Antioxidative Activity and Tyrosinase Inhibitory Activity of the Extract and Fractions from Arctium lappa Roots and Analysis of Phenolic Compounds (우엉 뿌리 추출물의 항산화 및 Tyrosinase 저해 활성과 Phenolic Compound 분석)

  • In this study, we investigated on antioxidative activities and tyrosinase inhibitory activities of methanol extract and its fractions from roots of Arctium lappa. The total phenolic compound and flavonoid content of the ethylacetate fraction was found to be 818.29 mg/g and 360.59 mg/g as the highest content. In the measurement of DPPH radical scavenging ability and tyrosinase inhibitory activity, the ethylacetate fraction was higher than the other fractions and the extract. In addition, comparative analysis of phenolic compounds by liquid chromatography-mass spectrometry (MS)/MS system under the multiple-reaction monitoring (MRM) with negative-ion electrospray ionization mode. The main phenolic compounds in the extract and fractions of roots from Arctium lappa were cynarin and chlorogenic acid. The main phenolic compound of the ethylacetate fraction was cynarin. n-Butanol fraction had a significantly higher chlorogenic acid content than other samples. In conclusion, DPPH radical scavenging ability and tyrosinase inhibitory activity of the cynarin-riched ethylacetate fraction showed the highest activity.

Antibacterial Activity against Pathogens of Acne and Tyrosinase Inhibitory Activity of Extract and Fractions from Bark of Prunus sargentii (산벚나무 수피 추출물 및 용매 분획물의 여드름 원인균에 대한 항균활성과 Tyrosinase 저해 활성)

  • In this study, we investigated on antioxidative activity, antibacterial activity against pathogens of acne and tyrosinase inhibitory activity in 75% EtOH extract and its fractions from bark of Prunus sargen tii. The total polyphenol and flavonoidcontent of the EtOAc fraction were found to be 378.0 mg/g and 67.5 mg/g as the highest content. In the measurement of DPPH radical scavenging ability, EtOAc and BuOH fraction were exhibited stronger scavenging ability than the other fractions and 75% EtOH extract. In antibacterial activity by disc diffusion assay against pathogen of acne, antibacterial activity of the EtOAc fraction and 75% EtOH extract was stronger than the other fractions. Especially, the EtOAc fraction was the highest effective fraction in the antibacterial activity. Moreover, tyrosinase inhibitory activity of EtOAc fraction, BuOH fraction and 75% EtOH extract was higher than the other fractions. In particular, tyrosinase inhibitory activity of the EtOAc fraction showed higher activity than ascorbic acid used as positive control.

Tyrosinase Inhibitory Activity and Melanin Production Inhibitory Activity of the Extract and Fractions from Paeoniae Radix (작약 메탄올 추출물 및 분획물의 Tyrosinase 저해 및 Melanin 생성 억제활성)

  • We investigated antioxidative activity, tyrosinase inhibitory activity, and melanin production inhibitory activity of the methanol extract of Paeoniae Radix and its fractions. The total polyphenol content of the extract was 73.45 mg/g, and content of the ethyl acetate fraction was 514.50 mg/g as the highest content of fractions. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate was 3.86 ${\mu}g/ml$ as a result of greater activity in the positive control (ascorbic acid). Moreover, tyrosinase inhibitory activity of the ethyl acetate fraction showed higher activity than arbutin used as a positive control. In the cytotoxicity measurement by MTT assay, the extract and fractions were exhibited cell viabilities of 76.96~157.26% against Raw 264.7 and B16F10 cell in concentration of 10~100 ${\mu}g/ml$. In nontoxic concentration range, the ethyl acetate fraction showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-stimulated B16F10 cell. As a result, the ethyl acetate fraction of the methanol extract from Paeoniae Radix could be applicable to functional materials for skin-whitening agents.

