A study on the Immune Response of Salmonella Typhi Inactivated by Gamma Ray (전리방사선(電離放射線)에 의(依)하여 불활화(不活化)된 Salmonella typhi의 면역반응(免疫反應) 연구(硏究))

  • For the preparation of non-infective vaccine against typhoid fever, the authors had carried out an investigation on the possibilities of using $^{60}Co\;{\gamma}$-ray irradiation for the inactivation of Salmonella typhi Ty 2. The following results were obtained. 1) Chicks shortly after hatching were higher susceptible than mice to intracerebral injection with S. typhi. 2) In safety test on experimental animal, $^{60}Co\;{\gamma}$-ray irradiated vaccines demonstrated no clinical symptom while the phenol treated vaccines revealed several cases of narcosis by intraperitoneal injection in chicks. 3) The rabbit antisera immunized with $^{60}Co\;{\gamma}$-ray irradiated and phenol treated vaccine revealed almost same agglutinin titers to each other. 4) In potency test in chicks, there was no significance in the protective capacity between $^{60}Co\;{\gamma}$-ray irradiated and phenol treated vaccines.

Experimental study on Methods of Fever Control -Effects of ice-pack application on different body areas.- (냉찜질부위에 따른 열 조절 효과에 관한 실험적 연구)

  • The purpose of this study is to observe the effects of ice-pack application on different body areas and to note which body area produces the best results according to age, room temperature, sex and disease. Seventeen patients, including six men and eleven women, hospitalized at Seoul National University Hospital from May 22 to August 29, 1971, ranging in age from 15 through 60 years with an oral temperature of 38℃ or over, were chosen for this study. They were observed with regard to the effect of the ice-packs after applications for 30, 45 or 60 minutes. Four ice-packs, 7.6inch by 9 inch, were applied on the chest and the abdominal surface in the control group and the same size and number of packs on the axillae and political regions in the experimental group. The study results were reviewed in a statistical method to obtain the following findings 1. There was no significant difference in the change of the body temperature between the control group and the experimental group after applying the packs for 30, 45 or 60 minutes. 2. Individually, there was a significant difference in the change of body temperature between before and after application for 30, 45 or 60 minutes. 3. The difference of room temperature also had no merle influence on the body temperature taken immediately before applying the ice-packs. Even when ice-packs were applied for 30, 45 or 60 minutes, the room temperature did not influence the reduction of temperature in the patient. 4. There was a significant difference in the change of the body temperature, according to the sex, after application for 30, 45 or 60 minutes. Male patient's body temperature decreased far more noticeably than the female patients and the degree of reduction became more pronounced the longer the packs were applied. 5. In regard to age, in adults, the degree of reduction in the body temperature by the application of ice-packs was similar after appling them for 30, 45 or 60 minutes and did not depend upon the length of application. 6. Regardless of the disease, there was no great difference in the reduction of body temperature when the ice-packs were applied for 30 or 45 minutes, except for patients with typhoid fever when a striking decrease in the fever resulted after a 60 minute application of the ice-pack.

Reconstruction of the Acquired Facial Deformity due to Cancrum Oris Sequelae (괴저성 구내염 후유증에 의한 후천성 안면 변형의 재건)

  • Acquired facial deformities following cancrum oris sequelae manifested variably according to the nature of tissue necrosis. In cases that tissue loss extends over a wide area of the face, or the tissue nature is different due to congenital facial cleft, it is difficult to reconstruct with a single operation. As cancrum oris has virtually disappeared from our country, clinical report of reconstruction is also rare. We report 5 cases of facial deformities following cancrum oris sequelae. Since 1988, five adult patients(4 female and 1 male) were treated by authors. These patients, with an age ranged from 47 to 58 years, all suffered from acquired facial cleft such as facial mutilation, asymmetry. The stages of operation were from 1 to maximum of 5 operations. All surgeries achieved satisfactory results after a long-term follow-up. But one surgery in the case of palatal mucosal flap for the coverage of reconstructed maxilla alveolar bone resulted tissue sloughness, followed by osteomyelitis. Those were debrided and discarded. In conclusion, all sites of deformities were positioned around one of the oral commissures. In their past medical history, they have been suffering from measles, typhoid fever and unknown febrile illness. We diagnosed the acquired facial deformity following cancrum oris sequelae. The reconstruction of acquired facial deformity following cancrum oris sequelae were difficult due to extensive multiple tissue defects. Therefore multiple staged operations were inevitable. The authors reconstructed 5 cases of simple and complex form of facial deformity with minimum staged operations. All patients were satisfied functionally and cosmetically.

