합성 ABA 유도체의 벼 유묘 생장저해 작용 (Synthesis of Abscisic Acid Analogs and Their Biological Activity on Growth of Rice Seedling)

  • 천연의 (S)-(+)-ABA 보다 우수한 생육저해 활성을 갖는 ABA 유도체를 개발하기 위한 연구의 일환으로 p-hydroxy methyl cinnamate와 umbelliferone을 (S)-(+)-2-cis, 4-trans-ABA에 ester 결합시켜서 ABA-methyl cinnamate ester(AC)와 ABA-umbelliferone ester(AU) 화합물을 각각 83%와 78%의 높은 수율로 얻었다. 이들의 벼 유묘 생육저해 활성은 (+)-ABA에 비해서 AC가 $3{\sim}10$배, AU가 $10{\sim}30$배 정도로 훨씬 더 강한 활성을 보였다.

Determination of Biosynthetic Pathway of Decursin in Hairy Root Culture of Angelica gigas

  • To establish the biosynthetic pathway of decursin in Angelica gigas Nakai, feeding experiment with stable isotope-labeled precursors were conducted. Umbelliferone and decursin were labeled with deuterium at C-3. The umbelliferone, the decursin, and other commercially available putative precursors, L-phenylalanine-ring-$d_5$ and trans-cinnamic acid-$d_7$, were fed to the hairy root culture of A. gigas. Each deuterated compound was incorporated into decursinol, decursinol angelate, and decursin as determined by mass spectrometric analysis. These findings confirmed the coumarin biosynthesis pathway sequence is composed of phenylalanine, cinnamic acid, umbelliferone, decursinol, and decursin.

Effects of Nodakenin, Columbianadin, and Umbelliferone Isolated from the Roots of Angelica decursiva on the Gene Expression and Production of MUC5AC Mucin from Human Airway Epithelial NCI-H292 Cells

  • Angelica decursiva has been utilised as remedy for controlling the airway inflammatory diseases in folk medicine. We investigated whether nodakenin, columbianadin, and umbelliferone isolated from the roots of Angelica decursiva inhibit the gene expression and production of MUC5AC mucin from human airway epithelial cells. Confluent NCI-H292 cells were pretreated with nodakenin, columbianadin or umbelliferone for 30 min and then stimulated with epidermal growth factor (EGF), phorbol 12-myristate 13-acetate (PMA) or tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) for 24 h. The MUC5AC mucin gene expression was measured by reverse transcription - polymerase chain reaction (RT-PCR). Production of MUC5AC mucin protein was measured by enzyme-linked immunosorbent assay (ELISA). The results were as follows: (1) Nodakenin did not affect the expression of MUC5AC mucin gene induced by EGF, PMA or $TNF-{\alpha}$. Columbianadin inhibited the expression of MUC5AC mucin gene induced by EGF or PMA. However, umbelliferone inhibited the expression of MUC5AC mucin gene induced by EGF, PMA or $TNF-{\alpha}$; (2) Nodakenin also did not affect the production of MUC5AC mucin protein induced by EGF, PMA or $TNF-{\alpha}$. Columbianadin inhibited the production of MUC5AC mucin protein induced by PMA. However, umbelliferone inhibited the production of MUC5AC mucin protein induced by EGF, PMA or $TNF-{\alpha}$. These results suggest that, among the three compounds investigated, umbelliferone only inhibits the gene expression and production of MUC5AC mucin stimulated by various inducers, by directly acting on airway epithelial cells, and the results might explain the traditional use of Angelica decursiva as remedy for diverse inflammatory pulmonary diseases.

  • A cytotoxic coumarin against L1210 cell was isolated from the unripe fruit of Poncirus trifoliata ($ED_{5}$) = 10.2 $\mu$ g/ml. Its structure was identified as aurapten, 7-geranyloxycoumarin. Hydrolysis of the substance gave umbelliferone and geraniol. Only geraniol showed the cytotoxic activity ($ED_{53}$ = 6.5 $\mu$g/ml) while umbelliferone and its methyl or allyl derivatives were not active.

