Effect of autogenous Escherichia coli vaccine in pig (돼지에서 대장균 자가백신 효과)

  • This study was performed to investigate the immunogenicity of autogenous E coli vaccines and their preventive effects on diarrhea in suckling piglets. Autogenous E coli live and killed vaccines were made from the E coli strains isolated from piglets showing diarrhea in field. In group I, pregnant sows were administered with live and killed vaccines at 4 and 2 weeks before parturition, respectively, Killed vaccines were administered twice to pregnant sows at 4 and 2 weeks before parturition in group II, and saline instead of autogenous E coli vaccines was administered to pregnant sows in group III for the control. After parturition, antibody titers in colostrum and milk from sows, incidence of diarrhea in suckling piglets, and immunoreactivity in the ileum of piglets from each treatment group were examined. The results were as follows ; 1. Sixty-two strains of E coli were isolated from suckling piglets with diarrhea. Of the strains, K88 pilus and K99 pilus antigens were identified in 6(9.8%) and 4(6.5%), respectively. Molecular weights of K88 and K99 pilus were 27,500 and 18,500 daltons, respectively. 2. Antibody titers in colostrum from sows after parturition were 1 : 512 to 1 : 1,024 in group I, 1.256 to 1.512 in group II, and 1 : 4 to 1 : 16 in group III. 3. The incidences of diarrhea In suckling piglets of group I, II and III were 3.3%, 9.4% and 21.4%, respectively. 4. When the immunoreactivity in the ileum of piglets from each group was examined, the proportion of IgG-immunoreactivity cells in group I or II was higher than that in group III. In conclusion, administration of autogenous E coli vaccines to pregnant sows before parturition can be an effective way to prevent diarrhea in suckling piglets.

Development of antibodies after foot and mouth disease vaccination in pigs (돼지에서 구제역 백신 접종 후 항체가 변화 조사)

  • Three serotypes (O+A+Asia1 type) of the foot-and-mouth disease (FMD) vaccine were injected into cloven-hoofed animals in Korea after the nationwide spread of FMD at the end of 2010. This study was conducted to investigate FMD antibody development after FMD vaccination, and to determine whether there was a significant correlation between the antibody titer of the sow and the antibody titer of the growing pigs. The antibody titer (percentage inhibition [PI] titer) of the sow (gilt) after FMD vaccination was maintained at a level higher than 50 (P<0.05) for 5 months. A higher PI titer for the 1-month-old growing pigs corresponded with greater inhibition of the PI titer of the vaccinated growing pigs (P<0.05). A negative correlation (P<0.05) between the PI titer of the 1-month-old growing pigs and the PI titer of 3-month-old growing pigs, 4-month-old growing pigs after FMD vaccination at 2 months, 3months was identified, with a coefficient of determination ($R^2$) of 0.274. Thus the PI titer of the growing pigs was inhibited to a greater degree when vaccination was performed at 2 months of age than at 3 months. However, many other factors likely influence growing pigs' PI titer in addition to the PI titer of the sow and age at vaccination, given that the coefficient of determination was somewhat lower.

Immunologic effects of somatic antigens of house dust mite (Dermatophagoides pteronyssinus) against canine sarcoptic mite (Sarcoptes scabiei var. canis) infestation (집먼지진드기 체항원을 이용한 개 옴 감염증에 대한 면역효과)

  • Canine sarcoptic mite (Sarcoptes scabiei var. canis) burrow usually in the stratum corneum of the skin of dogs and rabbits. Antigens from the burrowing mites induce cutaneous inflammatory reaction and humoral and cell-mediated immune response in the host. The effect of immunization induced by somatic antigens of house dust mite (Dermatophagoides spp.) has been evaluated to control the canine sarcoptic mite in this experiment. Twelve common antigens (187, 142, 126, 120, 109, 92, 80, 68, 51, 30, 25, 17 kDa) were found using SDS-PAGE with silver staining and Western blot between canine sarcoptic mite and house dust mite. In order to evaluate the immunologic effect of these common antigens 10 New Zealand white rabbits were divided as 4 groups such as negative control (group I), positive challenged control (group II), vaccinated (group III), and vaccinated-challenged (group IV) groups. Group II was artificially infested with about 1,000 canine sarcoptic mites and group III and IV were immunized with somatic antigens of house dust mite. In addition group IV was artificially infested with about 1,000 canine sarcoptic mites and group II, IV were treated with ivermectin. At the 8 weeks of the vaccination with common antigen, the antibody titers of all groups of II, III and IV had been increased. Both infestation score and live canine sarcoptic mite counts of group IV were lower than group III. Infestation score of group II become 0 by 2 weeks and group IV by 4 weeks after infestation. These results suggest that house dust mite, which is easy to culture in vitro, can be a vaccine candidate for protection of canine sarcoptic mite infestation.

