Histopathologic Studies on the Brain and Lymphoid Organs in Hog Cholera III. Encephalitis of Pigs and Rabbits (Hog Cholera 병돈(病豚)의 뇌(腦) 및 임파장기(淋巴臟器)에 관한 병리조직학적(病理組織學的) 연구(硏究) III. 뇌염소견(腦炎所見)에 대하여)

  • This study was undertaken to clarify the histopathological changes of pigs naturally infected with hog cholera. Microscopic observations of the brain as well as clinical observations were carried out for the naturally and experimentally infected pigs with hog cholera viruses. Electron microscopically the vascular cuffings of the brain were also observed in the experiment. In addition, clinical and pathological studios were carried out in the rabbits inoculated with ALD, lapinized and isolated strain of hog cholera virus, respectively. The results obtained are as follows; Vascular cuffing of the brain was observed in about 97% of the natural cases and all of the experimental cases. Among the 496 cuffed blood vessels of natural cases, intramural cuffing (82.9%), intramural and perivascular cuffing (11.9%) and perivascular cuffing (5.2%) were seen, respectively. Electron microscopic findings on cuffed blood vessels of the brain were endothelial cellular degeneration, intramural separation and vacuolation, perivascular vacuolation and infiltration of pleomorphic lymphoid cells. In the rabbits dosed with tissue suspension of the lymphoid organs and isolated hog cholera virus, high body temperature and vascular cuffing of the brain were observed, meanwhile these changes were more significant in pre-injected cased with indian ink.

  • 1, 1-dichloroethylene (DCE) is well known hepatotoxicant as a model acute hepatotoxicity and selectively injure the bile canalicular membrane of centrilobular hepatocytes. In this study, we investigated hepatic gene expression and histopathological changes in response to DCE treatment. DCE was administered once daily at 20 mg/kg up to 14 days via intraperitoneal injection. Five mice were used in each test group and were sacrificed at 1, 7, and 14 days. Serum biochemical and histopathological analysis were performed for evaluation of hepatotoxicity level. Direct bilirubin and total bilirubin activities were slightly elevated in treated group at 7 days. DCE treatment for 7 days resulted in centrilobular hepatocyte hypertrophy and hepatocyte vacuolation, and mild hepatocyte vacuolation and high hepatocyte basophilia were observed in 14 days treated group. One hundred twenty three up-regulated genes and 445 down-regulated genes with over 2-fold changes between treated and control group at each time point were used for pathway analysis. These data may contribute in understanding the molecular mechanism DCE-induced hepatotoxicity.

  • A 7-year-old Maltese was presented to a veterinary clinic for a history of anorexia, soft feces, and anemia. During abdominal sonography and computed tomography imaging, splenic mass was suspected. Laparotomy revealed the bloody ascites, and the enlarged spleen containing a large spherical mass. Microscopically, the splenic parenchyma was replaced by neoplastic cells with cytoplasmic vacuolation and abundant myxoid matrix. Cellular vacuolation and myxoid matrix were positive to Oil-Red-O and Alcian-blue stains. The neoplastic cells were positive to vimentin on immunohistochemistry. The case was diagnosed as myxoid liposarcoma. Based on a poor prognosis, the dog was euthanized three weeks after splenectomy.

  • Purpose : To observe the histopathological changes in the pulp tissues of mandibular molars in streptozotocin-induced diabetic rats after irradiation. Materials and Methods : The male Sprague-Dawley rats weighing approximately 250 gm were divided into four groups: control, diabetes, irradiation, and diabetes-irradiation groups. Diabetes mellitus was induced in the rats by injecting streptozotocin. Rats in control and irradiation groups were injected with citrate buffer only. After 5 days, the head and neck region of the rats in irradiation and diabetes-irradiation groups were irradiated with a single absorbed dose of 10 Gy. All the rats were sacrificed at 3, 7, 14, 21, and 28 days after irradiation. The specimen including the mandibular molars were sectioned and observed using a histopathological method. Results : In the diabetes group, capillary dilatation was observed. However, there was no obvious morphologic alteration of the odontoblasts. In the irradiation group, generalized necrosis of the dental pulp tissues was observed. Vacuolation of the odontoblasts and dilatation of the capillaries were noted in the early experimental phases. In the diabetes-irradiation group, generalized degeneration of the dental pulp tissues was observed. Vacuolation of the dental pulp cells and the odontoblasts was noted in the late experimental phases. Conclusion : This experiment suggests that dilatation of the capillaries in the dental pulp tissues is induced by diabetic state, and generalized degeneration of the dental pulp tissues is induced by irradiation of the diabetic group.

  • To elucidate the effect of temperature on the fruit body growth and structure of pleurotus ostreatus cultivar Chunchu No.2, microstructures were observed with scanning- and transmission electron microscope. Pileocystidia were well developed on the surface of pileus in the fruit body cultivated at $7^{\circ}C$. As the increasing temperature for fruit body development, thickness of pileus, thickness of stipes and length of stipes shown thin, thick and long, respectively. Color of pileus was also changes to whitish grown under high temperature. Physical structure shown as hardness and gumminess of stipes grown at high temperature, fruit body were soften than that of low temperature. Microstructures of fruit body grown at high temperature shown fast-discharge of basidiospore and sytoms aging. vacuolation in the cell observed at mycelium grown under high temperature. Optimum temperature for vegetative growth of fruit bodies were $15^{\circ}C$, and shown regular arrangement of mycelium on the surface of stipes.