  • Two quinazolone alkaloids, (+)-febrifugine (1) and isofebrifugine (2), along with three coumarin derivatives, 6-hydroxy coumarin (3), skimmin (5), and $umbelliferone-7-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}4)-{\beta}-D-glucopyranoside$ (6), were isolated from the roots of Hydrangea chinensis. Compound 6 is a new compound. In addition, umbelliferone (4), linoleic acid (7), two steroidal glycosides (8, 9), three furfural derivatives (10-12), and butyl-$\beta$-D-fructofuranoside (13) were isolated from the leaves of the same plant. The structures of all isolates were elucidated by spectral methods.

식방풍중의 Coumarin성분의 확인 및 정량 (Analysis of the Coumarin Constituents in Peucedanii Radix)

  • A new method for the analysis of coumarin constituents in the roots of Peucedanum japonicum by high performance liquid chromatography was established. Among two coumarin constituents identified, peucedanol was confirmed to be applicable to a standard compound. A reversed phase system with a ${\mu}Bondapak$ C_{18}$, column using $H_2O-MeOH=5\;:\;4$ as a moble phase was developed. Peucedanol and a minor constituent, umbelliferone were detected at 333nm and the analysis was successfully carried out within 20 min.

  • From the root of Angelica decursiva-albiflora Yook, which has been used as a folk medicine for a sedative, analgesic and expectorant, four free coumarins, e.g., decursidin(I), decursin(II), umbelliferone(III) and nodakenetin(IV), and two coumarin glycosides, e.g., nodakenin(V) and decuroside I(VI) were isolate. The cytotoxicity of nodakenin(V) against L-1210 leukemia cells was less effective than cisplatin, but in the nephrotoxicity against rabbit kidney proximal tubular cell nodakenin(V) showed remarkably less nephrotoxicant than cisplatin.

연부(軟腐)고구마의 생화학적(生化學的) 연구(硏究) (Biochemical Study on Soft-rotted Sweetpotatoes)

  • Rhizopus nigricans에 감염(感染)된 고구마의 생화학적(生化學的) 성질(性質)을 구명(究明)하기 위하여 연부(軟腐)고구마로부터 이상대사산물(異常代謝産物)을 분리(分離)하고 그 성질(性質)에 관(關)하여 실험(實驗)한 결과(結果)를 다음과 같이 요약(要約)한다. 1. 건전(健全)한 고구마에는 없는 이상대사산물(異常代謝産物)로서 19개(個)의 Ehrlich's 시약(試藥)에 대(對)한 양성반응성질(陽性反應性質)과 3개(個)의 형광성물질(螢光性物質)의 존재(存在)를 박층(薄層)크로마토그라프로 확인(確認)하였다. 2. 이상대사산물중(異常代謝産物中)에는 그 생성량(生成量)이 가장 많은 물질(物質)로서 Ipomeamarone과 Umbelliferone을 분리(分離)하고 이들의 정제(精製)와 동정(同定)을 행(行)하였다. 3. Ipomeamarone과 Umbelliferone의 경시적(經時的) 생성량(生成量)을 관찰(觀察)하고 이들의 가능(可能)한 생합성(生合成) 경로(經路)에 관(關)하여 고찰(考察)하였다. 4. 연부(軟腐)고구마의 이상대사산물중(異常代謝産物中) Ipomeamarone과 이의 유사물질(類似物質)은 ${\alpha}-amylase$에 대(對)한 활성(活性)을 저해(抵害)하였으며 그 저해형(抵害型)은 비경쟁적(非競爭的)이었다. 5. 연부(軟腐)고구마로부터 분리한 Ipomeamarone은 Yeast의 호흡(呼吸)을 촉진(促進)시켰으며 산화적(酸化的) 인산화반응(燐酸化反應)의 연합해제제(聯合解除劑)인 2, 4-dinitrophenol과 동일(同一)한 효과(效果)를 나타내었다.