The Isolation of the Inhibitory Constitutents on Melanin Polymer Formation from the Leaves of Cercis chinensis (박태기나무의 잎으로부터 피부멜라닌 색소생성 억제성분의 분리)

  • Tyrosinase plays an important role in the process of melanin polymer biosynthesis. Therefore, the enzyme inhibitors have been of great concern as cosmetics to have skin-whitening effects on the local hyperpigmentation. During the search for new inhibitory compounds on melanin polymer biosynthesis from natural sources, MeOH extracts of 589 higher plants were tested for the inhibitory effect on tyrosinase activity by the muschroom tyrosinase assay in vitro. Among plants tested, the leaves of Cercis chinensis exhibited potent inhibitory effect on mushroom tyrosinase activity. Subsequently seven active compounds were isolated from the ethyl acetate soluble part of acetone extract of the leaves of C. chinensis by the activity guided fractionation monitoring the inhibitory effect on tyrosinase activity. Their chemical structures were identified as $kaempferol-3-0-{\alpha}-L-rhamnoside$, quercitrin, $myricetin-3-0-{\alpha}-L-rhamnoside$, myricetin-3-0-(2'-O-galloyl)- ${\alpha}$ -L-rhamopyranoside (desmanthin), (-)-epicatechin-3-0-gallate, (-)-epigallocatechin-3-0-gallate, and methyl gallate on the basis of the speculation of spectral data and chemical reaction. Among the flavonol rhamnosides, myricetin-3-0-(2'-O-galloyl)- -L-rhamnoside(desmanthin) showed most potent inhibitory effect on tyrosinase activity and the structure of B-ring in flavonol moiety was related to the activity. (-)-Epigallocatechin-3-O-gallate having pyrogallol group in flavan-3-ol moiety exhibited more potent inhibitory effect than (-)-epicatechin-3-0-gallate having catechol group in flavan-3-ol moiety on mushroom tyrosinase activity.

Inhibitory Effects of Siegesbeckia Herba Extracts on the Melanin Production and Tyrosinase Activity in B16F10 cells (희첨 추출물이 B16F10 세포에서 Melanin 생성과 Tyrosinase 활성에 미치는 억제효과)

  • Objective : Siegesbeckia Herba can treat various skin disease by expelling wind and removing dampness and clearing away heat and toxic material effects. This study was designed to investigate effects of Siegesbeckia Herba Extracts(SHE) on skin elasticity and whitening using B16F10 cell lines. Method : In this experiment, We observed effect of SHE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : 1. SHE treated group showed decreased cell viability rates significantly compared with non-treated group. More than SHE $250{\mu}g/ml$, $500{\mu}g/ml$ and $1,000{\mu}g/ml$ of treated groups were lower levels of melanin synthesis respectively. 2. SHE significantly showed tyrosinase inhibitory activity in vitro, SHE increased tyrosinase inhibitory activity and elastase inhibitory activity in B16F10 cells, and tyrosinase inhibitory activity in vitro. 3. Tyrosinase inhibitory activity and elastase inhibitory activity in B16F10 cells, tyrosinase inhibitory activity in vitro were not accepted statistical significance compared with non-treated group. 4. SHE treated group showed increased SOD-like activity rates significantly compared with non-treated group. More than SHE $250{\mu}g/ml$, $500{\mu}g/ml$ and $1,000{\mu}g/ml$ of treated groups were lower levels of melanin synthesis respectively. Conclusion : These results suggest that SHE can inhibit melanin synthesis and tyrosinase inhibtory activity. So, We suggest that SHE can be maintained skin whitening.

Tyrosinase Inhibitory Prenylated Flavonoids from Sophora flavescens

  • For the purpose of the development of a skin-whitening agent, Sophora flavescens was evaluated for tyrosinase inhibitory activity and its active principles were identified followed activity-guided isolation. The ethanol extract and dichloromethane fraction from S. flavescens showed significant inhibition of mushroom tyrosinase. From the dichloromethane fraction, three known prenylated flavonoids, sophoraflavanone G, kuraridin, and kurarinone, were isolated. Compared with kojic acid ($IC_50$=20.5 $\mu$M), these compounds possessed more potent tyrosinase inhibitory activity. (omitted)

The Inhibitory Effects of Nelumbo nucifera Gaertner Extract on Melanogenesis (연자육 추출물의 멜라닌 합성 저해효과)

