A report on 17 unrecorded bacterial species in Korea isolated from Lakes Soyang and Chungju in 2016

  • As a part of the research program 'Survey of freshwater organisms and specimen collection', freshwater samples were collected from Lakes Soyang and Chungju in 2016. Hundreds of bacterial strains were isolated from the samples and were identified based on 16S rRNA gene sequences. Among the bacterial isolates, strains showing higher than 98.7% sequence similarity with validly published bacterial species not reported in Korea were selected as unrecorded bacterial species. Based on 16S rRNA gene sequence similarity, 17 strains were identified as unrecorded bacterial species in Korea. The 17 bacterial strains were phylogenetically diverse and belonged to four phyla, seven classes, 13 orders, 14 families, and 16 genera. At generic level, the unreported species were affiliated with Caulobacter, Paracoccus, and Mesorhizobium of the class Alphaproteobacteria, Deefgea, Undibacterium, Chitinimonas, Inhella, and Sphaerotilus of the class Betaproteobacteria, Vibrio and Cellvibrio of the class Gammaproteobacteria, Sanguibacter and Clavibacter of the phylum Actinobacteria, Lactococcus of the phylum Firmicutes, Deinococcus of the class Deinococci, and Chryseobacterium and Flavobacterium of the phylum Bacteroidetes. The unreported species were further characterized by examining Gram reaction, colony and cell morphology, biochemical properties, and phylogenetic position. Eighteen unreported radiation-resistant bacterial species isolated from Korea in 2018

  • In 2018, as a subset study to discover indigenous prokaryotic species in Korea, a total of 18 unreported bacterial strains were discovered. From the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade, it was determined that each strain belonged an independent and predefined bacterial species. There were no official report that these 18 species were previously described in Korea; therefore, one strain of Williamsia, one strain of Rhodococcus, three strains of Microbacterium, three strains of Agromyces, one strain of Arthrobacter, one strain of Paeniglutamicibacter, one strain of Pseudarthrobacter, one strain of Nocardioides, one strain of Fibrella, one strain of Hymenobacter, one strain of Deinococcus, two strains of Fictibacillus, and one strain of Paenibacillus are described as unreported bacterial species in Korea.   • Culturable bacterial diversity was investigated using freshwater and sediment samples collected from the North Han River basin in 2017, as a part of the research program 'Survey of freshwater organisms and specimen collection'. Over a thousand bacterial strains were isolated from the samples and identified based on 16S rRNA gene sequences. Among the bacterial isolates, 22 strains showing higher than 98.7% sequence similarity with validly published bacterial species, but not reported in Korea, were classified as unrecorded species in Korea. The 22 bacterial strains were phylogenetically diverse and assigned to 6 classes, 11 orders, 15 families, and 21 different genera. At the generic level, the unreported species were affiliated with Flavobacterium of the class Flavobacteria, Flexibacter of the class Cytophagia, Blastomonas, Brevundimonas, Elstera, Rhizobium, Roseomonas, Sphingomonas, and Xanthobacter of the class Alphaproteobacteria, Albidiferax, Cupriavidus, Curvibacter, Ferribacterium, Hydrogenophaga, Iodobacter, Limnohabitans, Polaromonas, Undibacterium, and Variovorax of the class Betaproteobacteria, Pseudomonas of the class Gammaproteobacteria, and Arcobacter of the class Epsilonproteobacteria. The unreported bacterial species were further characterized by examining Gram reaction, colonial and cellular morphology, and biochemical properties. The detailed descriptions of 22 strains of the unreported bacterial species are also provided.

A report of 31 unrecorded bacterial species isolated from freshwater

  • A total of 31 bacterial strains were isolated from the Geum River basin in the Republic of Korea during our investigation of indigenous prokaryotic species. The isolated bacterial strains had high 16S rRNA gene sequence similarity (>98.7%) with those of validly published bacterial species, which have not been reported in Republic of Korea. The 31 bacterial strains were phylogenetically diverse and assigned to 4 phyla, 8 classes, 18 orders, 21 families, and 27 genera. At the genus level, the unreported species were affiliated with Kineococcus, Pedococcus, Rhodoluna, Salinibacterium, Rhodoluna, Arthrobacter, Williamsia, Nakamurella, Nocardioides of the class Actinobacteria, Patulibacter of the class Thermoleophilia, Pontibacter, Hymenobacter of the class Cytophagia, Flavobacterium of the class Flavobacteriia, Geomicrobium of the class Bacilli, Brevundimonas, Gellertiella, Rhizobium, Paracoccus, Taonella, Sphingomonas of the class Alphaproteobacteria, Burkholderia, Polaromonas, Hydrogenophaga, Chitinilyticum, Azospira, Zoogloea of the class Betaproteobacteria, and Pseudomonas of the class Gammaproteobacteria. The unreported bacterial species were further characterized by examining their morphological, cultural, physiological, and biochemical properties.   • As a part of the research project "Survey of freshwater organisms and specimen collection," freshwater samples were collected from the Nakdong River. Among the bacterial isolates, we selected strains that showed higher than 98.7% 16S rRNA gene sequence similarity with confirmed bacterial species previously unreported in South Korea. The 14 new records to South Korea were phylogenetically diverse and belonged to four phyla, six classes, 11 orders, and 14 genera. At the genus level, these species were found to be affiliated with Reyranella, Ferrovibrio, Brevundimonas, and Aquidulcibacter of the class Alphaproteobacteria; Pseudomonas, Cellvibrio, and Photobacterium of the class Gammaproteobacteria; Paenibacillus and Bacillus of the phylum Firmicutes; Chryseobacterium, Flavobacterium, Pedobacter of the phylum Bacteroidetes; and Actinomadura and Leifsonia of the phylum Actinobacteria. These species were further characterized by examining their Gram reaction, colony and cell morphologies, biochemical properties, and phylogenetic positions. The detailed descriptions of these 14 previously unreported species are provided.

