Regeneration Studies in Grateloupia filicina (J.V. Lamouroux) C. Agardh - An Important Carrageenophyte and Edible Seaweed

  • Grateloupia filicina (J.V. Lamouroux) C. Agardh (Halymeniaceae, Cryptonemiales, Rhodophyta) is an edible seaweed as well as an important source of carrageenan. In the present study, attempt has been made to develop a suitable protocol for effective regeneration of the seaweed and the rapid multiplication of the desired varieties. The young upright thallus of G. filicina was grown in axenic culture using both solid and liquid media. The various media tested were f/2, Provasoli’s Enriched Seawater (PES) and Enriched Seawater (ESW). The effect of glycerol (as a carbon source) and various plant growth regulators i.e. auxin (NAA) and cytokinins (Kinetin and BA) were tested. Although, regeneration of young thalli was observed from the cut ends in all the media, better growth was found in f/2, PES, f/2 (0.5% Glycerol), f/2 (NAA ${10^{-5}}_M)\;and\;f/2\;(BA\;{10^{-6}}_M$). On the other hand callusing was observed only in solid media supplemented with low concentration of Glycerol (0.5%) in f/2, NAA ${10^{-5}}_M\;in\;f/2,\;PES\;and\;BA\;{10^{-5}}_M$ in f/2. Young thalli were developed from the callus sub culture after 40 days of inoculation.

시호 모상근 배양에서 생장과 시호 사포닌 생합성에 미치는 배지와 IAA 및 IBA의 영향 (Effects of IAA, IBA, and Media on Growth and Saikosaponin Biosynthesis in Bupleurum falcatum Hairy Root Culture)

  • 시호 모상근 (BFHR2 클론) 배양에서 생장과 saikosaponin 생산에 미치는 기본배지, 배지의 다량무기염농도 변화 및 IAA와 IBA의 효과를 조사하였다. 조사한 배양액 중 RCM 배지에 다량무기염농도를 2배로 증가시킨 배지에서 생장이 가장 양호하였다. 또한 2RCM배지에서는 saikosaponin인 a, c와 d의 생성을 확인하였으며, 반면에 MS배지에서는 saikosaponin의 생성이 극히 저조하였다. IBA는 0.5mg/L에서 약 60%의 생장촉진효과를 보였으나, IAA와 IBA두 가지 모두 0.01~5mg/L 까지의 농도에서 뿌리형태의 외적인 변화 없이 saikosaponin 생성을 강하게 억제하는 작용을 나타내었다.

Studies on Conditioned Media in Human Cells: Evaluation Using Various Cell and Culture Conditions, Animal Disease Models

  • In the last several decades, cell therapy research has increased worldwide. Many studies have been conducted on cell therapy, and have revealed that transplanted cells did not survive for long, and implanted cells remained inactive causing immune rejection depending on the patient's condition. Therefore, studies on cell-free therapy need to be conducted. To overcome these limitations, an alternative is the use of supernatant from cells, called "conditioned media (CM)." During in vitro cell culture, culture media supply nutrients to maintain cell characteristics and viability. In the culture, cells not only consume nutrients but also release beneficial proteins and substances, which are called "secretome." CM from cells can be stored for a long time and is easy to handle. Moreover, secretome in CM can also be measured; exact amount of secretome is important to set the standard value for disease treatment. Here, we reviewed studies on CM and confirmed that various secretomes from CM were identified in these studies. Moreover, these findings could benefit cell and animal studies in future. In conclusion, CM could be a potential candidate for an alternative to cell therapy.