  • In order to develop new skin whitening agents, we prepared the $CH_2Cl_2$ layer (NGC) and BuOH layer (NGB) of 75% EtOH extract of the Nelumbinis nucifera Gaertner. We measured their tyrosinase inhibitory activity in vitro and melanin synthesis inhibitory activity in B16-F1 melanoma cells. They did not show inhibitory activity against mushroom tyrosinase but showed melanin synthesis inhibitory activity in a dose-dependent manner. In a melanin synthesis inhibition assay, NGC and NGB suppressed melanin production up to 52% and 46% at a concentration of $100{\mu}g/mL$, respectively. To elucidate the mechanism of the inhibitory effects of NGC and NGB on melanogenesis, we measured the expression of melanogenesis-related proteins by western blot assay. As a result, NGC suppressed the expression of tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), phosphorylated cAMP responsive element binding (p-CREB) protein, and microphthalmia associated transcription factor (MITF). And NGB inhibited the protein expression of tyrosinase and MITF, but had no significant effect on TRP-1, TRP-2, and p-CREB expression. Moreover, NGB increased the expression of phosphorylated extracellular signal-regulated kinase (p-ERK). In addition, we examined the inhibitory effect on the glycosylation of tyrosinase. As a result, NGC and NGB inhibited the activity of ${\alpha}$-glucosidase in vitro and the glycosylation of tyrosinase in B16-F1 melanoma cells. From these results, we concluded that NGC and NGB could be used as active ingredients for skin whitening.

Tyrosinase Inhibitory Activity and Melanin Production Inhibitory Activity of Taraxinic Acid from Taraxacum coreanum (흰민들레(Taraxacum coreanum)에서 분리한 taraxinic acid의 tyrosinase 활성저해 및 melanin 생성저해 효과)

  • This study was to investigate the Taraxinic acid from Taraxacum coreanum on tyrosinase activity and melanogenesis. In B16BL6 cell, Taraxinic acid did not show cytotoxicity even at concentrations of up to 100 ㎍/mL. In addition, tyrosinase inhibitory activity and melanogenesis inhibitory activity were confirmed by stimulation with α-melanocyte stimulating hormone (α-MSH) in the presence of taraxinic acid. Taraxinic acid was added to cells at concentrations of 10, 50 and 100 ㎍/mL and treated for 48 hours to confirm tyrosinase inhibitory activity and melanin production. The tyrosinase inhibitory activity increased in proportion to the amount of the sample, and showed an inhibitory activity of about 54.5% at a concentration of 50 ㎍/mL. Melanin production decreased in proportion to the sample amount, and it was about 62.2% at the concentration of 10 ㎍/mL. From the above results, it was found that Taraxinic acid had higher tyrosinase activity and melanin synthesis inhibitory activity in melanocyte than arbutin. The results suggest that Taraxinic acid can be utilized in natural whitening cosmetics.

Screening of Inhibitory Effect of Edible Mushrooms on Tyrosinase and Isolation of Active Component (한국산 식용버섯류의 Tyrosinase 활성 저해 검색 및 그 유효성분 분리)

  • For the purpose of isolation and screening of tyrosinase inhibitory activity from edible mushrooms, Pleurotus ostreatus, Auricularia auricula-Judae, Umbilicaria esculenta, Agaricus bisporus, Flammuline velutipes, Lentinus edodes, Ganoderma lucidum, and Coriouls versicolor were examined by tracing inhibitory activities against tyrosinase, utilizing L-3,4-dihydroxyphenylalanine (L-DOPA) as a substrate. Among the eight edible mushrooms tested, Umbilicaria esculenta showed potent enzyme inhibitory activities above 7804% against tyrosinase in ethylacetate (EtOAc) extracts. Ganoderma lucidum and Agaricus bisporus showed inhibitory activities of 67.3% and 51.5% in water extracts. EtOAc extracts of Umbilicaria esculenta was fractionated from silicagel column chromatography and one fraction showed the most inhibitory activity of 60.9%. The three bands (Rf=0.38, 0.27, 0.19) were isolated from preparative TLC of the fraction for purification and identified as mixtures of orsellinate, methyl orsellinate, methyl lecanorate, and methyl gyrophorate by high pressure liquid chromatography (HPLC), ultravisible spectrophotometer (UV), mass spectrophotometer (Mass), nuclear magnetic resonance spectrometer (NMR).

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).