  • Major advances in positive-sense RNA virus research have been facilitated by the development of reverse genetics systems. These systems consist of an infectious cDNA clone that encompasses the genome of the virus in question. This clone is then used as a template for the subsequent synthesis of infectious RNA for the generation of synthetic viruses. However, the construction of infectious cDNA for the Japanese encephalitis virus (JEV) has been repeatedly thwarted by the instability of its cDNA. As JEV is an important human pathogen that causes permanent neuropsychiatric sequelae and even fatal disease, a reliable reverse genetics system for this virus is highly desirable. The availability of this tool would greatly and the development of effective vaccines as well as facilitate studies into the basic biology of the virus, including the molecular mechanisms of viral replication, neurovirulence, and pathogenesis. We have successfully constructed a genetically stable infectious JEV cDNA containing full-length viral RNA genome. Synthetic RNA transcripts generated in vitro from the cDNA were highly infectious upon transfection into susceptible cells, and the cDNA remained stable after it had been propagated in E. coli for 180 generations. Using this infectious JEV cDNA, we have successfully expressed a variety of reporter genes from the full-length genomic and various subgenomic RNAs in vitro transcribed from functional JEV cDNAS. In summary, we have developed a reverse genetics system for JEV that will greatly facilitate the research on this virus in a variety of different fields. It will also be useful as a heterologous gene expression vector and aid the development of a vaccine against JEV.

Pre-vaccination Prevalence and Genotype Distribution of Human Papillomavirus Infection among Women from Urban Tunis: a Cross-sectional Study

  • Background: To estimate the pre-vaccination distribution of human papillomavirus (HPV) types among women from urban Tunis. Materials and Methods: A total of 611 women aged 18-69 years were enrolled in three local gynaecological outpatient departments. All underwent a gynaecological examination with Pap test and dry swab for HPV detection and typing performed by linear array genotyping test (Roche). Cytological examination was conducted on conventional Pap smears. Results: HPV DNA was found in 6.5% of the women; the most frequent HPV types were HPV 16 and HPV 11 at 3.27% and 1.96%, respectively. The second most frequent high risk (HR) HPV type was HPV 58 (0.82%) followed by HPV 18, HPV 31 and HPV 33 found in only 0.33% of women. Single infections with HPV types, targeted by the quadrivalent vaccine (6, 11, 16, and 18), were detected in 3.6 % of the study patients (55% of positive women). HPV infection was found in 3.83% of women with normal cytology and in 47.4% of women with cytological abnormalities. No statistically significant trend in prevalence by age group emerged for any HPV type or for high or low risk types. Conclusions: These data show a relatively low prevalence of HPV infection in women from urban Tunis with a high proportion of HPV16 and HPV58. This should be considered in the upcoming screening programs and vaccination strategy.

Correlation of Microvessel Density with Nuclear Pleomorphism, Mitotic Count and Vascular Invasion in Breast and Prostate Cancers at Preclinical and Clinical Levels