DPF의 PM 포집효율 예측을 위한 다양한 다공성 매질 유동장 모델 해석 (A Detailed Examination of Various Porous Media Flow Models for Collection Efficiency and Pressure Drop of Diesel Particulate Filter)

  • In the present study a detailed examination of various porous media models for predicting filtration efficiency and pressure drop of diesel particulate filter (DPF), such as sphere-in-cell and constricted tube models, are attempted. In order for demonstrating their validities of correct estimation on permeability, geometry of property configurations common in commercial cordierite DPFs are correlated to the porous media flow models, and validations of predicted filtration efficiencies due to the use of different unit collectors are made with experiments. The result shows that the porosity, pore size and permeability of cordierite DPF can be successfully correlated by Kuwabara flow field with correction factor of 0.6. The unit collector efficiency predicted by sphere-in-cell model agrees very well with measurements in accumulation mode, whereas that by constricted tube model with significant prediction error.

A Study on Radiation Risk Recognition Aided System Visualizing Risk Information by CG

  • The technology of Computer Graphics (CG) has been in great progress for almost 20 years and has proven to be a valuable tool for a broad variety of fields, including nuclear engineering. To work in any hazardous environment for example radiation field is particularly challenging because the danger is not always visually apparent. In this study as the application of CG to nuclear engineering field, we proposed to develop a radiation risk recognition aided system in which various radiation information; radiation risks, radiation distribution, hazard information and so on, were visualized by CG. The system used the server and client system. In the server there were two parts; one (main-server) was the database part having various data and the other (sub-server) was the visualization part visualizing the human phantom by POV-Ray. In the client there was the input and output part. The outputs from the system were various radiation information represented by coloring, circle graph and line graph intuitionally. The system is useful for a broad range of activities including radiation protection, radiation management, dose minimization, and demonstration to the public.

트랜스미디어 스토리텔링의 개념과 특성 (Concept and Properties of Transmedia Storytelling)

  • 트랜스미디어 스토리텔링은 다양한 미디어 플랫폼에 기반한 다수의 콘텐츠가 하나의 세계를 구현하는 것으로, 디지털 컨버전스 시대에 새롭게 나타난 양식을 말한다. 하나의 세계를 창조해 내기 위해 각기 다른 미디어가 개별적인 세계를 유기적으로 창출하여 다양한 의미를 생산해 내는 트랜스미디어 스토리텔링은 창작자의 영역을 넘어 소비자의 영역에까지 변화를 일으킨다. 트랜스미디어 스토리텔링의 첫 번째 특성은 창작의 공동 제작과 향유의 집단 지성이다. 두 번째 특성은 교차와 결합을 통한 스토리 분화로 소비자에게 새로운 경험과 통찰을 제시하는 것이다. 마지막 특성으로는 세계 구축을 위한 관계 맺기를 통해 콘텐츠가 수렴되고 발산되는 과정을 들 수 있다. 트랜스미디어 스토리텔링은 창작자와 소비자들이 만나는 여러 지점을 통해 모아지고 전개되는 것이다. 이러한 트랜스미디어 스토리텔링의 특성은 다양한 미디어 플랫폼을 통해 구축되는 트랜스미디어 콘텐츠를 분석하는 기반으로 작용할 것이다.

  • Multimedia systems present information by various media, for example, video, sound, music, animation, movie, etc., in addition to the text which has long been used for conveying the information. Among many multimedia applications, the multimedia information retrieval systems commercialized in the forms of multimedia encyclopedia CD-ROMs, benefited from various media for their ability to present information in an efficient and complete way. But using several media, on the other hand, may cause end users' confusion and furthermore, poorly designed user interface often exacerbates the situation. In this study, the multimedia systems were studied from the standpoint of usability. The conceptual framework of the user interface of the multimedia system was newly defined. And 100 initial variables for user interface design of general multimedia systems were suggested through literature survey and expert opinions based upon the framework developed. Among various application areas, the multimedia information retrieval systems were chosen for investigation, and 36 variables particularly relevant to user interface of the multimedia information retrieval systems were selected. According to the sequential research strategy, the variables that were considered to be most important were finally selected through a screening stage. A part of selected variables were verified through a human factors experiment as the first step of sequential research. Based upon the result of the experiment, guidelines for user interface design were provided. For future study, the variables remained will be Investigated and the study will expand to another application areas.