Evaluation of antibacterial and therapeutic effects of egg-white lysozyme against Salmonella Typhimurium in ICR mice infected with Salmonella Typhimurium (난백 유래 라이소자임의 마우스 살모넬라증에 대한 항균 및 치료 효과)

  • Salmonellosis is a major bacterial zoonosis that causes self-limited enteritis in animals and foodborne disease and typhoid fever in humans. Recently, multi-drug-resistant strains of Salmonella spp. have increased and caused more serious problems in public health. The present study investigated the antibacterial effects of egg-white lysozyme (EWL) against Salmonella (S.) Typhimurium and the therapeutic effects of EWL for murine salmonellosis. Evaluation of the antibacterial effects of EWL against S. Typhimurium revealed a minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of EWL of 6.25 and $300{\mu}g/mL$, respectively. In the bacterial growth inhibition test, EWL at 300 (p < 0.05) and $600{\mu}g/mL$ (p < 0.01) significantly inhibited the growth of S. Typhimurium at 4 h post-incubation. EWL administration at MIC (LYS-1), MBC (LYS-2) and $2{\times}MBC$ (LYS-3) for 14 days resulted in mortality of mice infected with S. Typhimurium of 70, 40 and 10%, respectively, while that of control mice (CON) was 90%. Counts of S. Typhimurium in murine spleens were significantly lower in LYS-2 and LYS-3 than CON (p < 0.05). The results of this study indicate that EWL has the potential for treatment of ICR mice infected with S. Typhimurium.

Cloning and Nucleotide Sequence Analysis of the aroA Gene from Salmonella typhi KNIH100 (Salmonella typhi KNIH100으로부터 aroA 유전자의 클로닝과 염기서열 분석)

  • Salmonella typhi is one of important causes of human enteric infections. S. typhi KNIH100 was isolated from a patient of typhoid fever in Korea. We cloned a 5.0 kb SalⅠ fragment containing the aroA gene encoding a 5-enolpyruvylshikimate-3-phosphate synthetase from chromosomal DNA of this strain. This recombinant plasmid was named pSAL80. E. coli CGSC2829, an aroA- mutant, was not grown on the M9 minimal medium but E. coli CGSC2829 (pSAL80) was grown on the M9 minimal medium. The aroA gene was composed of 1,284 base pairs with ATG initiation codon and TAA termination codon. Sequence comparison of the aroA gene exhibited 99%, 98%, and 77% identity with those of S. typhi Ty2, S. typhimurium, and E. coli respectively. As in the cases of Shigella sonnei and E. coli, the serC and aroA genes lie in a single operonic structure.

Methods for Coating the Killed Whole Cell Antigens of Salmonella typhi in Enzyme-linked Immunosorbent Assay (효소면역측정법을 위한 장티푸스 균체항원의 부착방법)

  • The advantages of enzyme-linked immunosorbent assay(ELISA) are its senstivity and simplicity in detecting IgG, IgM and IgA antibody. To apply ELISA to diagnosis of typhoid fever, antigen such as lipopolysaccharide of Salmonella typhi or killed whole cell must be coated on solid phase. It is easy to coat lipopolysaccharide on ELISA plate but troublesome to purify it. As it is easy to obtain the killed whole cells, the development of the appropriate method by which those antigens of S. typhi are optimally coated on solid phase is needed. To establish the appropriate method, carbonate buffer, methanol or poly-L-lysine was applied as binding substance on polystyrene or polyvinylchloride plate as solid phase when the killed whole cell antigens of S. typhi varided as follows: $10^6$, $10^7$, $10^8$ and $10^9\;cell/ml$. The criteria of the optimal method were determined as follows: 1. The optical density of positive sera is above 1.0(0.6 in IgM) at 1:10 serum dilution and is 0.3(0.2 in IgM) higher than that of negative sera: 2. The O.D. of sera is flat or lowering according to serum dilution: 3. It must be that the O.D. of negative sera is lower than 0.2 at the point of serum dilution where the O.D. of positive sera is higher than 1.0(0.5 in IgM). The results obtained were summarized as follows: 1. The methods which fitted the above criteria were to use poly-L-lysine as binding substance, polyvinylchloride plate as solid phase and $10^7\;cell/ml$ as antigen concentration of S. typhi(poly-L-lysine/polyvinylchloride/$10^7$) and poly-L-lysine/polyvinylchloride/$10^8$ in detecting IgG antibody, methanol/polystyrene/$10^9$, poly-L-lysine/polyvinylchloride/$10^8$ and poly-L-lysine/polyvinylchloride/$10^9$ in IgM and carbonate buffer/polystyrene/$10^8$, carbonate buffer/polystyrene/$10^9$, methanol/polystyrene/$10^8$, methanol/polyvinylchloride/$10^8$, methanol/polyvinylchloride/$10^9$, poly-L-lysine/polyvinylchloride/$10^8$ and poly-L-lysine/polyvinylchloride/$10^9$ in IgA. 2. The coaling method using poly-L-lysine, polyvinylchloride plate and $10^8\;cell/ml$ was best to assay IgG, IgM and IgA antibody all in one. By this method, to assay the each immunoglobulin calss with an appropriate fixed serum dilution, 1:320 dilution was best.