  • In 2020, a total of 12 bacterial strains were isolated from soil after a comprehensive investigation of indigenous prokaryotic species in Korea. It was determined that each strain belonged to independent and predefined bacterial species, with high 16S rRNA gene sequence similarity (>98.7%) and formation of a robust phylogenetic clade with the closest species. This study identified four families in the phylum Actinobacteria, two families in the phylum Proteobacteria, one family in the phylum Bacteroidetes one family in the phylum Firmicutes; and four species in the family Nocardiaceae, two species in the family Nocardioidaceae, one species in the family Cellulomonadaceae, one species in the family Hymenobacter, one species in the family Methylobacteriaceae, one species in the family Microbacteriaceae, one species in the family Bacillaceae and one species in the family Sphingomonadaceae. There is no official report of these 12 species in Korea, so they are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are included in the species description section.

A report on 29 unrecorded bacterial species isolated from the Nakdonggang River, Republic of Korea

  • As part of the research program "Freshwater Prokaryotic Organisms Research and Discovery," freshwater samples were collected from the Nakdonggang River. After plating the samples on several culture media and incubating aerobically, approximately 900 bacterial strains were isolated and identified using 16S rRNA gene sequences. Among the bacterial isolates showing higher than 98.7% 16S rRNA gene sequence similarity with those of already confirmed bacterial species previously unreported in Korea, 29 strains were selected. These strains were phylogenetically diverse and belonged to 3 phyla, 6 classes, 13 orders, and 21 genera. At the genus level, these previously unreported species were found to be affiliated with Novosphingobium, Sphingomonas, Polymorphobacter, Croceibacterium, Devosia, Endobacterium, Agaricicola, Bradyrhizobium, Paracoccus, and Pseudotabrizicola of the class Alphaproteobacteria; Undibacterium, Azonexus, and Dechloromonas of the class Betaproteobacteria; Acinetobacter and Budvicia of the class Gammaproteobacteria; Streptomyces, Nocardioides, Mycobacterium, and Cellulomonas of the phylum Actinomycetota; Flavobacterium and Pedobacter of the phylum Bacteroidota. These species were further characterized by examining their Gram reaction, colony and cell morphologies, biochemical properties, and phylogenetic positions. Report on 14 unrecorded bacterial species in Korea that belong to the phyla Bacteroidetes and Deinococcus-Thermus

  • As a result of the research project 'Survey and excavation of Korean indigenous species' to secure unrecorded species in Korea, a total of 14 unreported bacterial strains assigned to the phyla Deinococcus-Thermus and Bacteroidetes were isolated from various environmental habitats all around Korea. On the basis of 16S rRNA gene sequence similarity and affiliation to the clade with the closest species, it was concluded that the isolates represent deep groups of the phyla Deinococcus-Thermus and Bacteroidetes. There have been no records about these 14 unreported species in Korea; therefore 2 species of 2 genera in the class Deinococci within the phylum Deinococcus-Thermus, and 11 species of 9 genera in the class Flavobacteriia and one species in one genus in the class Sphigobacteriia within the phylum Bacteroidetes are described as unreported species found in Korea. Gram staining reaction, morphological and other biochemical characteristics are described in the species description section.

A report of 28 unrecorded bacterial species in Korea, isolated from freshwater and sediment of the Han River watershed in 2020

  • To obtain unrecorded freshwater bacterial species in Korea, water and sediment samples were collected from streams, lakes, and wetland of the Han River watershed in 2020. Approximately 800 bacterial strains were isolated on R2A agar after aerobic or anaerobic incubation, and identified using 16S rRNA gene sequences. A total of 28 strains, with ≥98.7% 16S rRNA gene sequence similarity with validly published bacterial species but not reported in Korea, were determined to be unrecorded bacterial species in Korea. The unrecorded bacterial strains were phylogenetically diverse and belonged to four phyla, eight classes, 13 orders, 19 families, and 25 genera. The unreported species were assigned to Acetobacter, Alsobacter, Mesorhizobium, Prosthecomicrobium, and Microvirga of the class Alphaproteobacteria; Vogesella, Formosimonas, Aquincola, Massilia, Acidovorax, and Brachymonas of the class Betaproteobacteria; Pseudoxanthomonas, Thermomonas, Lysobacter, Enterobacter, Kosakonia, and Acinetobacter of the class Gammaproteobacteria; Sulfuricurvum of the class Epsilonproteobacteria; Mycolicibacterium, Agromyces, Phycicoccus, and Microbacterium of the class Actinobacteria; Paenibacillus of the class Bacilli; Clostridium of the class Clostridia; and Flavobacterium of the class Flavobacteriia. The details of the unreported species, including Gram reaction, colony and cell morphology, biochemical characteristics, and phylogenetic position are also provided in the description of the strains.