폐펄라이트와 폐암면을 재활용한 혼합배지가 포인세티아 'Freedom Red'의 생육에 미치는 영향 (Effect of Various Mixtures of Used Perlite and Rockwool Slabs on Growth of Euphorbia pulcherrima 'Freedom Red' in a Mat Subirrigation System)

  • 본 연구는 배지 구성물질로서 폐펄라이트와 폐암면 입자를 함유한 다양한 생육배지에서 포인세티아의 생산 가능성을 구명하기 위하여 수행되었다. 두 가지 폐배지를 위주로 1:1과 1:3을 포함한 다양한 부피비로 혼합한 10가지 조합으로 실험하였으며, 25% 폐펄라이트:75% 피트모스의 혼합배지를 대조구로 하였다. 초장, 근장, 최대 가지 길이, 뿌리수, 엽면적, 포엽장과 포엽폭은 폐펄라이트를 함유한 배지에서 대조구에 비해 큰 유의차를 보였다. 하지만 경경과 포엽수는 차이가 없었다. 본 결과로 폐펄라이트와 폐배지의 포인세티아 식물의 재배 시 재활용의 가능성이 입증되었다.

Rhodococcus sp. TK6가 생산하는 Cyclohexanol Dehydrogenase의 동위효소 (Cyclohexanol Dehydrogenase isozymes produced by Rhodococcus sp. TK6)

  • TK6 was able to produce NAD+ dependent cyclohexanol dehydrogenase(CDH). The production of CDH was increased rapidly at the logarithmic phase and maintained constantly after that. In order to investigate the inductive production of CDH by various substrates, the bacteria were grown in the media containing alicyclic hydrocarbons and various alcohols as a sole crabon souce. CDH was induced most actively by cyclohexanol. Cyclohexanone and cyclohexane-1,2-diol also induced remarkable amount of CDH but it was induced weakly by 1-propanol, 1-butanol, 1-pentanol, 1-hexanol, 2-propanol, and 2-methyl-1-propanol. The dehydrogenase of the bacteria grown in the media containing cyclohexanol were weakly active for various alcohols, but the dehydrogenase activity for cyclohexane-1,2-diol was twice as much as that for cyclohexanol. Activity staining on PAGE of the cell free extract of Rhodococcus sp. TK6 grown in the media containing cyclohexanol reveals at least sever isozyme bands of CDH and we nominated the four major activity bands as CDH I, II, III, and IV. CDH I was strongly induced by cyclohexanol, cyclohexane-1,2-diok, but its activity was specific to cyclohexane-1,2-diol and 1-pentanol. CDH IV was strongly induced by cyclohexanol and cyclohexane-1,2-diol, and its activity was very specific to cyclohexane-1,2-diol.

인스타그램에 나타난 멀티 페르소나 패션이미지에 관한 연구 - "부캐" 사례를 중심으로 - (A study on multi-persona fashion images in Instagram - Focusing on the case of "secondary-characters" -)

  • The aim of this study was to analyze the semantic network structure of keywords and the visual composition of images extracted from Instagram in relation to the multi-persona phenomenon with in fashion imagery, which has recently been attracting attention. To this end, the concept of a 'secondary character', which forms a separate identity from a 'main character' on various social media platforms as well as on the airwaves, was considered as the spread of multi-persona and #SecondaryCharacter on Instagram was investigated. 3,801 keywords were collected after crawling the data using Python and morphological analysis was undertaken using KoNLP. The semantic network structure was then examined by conducting a CONCOR analysis using UCINET and Netdraw to determine the top 50 keywords. The results were then classified into a total of 6 clusters. In accordance with the meaning and context of the keywords included in each cluster, group names were assigned : virtual characters, relationship with the main character, hobbies, daily record, N-job person, media and marketing. Image analysis considered the technical, compositional, and social styles of the media based on Gillian Rose's visual analysis method. The results determined that Instagram uses fashion images that virtualize one's face to produce multi-persona representation s that show various occupations, describe different types of hobbies, and depict situations pertaining to various social roles.
