAR 유전자 마커 개발)

  • Objectives : Due to the morphological similarity and frequent occurrence of intermediate forms as well as morphological variations of aerial part, the correct identification between Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma is very difficult. To develop a reliable method for correct identification and improving the quality standards of Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma, we analyzed RAPD and developed SCAR marker. Methods : To amplify target DNA at the genomic level, 32 Operon 10-mer random primers were applied with four Rheum species, R. officinale, R. palmatum, R. tanguticum and R. undulatum. The nucleotide sequences were determined and species-specific primers were prepared depending on the species-specific RAPD amplicons after subcloned into the pGEM-Teasy vector. To develop the SCAR markers, species-specific PCR amplification and multiplex-PCR were carried out using the single species-specific primer pairs and combinations of them, respectively. Results : We used RAPD analysis of four Rheum plant species to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed two SCAR markers that amplified 314 bp and 390 bp DNA fragments in only R. undulatum but not in R. officinale, R. palmatum, R. tanguticum and R. undulatum, for distinguishing Rhei Undulatai Rhizoma and Rhei Radix et Rhizoma. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. Conclusions : These genetic markers can be used for the efficient discrimination of plants species and commercial herbal medicines between Rhei Undulatai Rhizoma and Rhei Radix et Rhizoma, to ultimately prevent indiscriminate distribution and prescription of these herbal medicines.