  • The purpose of this study was to evaluate the effect of a subsequent infection of PCV2 on piglets with PEDV. In clinical signs, the signs observed in dual-infected with PEDV and PCV2 piglets and alone infected with PEDV piglets ranged from diarrhoea to vomiting and dehydration. Dual-infected piglets developed signs of anorexia, vomiting and watery diarrhoea within 12 hpi. Nevertheless alone -infected piglets caused pasty diarrhea at first. In mortality, dual infections showed 25%, but alone -infections showed 8.3%, respectively. In gross findings, piglets dual-infected with PEDV and PCV2 appeared the severe findings of congestion, distension of lumen, milder curdes of undigested milk in stomach than those of single-infected piglets. In histopathological findings, piglets of dual-infection group appeared the more severe findings of villous atrophy and fusion, congesion, exfoliation, vacuolation, squamation, loss of cilia and proliferation of crypt. Significant (P<0.05) decrease in VH:CD ratio in dually infected piglets compared to piglets from alone-PEDV infections. In immunohistochemical findings, strong hybridization signals in dual-infected piglets observed moderate to severe villous atrophy or vacuolation with positive cells arranged continuously over the villi. In the lumen, exfoliated enterocytes were strongly positive in dual-infected piglets. A number of PEDV-positive cells in dual-infected pigs were significantly higher than that in alone PEDV-infected piglets.

Histopathological Observations on the Renal Injury in Rats Administered with Enrofloxacin and Oxalate (Oxalate와 Enrofloxacin 투여한 랫트신장에 대한 병리조직학적관찰)

  • To investigate the renal effects of enrofloxacin administration on rats induced with dehydration or hyperoxaluria, male rats were treated with enrofloxacin of 50 mg to 500 mg/kg b.w.. The microscopical observations of kidney and urine sediment were carried out in the experimental groups. The result obtained were as follows; The male rats deprived of water for 72 hours and administered with enrofloxacin. As enrofloxacin administration dose was increased, clinical signs such as loss of appetite, depression, weakness, and loss of urine output became more severe. In the histopathological findings, there were hyperemia and hemorrhage in renal cortex, vacuolation and necrosis of renal tubular epithelia, proteinous casts within renal tubules. The male rats were orally administered with sodium oxalate and injected with enrofloxacin for 7days. As enrofloxacin administration dose was increased, clinical signs such as the loss of appetite and water consumption, and weakness became more severe. In the histopathological findings, there are hemorrhage of glomeruli and cortical hyperemia, vacuolation and necrosis of tubular epithelia, proteinous casts in renal tubules. In the microscopical findings of urine sediment, there are calcium oxalate crystal (diamond-like type) and magnesium ammonium phosphate crystals (rhomboid). The male rats were intraperitoneally injected with sodium oxalate and administered with enrofloxacin for 7days. As enrofloxacin administration dose was increased, clinical signs such as the loss of appetite and water consumption, weakness were more severe. In the histopathological findings, there were hyperemia and hemorrhage in both glomeruli and renal cortex. Severe necrosis of renal tubular epithelia, bluish materials within renal tubules were also found. In the microscopical findings of urine sediment, there were many calcium oxalate crystals. The present results suggest that enrofloxacin has some injurious effects in rats having dehydration or hyperoxaluria, and clinically, we should consider these renal injury effects when we use enrofloxacin in patients accompanied renal disease, dehydration and hyperoxaluria conditions.

Prevalence and pathologic study of porcine epidemic diarrhea in Jeju (제주도의 돼지유행성설사 발생 상황과 병리학적 고찰)

  • From April 2014 to September 2015, 153 piglets from 52 farms in Jeju were diagnosed with porcine epidemic diarrhea (PED). The major PED cases were focused on suckling piglets (144 piglets, 94.1%), particularly in 1-7-day-old piglets. Histopathologically, severe villous atrophy was observed in the small intestine, especially in the jejunum and ileum. The mean villous height to crypt depth ratios of the jejunum and ileum were 1.4:1 and 1.5:1, respectively. The major histopathologic findings of the small intestine were cytoplasmic vacuolation, cuboidalization, squamation, and exfoliation of the mucosal enterocytes in the villi. The cytoplasmic vacuolations in the enterocytes were the most prevalent lesions in the small intestine and were more severe in the ileum than in the jejunum. According to immunohistochemistry methods, the PED virus (PEDV) antigens were presented in the cytoplasms of the enterocytes, and were distributed more prevalently in the ileum than in the jejunum. PEDV antigens were also detected in the colon of 26 piglets (19.5%). Sequence comparison and phylogenetic analysis indicated that 12 PEDV had more than a 98.9% homology with each other. These PEDV strains were highly homologous with the genogroup 2 North American group.

  • Ultrastructural changes of tissues caused by Bacillus thuringiensis var. kurstaki $\delta$-endotoxin intoxication of Hyphantria cunea were observed by transmission electron microscopy. Bt $\delta$-endotoxin crystals induced the disruption of microvilli, vacuolation of cytoplasm, changes in the cisternae of the endoplasmic reticulum, disappearance of basal striations, loss of ribosomes, and changes in the configurations of mitochondria in the columnar cell of midgut. The fat body cells also showed spherical endoplasmic reticulum and distorted mitochondria, and then the cells were destroyed.