  • Background: Tumor angiogenesis correlates with recurrence and appears to be a prognostic factor for both breast and prostate cancers. In the present study, we aimed to investigate the correlation of microvessel density (MVD), a measure of angiogenesis, with nuclear pleomorphism, mitotic count, and vascular invasion in breast and prostate cancers at preclinical and clinical levels. Methods: Samples from xenograft tumors of luminal B breast cancer and prostate adenocarcinoma, established by BT-474 and PC-3 cell lines, respectively, and commensurate human paraffin-embedded blocks were obtained. To determine MVD, specimens were immunostained for CD-34. Nuclear pleomorphism, mitotic count, and vascular invasion were determined using hematoxylin and eosin (H&E)-stained slides. Results: MVD showed significant correlations with nuclear pleomorphism (r=0.68, P=0.03) and vascular invasion (r=0.77, P=0.009) in breast cancer. In prostate cancer, MVD was significantly correlated with nuclear pleomorphism (r=0.75, P=0.013) and mitotic count (r=0.75, P=0.012). In the breast cancer xenograft model, a significant correlation was observed between MVD and vascular invasion (r=0.87, P=0.011). In the prostate cancer xenograft model, MVD was significantly correlated with all three parameters (nuclear pleomorphism, r=0.95, P=0.001; mitotic count, r=0.91, P=0.001; and vascular invasion, r=0.79, P=0.017; respectively). Conclusions: Our results demonstrate that MVD is correlated with nuclear pleomorphism, mitotic count, and vascular invasion at both preclinical and clinical levels. This study therefore supports the predictive value of MVD in breast and prostate cancers.

Enhanced CEA-specific Immune Responses by Tat-LLO Fusion Protein (Tat-LLO 융합 단백질에 의한 CEA 특이 항종양 면역 반응의 증가)

  • Background: Carcinoembryonic antigen (CEA) is well-known soluble tumor marker frequently detectable in peripheral blood of carcinoma patients and considered as good target for antigen-specific immunotherapy. However, it is known that the induction of immune response to CEA is very difficult because CEA is a self-antigen expressed in fetal cells and weakly expressed in normal colorectal epithelial cells. To enhance anti-tumor immunity specific for CEA, recombinant CEA protein was modified using listeriolysin O (LLO) for endosomal lysis and trans activator of transcription (Tat) domain for transducing extracellular proteins into cytoplasm. Methods: After immunization using dendritic cells pulsed with Tat-CEA, both Tat-CEA and LLO, and both Tat-CEA and Tat-LLO, antibody titer to CEA and LLO, cytotoxic T lymphocyte activity and the frequency of IFN-${\gamma}$ producing T lymphocytes were measured. Results: Immunization using DC pulsed with both Tat-CEA and Tat-LLO protein showed the increasement of production of CEA-specific antibody in serum, cytotoxic T lymphocyte activity, the frequency of IFN-${\gamma}$ secreting T cells, compared with DC pulsed with both Tat-CEA and LLO. Furthermore the ratio of CD8+T cell to $CD4^+$ cell among CEA-specific T cells was increased in group pulsed with both Tat-CEA and Tat-LLO. Conclusion: These results suggested that DC vaccine using Tat-LLO could be used for the development of effective immunotherapy for the treatment of tumor.

  • This research attempted to identify the factors associated with the intention of human papilloma virus vaccination among mothers of elementary school daughters. The subject of study is 132 mothers of elementary school students. Data collection was conducted from July 1 to August 30 in 2017, and analyzed using SPSS 23.0 Program. Results showed that 94.7% children of the participants had not been vaccinated, whose reasons were the low knowledge about HPV vaccination, concerns about its side effects, burden for cost. The attitude toward HPV vaccination appeared to be the most important predictor of vaccination, which was followed by perceived behavior control and subjective norm as meaningful predictors, as well. Based on these results, further researches should be conducted to develop educational programs, which are able to offer exact information about the effect and side effect of HPV vaccination, and verify the effectiveness.

  • The trial and success of malicious cyber attacks has been increased rapidly with spreading of Internet and the activation of a internet shopping mall and the supply of an online, or an offline internet, so it is expected to make a problem more and more. The goal of intrusion detection is to identify unauthorized use, misuse, and abuse of computer systems by both system insiders and external penetrators in real time. In fact, the general security system based on Internet couldn't cope with the attack properly, if ever. other regular systems have depended on common vaccine softwares to cope with the attack. But in this paper, we will use the positive selection and negative selection mechanism of T-cell, which is the biologically distributed autonomous system, to develop the self/nonself recognition algorithm and AIS (Artificial Immune System) that is easy to be concrete on the artificial system. For making it come true, we will apply AIS to the network environment, which is a computer security system.