Evaluation of Antibacterial and Therapeutic Effects of a Sodium salts Mixture against Salmonella typhimurium in Murine Salmonellosis (나트륨 염 복합조성물의 마우스 살모넬라증에 대한 항균 및 치료효과)

  • Salmonellosis is a major bacterial zoonosis that causes self-limited enteritis to fatal infection in animals and food-borne infection and typhoid fever in humans. Multidrug-resistant strains of Salmonella spp. has increased over the last several decades and recently causes more serious problems in public health. The present study was investigated bacteriocidal effects of sodium chlorate, sodium azide, sodium cyanide, and sodium salts mixture containing sodium chlorate, sodium azide, and sodium cyanide on infection with S. typhimurium in macrophage RAW 264.7 cells, and antibacterial effects of sodium salts mixture for murine salmonellosis. In infection assay of S. typhimurium in RAW 264.7 cells, bacterial survival rates within macrophage in all treated groups was significantly reduced comparing to that of the control group with the passage of incubation time. Administration of sodium salts mixture showed a therapeutic effect for S. typhimurium infected ICR mice. The mortality of mice treated with sodium salts mixture was 70% until 12 days, while that of control mice was 100% until 9 days after S. typhimurium infection. The results of this study strongly indicate that sodium salts mixture has a potency treatment for murine salmonellosis.

Identification and Characterization of Genes Involved in Cysteine Auxotrophy in Salmonella typhi (Salmonella typhi의 시스테인 영양요구성에 관여하는 유전자의 동정 및 특성 연구)

  • In spite of long research period for Salmonella typhi, little information is known about the pathogenesis mechanism of human typhoid fever caused by S. typhi due to lack of infection model in animals. A wild-type of S. typhi Ty2 strain requires cysteine to grow on minimal media. We hypothesized that this cysteine requirement may restrict colonization of S. typhi in animals during infection process. Among the S. typhi strains carrying Salmonella typhimurium genomic library, we have isolated three S. typhi transformants growing on minimal media without cysteine. Although there were three ORFs in DNA of pBP71, the STM1490 ORF complemented cysteine auxotrophy of S. typhi. Analysis of the deduced amino acid sequence of the STM1490 homolog in S. typhi revealed that there are differences in two amino acids. Plasmids containing amino acid substitutions in STM1490 supported S. typhi growth on minimal media without cysteine, indicating irrelevance of these two amino acids to STM1490 function. These results tells us that there are other factors or systems involved in cysteine requirement of S. typhi.

Accurate determination of chloramphenicol in meat by isotope dilution liquid chromatography mass spectrometry (ID-LC/MS) (동위원소희석 액체크로마토그래피-질량분석법을 이용한 육류 중 클로람페니콜의 정밀한 정량 분석에 관한 연구)

  • Chloramphenicol is one of the most effective antibiotics for treatment of food-producing animals for typhoid fever. However, it has been reported that it caused severe side effects such as aplastic anemia in human, therefore the use of chloramphenicol for food-producing animal is prohibited by European Union and other countries. In this study, the analytical method using isotope dilution liquid chromatography-mass spectrometry (ID-LC/MS) was established for accurate determination of chloramphenicol in meat. Chloramphenicol was extracted with ethylacetate from porcine and solid phase extraction cartridge was used for enhancing the recovery. The residue of chloramphenicol in porcine was analyzed using the liquid chromatography mass spectrometer (LC/MS) interfaced with electrospray ionization source. Analysis was performed in negative mode with selected reaction mornitoring mode at m/z 321${\rightarrow}$257 of $[M-H]^-$ ${\rightarrow}$ $[M-H-(HCOCl)]^-$ and m/z 326 ${\rightarrow}$ 262 channels for its isotope. The established method was tested using fortified samples at the level of 0.2 1, 10, $25\;{\mu}g$/kg and analytical results agreed with the gravimetrically fortified values within their uncertainties. This method was validated by analyzing a certified reference material, BCR 445, from IRMM (Institute for Reference Materials and Measurement). Our measurement values agreed with the certified value within their uncertainties. The uncertainty of our measured value was much lower than that the certified value from IRMM.
